포도상구균 장내 C 형 변이독소 (SEC mutant)의 면역원성에 대한 연구 (Immunogenicity of staphylococcal enterotoxin C mutant antigen in mice and dairy cows)

  • Mastitis is one of the most significant cause of economic loss to the dairy industry. Especially, Staphylococcus aureus is a major contagious mastitis-causing pathogen in dairy cattle. Because of its high transmission rate and resistance to antibiotic therapy, staphylococcal mastitis presents a constant threat to the dairy industry. Staphylococcal enterotoxin C(SEC) produced by S aureus has been known as one of superantigens which are able to stimulate a large proportion of T lymphocytes independently of their antigenic specificity. In this experiment, we have conducted preliminary studies with mice and lactating cows to evaluate the immunogenicity and safety of the experimental vaccine consists of SEC mutant antigen on controlling the bovine mastitis associated with S aureus infections. The average value of somatic cell counts in quarter milk, isolation rate of S aureus were consistently decreased in SEC-SER vaccinated groups, whereas antibody titers were highly increased in SEC-SER vaccinated groups. Peripheral blood were also collected from the lactating cows to determine the proportion of leukocyte subpopulation associated with humoral immunity(HI) and cell mediated immunity(CMI). Proportion of leukocyte subpopulation expressing $BoCD2^+$(total T lymphocyte), $BoCD4^+$(T helper cell), $BoCD8^+$(T cytotoxic/suppressor cell) and NonT/NonB lymphocyte which are involved in CMI in SEC-SER vaccinated groups were decreased for the initial stage after first vaccination and then increased from ten weeks after first vaccination maintaining elevated level till 14 weeks after vaccination. In contrast, proportion of monocyte, MHC class II and B lymphocyte which are associated with the production of primary immune response in SEC-SER vaccinated groups were increased for the initial period and then decreased from ten weeks after first vaccination. We present evidence that vaccination of SEC-SER mutant antigen in lactating cows induced a significant proliferation of bovine T lymphocytes. These results suggest that SEC-SER mutant antigen used in this experiment might be one of potential immunogen in developing innovative vaccine against bovine IMI associated with S aureus. Additional challenge trials should be carried out to evaluate substantial protection against S aureus under the commercial farm conditions.

  • 마크로파지의 활성을 조사하기 위해 carbon clearance time을 조사한 결과 AEY 처리구가 control이나 CEY 처리구에 비해 짧았다. 특히 AEY 500$\mu\textrm{g}$ 처리에서 carbon clearance 시간이 5분으로 control에서 9.42분, CEY(250$\mu\textrm{g}$ 처리)에서 9.01분보다 유의성 있는 감소를 보였다. AEY 처리에 의해 응집소가와 용혈소가가 다소증가되었다. SRBC처리 1일째와 3일째에 250$\mu\textrm{g}$ 처리구를 비교해 보면, control, CEY, AEY의 응집소가는 각각 5.50, 5.63, 6.00 및 5.25, 5.38, 5.50로 용혈소가는 각각 4.75, 5.38, 5.50 및 4.25, 5.63, 5.63으로 AEY 처리구가 control, CEY 처리구에 비해 면역 활성이 있었지만 유의성은 없었다.

곤충유래 항균 펩티드의 의약학적 적용 (Pharmaco-medical Application of Antimicrobial Peptides Derived from Insect)

