Changes in the $Ca^{2+}-,\;Mg^{2+}-dependent$ Adenosine Triphosphatase Activity and Ultrastructure of Marine Fishes by Partial Freezing -I. Denaturation of Yellowtail Myofibrillar ATPase During Cold Storage- (해산어의 부분동결에 의한 $Ca^{2+}-,\;Mg^{2+}-dependent$ Adenosine Triphosphatase 활성 및 근섬유의 미세구조의 변화 -I. 저온저장에 의한 방어 근원섬유 단백질의 변성-)

  • Myofibrillar protein(myofibil) was prepared from Yellowtail fish (Seriola quinqueradiata), and then, it was stored at $0^{\circ}C$(ice-cooling), $-3.5^{\circ}C$(partial freezing) and $-20^{\circ}C$(freezing). Another myofibrils were prepared from the fish stored with ice-cooling, partial freezing and freezing for a week as the maximum. Denaturation of muscle protein during the storage was investigated by the measurement of $Ca^{2+}-$ and $Mg^{2+}-ATPase$ activity. Specific activity of $Ca^{2+}-\;and\;Mg^{2+}-ATPase$ associated with Yellowtail myofibrils was 0.155 and $0.149\;{\mu}\;mole$ Pi/min/mg of protein, respectively, before storgae. ATPase activity of myofibils did not show any significant difference between $0^{\circ}C$ and $-3.5^{\circ}C$ whereas it was decreased faster at $-20^{\circ}C$ than at $0^{\circ}C$ or $-3.5^{\circ}C$. ATPase activity of myofibirls extracted from the fish stored for a week was 1.2-1.8 times higher than myofibils stored with ice-cooling or partial freezing while it was 2.5-3 times higher than that with freezing. Apparent denaturation constant of $Ca^{2+}-ATPase$ of myofibrils was between 0.48-0.65, and it was 2-3 times higher than that of $Mg^{2+}-ATPase$. The constant of myofibrils extracted from the fish did not show significant difference between $Ca^{2+}-\;and\;Mg^{2+}-ATPase$.

  • Thermal energy storage(TES) cooling system using cheaper electricity of off-peak time has been applied to relief a significant portion of the peak demand of electricity during the daytime in summer. Slurry ice type thermal energy storage cooling system is one kind of more efficient ice-thermal energy storage cooling system than Ice-on-Coil type or Encapsulated type TES cooling system, even though, which are more popular TES system. This study is experimented to observe flow pattern and formation of slurry ice in reversing flow to improve efficiency of heat transfer between fluid and freezing tube.

  • Histological changes of the abalone muscle occurring in the process of freezing as well as thawing were investigated, and its results were compared to the histological structure of fresh muscle. The muscles of the abalone were mainly composed of smooth muscle fibres, and its morphological structures is similar to those of the top shell. Mechanical damgage of the muscle fibres caused by freezing were not observed while a number of small ice crystals were found between the muscle fibres. The damage by thawing was not much remakable compare to the destruction of muscle fibres of fish.

  • The earth structures and in-ground LNG tank, and buildings can be constructed with using artificial freezing method on the reclaimed land. In this study, upon freezing a saturated soil in a closed-system from the top, a considerable pressure was developed. The pressure is the result of the surface energy of a curved ice-water interface. The most significant of these parameters will have the greatest effect on the classification. In order to establish frost-susceptibility criteria based on frost heaving expansion pressure, more soils have to be tested. This study was initiated to investigate the soils frost heaving expansion pressure and moisture characteristics resulting from freezing and freezing-thawing cycle process. Weathered granite soils, sandy soil, sandy soil were used in the laboratory freezing test subjected to thermal gradients under closed- systems.

