Novel variants of IDS gene, c.1224_1225insC, and recombinant variant of IDS gene, c.418+495_1006+1304del, in Two Families with Mucopolysaccharidosis type II

  • In this report, the phenotypes of three patients from two families with mucopolysaccharidosis type II (MPS II) are compared: a novel variant and recombinant variant of IDS gene. The results of urine in patients showed a pronounced increase in glycosaminoglycan excretion with decreased iduronate-2-sulfatase enzyme activity in leukocyte, leading to a diagnosis of MPS II. A patient has a novel variant with 1 bp small insertion, c.1224_1225insC in exon 9, which caused frameshifts with a premature stop codon, and two patients have a recombination variant, c.418+495_1006+1304del, leading to the loss of exons 4, 5, 6, and 7 in genomic DNA, which is relatively common in Korean patients. They had different phenotypes even in the same mutation. The patients have now been enzyme replacement therapy with a significant decrease in glycosaminoglycan excretion. Further study on residual enzyme activity, as well as experience with more cases, may shed light on the relationship between phenotypes in MPS II and gene mutations.

  • Hypertension is a complex disease with strong genetic influences. Essential hypertension has been shown to be associated with insulin resistance. To clarify the genetic basis of insulin resistance in Hypertension, case-control association studies were performed to examine candidate genes for insulin resistance in hypertension. Polymorphisms investigated were the BstO I polymorphism of the $\beta$3-adrenergic receptor (ADRB3) gene, the Xba I Polymorphism of the glycogen synthase (GSY) gene, the Dde I polymorphism of the protein phosphatase 1 G subuit (PP1G) gene, the BstE II polymorphism of the glucagon receptor (GCG-R) gene, the Pst 1 polymorphism of the insulin (INS) gene and the Acc I polymorphism of the glucokinase (GCK) gene. No significant differences were observed in the distribution of alleles and genotypes of the ADRB3, GSY PP1G, GCG-R, INS, and GCK genes between hypertensive and normotensive groups. Although the frequencies in each of these polymorphisms were not significantly different between essential hypertensive and normotensive individuals, our results may provide additional information for linkage analysis and associative studies of disorders in carbohydrate metabolism or in cardiovascular disease.

  • It is now generally accepted that a phosphoinositide cycle is involved in the transduction of a variety of signals in plant cells. In animal cells, the hydrolysis of phosphatidyl-4,5-bisphosphate catalysed by phosphatidylinositol - specific phospholipase C yields to D-myo-inositol - 1,4,5-trisphosphate and diacylglycerol, which are well known second messengers. The binding of InsP$_3$to a receptor located on the endoplasmic reticulum triggers a calcium release from the endoplasmic reticulum. We have detected and partially characterised key components of phosphoinositide signaling. First, tobacco microsomal fraction and plasma membrane PI-PLC. Consecutively, using a radioligand binding assay we have identified a $Ca^{2+}$ -dependent high affinity InsP$_3$binding site in microsomal membrane fraction vesicle preparation and then we have measured inositol-1,4,5-trisphosphate induced calcium release from tobacco microsomal fraction. These findings suggest that phosphoinositide signaling system is present and operates in the tobacco suspension culture.e.

Upregulation of Kruppel-like Factor 4 Gene expression by Allomyrina dichotoma Hemolymph in the INS-1 Pancreatic β-cells

  • The hemolymph of Korean rhinoceros Allomyrina dichotoma consists of blood and lymph in which various kinds of proteins function physiologically. We have previously demonstrated that A. dichotoma hemolymph has the potential to treatment and prevent diabetes through activating transcription factor 3-gene (ATF3) regulation. In this study, we investigate the expression of Kruppel-like factor 4 (KLF4) in A. dichotoma hemolymph-treated INS-1 pancreatic β-cells. The new findings show that A. dichotoma hemolymph, which upregulates KLF4 gene expression in a dose-dependent and time-dependent manner. In addition, hemolymph combine with mild endoplasmic reticulum (ER) stress, which also differentially regulates KLF4 gene expression. These results may provide insights to KLF4 gene-related disease therapies through KLF4 gene regulation.

  • This paper describes the development of a navigation HILS (hardware in the loop simulation) system for an integrated navigation performance analysis of a large diameter unmanned underwater vehicle (LDUUV). The HILS system was used for the performance analysis of the LDUUV. When a conventional HILS system is used, it is not possible to calculate the velocity and position using an inertial navigation system (INS). To cope with this problem, an external acceleration was generated. To evaluate the proposed method, we compare the results of a Monte Carlo simulation and navigation HILS experiment.