  • 현재까지 곤충 항균 펩티드는 1980년에 세크로피아나방(Hyalophora cecropia) 번데기의 혈림프로부터 세크로핀(cecropin)이 처음으로 정제된 이후로 150개 이상의 펩티드가 분리되어 특성들이 보고되어 왔다. 그러므로 곤충은 항균 펩티드 선발을 위한 좋은 재료로서 고려되어 왔다. 곤충 항균 펩티드는 분자량이 작으며 양전하를 띠고 다양한 길이와 서열 및 구조를 갖는 양친매성의 특징을 갖는다. 곤충 항균 펩티드는 박테리아, 진균, 기생충, 그리고 바이러스와 같은 병원체들의 침입에 대항하여 곤충의 선천성 면역체계에서 중요한 역할을 수행한다. 대부분의 곤충 항균 펩티드들은 상처가 나거나 면역화 시 지방체와 다른 특정 조직들에서 유도 합성된다. 이어서 그 항균 펩티드들은 미생물들에 대항하여 작용하기 위해 혈림프로 분비되어 나온다. 이들 펩티드들은 항암활성을 포함하여 다양한 미생물들에 대해 광범위한 항균활성을 나타낸다. 곤충 항균 펩티드는 구조 및 서열상의 특징들에 기초하여 크게 4개의 패밀리로 나누어질 수 있다. 다시 말해서 α-나선형 펩티드, 시스테인-풍부 펩티드, 프롤린-풍부 펩티드, 그리고 글리신-풍부 펩티드/단백질이 그것이다. 예를 들면, 세크로핀, 곤충 디펜신(defensin), 프롤린-풍부 펩티드, 그리고 아타신(attacin)이 일반적인 곤충 항균 펩티드들인데, 글로베린(gloverin)과 모리신(moricin)은 나비목 종들에서만 확인되어 왔다. 본 총설에서는 곤충의 항균 펩티드들에 초점을 맞추어 곤충 항균 펩티드들의 적용 가능성 및 방향과 함께 현재의 지식들과 최근의 진전된 사항들에 대하여 논의하고자 한다.

누에로부터 핵다각체병 바이러스 방어관련 유전자 정보 분석 (Identification of Antiviral-related Genes Up-regulated in Response to Bombyx mori Nucleopolyhedrovirus)

  • 누에 BmNPV는 잠사업에 있어서 가장 위해한 바이러스로써 익히 보고되었으며, 종종 잠사업의 심각한 경제적 손실을 야기하기도 한다. 곤충의 박테리아, 곰팡이 그리고 원생동물과 같은 다양한 병원체에 대응하는 곤충의 생체 방어기작에 대한 연구가 많이 알려져 있지만, 항바이러스 기작에 대한 연구는 매우 부족한 실정이다. 따라서 본 연구에서는 누에서 처음으로 누에의 BmNPV에 대한 생체방어 관련 유전자를 선발하기 위하여, 누에에 인위적으로 BmNPV를 주사하여 면역을 유도한 다음, 이로부터 cDNA 유전자은행을 제작하였다. 제작된 cDNA 유전자은행으로부터 무작위로 3,332개의 cDNA 클론을 선발하여 정상 누에에 비하여 BmNPV에 의해 면역이 유도된 누에에서 차별화 발현되는 109종의 잠정 항바이러스 유전자 클론을 차별화선별법에 의해서 분리하였다. 본 연구를 통해 확보한 109개의 유전자 정보는 누에의 바이러스에 대한 면역반응뿐만 아니라 최근에 개발된 누에 형질전환 기술을 이용하여 BmNPV 저항성 누에 품종을 개발하는데 중요한 기초 자료를 제공할 것으로 기대되며 또한, 인간의 중요한 항바이러스제 개발을 위한 모델 곤충으로써 누에를 이용하는데 기초 자료로 활용될 것으로도 기대된다.

The effect of multi-strain probiotics as feed additives on performance, immunity, expression of nutrient transporter genes and gut morphometry in broiler chickens

  • Objective: This study was conducted to investigate the effects of dietary multi-strain probiotic (MSP) (Bacillus coagulans Unique IS2 + Bacillus subtillis UBBS14 + Saccharomyces boulardii Unique 28) on performance, gut morphology and expression of nutrient transporter related genes in broiler chickens. Methods: A total of 256 (4×8×8) day-old CARIBRO Vishal commercial broiler chicks of uniform body weight were randomly distributed into four treatments with 8 replicates each and having eight chicks in each replicate. Four dietary treatments were T1 (negative control-basal diet), T2 (positive control-antibiotic bacitracin methylene disalicylate at 20 mg/kg diet), T3 (MSP at 107 colony-forming unit [CFU]/g feed), and T4 (MSP at 108 CFU/g feed). Results: During 3 to 6 weeks and 0 to 6 weeks, the body weight gain increased significantly (p<0.05) in T3 and T4 groups. The feed intake significantly (p<0.05) reduced from T1 to T3 during 0 to 3 weeks and the feed conversion ratio also significantly (p<0.05) improved in T3 and T4 during 0 to 6 weeks. The humoral and cell mediated immune response and the weight of immune organs were also significantly (p<0.05) improved in T3 and T4. However, significant (p<0.05) dietary effects were observed on intestinal histo-morphometry of ileum in T3 followed by T4 and T2. At 14 d post hatch, the relative gene expression of glucose transporter (GLUT5), sodium-dependent glucose transporter (SGLT1) and peptide transporter (PepT1) showed a significant (p<0.05) up-regulating pattern in T2, T3, and T4. Whereas, at 21 d post hatch, the gene expression of SGLT1 and PepT1 was significantly (p<0.05) downregulated in MSP supplemented treatments T3 and T4. Conclusion: The supplementation of MSP at 107 CFU/g diet showed significant effects with improved performance, immune response, gut morphology and expression of nutrient transporter genes. Thus, the MSP could be a suitable alternative to antibiotic growth promoters in chicken diets.

