  • Saccharomyces cerevisiae was immobilized by incubating iron oxides with calcium alginate, and by polyacrylamide entrapment to use repeatedly for the conversion of glucose to ethanol. Magnetic and non-magnetic immobilized yeast and polyacrylamide immobilized yeast were compared with the native yeast a batch-fermentation of ethanol from glucose. Three kinds of immobilized yeast tended almost identically, having ethanol productivity as well as the final yield about the same to what was found for the native yeast. The long-term operational stability of three kinds of immobilized yeast were significant difference according as immobilized yeast activation or non-activation before ethanol fermentation. In the non-activation they lost their activity of fermentation rapidly in the beginning stage an slower at a later stage. On the other hand, in the activation with nutrient media, their activities were increased to some extent and stable in the later stage. The cell count of three kinds of immobilized yeast after activiation by incubating nutrient media, increased by a factor of about 45 to 48, whereas the fermenting capacity increased by a factor of 174 to 178. In the prearation of immobilized biocatalysts, magnetic matter does not seem to have any adverse affect on the properties of the microorganism. The immobilized biocatalysts by utilizing magnetic matter have some advantages, especially in application of viscous media or insoluble particle-containing media, for this work was linked with microbial utilization of environmental wastes and elimination of envirnmental pollutant.

Toluene Removal and Microbial Growth of Candida tropicalis Immobilized with Polymer Media in Airlift Bioreactors (효모 Candida tropicalis 고정화 담체를 이용한 Airlift 미생물반응기의 톨루엔 제거 및 미생물 성장)

  • This study was conducted to improve biological degradation efficiency of toluene as a model volatile organic compound (VOC) using yeast Candida tropicalis and to suggest an effective method for bioreactor operation. The yeast strain was immobilized with polyethylene glycol (PEG), alginate, and powdered activated carbon (PAC). The yeast-immobilized polymer media were used as fluidized materials in an airlift bioreactor. Polymer media without PAC were also made and operated in another airlift bioreactor. The two bioreactors showed toluene removal efficiencies ranging 80-96% at loading rates of $10-35 g/m^3-hr$, and the bioreactor containing the polymer media with PAC achieved higher removal efficiency. Protein contents in the liquid phase showed that the bioreactor using the yeast-immobilized polymer media with PAC had a higher rate of microbial growth initially than that without PAC. In addition, the microbial growth rate inside of the polymer media with PAC was five times higher than that without PAC. Consequently, the polymer media containing the yeast strain and PAC could enhance removal efficiencies for VOCs, and the immobilization method improve microbial activity and stability for a long-term operation of biological systems.

Ammonium Ion Effects and Its In Situ Removal by Using Immobilized Adsorbent in Hybridoma Cell Culture (하이브리도마 세포배양에서 암모늄 이온의 영향 및 고정화 흡착제에 의한 암모늄 이온의 동시제거)

  • The effects of ammonium ion on cell growth kinetics, monoclonal antibody productivity, and cell metabolism of hybridoma cells were investigated. The mouse-mouse hybridoma cell line VlIIH-8 producing mouse IgG2a was used as a model system. Ammonium ion showed an inhibitory effect on cell growth and monoclonal antibody production. New immobilized adsorbents were developed for the reduction of the inhibitory effect of ammonium ion. The ammonium ion selective zeolite, Phillipsite-Gismondine was entrapped in calcium alginate bead or in dialysis membrane and applied to the hybridoma cell culture system for the in situ removal of ammonium ion from culture media. The effects of ammonium the both serum supplemented and serum free media on the cell growth were studied by applying immobilized adsorbents of calcium alginate bead type. The results demonstrated a substantial enhancement in cell growth. Applying immobilized adsorbents of dialysis membrane type to serum supplemented media also resulted in the stimulation of cell growth, cell viability and monoclonal antibody production.

  • Biofiltration was successfully applied to treat a mixture of volatile organic compounds(benzene, xylene) from contaminated air stream. Immobilized Ps. oleovorans biofilter was evaluated for its value in simultaneous removal of benzene and xylene from waste air stream. The variety of operating conditions were tested to evaluate important factors such as space velocity, pH, water content, etc.

Immobilization Imparts Stability to Watermelon Urease to Work in Water Miscible Organic Media

  • The behaviour of alginate immobilized and soluble watermelon (Citrullus vulgaris) urease in water miscible organic solvents like, acetonitrile, dimethylformamide (DMF), ethanol, methanol, and propanol is described. The organic solvents exhibited a concentration dependent inhibitory effect on both the immobilized and the soluble urease in the presence of urea. Pretreatment of soluble enzyme preparations with organic solvents in the absence of substrate for 10 min at $30^{\circ}C$ led to rapid loss in the activity, while similar pretreatment of immobilized urease with 50% (v/v) of ethanol, propanol, and acetonitrile was ineffective. Time-dependent inactivation of immobilized urease, both in the presence and in the absence of urea, revealed stability for longer duration of time even at very high concentration of organic solvents. The soluble enzyme, on the other hand, was rapidly inactivated even at fairly lower concentrations. The results suggest that the immobilization of watermelon urease in calcium alginate make it suitable for its application in organic media. The observations are discussed.