  • If the temperature of free water drops below the freezing point the water turns into ice and its volume increases. Analyzing of the results in test, it is hoped that these is broadly used in engineering plan. The compressive strength of frozen soils and the unconfined compressive strength of the ice. The creep strength depends on factors including creep ratio, time, strain and temperature. For a linear decreases in temperature, strength predicts an exponential increasing. The relationships between dry unit weight and compressive strength, water content and freezing with compacted soil samples, have been analyzed to understand the strength of frozen soil. Therefore, it is thankful that the results of analsis shall find useful application as a framework for generalizing experimental information as well as a basis for solving various frozen soil mechanics problems.

  • Two approaches for the fabrication of tailored powder composites with specially distributed pore-grain structure and chemical composition are investigated. Electrophoretic Deposition (EPD) followed by microwave sintering is employed to obtain functionally graded materials (FGM) by in-situ controlling the deposition bath suspension composition. $Al_2O_3/ZrO_2$ and zeolite FGM are successfully synthesized using this technique. In order to fabricate an aligned porous structure, unidirectional freezing followed by freeze drying and sintering is employed. By controlling the temperature gradient during freezing of powder slurry, a unidirectional ice-ceramic structure is obtained. The frozen specimen is then subjected to freeze drying to sublimate the ice. The obtained capillary-porous ceramic specimen is consolidated by sintering. The sintering of the graded structure is modeled by the continuum theory of sintering.

Isolation and Identification of Ice Nucleation Active Fusarium Strains from Rapid Apple Declined Trees in Korea

  • In biological particles such as Fusarium species, ice nucleation activity (INA) has been observed. Fusarium strains isolated from apple declined trees in Korea were identified with a multilocus sequence analysis using the tef1 and rpb1 genes. Droplet-freezing and tube-freezing assays were used to determine the INA of the strains, using Pseudomonas syringae pv. syringae KACC 21200 as a positive control and resulting in seven INA+ fungal strains that were identified as F. tricinctum (KNUF-21-F17, KNUF-21-F18, KNUF-21-F29, KNUF-21-F32, KNUF-21-F38, KNUF-21-F43, and KNUF-21-F44). The effect of Fusarium INA+ KNUF-21-F29 was compared to that of INA- strains on Chrysanthemum morifolium cv. Shinma explants. A higher callus formation and no-shoot formation were observed, suggesting that fungal INA could play a role in cold injuries and be a factor to consider in rapid apple decline. To the best of our knowledge, this is the first report of INA fungal strains isolated in Korea.

Proposal of a Black Ice Detection Method Using Vehicle Sensors to Reduce Traffic Accidents (교통사고 경감을 위한 차량 센서를 사용한 블랙아이스 탐지 방법 제안)

  • As the invention of automobiles and construction of roads for vehicles began, the occurrence of traffic accidents began to increase. Accordingly, efforts were made to prevent traffic accidents by changing the road construction method and using signal systems such as traffic lights, but until now, numerous human and property damages have occurred every year due to traffic accidents caused by freezing of the road due to bad weather. In this paper, we propose a method of transmitting ice detection data detected using vehicle sensor data to vehicle navigation to reduce traffic accidents caused by road freezing.

Freezing Behaviors of Frozen Foods Determined by $^1H$ NMR and DSC

  • The freezing patterns of commercial frozen foods were characterized by using proton nuclear magnetic resonance ($^1H$ NMR) relaxometry and differential scanning calorimetry (DSC). The liquid-like components like unfrozen water were investigated as a function of temperature (10 to $-40^{\circ}C$) and then compared with the unfrozen water content measured by DSC. The formation of ice crystals and the reduction of water in the foods during freezing were readily observed as a loss of the NMR signal intensity. The proton NMR relaxation measurement showed that the decreasing pattern of the liquid-like components varied depending on the samples even though they exhibited the same onset temperature of ice formation at around $0^{\circ}C$. When compared with the unfrozen water content obtained by the DSC, the NMR and DSC results could be closely correlated at the temperature above $-20^{\circ}C$. However, the distinct divergence in the values between 2 methods was observed with further decreasing temperatures probably due to the solid glass formation which was not detected by DSC.