SARS-CoV-2 mRNA Vaccine Elicits Sustained T Cell Responses Against the Omicron Variant in Adolescents

  • Vaccination against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been acknowledged as an effective mean of preventing infection and hospitalization. However, the emergence of highly transmissible SARS-CoV-2 variants of concern (VOCs) has led to substantial increase in infections among children and adolescents. Vaccine-induced immunity and longevity have not been well defined in this population. Therefore, we aimed to analyze humoral and cellular immune responses against ancestral and SARS-CoV-2 variants after two shots of the BNT162b2 vaccine in healthy adolescents. Although vaccination induced a robust increase of spike-specific binding Abs and neutralizing Abs against the ancestral and SARS-CoV-2 variants, the neutralizing activity against the Omicron variant was significantly low. On the contrary, vaccine-induced memory CD4+ T cells exhibited substantial responses against both ancestral and Omicron spike proteins. Notably, CD4+ T cell responses against both ancestral and Omicron strains were preserved at 3 months after two shots of the BNT162b2 vaccine without waning. Polyfunctionality of vaccine-induced memory T cells was also preserved in response to Omicron spike protein. The present findings characterize the protective immunity of vaccination for adolescents in the era of continuous emergence of variants/subvariants.

Immunization of Mice with Recombinant Brucella abortus Organic Hydroperoxide Resistance (Ohr) Protein Protects Against a Virulent Brucella abortus 544 Infection

  • In this study, the Brucella abortus ohr gene coding for an organic hydroperoxide resistance protein (Ohr) was cloned into a maltose fusion protein expression system (pMAL), inserted into Escherichia coli, and purified, and its immunogenicity was evaluated by western blot analysis using Brucella-positive mouse sera. The purified recombinant Ohr (rOhr) was treated with adjuvant and injected intraperitoneally into BALB/c mice. A protective immune response analysis revealed that rOhr induced a significant increase in both the IgG1 and IgG2a titers, and IgG2a reached a higher level than IgG1 after the second and third immunizations. Additionally, immunization with rOhr induced high production of IFN-γ as well as proinflammatory cytokines such as TNF, MCP-1, IL-12p70, and IL-6, but a lesser amount of IL-10, suggesting that rOhr predominantly elicited a cell-mediated immune response. In addition, immunization with rOhr caused a significantly higher degree of protection against a virulent B. abortus infection compared with a positive control group consisting of mice immunized with maltose-binding protein. These findings showed that B. abortus rOhr was able to induce both humoral and cell-mediated immunity in mice, which suggested that this recombinant protein could be a potential vaccine candidate for animal brucellosis.

