콩의 발아 전 침종처리가 발아에 미치는 영향 (Effect of Pre-Germination by Treatment of Soaking on Germination of Soybean)

  • 우리나라에서 육성된 콩 품종 90개에 대하여 종자의 발아 전 침종처리에 따른 발아능과 발아율을 조사한 결과는 다음과 같다. 침수량과 침종일수에 따른 콩 종자의 발아력은 침수량이 증가하고, 침종일수가 증가할수록 낮아졌는데, 침수량 90ml와 3일간 침종에서 품종간의 차이가 크게 나타났다. 종자의 수분흡수는 침종 후 6시간까지 급격하게 증가한 후 완만하게 증가하다가 24시간 이후부터는 줄어들었는데, 나물콩의 수분 흡수가 가장 낮았고, 밥밑콩의 수분함량이 가장 낮았다. 용존산소량은 침종 후 3시간까지는 급격히 감소하다가 그 이후에는 완만하게 감소하였는데, 풋콩 및 올콩은 침종 3시간 후부터 용존산소량이 가장 낮았으나 24시간이후에는 다른 용도의 콩에 비해 높아졌다. 전기전도율과 TDS는 침종 후 시간이 경과함에 따라 증가하였는데 풋콩 및 올콩이 가장 높았고, 다음으로 장류콩, 밥밑콩이 높았으며 종자의 크기가 작은 나물콩이 가장 낮았다. 침종 후 발아율이 높은 품종은 장류콩에서는 장수콩, 나물콩에서는 소백나물콩, 밥밑콩에서는 선흑콩, 그리고 풋콩 및 올콩에서는 선록콩이었으며, 발아율이 낮은 품종은 장류콩에서는 익산과 송학콩, 나물콩에서는 방사콩, 밥밑콩에서는 진율콩, 풋콩 및 올콩에서는 신록콩이었다. 전기전도율과 TDS가 낮은 품종들은 발아율이 높았고, 높은 품종은 발아율이 낮은 경향이었다. 종자의 수분흡수가 낮은 품종들은 발아율이 높아 수분흡수와 발아율은 부의 상관이 있었다. 종자의 무게가 높은 품종은 전기전도율과 TDS가 높았으며, 종자의 무게가 낮은 품종은 전기전도율과 TDS도 낮아 정의 상관이 있었다.

Identification of Subspecies-specific STS Markers and Their Association with Segregation Distortion in Rice(Oryza sativa L.)

  • Two subspecies, japonica and indica, have been reported in rice, which differ in several ecotypic traits. However, reproductive barriers in hybrid progenies between subspecies have been major obstacles in breeding programs using inter-subspecific hybridization. As the first step to elucidate the reproductive barriers, we developed subspecies-specific(SS) STS markers in this study. A total of 765 STS primers were designed through comparing DNA sequences at every $2{\sim}3$cM interval between japonica and indica rices, which are available at Web DBs such as IRGSP, NCBI, TIGR, and GRAMENE, and tested for subspecies-specificity using 15 indica and 15 japonica varieties of diverse origin. Of them, 67 STS markers were identified as SS STS markers and their subspecies-specificity scores were estimated. The SS markers were dispersed throughout the genome along chromosomes. Of them, 64 SS markers were mapped on an RIL population derived from a Dasanbyeo(indica)/TR22183(japonica) cross. Genomic inclination of RILs was evaluated based on the genotyping with different types of markers. Association test between markers and segregation distortion revealed that segregation distortion might not be the cause of generating SS markers. The SS markers will be applicable to estimate the genomic inclination of varieties or lines and to study the differentiation of indica and japonica, and ultimately to breed true hybrid rice varieties in which desirable characters from both subspecies are recombined.

