Th2 세포에서 IL-12에 의한 IL-18R ${\alpha}$의 발현유지 및 IL-18 자극에 의한 GATA-3의 유도 (IL-18R ${\alpha}$ Mediated GATA-3 Induction by Th2 Cells: IL-12 Supports IL-18R ${\alpha}$ Expression in Th2 Cells)

  • Background: IL-18 was originally cloned as a IFN-${\gamma}$ inducing factor in primed T cells. In synergy with IL-12, IL-18 has been shown to induce strikingly high levels of IFN-${\gamma}$ production by T cells and to enhance Th1 development. Also this cytokine exerts induction of Th2 development through IL-4 induction. Methods: Resting $CD4^+$ T cells were sorted by negative selection and activated by anti-CD3 plus anti-CD28 Ab. Expression of IL-12 binding sites, IL-18 binding sites, IL-18R ${\alpha}$, and GATA-3 mRNA were analysed by FACS and RT-PCR, respectively. Results: Resting $CD4^+$ T cells expressed IL-18R ${\alpha}$ chain but not IL-18 binding sites, suggesting a lack of IL-18R ${\beta}$ expression. IL-18R ${\alpha}$ was maintained on the Th1 and Th2 committed cells. IL-18 binding sites were induced on the Th1 but not Th2 cells. Exposure of these cells to IL-18 led to up-regulation of GATA-3 mRNA expression only in Th2 committed cells. To elucidate the relationship between IL-18R ${\alpha}$ expression and GATA-3 induction by IL-18, Th1 and Th2 committed cells were further cultured in medium with or without IL-12 for 2 days. IL-12 binding sites were maintained on the Th1 and Th2 cells regardless of IL-12 treatment, but IL-18R a expression was rapidly down-regulated on the IL12-untreated Th2 cells which did not induce GATA-3 mRNA expression followed by IL-18 stimulation. Conclusion: IL-12 supports expression of IL-18R ${\alpha}$ and GATA-3 mRNA expression was induced by IL-18 through IL-18R ${\alpha}$ without expression of IL-18 binding site in Th2 cells.

  • Tuna cooking juice concentrate (TCJC) is by-produced during the canning processing of skipjack tuna Katsuwonus pelamis and it is well known that TCJC contains various nutritional components. Therefore, the immuno-stimulating activity of TCJC was investigated using macrophage RAW 264.7 cell line and the spleen cell isolated from BALB/c mice. The TCJC increased the production of IL-6, TNF-${\alpha}$, and IL-$1{\beta}$ in a dose-dependent manner compared to the control in RAW 264.7 cells without any toxicity. In particular, the production of TNF-${\alpha}$ was increased over 300-fold. The production of both Th1 cytokine (such as IFN-${\gamma}$, TNF-${\alpha}$, IL-2, and IL-12) and Th2 cytokine (IL-4, IL-6, IL-10) was also increased by TCJC treatment in splenocytes. Moreover, the TCJC increased the splenocyte proliferation in a concentration-dependent manner compared to control. These results indicate that TCJC may enhance immune function by promoting various cytokine production.

  • KASI is planning a large telescope project to acquire 10% share of the GMT (Giant Magellan Telescope) extremely large telescope as one of national R&D project. By participating in GMT, we expect to provide the best environment for observational astronomers in Korea and give opportunity to upgrade the current research and instrumentation level to world-leading class. In this contribution, we will report the concept of the GMT project and Korean participation plan as well as the current status of fundraising activity.

The Effect of Adiponectin on the Regulation of Filaggrin Expression in Normal Human Epidermal Keratinocytes

  • Background: Adiponectin, an adipokine secreted from adipocytes, affects energy metabolism and also shows anti-diabetic and anti-inflammatory properties. Recent studies have reported that adiponectin plays a role in regulating skin inflammation. Objective: This study aimed to investigate the effect of adiponectin on the expression of filaggrin (FLG) in normal human epidermal keratinocytes (NHEKs). Methods: NHEKs were serum-starved for 6h before being treated with adiponectin. Afterward, cell viability was assessed by MTT assay. We also treated with calcium, interleukin (IL)-4, and IL-13 to provide positive and negative comparative controls, respectively. Gene mRNA expression was quantified using real time reverse transcription polymerase chain reaction, and protein expression was evaluated using Western blot. To evaluate the relationship among mitogen-activated protein kinases (MAPKs), activator protein 1 (AP-1), and FLG, we also treated cells with inhibitors for MAPKs JNK, p38, and ERK1/2. Results: FLG and FLG-2 mRNA expression in NHEKs significantly increased after treatment with $10{\mu}g/ml$ adiponectin. Adiponectin also restored FLG and FLG-2 mRNA expression that was otherwise inhibited by treatment with IL-4 and IL-13. Adiponectin induced FLG expression via AP-1 and MAPK signaling. Conclusion: Adiponectin positively regulated the expression of FLG and could be useful as a therapeutic agent to control diseases related to disrupted skin barrier function.

