수중운동이 스테로이드를 투여한 흰쥐의 골격근 무게 및 단백질 함량에 미치는 효과 (The Effect of Aquatic-Exercise on Skeletal Muscle Weight and Myofibrillar Protein Content of Rats with Steroids Injection)

  • The purpose of this study was to know the effect of aquatic-exercise on muscle atrophy which induced by steroid injection. The forty-eight Sparague-Dawley adult male rats were assigned to the 4 groups; Group I (distilled water injection), Group II (steroid injection), Group III (distilled water injection with aquatic exercise), Group IV (steroid injection with aquatic exercise). We observed their body weight, muscle relative weight, myofibrillar protein content. The results of this study were as follows; 1. Body weight was decreased rapidly on steroid injection groups in comparison with distilled water injection groups after 2 weeks (p<0.01), but that was almost recovered as before test on steroid injection group with aquatic exercise. 2. The relative weight of gastrocnemius muscles was decreased on steroid injection groups in comparison with distilled water injection groups. however, Decrease of it on steroid injection group with aquatic exercise for steroid injection group was fallen. There was very significant difference after 4 weeks (p<0.01). 3. Myofibrillar protein content of gastrocnemius muscles was decreased on steroid injection groups in comparison with distilled water injection groups. however, Decrease of it on steroid injection group with aquatic exercise for steroid injection group was fallen. There was significant difference after 4 weeks (p<0.05).

Activator 사용전후 부정교합 유형에 따른 각 근육 활성도의 변화에 관한 연구 (AN ELECTROMYOGRAPHIC STUDY ON THE CHANGES IN MUSCLE ACTIVITY IN DIFFERENT MALOCCLUSION TYPES FOLLOWING ACTIVATOR THERAPY)

  • 본 연구는 II급 및 III급 부정교합자를 대상으로 activator사용에 따른 각 근육의 적응 및 변화양상을 알아보고자 시행되었다. 본 연구의 대상은 성장 발육기에 있는 아동중 II급 부정교합자 15명과 III급 부정교합자 17명으로 기능적 요인을 갖고있는 자로 선정하였고, 연구방법은 전측두근(T.A.), 교근(M.M.), 후측두근(T.P.), 악이복근 전복(D.A.)의 활성도를 알아보고자 $Biopak^{TM}$ program 과 쌍극표면전극을 이용하여 activator 치료 시작시기와 사용후 6-12개월 사이의 두차례에 걸쳐 안정위, 연하시, clenching등 세가지 조건에서 근활성도를 측정하여 비교해본 결과 다음과 같은 결론을 얻었다. II급 및 III급 부정교합자와 정상교합자의 근활성도를 비교한 결과 안정위시와 연하시 유의한 차이를 보이지 않았으나 clenching시 교근과 후측두근에서는 차이를 나타내었다. 치료전 근활성도가 정상보다 작은 군은 유의성 있는 증가를, 큰 군은 유의성 있는 감소를 보이는 등 대부분의 근육이 일정한 경향을 나타내었고, 이러한 경향은 III급 부정교합군에서 전측두근(연하시), 후측두근(clenching시), 악이복근 전복(연하시, clenching시)을 제외하고 모두에서 보였으며 II급 부정교합군에서는 전측두근(연하시), 교근(안정위, 연하시), 후측두근(clenching시)에서만 관찰되었다. 그러나 이러한 경향은 clenching시에서 감소되었다. II급 부정교합군과 III급 부정교합군과의 근활성도 비교에서 안정위시는 III급 부정교합군의 근활성도 증가율이 II급 부정교합군보다 우세했고, 연하시에는 교근을 제외하고 II급 부정교합군이, clenching시에는 역시 II급 부정교합군이 악이복근 전복을 제외하고 우세하게 나타났다.

저강도 격일 운동과 매일 운동이 쥐 뒷다리근에 미치는 근 비대 효과 비교 (Comparison of Hypertrophic Effects of Low-Intensity Exercise on Rat Hindlimb Muscles between Every Other Day Exercise and Everyday Exercise)

  • Purpose: The purpose of this study was to compare the hypertrophic effects of low-intensity exercise on weight, myofibrillar protein content and Type I, II fiber cross-sectional area of hindlimb muscles of rats between every other day exercise and every day exercise. Methods: Adult male Sprague-Dawley rats were assigned to 1 of 3 groups: control group (C, n=6), experimental group 1 (E1, n=7) and experimental group 2 (E2, n=7). Rats in E1 group had 7 sessions (every other day) and those in E2 group had 14 sessions (every day) of exercise in which they ran on a treadmill for 30 min/day at 10 m/min. Results: Muscle weight, cross-sectional area of type I fiber and myofibrillar protein content of soleus and myofibrillar protein content of plantaris in E1 group, and myofibrillar protein content of soleus and cross-sectional area of type I fiber of plantaris in E2 group were greater than those in C group. Cross-sectional area of type I fiber of soleus of E1 group was higher than E2 group while cross-sectional area of type I fiber of plantaris of E2 group was higher than E1 group. Conclusion: Hypertrophy of hindlimb muscles occurs from every other day exercise similar to every day exercise.