Screening of Selected Korean Sweetpotato (Ipomoea batatas) Varieties for Fusarium Storage Root Rot (Fusarium solani) Resistance

  • A common post-harvest disease of sweetpotato tuber is root rot caused by Fusarium solani in Korea as well as the other countries. Storage root rot disease was monitored earlier on sweetpotato (Ipomoea batatas) in storehouses of different locations in Korea. In the present study, an isolate SPL16124 was choosen and collected from Sweetpotato Research Lab., Bioenergy Crop Research Institute, NICS, Muan, Korea, and confirmed the identification as Fusarium solani by conidial and molecular phylogenetic analysis (internal transcribed spacer ITS and translation elongation factor EF 1-α gene sequences). The isolate was cultured on potato dextrose agar, and conidiation was induced. The fungus was screened for Fusarium root rot on tuber of 14 different varieties. Among the tested variety, Yenjami, Singeonmi, Daeyumi, and Sinjami showed resistant to root rot disease. Additionally, the pathogen was tested for pathogenicity on stalks of these varieties. No symptom was observed on the stalk, and it was confirmed that the disease is tissue specific.

Physiological responses of selected Philippine upland rice genotypes evaluated using drought and salinity stress

  • Screening for drought and salinity tolerance was undertaken for selected Philippine upland rice landraces during germinative and seedling stages to identify varieties which can potentially be grown in marginally dry and saline soils. While increasing PEG and NaCl concentrations caused obvious signs of injury to all rice genotypes, considerable varietal differences were noted in the nature of responses providing evidence that these genotypes possess broad intraspecific genetic variations for drought and salt tolerance. Inconsistent responses of these varieties during both growth stages highlight complexities involved in stress responses and underscore the futility of utilizing a single stage in the rice plant's life cycle for physiological screening. Notwithstanding these perplexing responses, G_Katiil and Ml-Pilit Tapul were observed to thrive relatively well despite increased salt and drought stress during early growth stages and may therefore possess genes needed in crop improvement efforts for drought and salinity tolerance. While these results do not reflect the entire spectrum of adaptive expression to drought and salinity stress during the life cycle of the upland rice plant, they nonetheless provide an easy, reliable and reproducible method for preliminary identification of drought and salt tolerant rice varieties.

Genetic Variability Based on Randomly Amplified Polymorphic DNA in Kacip Fatimah (Labisia pumila Benth & Hook f) collected from Melaka and Negeri Sembilan States of Malaysia

  • In Malaysia, Labisia pumila Benth & Hook f, popularly known as 'Kacip Fatimah' has been used traditionally to treat various elements of the woman's health in Malay community. The objective of this study was to develop randomly amplified polymorphic DNA (RAPD) based DNA markers for the identification of L. pumila and to distinguish its three varieties from each other. Total DNA from nine accessions of L. pumila was extracted by CTAB method and polymerase chain reactions (PCR) were carried out to amplify the segments of DNA using different primers to develop DNA barcode using RAPD technique. To find out variety-specific DNA marker/s, twenty different 10-mer primer sequences with annealing temperature from 36-$40^{\circ}C$ were evaluated in triplicate. Out of 20 random primers, two primers (OPA-1 and OPA-2/A10) were selected which produced reliable RAPD band patterns. To have DNA based handle, two RAPD amplification products were cloned and sequenced to determine the identity of the DNA. RAPD analysis using two random primers generated 72 discrete bands ranging in size 200 bp-3,000 bp. Fifty nine of these were polymorphic loci (82%) and thirteen were non-polymorphic loci (18%). A total of 32 bands polymorphic loci (72%) were amplified with primer OPA-1 and analyzed by cluster analysis and UPGMA (Unweighted Pair Group Method with Arithmetic) to present a dendogram depicting the degree of genetic relationship among nine accessions of L. pumila. Our results shows the reasonable genetic diversity among the L. pumila varieties and within varieties; and two RAPD marker sequences obtained could be used to identify L. pumila at species level.