Participation of IL-1β in temporomandibular nociception in rats with CFA-induced inflammation

  • The aim of the present study was to develop an animal model for evaluation of temporomandibular (TMJ) nociception under TMJ inflammation. We also investigated the participation of $IL-1{\beta}$ in inflammation-induced TMJ nociception. Experiments were carried out using male Sprague-Dawley rats. Intra-articular injection of 3% formalin was administered to evaluate hyperalgesia 3 days after CFA injection. Intra-articular injection of 3% formalin did not produce nociceptive behavior in normal rats. Although intra-articular injection of 3 doses of CFA produced TMJ inflammation, only 1:3 diluted CFA produced hyperalgesia when formalin was injected intra-articularly 3 days after CFA injection. Co-administration of IL-1 receptor inhibitor with formalin into the TMJ cavity 3 days after CFA injection was performed. Co-administration of IL-1 receptor inhibitor significantly inhibited formalin-induced hyperalgesia in rats with CFA-induced TMJ inflammation. These results suggested that intra-articular injection of formalin produced hyperalgesia under chronic TMJ inflammation. Moreover, $IL-1{\beta}$ plays an important role in TMJ hyperalgesia under chronic inflammation and blockade of $IL-1{\beta}$ is a potential therapeutic target for inflammatory TMJ pain.

Genetic Variants in Interleukin-2 and Risk of Lymphoma among Children in Korea

  • To estimate the genetic susceptibility for childhood lymphoma, we conducted an association study for 23 cases and 148 controls. Total 1536 tag single nucleotide polymorphisms (SNPs) were selected in 138 candidate gene regions related to immune responses, apoptosis, the cell cycle, and DNA repair. Twelve SNPs were significantly associated with the risk of lymphoma ($P_{trend}$ <0.05) in six genes ($IL1RN$, $IL2$, $IL12RB1$, $JAK3$, $TNFRSF13B$, and $XRCC3$). The most significant association was seen for $IL2$ variant rs2069762 ($OR_{TG+GG}$ vs. TT=3.43 (1.29-9.11), $P_{trend}$=0.002, min$P$=0.005). These findings suggest that common genetic variants in $IL2$ might play a role in the pathogenesis of childhood lymphoma.

  • Sopungchungyoung-tang (SCT) has been widely used in Korea as a treatment of atopic dermatitis. SCT consists of Talcum, Rehmannia glutinosa, Angelica sinensis, Paeonia lactiflora, Cnidium officinale, Ledebouriella divaricata, Schizonepeta tenuifolia, Scutellaria baicalensis, Glycyrrhiza uralensis, Mentha arvensis, Cordyceps cicadae. We examined the immunological effect of SCT in vitro. We studied about the effect of SCT on Th cells' differentiation. In the case of CD4 T cells under neutral condition where there was only rIL-2 stimulus, SCT inhibited IFN-γ secretion by 70-80 %. Likewise, SCT also inhibited the IL-4 secretion of neutral Th cells by 85-90 %. We also experimented with the polarized Th1 cells/ Th2 cells and their production of IFN-γ and IL-4, respectively. There also were inhibitory effects on the polarized cells like there was on neutral cells, they were not as strong on the polarized cells. Under Th1 polarized condition, SCT acted dose-dependently, while in Th2 cells, the IL-4 production was inversely proportional to the doses of SCT. From the current study, it can be concluded that SCT exerts inhibitory effects on cytokine production without interfering with immune cells' activity. The result that SCT inhibits IFN-γ and IL-4 confirms that it does have a effect on immunomodulation.

  • This paper deals with the ways that can improve the reliability of the course evaluation system based on reformation of the system and design of user interface for the practical applications. In the reformation of the system aspect, there are the improvements of the course evaluation questionnaires and the feedback of course evaluation to the lecturers. In the design aspect, a computer screen of the graphic user interface is redesigned to reflect the reformation of the system aspect issues. The results show the effectiveness in the reliability improvement.

근병증을 동반한 콩팥성전신섬유화증 (Nephrogenic Systemic Fibrosis with Myopathy)

  • Nephrogenic systemic fibrosis (NSF) is a systemic disease that affects the skin and other tissues in patients with renal insufficiency and exposure to gadolinium-containing contrast. A 55-year-old woman with end-stage renal disease on hemodialysis was consulted for progressive general weakness. After she had undergone multiple MRIs with gadolinium-containing contrast media, muscle weakness and skin lesions were developed. Her skin and muscle biopsy specimens showed CD34+ fibroblast entrapping collagen bundles. There are few reports of NSF with myopathy.