흰쥐의 만성 알콜성 근위축에 시호소간산(柴胡疎肝散)이 미치는 효과 (The Effects of Shihosogan-san on Alcohol-induced Muscle Atrophy in Rats)

  • Chronic or acute alcohol abuse often leads to liver injury associated with alcoholic hepatitis, liver fibrosis, cirrhosis, and liver cancer. In addition to the liver, alcohol abuse also induces a variety of other tissue injuries including pancreatitis, cardiomyopathy, neurotoxicity and muscle loss. Chronic skeletal muscle myopathy, independent of peripheral neuropathy, is well recognised in alcoholic patients. Several mechanisms may be involved in the pathogenesis of alcoholic myopathy. Ethanol is a potent inhibitor of muscle protein synthesis. Gastrocnemius and plantaris muscles are Type II fiber-predominant and usually considered representative of the musculature as a whole. Whereas, soleus muscle is Type I fiber predominant. Shihosogan-san is a traditional Korean medicine that is widely employed to treat indigestion and liver diseases. Muscle diseases are often related to liver diseases and conditions. We therefore tested the hypothesis that treatment with Shihosogan-san could ameliorate the ethanol-induced changes in muscle protein synthesis. Young male Sprague-Dawley rats were orally given 25% ethanol (5ml/kg, body weight) daily with Ethanol for 28 days. Normal group was similarly administrated with saline. In Shihosogan-san treated group, rats were orally administrated Shihosogan-san extract, and rats of EtOH group were given with the vehicle only. After 4 week, the morphology of gastrocnemius and plantaris muscles were assessed by hematoxylin and eosin staining. For comparative purposes, liver function was also investigated. The muscles from rats of EtOH group displayed a significant reduction in average cross section area compared to Normal group. Shihosogan-san treated group had increased fiber compared to the EtOH group. Moreover, Shihosogan-san treated group compared with EtOH group showed significantly decreased pro-apoptotic BAX expression and increased anti-apoptotic Bcl-2 expression. In conclusion, Shihosogan-san extract showed ameliorating effects on chronic alcohol toxicity in skeletal muscle.

대금음자(對金飮子)가 흰쥐의 만성 알콜성 근위축에 미치는 영향 (The Effects of Daekumeumja on Alcohol-induced Muscle Atrophy in Rats)

  • Chronic alcoholic myopathy is one of the most common skeletal muscle disorders. It is characterized by a reduction in the entire skeletal musculature, skeletal muscle weakness, and difficulties in gait. Patients with alcoholic hepatitis and cirrhosis have severe muscle loss that contributes to worsening outcome. Although the myopathy selectively affects Type II (fast twitch, glycolytic, anaerobic) skeletal muscle fibers, total skeletal musculature is reduced. The severity of the muscle atrophy is proportional to the duration and amount of alcohol consumed and leads to decreased muscle strength. The mechanisms for the myopathy are generally unknown but it is not due to overt nutritional deficiency, nor due to either neuropathy or severe liver disease. Skeletal muscle mass and protein content are maintained by a balance between protein synthesis and breakdown and in vivo animal models studies have shown that ethanol inhibits skeletal muscle protein synthesis. Daekumeumja is a traditional Korean medicine that is widely employed to treat various alcohol-induced diseases. Muscle diseases are often related to liver diseases and conditions. The main objective of this study was to assess that Daekumeumja extract could have protective effect against alcoholic myopathy in a Sprague-Dawley rat model. Rats were orally given 25% ethanol (5ml/kg, body weight) for 8 weeks. After 30 minutes, rats were administrated with Daekumeumja extract. Controls were similarly administrated with the vehicle alone. The weights of gastrocnemius, soleus and plantaris muscles were assessed and the morphologic changes of gastrocnemius and plantaris muscles were also assessed by hematoxylin and eosin staining. In results, The muscles from ethanol treated rats displayed a significant reduction in muscle weight and average cross section area compared to Normal group. Daekumeumja extract treated group showed increased muscle weight and muscle fiber compared to the ethanol treated group. It was concluded that Daekumeumja extract showed ameliorating effects on chronic alcohol myopathy in skeletal muscle.