Four Dammarane Triterpenes and Their Inhibitory Properties Against Eight Receptor Tyrosine Kinases

  • In recent years, tyrosine kinases (TKs) have been the target to combat cancers, and most of the developed inhibitors are of synthetic origin. Natural compounds that have the properties as the TK's inhibitors are very limited. This paper described the isolation of a new dammarane triterpene from the tree bark of Sandoricum koetjape, along with three known related dammaranes from the damar resin of Shorea javanica, as well as their inhibitory properties against eight receptor TKs (RTKs: EGFR, HER2, HER4, IGF1R, InsR, KDR, PDGFRα, and PDGFRβ). Based on the NMR and mass spectral data the new compound was identified as (12β,20S)-12,20-dihydroxy-3,4-seco-dammaran-4,24-dien-3-oic acid (12β-hydroxydammarenolic acid) (1), while the three known compounds were identified as (20S)-20-hydroxy-3,4-seco-dammaran-4,24-dien-3-oic acid (dammarenolic acid) (2), (3β,20S)-3,20-dihydroxydammaran-24-ene (3), and (20S)-3-oxo-20-hydroxydammaran-24-ene (4). The tyrosine kinase assay of the four compounds resulted only 1 and 2 at concentration of 10 μM that had weak activity against EGFR and InsR, with their % inhibitory were 30%, 27% (1), 45%, and 32% (2), respectively. The results suggested that the presence of a linear carboxylic acid group in both compounds could be of significance to the inhibitory properties against the two RTKs.

Protopanaxadiol ameliorates palmitate-induced lipotoxicity and pancreatic β-cell dysfunction in INS-1 cells

  • Background: Free fatty acid-induced lipotoxicity is considered to play an important role in pancreatic β-cell dysfunction. The effect of ginsenosides on palmitic acid-induced pancreatic beta-cells cell death and failure of glucose-stimulated secretion of insulin (GSIS) was evaluated in this study. Methods: Enzyme-linked immunosorbent assay kit for a rat insulin was used to quantify glucose-stimulated insulin secretion. Protein expression was examined by western blotting analysis. Nuclear condensation was measured by staining with Hoechst 33342 stain. Apoptotic cell death was assessed by staining with Annexin V. Oil Red O staining was used to measure lipid accumulation. Results: We screened ginsenosides to prevent palmitic acid-induced cell death and impairment of GSIS in INS-1 pancreatic β-cells and identified protopanaxadiol (PPD) as a potential therapeutic agent. The protection effect of PPD was likely due to a reduction in apoptosis and lipid accumulation. PPD attenuated the palmitic acid-induced increase in the levels of B-cell lymphoma-2-associated X/B-cell lymphoma 2, poly (ADP-ribose) polymerase and cleaved caspase-3. Moreover, PPD prevented palmitic acid-induced impairment of insulin secretion, which was accompanied by an increase in the activation of phosphatidylinositol 3-kinase, peroxisome proliferator-activated receptor γ, insulin receptor substrate-2, serine-threonine kinase, and pancreatic and duodenal homeobox-1. Conclusion: Our results suggest that the protective effect of PPD on lipotoxicity and lipid accumulation induced by palmitic acid in pancreatic β-cells.

  • The geo-referencing accuracy of the images acquired by a UAV based multi-sensor system is affected by the accuracy of the mounting parameters involving the relationship between a camera and a GPS/INS system as well as the performance of a GPS/INS system. Therefore, the estimation of the accurate mounting parameters of a multi-sensor system is important. Currently, we are developing a low altitude multi-sensor system based on a UAV, which can monitor target areas in real time for rapid responses for emergency situations such as natural disasters and accidents. In this study, we suggest a system calibration method for the estimation of the mounting parameters of a multi-sensor system like our system. We also generate simulation data with the sensor specifications of our system, and derive an effective flight configuration and the number of ground control points for accurate and efficient system calibration by applying the proposed method to the simulated data. The experimental results indicate that the proposed method can estimate accurate mounting parameters using over five ground control points and flight configuration composed of six strips. In the near future, we plan to estimate mounting parameters of our system using the proposed method and evaluate the geo-referencing accuracy of the acquired sensory data.

Growth of Green Pepper(Capsicum annuum L.) in a Plastic Greenhouse Covered with Anti-dropping Plasma Film (방적성 Plasma 처리 필름으로 피복된 플라스틱온실의 풋고추 생육)

  • The Plasma film treated with a high electric voltage was developed to enhance flow down of condensation drops on inside plastic film. Arch type greenhouse framed with iron pipe of 25mm diameter defand 1.5mm thickness were covered with either the developed plasma film or surfactant film(control). Green pepper seedlings raised for 40 days in plug trays were transplanted at a density of 110cm by 30cm in each greenhouse. The mount of condensational water on film surface, generated by 7$0^{\circ}C$ water bath chimney systems and flew down, was collected and measured. The amount of collected water after 150 minutes was 2.56 mL.100c $m^{-2}$ and 0.94mL.100c $m^{-2}$ , respectively, in the plasma film and surfactant film-covered greenhouses. The amount of condensational water drops attached on the cover at 08:20 a.m. at 60 days filter covering was 0.34mL.100c $m^{02}$ and 0.32mL.100c $m^{-2}$ , respectively, in the plasma film- and surfactant film-covered greenhouses. Solar irradiance transmitted into greenhouse was 2.0% higher in the greenhouse covered with the plasma film tan that in the greenhouse covered with the surfactant film. Air temperature in the plasma film-covered greenhouse was higher than the surfactant film-covered greenhouse by 0.5$^{\circ}C$. However, there was no difference in relative humidity between the two greenhouse. Plant height, leaf area, dry weight and early yield showed no significant differences.s.