Distinct Humoral and Cellular Immunity Induced by Alternating Prime-boost Vaccination Using Plasmid DNA and Live Viral Vector Vaccines Expressing the E Protein of Dengue Virus Type 2

  • Background: Dengue virus, which belongs to the Flavivirus genus of the Flaviviridae family, causes fatal dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) with infection risk of 2.5 billion people worldwide. However, approved vaccines are still not available. Here, we explored the immune responses induced by alternating prime-boost vaccination using DNA vaccine, adenovirus, and vaccinia virus expressing E protein of dengue virus type 2 (DenV2). Methods: Following immunization with DNA vaccine (pDE), adenovirus (rAd-E), and/or vaccinia virus (VV-E) expressing E protein, E protein-specific IgG and its isotypes were determined by conventional ELISA. Intracellular CD154 and cytokine staining was used for enumerating CD4+ T cells specific for E protein. E protein-specific CD8+ T cell responses were evaluated by in vivo CTL killing activity and intracellular IFN-${\gamma}$ staining. Results: Among three constructs, VV-E induced the most potent IgG responses, Th1-type cytokine production by stimulated CD4+ T cells, and the CD8+ T cell response. Furthermore, when the three constructs were used for alternating prime-boost vaccination, the results revealed a different pattern of CD4+ and CD8+ T cell responses. i) Priming with VV-E induced higher E-specific IgG level but it was decreased rapidly. ii) Strong CD8+ T cell responses specific for E protein were induced when VV-E was used for the priming step, and such CD8+ T cell responses were significantly boosted with pDE. iii) Priming with rAd-E induced stronger CD4+ T cell responses which subsequently boosted with pDE to a greater extent than VV-E and rAd-E. Conclusion: These results indicate that priming with live viral vector vaccines could induce different patterns of E protein-specific CD4+ and CD8+ T cell responses which were significantly enhanced by booster vaccination with the DNA vaccine. Therefore, our observation will provide valuable information for the establishment of optimal prime-boost vaccination against DenV.

Potentiation of Th1-Type Immune Responses to Mycobacterium tuberculosis Antigens in Mice by Cationic Liposomes Combined with De-O-Acylated Lipooligosaccharide

  • Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis. Bacillus Calmette-$Gu\acute{e}rin$ (BCG) vaccine is the only TB vaccine currently available, but it is not sufficiently effective in preventing active pulmonary TB or adult infection. With the purpose of developing an improved vaccine against TB that can overcome the limitations of the current BCG vaccine, we investigated whether adjuvant formulations containing de-O-acylated lipooligosaccharide (dLOS) are capable of enhancing the immunogenicity and protective efficacy of TB subunit vaccines. The results revealed that the dLOS/dimethyl dioctadecyl ammonium bromide (DDA) adjuvant formulation significantly increased both humoral and Th1-type cellular responses to TB subunit vaccine that are composed of three antigens, Ag85A, ESAT-6, and HspX. The adjuvanted TB vaccine also effectively induced the Th1-type response in a BCG-primed mouse model, suggesting a potential as a booster vaccine. Finally, the dLOS/DDA-adjuvanted TB vaccine showed protective efficacy against M. tuberculosis infection in vitro and in vivo. These data indicate that the dLOS/DDA adjuvant enhances the Th1-type immunity and protective efficacy of the TB subunit vaccine, suggesting that it would be a promising adjuvant candidate for the development of a booster vaccine.

Defining B Cell Epitopes of Ovalbumin for the C57BL/6 Mice Immunized with Recombinant Mycobacterium smegmatis

  • Recombinant Mycobacterium smegmatis expressing ovalbumin was used to immunize C57BL/6(H-$2^b$) mice, and the humoral immunity against recombinant ovalbumin was analyzed. Antibodies were purified by denatured ovalbumin-conjugated affinity chromatography. The epitopes of the antibodies were screened with a random peptide library displayed on the tip of fUSE5 filamentous phage pIII minor coat proteins. Two peptides, IRLADR and SPGAEV, were selected predominantly by the recognition of purified antibodies using biopanning methods. The composition of the peptide sequence with the primary structure of OVA revealed that the peptide sequence analogizes to INEAGR, part of the $^{323}ISQAVHAAHAEINEAGR^{339}$ sequence previously reported as the antigenic determinant for murine Band also Th cell epitopes (I-$A^d$ binding). Also, the structures of these mimotopes obtained from restrained molecular dynamic computations resulted in the formation of a $\beta$-turn proven to be a secondary structure of the parent peptide within the ovalbumin molecule, enabling us to confirm the structural similarity. This study demonstrates that immunization with recombinant M. smegmatis can generate neutralizing antibodies identical with those induced by the administration of natural antigenic proteins and supports the potential use of mycobacteria as vaccine delivery vehicles.