한방사료 첨가제인 어보산의 효과 II. 어보산 첨가사료로 사육한 넙치의 육질에 대한 연구 (Utilization of Obosan (Dietary Herbs) II. Muscle Quality of Olive Flounder, Paralichthys olivaceus Fed with Diet Containing Obosan)

  • 양식어류의 육질을 개선시키기 위한 연구의 환으로, 한방사료 첨가제인 어보산(성암산업, 대한민국)으로 사육한 넙치(Paralichthys olivaceus)에 대한 관능 검사, 물성 및 정미성분 측정 등을 하여 육질을 평가하였다. 기호 검사 결과, 어보산 첨가군은 대조군에 비해 모든 항목에서 선호되었으며, 특히 어육의 질감 (P<0.05), 맛(P<0.01) 및 종합적인 선호도(P<0.01)에서 유의한 차이를 나타내었다. 식별 검사에서도 어보산 첨가군의 육질은 대조군에 비해 광택(P<0.01), 경도 (P<0.05), 탄력성(P<0.01) 및 촉촉한 정도 (P<0.05)가 뛰어나고 결 (P<0.05)이 고우며 맛난 맛(P<0.01) 이 강한 것으로 평가되었다. 어보산 첨가사료로 사육한 넙치의 근육 물성은 대조군에 비해 경도가 20.5% (P<0.01) , 탄력성이 9.3%(P<0.01) 그리고 응집성이 16.7% (P<0.05) 유의하게 높은 값을 보였다. 대조군 사료와 어보산 첨가사료로 사육한 넙치의 정미 성분을 비교 분석한 결과, 정미 물질로서 어육 추출물 중에 함유되어 있는 유리 아미노산 총량은 첨가군이 대조군에 비해 약 41.6% 더 많은 결과를 보였으며, 특히 유리 아미노산 중 어육의 맛난 맛과 관련있는glutamic acid, proline, glycine, alanine 및methionine 등의 총량은 첨가군이 대조군에 비해 약 38.6% 더 많게 나타났다. 핵산 관련물질 중 ATP의 양은 첨가군이 대조군에 비해 약 87.8%.더 많게 나타났음에도 불구하고 IMP의 양은 첨가군이 대조군에 비해 약 32.6%.더 적은 값을 보였다. 따라서 어보산은 넙치의 육질 개선에 효과적임을 알 수 있었다.

Peripheral metabotropic glutamate receptors differentially modulate mustard oil-induced craniofacial muscle pain in lightly anesthetized rats

  • The present study investigated the role of peripheral group I, II, and III metabotropic glutamate receptors (mGluRs) in mustard oil (MO)-induced nociceptive response in the masseter muscles of lightly anesthetized rats. Experiments were carried out on male Sprague-Dawley rats weighing 300-350 gm. After initial anesthesia with sodium pentobarbital (40 mg/kg, i.p.), one femoral vein was cannulated and connected to an infusion pump for intravenous infusion of sodium pentobarbital. The rate of infusion was adjusted to provide a constant level of anesthesia. MO (30 ${\mu}L$) was injected into the mid-region of the left masseter muscle via a 30-gauge needle over 10 seconds. After 30 mL injection of 5, 10, 15, or 20% MO into the masseter muscle, total number of hindpaw-shaking behavior was monitored. Intramuscular administration of MO significantly produced hindpawshaking behavior in a dose-dependent manner, as compared with the vehicle (mineral oil)-treated group. Intramuscular pretreatment with 10 or 100 ng DHPG, a group I mGluRs agonist, enhanced MO-induced hindpaw-shaking behavior, while APDC (20 or 200 ${\mu}g$), a group II mGluRs agonist, or L-AP4 (2 ${\mu}g$), a group III mGluRs agonist, significantly reduced MO-induced nociceptive behavior. The antinociception, produced by group II or III mGluRs agonists, was abolished by pretreatment with LY341495, a group II mGluRs antagonist, or CPPG, a group III mGluRs antagonist, res-pectively. Based on these observations, peripheral mGluRs differentially modulated MO-induced nociceptive behavior response in the craniofacial muscle pain and peripheral group II and III mGluRs agonists could be used in treatment of craniofacial muscle nociception.

Supplementation of guanidinoacetic acid and rumen-protected methionine increased growth performance and meat quality of Tan lambs

