  • Ginsan, a polysaccharide extracted from Panax ginseng. was earlier scrutinized for a biological-response modifier. We further studied the protective and restorative activity of Ginsan against sublethal dose irradiation owing to increase production of endogenous hematopoietic growth factors such as IL-1. TNF-${\alpha}$. IL-6, GM-CSF. Which induce strong redox-emzyme elevation. Exposing to radiation induces reactive oxygen species (ROS). which play an important causative role in radiation damage.   • Objectives The purpose of this study was to assess the efficacy of Chuna and Stylex device treatment in patients with genu varum. Methods 2 patients (21/male, 40/female) with genu varum were recruited in this study. Chuna treatment at pelvic, and stylex device treatment at both legs were applied to all participants. Outcomes were assessed using Q-angle (Quadriceos angle), TFA (Tibiovemoral angle), Distance of both sides patella. Results 1. Distance of both sides patella were decrease in all case. 2. Q-angel and TFA were decrease in case 1. but slight invrease in case 2. Conclusions This study showed that Chuna and Stlyex divice treatments could be used as an effective method for decrease distance of both sides patella. but Q-angle and TFA need more research on whether proper assessment as a measure of genu varum.

Characterization of the N-glycosylation of Recombinant IL-4 and IL-13 Proteins Using LC-MS/MS Analysis and the I-GPA Platform

  • Interleukin-4 (IL-4) and IL-13 are cytokines secreted by immune cells. Cytokines induce the proliferation of macrophages or promote the differentiation of secretory cells. The initiation and progression of allergic inflammatory diseases, such as asthma, are dependent on cytokines acting through related receptor complexes. IL-4 and IL-13 are N-glycoproteins. Glycan structures in glycoproteins play important roles in protein folding, protein stability, enzymatic function, inflammation, and cancer development. Therefore, the glycan structure of IL-4 and IL-13 needs to be elucidated in detail for the development of effective therapies. We report the first attempt to characterize the site-specific N-glycosylation of recombinant IL-4 and IL-13 via liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. The tandem mass spectra of intact N-glycopeptides were identified using the Integrated GlycoProteome Analyzer (I-GPA) platform, which can automatically and rapidly analyze multiple N-glycopeptides, including their glycan composition and amino acid sequences. The recombinant IL-4 and IL-13 were identified with amino acid sequence coverages of 84% and 96%, respectively. For IL-4, 52 glycoforms on one N-glycosylation site were identified and quantified. In IL-13, 232 N-glycopeptides from three N-glycosylation sites were characterized, with the site Asn52 being the most extensively glycosylated (~80%). The complex glycans were the most abundant glycan on IL-4 and IL-13 (~96% and 91%, respectively), and the biantennary glycans were the most abundant in both recombinant IL-4 and IL-13 proteins.

  • In order to evaluate the antiallergic effects of CSYJT, studies were done. We measured the cytotoxic activity for cytokines transcript expression, production of IL-4, IgE, $IFN-{\gamma}$, proliferation of B cell in HRF plus anti-CD40mAb plus rIL-4 stimulated murine splenic B cells. and cytokines transcript expression of IgE in Mast cell The results were obtained as follows: 1. CSYJT decreased the expression of IL-4 in mast cell significantly. 2. CSYJT decreased the production of IL-4 significantly. 3. CSYJT decreased the expression of IgE in mast cell significantly. 4. CSYJT decreased the production of IgE significantly. 5. CSYJT increased the appearance of $IFN-{\gamma}$. The facts above prove that CSYJT is effective against the allergy. Thus, I think that we should study on this continuously.

The Effects of Gudambopae-Tang on Changes of Cytokines in Allergic Rhinitis Model (거담보폐탕(去痰補肺湯)이 알레르기성 비염 모델 생쥐의 cytokine변화에 미치는 영향)

  • In order to investigate the effects of Gudambopae-Tang on allergic rhinitis, laboratory study about cytokines and OVA-specific IgE was perfomed in the present study. The results are summarized as follow; 1. IL4, IL-5 was increased in medicating group than control group. 2. $IFN-{\gamma}$ was increased in medicating group than control group. 3. total IgE and OVA-specific IgE was significant decreased in medicating group than control group, statistically significant. This study shows experimentally that Gudambopae-tang could reduce allergic reaction in allergic rhinitis. Advanced studies are required to investigate the mechanisms of herbal medicine in allergic rhinitis model.