1차종발생자료를 응용한 지리참조연산표준화 및 자료 품질 관리의 필요성 - 쇠무릎과 털쇠무릎의 적용 사례 - (Georeferencing of Primary Species Occurrence Data and Necessity of Data Quality Control - A Case Study of Two Varieties of Ox-Knee, Achyranthes bidentata Blume -)

  • 한약재 우슬로 알려진 쇠무릎과 털쇠무릎 두 변종 식물표본자료의 지리참조연산 표준화 작업을 통해, 1차종 발생데이터를 수집하면서 발생된 오류의 특성과 발생 원인을 조사하였다. 본 연구에서 시도한 재동정의 경우, 변종 수준의 동정에서는 쇠무릎의 경우 41%, 털쇠무릎의 경우 28%가 오동정으로 확인되었지만, 동정자의 동정신뢰도 수준은 변종의 동정 정확도와 무관하였다. 전체 303개 표본자료 중 71점의 지리참조연산을 실시하여 산출된 불확실성의 범위는 0.1297 km(최소)-32.4 km(최대), 평균 4,055m, 표준편차 5,772m로 확인되었다. 털쇠무릎은 한반도 중부에 넓게 분포하였으나, 쇠무릎은 경상남도와 전라남도를 포함한 남부해안 지역과 서해안 지역을 중심으로 분포하였다. 바이오클림(Bioclim) 분포모델을 적용한 결과, 쇠무릎의 최적생육지역은 남부지방 및 서부해안지역인 반면, 털쇠무릎의 경우 충청남도를 중심으로 한 중부지역임을 확인하였다. 두 분류군이 위도상으로 구분되는 가장 중요한 원인은 기후 인자 중 강수량보다는 기온인자로 확인되었다. 표본자료의 정보량을 판단하기 위해 지리참조연산결과 종의 전체적인 분포정보, 기후 정보 모델링을 통한 최적생육분포 면적 등의 정보량이 증가되었다. 본 연구에서 실시한 것과 같이 자료 품질 향상을 위해 최초 자료수집과 이후 입력까지 모든 관리 절차에서 발생하는 오류를 관리할 필요가 있다.

Single Nucleotide Polymorphism (SNP) Discovery and Kompetitive Allele-Specific PCR (KASP) Marker Development with Korean Japonica Rice Varieties

  • Genome resequencing by next-generation sequencing technology can reveal numerous single nucleotide polymorphisms (SNPs) within a closely-related cultivar group, which would enable the development of sufficient SNP markers for mapping and the identification of useful genes present in the cultivar group. We analyzed genome sequence data from 13 Korean japonica rice varieties and discovered 740,566 SNPs. The SNPs were distributed at 100-kbp intervals throughout the rice genome, although the SNP density was uneven among the chromosomes. Of the 740,566 SNPs, 1,014 SNP sites were selected on the basis of polymorphism information content (PIC) value higher than 0.4 per 200-kbp interval, and 506 of these SNPs were converted to Kompetitive Allele-Specific PCR (KASP) markers. The 506 KASP markers were tested for genotyping with the 13 sequenced Korean japonica rice varieties, and polymorphisms were detected in 400 KASP markers (79.1%) which would be suitable for genetic analysis and molecular breeding. Additionally, a genetic map comprising 205 KASP markers was successfully constructed with 188 $F_2$ progenies derived from a cross between the varieties, Junam and Nampyeong. In a phylogenetic analysis with 81 KASP markers, 13 Korean japonica varieties showed close genetic relationships and were divided into three groups. More KASP markers are being developed and these markers will be utilized in gene mapping, quantitative trait locus (QTL) analysis, marker-assisted selection and other strategies relevant to crop improvement.

분자표지자에 의한 지황 유전집단의 유전적 다양성 (Genetic Diversity of Rehmannia glutinosa Genotypes Assessed by Molecular Markers)

  • RAPD 분석을 이용하여 지황 육성 계통과 지역 수집종 들을 구분할 수 있는 분자표지자를 선발하고, 집단 간, 집단 내 유전적 다양성을 평가하기 위하여 본 실험을 수행하였다. 총 20개의 임의 primer를 이용하여 PCR 한결과, 육성 계통과 수집종 들을 구별할 수 있는 OPA-1 등 10개의 재현성과 다형성이 좋은 프라이머 들을 선발하였다. 특히 OPA-10, OPA-11 및 OPA-19는 고려지황과 지황1호를 다른 계통 및 수집종 들과 구별할 수 있었으며, 이들 프라이머를 이용하여 0.9 kb, 1.2 kb, 1.3 kb 및 1.4 kb등의 육성계통 특이적인 DNA 밴드들을 확보할 수 있었다. 한편 이들의 결과를 토대로 통계처리에 의한 유전분석 결과, 고려지황, 지황1호 및 일본지황은 집단 내 유사도가 높아 다른 집단들과 구별되었다. 결론적으로, RAPD 분석을 통한 결과는 지황의 유전적 다양성 이해와 특정 계통을 다른 계통 및 수집종 들과 구분할 수 있는 방법으로 이용될 수 있다.

