Penile Blood Change after Oral Medication of Korean Red Ginseng in Erectile Dysfunction Patients (발기부전환자에서 홍삼 복용후의 음경혈류와 발기력 변화)

  • The clinical efficacies of Korean Red Ginseng for improving sexual function has been reported only by self-reporting questionnaires.AVS-Penogram is a method evaluating natural penile blood flow changes by Gamma-camera after intravenous injection of Techmetium 99$\^$m/-RBC and also stimulating by erotic video tape. To more objectively evaluate the efficacies of Korean Red Ginseng for erectile function, we applied AVS-Penogram as a objective diagnostic tool. AVS-Penogram studies were done in 28 patients with erectile dysfunction after administration of Red Ginseng(19) or placebo(9) group. The pattern of AVS-Penogram before medication were normal type in 4, impossible in 2, unstable in 20, delayed in 1, and mixed in 1. The unstable type were the most prevalent observed in 71.4% of all patients. The patterns of AVS-penogram were 2.2. 13.1 in Red Ginseng group, while in placebo group normal 2, unstable 7. After 4 weeks in Red Ginseng group, there were 3 cases of improved penogram findings, while only 1 in placebo group. There were no statistical significancies in the type of penogram changes on both group. But the maximum radioactivity were improved in 11 patients(57.9%) in Red Ginseng group, while 2(22.2%) in placebo group. There were statistical significancies in comparing the maximal radioactivity of penogram on both group. These AVS-Penogram findings shows, encouraging results of by increased penile blood flow Korean Red Ginseng our medication. Further study is more needed to reconfirm these results by more concentrated active formulations of sapanonin St other active ginsenosides. These promising results will lead to develop a new remedy for erectile dysfunction by Korean Red Ginseng

Effect of Antioxidant Activity and Induction of DNA Damage on Human Gastric Cancer Cell by Rubus coreanus Miquel (복분자 추출물의 항산화활성 및 인간 위암 세포주에 대한 유전적 손상 유도)

  • Rubus coreanus Miquel (RCM), a type of red raspberry, grows wild in Korea and China and its unripe fruit is used as a folk medicine for the treatment of impotence and as a diuretic. RCM was extracted with methanol and then further fractionated it into for different types. In this study, we investigated the antioxidant activity of a RCM extract (ext.) and its fraction (fr.). DPPH free radical scavenging activity assay, total polyphenols contents, total flavonoids contents assay were used to analyze antioxidant activity. The DPPH free radical scavenging activity $(RC_{50}:1.67{\mu}g/ml)$ and total polyphenols contents $(546.25{\mu}g/mg)$ were higher in butanol fraction than in other fr. And total flavonoids contents was higher in ethylacetate fr. $(141.78{\mu}g/mg)$. We applied comet assay to measure the DNA damage in the individual cells and exposed time course at $IC_{50}$. Comet assay is a rapid and sensitive fluorescent microscopic method to examine DNA damage and repair at individual cell level. Anti-inflammatory Effects of Epimedii Herba Water Extract through Inhibition of Nuclear Factor-κB in RAW 264.7 Cells (RAW 264.7 세포에서 음양곽(淫羊藿) 물 추출물의 nuclear factor-κB 억제를 통한 항염증 효과)

  • Objectives : Epimedii Herba has been frequently used in Korean Traditional Medicine to treat impotence, spermatorrhoea, exophthalmos, and forgetfulness. Present study investigated anti-inflammatory effects of Epimedii Herba water extract (EWE) and attempted to elucidate molecular mechanisms involved. Methods : To explore anti-inflammatory effects of EWE, RAW 264.7 cells, a murine macrophage cell line, were pretreated with $10-100{\mu}g/m{\ell}$ of EWE, and then subsequently exposed to $1{\mu}g/m{\ell}$ of lipopolysaccharide (LPS). Levels of nitric oxide (NO), interleukin-6, $interleukin-1{\beta}$, and tumor necrosis $factor-{\alpha}$ were monitored in the medium. Expression levels of inducible nitric oxide synthase and cyclooxygenase-2 were determined by immunoblot and real-time PCR analyses. Signaling pathways related with nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) and mitogen-activated protein kinases were monitored to elucidate molecular mechanisms involved. Finally, the role of three flavonoid compounds in EWE on LPS-mediated NO production were investigated. Results : In conditioned medium, pretreatment of EWE ($100{\mu}g/m{\ell}$) significantly inhibited LPS-stimulated NO and pro-inflammatory cytokine production. In addition, EWE attenuated the expressions of inducible nitric oxide synthase and cyclooxygenase-2 by LPS. EWE prevented the phosphorylation and degradation of inhibitory ${\kappa}B{\alpha}$, nuclear translocation of $NF-{\kappa}B$, and DNA binding of $NF-{\kappa}B$, while EWE did not change the phosphorylation of mitogen-activated protein kinases by LPS. Moreover, icariin, icaritin, and quercetin partly, but significantly, inhibited the LPS-stimulated NO production. Conclusions : These results suggest that EWE has an ability to prevent inflammation in macrophages through inhibition of $NF-{\kappa}B$ signaling pathway.

  • To help understanding about hazard chemicals occurring newly in food, it has been reviewed characteristics, mechanism, food standards and analytical trend of the hazard chemicals occurring avoidably in food manufacturing process. The new hazard chemicals in food are classified by 4 regarding mechanism such as heat, fermentation, additives and unknowns. The new hazard chemicals by heat process are acrylamide, furan, HCAs and PAHs. By the fermentation, ethylcarbamate and biogenic amines are occurred. According to food additives, 3-MCPD, benzene and nitrosamines can be produced. And the last group is the illegal compounds including anti-impotence drug analogues and anti-obesity drug analogues. To analyze the new hazard chemicals in food, GC or LC with UVD, FLD or MSD are used mainly after sample pretreatment by LLE, SPE or headspace method.

Other Processed Products, Monitoring and the Exposed Dose Assessment of Heavy Metal, the Illegal Compounds (기타가공품의 중금속, 부정유해물질 모니터링 및 노출량 평가)

  • This study was performed for both evaluating the safety of other processed products and providing basic information for making the general standard for contaminants in the category of other processed products. We analyzed the contents of three heavy metals, thirty six anti-impotence drugs and their analogues, three anti-obesity drugs and their analogues, twenty eight steroid drugs and their analogues, collecting in Incheon Metropolitan City. Any illegal compound was not detected in those products. However the contents of lead, cadmium and mercury of those products were at the range of 0.001-13.390 mg/kg, 0.03-1.231 mg/kg and 0.001-0.650 mg/kg respectively. Because there are no standards of heavy metals against other processed products, we compared the analytical results with relevant standards of both S. Korea and foreign countries. As a result, two products exceeded the relevant standards of lead, and other two products exceeded the relevant standards of mercury. The relative hazards compared to PTWI of FAO/WHO (Codex), Seafoods-pajeonmix, Perilla seed powder exceeded PTWI standards 0.214. The compulsory standards of each food product are determined by the category of the products. Because there is no standard of heavy metals in the category of other processed products in S. Korea, any food products registered as other processed product by manufacturer are free with those standards. Abusing similar problems on the categorization of food products could cause consumers' health problem. To prevent these problems, detail regulations on the categorization of food products have to be introduced.