Bioconversion of progesterone by immobilized aspergillus phoenicis (고정화된 aspergillus phoenicis를 이용한 progesterone 전환)

  • Progestrone bioconversion by immobilized Aspergillus phoenicis was studied. Progesterone was converted into 11$\alpha$-hydroxyprogesterone and 3-minor byproducts. Whole cells of A. phoenicis were immobillized by enreappment with calcium-alginate, K-carrageenan, or polyacrylamide. Of these materials tested, cell immobilized in $Ca^{2+}$ -alginate gels showed the highest activity for 11$\alpha$-hydroxylation of progesterone. In the case of mycelia immobilized in $Ca^{2+}$-alginate, futher progressing hydroxylation of 11$\alpha$-hydroxyprogesterone was greatly reduced. Spores of A. phoenicis which were immobillized with $Ca^{2+}$-alginate and germinatedin situ for 25 hours showed higher 11$\alpha$-hydroxylase activity than those of entrapped whole mycelia and maintained initial enzyme activity for all 8 times of repeated use. After 16 times of reuse, the activity was declined 30% or more. When culture media and $Zn^{2+}$ were introduced into the reaction media, the activity of the immobilized mycelia which had been lowered due to many times of reuse was effectively reactivated.

Complex odor removal in pilot-scale biofilter with microorganisms immobilized on polymer gel media (미생물 포괄고정화 담체를 적용한 파일럿 스케일 바이오필터에서의 복합악취 제거)

  • A pilot-scale biofilter was constructed to discover degradation characteristics of the complex odor discharged from Ansan wastewater treatment plant. Candida tropicalis for volatile organic compounds, sulfur oxidizing bacteria(SOB) for hydrogen sulfide, and bacteria extracted from feces soil were immobilized on a polymer gel media. According to this study, the EBCT was varied from 36 sec to 18 sec. Toluene was removed as 80% along the variations, but it was recovered as 100% within 1 week. All benzene and xylene were removed during the operation while the efficiency of hydrogen sulfur was temporary decreased at 18 sec of EBCT, thereafter it was recovered to 100% within a week. The maximum elimination capacities of the benzene, toluene, xylene, and hydrogen sulfur were 6.6 g/$m^{3}$/hr, 31.7 g/$m^{3}$/hr, 7.8 g/$m^{3}$/hr, and 133.6 g/$m^{3}$/hr, respectively. There were merits on removal both organic and inorganic complex odor using the pilot-scale biofilter embedded with microorganisms immobilized on polymer gel media.

Stabile Fermentation of Citric Acid Using Immobilized Saccharomycopsis lipolytica

  • The effects of media composition on citric acid fermentation using surface immobilized Saccharomycopsis lipolytica were studied. The use of the standard medium for these organisms resulted in rapid decrease of citric acid production and a transformation of immobilized cell morphologies from a yeast-type to a mycelium-type. When the standard medium was enriched with vitamins, trace minerals, a growth factor and ammonium to form a Vigorous Stationary Phase (VSP) fermentation type medium, relatively stable citric acid production (10 mg/lㆍh) was obtained. Using the VSP type medium, the surface immobilized cells also retained their yeast-type form.

Elimination capacities of toluene and ammonia in the bio-filter system depending on type of media (담체 종류에 따른 바이오필터의 톨루엔과 암모니아 분해능 평가)

  • Contribution of immobilized media with bacteria to the odor removal was evaluated in a lab scale bio-filter compared to that with sponge or ceramic media without the immobilized bacteria. Candida tropicalis for volatile organic compounds and ammonium oxidizing bacteria (AOB) for inorganic compounds were used as seeds in lab-scale bio-reactors. Three different type of media in the bio-reactors that immobilized bioreactor (IBR), sponge bioreactor (SBR), and ceramic bioreactor (CBR) were examined, respectively. An empty bed contact time (EBCT) of the bio-filters was fixed as 60 seconds, and the inlet concentration of toluene was changed from 20 ppm to 200 ppm to observe the removal efficiency depending on the concentrations. As a result, the maximum elimination capacities of IBR, SBR, and CBR were 166 $g/m^3/hr$, 138 $g/m^3/hr$, and 138 $g/m^3/hr$, respectively. In addition, toluene as an organic compound and ammonia as an inorganic compound were applied together with different inlet concentrations varied from 80 ppm to 250 ppm of toluene and from 2.5 ppm to 40 ppm of ammonia. The toluene maximum elimination capacities in IBR, SBR, and CBR were 97.4 $g/m^3/hr$, 59.5 $g/m^3/hr$, and 81.9 $g/m^3/hr$, respectively. The ammonia maximum elimination capacities were reached as 7.2 $g/m^3/hr$ in IBR, 6.6 $g/m^3/hr$ in SBR, and 7.0 $g/m^3/hr$ in CBR.

Application of AuNPs immobilized on UV Cross-linked P4VP Thin Film as pH Nanosensors (pH 나노센서로의 응용을 위한 UV-가교 P4VP 박막에 고정한 금 나노입자의 특성)

  • In this report, we describe the use of gold nanoparticles (AuNPs) immobilized on pH. responsive, cross-linked poly(4-vinylpyridine) (P4VP) thin films, as a potential application for pH nanosensors. The methodology is based on the variation in surface plasmon resonance of immobilized AuNPs with changing the interparticle distances, caused by the swelling/deswelling of the pH responsive P4VP polymer films. The change in optical properties of the immobilized AuNPs in response to the pH of surrounding media was investigated by a simple yet powerful tool; UV-vis absorption spectroscopy. The swelling of the P4VP chains at pH 2 causes an increase in the interparticle distances of immobilized AUNPS ($\sim20nm$) and hence leads to a blue shift of 48 nm in their surface plasmon resonance band peak. On the other hand, when the surrounding media was altered from pH 2 to 10, a red shift of absorption maxima was observed. The changes were rapid, and the effect was reversible. This system could prove to be useful in fabricating nanosensors for detecting the pH or pH changes of surrounding aqueous medium.