'개량머루', '거봉' 및 '홍이슬' 품종의 anthocyanin 조성과 특징 (Anthocyanin Composition and Characteristics of 'Gaeryangmeoru', 'Kyoho', and 'Hongisul' Grape Varieties)

  • 본 연구는 국내에서 재배중인 '개량머루', '거봉', '홍이슬' 품종의 anthocyanin 조성을 UPLC-ESI-MS/MS를 이용하여 조사하였다. '개량머루'는 착색에 불량하게 작용하는 기상 조건에서도 착색이 우수한 흑포도 품종이고, '거봉'과 '홍이슬'은 교잡종 포도로 각각 검정색과 분홍색 과피를 갖고 있다. 과피에서 추출된 anthocyanin의 조성은 UPLC-ESI-MS/MS를 이용하여 분석하였다. 각 품종에서 검출된 anthocyanin은 '개량머루'와 '거봉'에서는 각각 25가지, '홍이슬'에서는 21가지였고, 각 품종에서 검출량이 많았던 anthocyanin은 8(개량머루), 15(거봉), 5(홍이슬)가지였다. 세 품종 모두 mono-glucoside가 di-glucoside보다 많았으며, '개량머루'와 '거봉'은 malvidin이 많았던 반면, '홍이슬'은 cyanidin이 가장 많았다. '개량머루'는 acylated anthocyanin(2.0%)은 거의 존재하지 않았고 '거봉'과 '홍이슬'에서는 p-coumaric acid의 acylation이 가장 많았다. Cyanidin feruloyl glucoside가 '홍이슬'에서만 검출되어 품종 지표로 활용이 가능하였다. 결론적으로 anthocyanin의 조성을 anthocyanidin, 결합된 당의 개수, acylation 및 특정 anthocyanin을 기준으로 분류하여 품종의 특징이 확인되었다. 이러한 품종별 anthocyanin 특성을 이용하여 '개량머루'가 Vitis amurensis 혹은 그 교잡종의 가능성과 '홍이슬'에서 품종 구별의 기준으로 활용 가능성이 확인되었다.

마황(麻黃)과 그 위품의 내부형태감별 (The Anatomical Identification-key of Ephedrae Herba and its adulterant)

  • Objectives : Ephedra regeliana, although similar to the other E. sinica, E. intermedia, and E. equisetina, is not authorized by The Korean Pharmacopoeia and yet has also been imported under the name of 'Ma-Hwang'. The aim of this study was to perform a comparative anatomical evaluation of Ephedrae Sinicae Herba (ESH), Ephedrae Intermedicae Herba (EIH), Ephedrae Equisetinae Herba (EEH) and Ephedrae Regelianae Herba (ERH). Methods : Permanent paraffin-embedded specimens were produced using the paraffin-tertiary butyl alcohol (TBA) method, to visualize their anatomical features via light microscopy. Results : ESH and ERH transverse sections were in the shape of a long ellipse, while those of EIH and EEH were in the shape of a circle. These anatomical characteristics substantiated the results of external morphological examination. The sections revealed that each of the four samples showed significantly different cortexes. The cortex of ESH was wider than two thirds of the section, unlike that of the other samples which was relatively narrow. The xylems of ERH, EIH, and EEH demonstrated interconnectivity with other adjacent xylems, contrary to those in ESH. Therefore, the characteristic development of the xylems over time was used to distinguish ERH from the other. Furthermore, parenchymal morphology in the central cylinder was also a good criterion to differentiate between EIH and EEH, since EIH demonstrated parenchymal cells that were shaped like a rounded-triangle, whereas EEH parenchymal cells were chrysanthemum-shaped. Conclusions : The identification-keys established in this study would facilitate the accurate identification of microscopic features of the four varieties of 'Ma-Hwang'.