  • Objective: Tan lambs (n = 36, 3 mo old, 19.1±0.53 kg) were used to assess effects of dietary guanidinoacetic acid (GAA) and rumen-protected methionine (RPM) on growth performance, carcass traits, meat quality, and serum parameters. Methods: Lambs were randomly assigned to three treatment groups, with 6 pens per group and 2 lambs per pen. Dietary treatments were: basal diet alone (I); basal diet supplemented with 0.08% GAA+0.06% RPM (II); and basal diet supplemented with 0.08% GAA+0.08% RPM (III). Diets were provided three times a day for 90 d. Intake per pen was recorded daily and individual lamb body weight (BW) was measured monthly. Carcass traits were measured after slaughter and meat quality at the end of the experiment, blood samples were taken on a subgroup of lambs for analysis of indicators mostly related to protein metabolism. Results: Final BW and average daily gain for the first and second month, and for the entire experiment were greater in Treatment II compared to Treatment I (p<0.05), whereas feed to gain ratio was lower (p<0.05). Treatment II had the optimal dressing percentage and net meat weight proportion, as well as crude protein and intramuscular fat concentrations in muscles. Treatment II improved meat quality, as indicated by the greater water holding capacity, pH after 45 min and 48 h, and lower shear force and cooking loss. Dietary supplementation of GAA and RPM also increased the meat color a* and b* values at 24 h. Finally, Treatment II increased total protein, and serum concentrations of albumin and creatinine, but decreased serum urea nitrogen concentrations, indicating improved protein efficiency. Conclusion: In this study, 0.08% GAA+0.06% RPM supplementation improved growth performance and meat quality of Tan lambs.

S.O.T CATEGORY II BLOCKING이 복근지구력과 어깨 통증에 미치는 영향 (THE EFFECT OF SACROOCCIPITAL TECHNIQUE CATEGORY II BLOCKING ON ABDOMINAL MUSCLE ENDURANCE AND SHOULDER PAIN)

  • 본 연구는 카이로프랙틱 기법 중 누운 자세에서 시술되어지는 S.O.T category II 골반 블록 중재가 골반의 불안정으로 인한 기울어짐을 회복시킴으로써 이에 따른 어깨통증을 줄이고 복근의 지구력을 향상시킬 수 있는지 알아보고자 한 것이다. 이 실험을 위해 엉치엉덩관절의 불안정을 가지고 있으며 허리의 통증이 있는 총 50명의 참여자가 실험군과 대조군에 각 25명씩 무작위 할당되었다. 실험군은 category II 블록 중재를 받았고, 대조군은 거짓블록 중재를 받았으며, 이들 두 집단을 통해 중재 전, 중재 후, 그리고 중재 2주 후에 복근지구력과 어깨통증을 측정하여 그 결과를 반복측정 이원분산분석법으로 분석하였다. 분석결과 S.O.T category II 골반 블록 중재 후 실험집단에서 복근지구력이 유의한 증가를 보였으며 중재가 끝난 2주 후에도 유지 되었다. 또한 어깨통증의 경우에도 실험집단에서 통증의 감소를 보였으며 역시 중재가 끝난 2주 후에도 유의한 감소를 보였다. 이상의 연구 결과를 종합해 볼 때 S.O.T category II 골반 블록 중재는 골반의 안정성을 유도하여 복근지구력을 증진시키고, 어깨통증을 감소시키며 중재 후에도 지속효과가 있는 것으로 나타남으로, 골반의 불안정성이 원인이 되는 어깨통증이나 복근지구력을 회복시킬 수 있는 중재법으로 활용되어질 수 있을 것으로 사료된다.

스테로이드 치료 후 회복기의 규칙적인 운동이 쥐의 스테로이드 유발성 위축 뒷다리근에 미치는 영향 (Effect of regular exercise during recovery period following steroid treatment on the atrophied hindlimb muscles induced by steroid in rats)

  • This study was conducted to determine whether low intensity regular exercise following steroid treatment could attenuate steroid-induced muscle atrophy. Thirty-eight Sprague-Dawley rats weighing $165{\sim}175g$ were divided into six groups ; control group(C), dexamethasone administration group(D), sedentary normal saline administration group(C+Se), exercise after normal saline administration group(C+Ex), sedentary group after dexamethasone administration(D+Se), exercise group after dexamethasone administration(D+Ex). Either dexamethasone(5mg/kg) or normal saline was injected for 7days accordingly. Exercise was started at 10m/min on the $10^{\circ}$ grade treadmill and gradually increased up to 15m/min by the 7th day for 60minutes/day($20min{\times}3$). The data were analyzed by Kruskal-Wallis test and Mann-Whitney U test using the SPSS WIN 9.0 program. Body weight, muscle weight and myofibrillar protein content of both plantaris and gastrocnemius, Type I, II muscle fiber cross-sectional area of plantaris, and Type II muscle fiber cross-sectional area of gastrocnemius in D group were significantly lower than those of C group(p<0.05) respectively. Hindlimb muscle weight, myofibrillar protein content of both plantaris and gastrocnemius. Type I muscle fiber cross-sectional area of soleus and Type I, II muscle fiber cross-sectional area of plantaris in D+Ex group tended to increase compared to those of D+Se group. Myofibrillar protein content of both plantaris and gastrocnemius, Type I muscle fiber cross-sectional area of plantaris in D+Ex group tended to increase compared to those of C+Se group. Based on these results, it is suggested that regular low-intensity exercise during recovery period after steroid treatment might facilitate the recovery from steroid-induced muscle atrophy.
