Molecular analysis of chicken interferon-alpha inducible protein 6 gene and transcriptional regulation

  • Interferon-alpha inducible protein 6 (IFI6) is an interferon-stimulated gene (ISG), belonging to the FAM14 family of proteins and is localized in the mitochondrial membrane, where it plays a role in apoptosis. Transcriptional regulation of this gene is poorly understood in the context of inflammation by intracellular nucleic acid-sensing receptors and pathological conditions caused by viral infection. In this study, chicken IFI6 (chIFI6) was identified and studied for its molecular features and transcriptional regulation in chicken cells and tissues, i.e., lungs, spleens, and tracheas from highly pathogenic avian influenza virus (HPAIV)-infected chickens. The chIFI6-coding sequences contained 1638 nucleotides encoding 107 amino acids in three exons, whereas the duck IFI6-coding sequences contained 495 nucleotides encoding 107 amino acids. IFI6 proteins from chickens, ducks, and quail contain an IF6/IF27-like superfamily domain. Expression of chIFI6 was higher in HPAIV-infected White Leghorn chicken lungs, spleens, and tracheas than in mock-infected controls. TLR3 signals regulate the transcription of chIFI6 in chicken DF-1 cells via the NF-κB and JNK signaling pathways, indicating that multiple signaling pathways differentially contribute to the transcription of chIFI6. Further research is needed to unravel the molecular mechanisms underlying IFI6 transcription, as well as the involvement of chIFI6 in the pathogenesis of HPAIV in chickens.

IL-17 Imbalance Promotes the Pyroptosis in Immune-Mediated Liver Injury Through STAT3-IFI16 Axis

  • Autoimmune hepatitis (AIH) affects all age group and occurs mainly in women. Pyroptosis is a novel programmed cell death featured with cell bursting and release of proinflammatory cytokines. A deeper understanding of AIH pathogenesis will contribute to novel therapy for AIH patients. Here, we aimed to investigate the role of IL-17 in immune-mediated liver injury. The levels of cytokines were measured by ELISA, and mRNA levels of STAT3 and IFN gamma-inducible protein 16 (IFI16) were detected by PCR. Expressions of STAT3, IFI16, gasdermin D and cleaved caspase-1 were measured by western-blotting. Immunohistochemical staining and transmission electron microscopy were applied to evaluate liver histopathological changes of the treated mice. Our results showed that the levels of IFI16 was increased in hepatocytes treated with IL-17 protein, and further elevated after STAT3-overexpressed (STAT3-OE) lentivirus treatment. The levels of IFI16 were reduced in hepatocytes treated with IL-17 neutralizing Ab (nAb), but were significantly increased after STAT3-OE treatment. Pyroptosis was observed in hepatocytes treated with IL-17 protein, and further cell damage was observed after STAT3-OE lentivirus treatment. Liver damage was alleviated in mice treated with IL-17 nAb, however sever damage was experienced after STAT3-OE lentivirus treatment. A binding interaction between IFI16 and STAT3 was detected in IL-17 treated hepatocytes. Glutathione transaminase activity was enhanced in concanavalin A-induced AIH mice compared to the control group (p<0.01). IL-17 plays an important role in activating STAT3 and up-regulating IFI16, which may promote the pyroptosis in AIH-related liver injury through STAT3-IFI16 axis.

Association between Interferon-Inducible Protein 6 (IFI6) Polymorphisms and Hepatitis B Virus Clearance

  • CD8+T cells are key factors mediating hepatitis B virus (HBV) clearance. However, these cells are killed through HBV-induced apoptosis during the antigen-presenting period in HBV-induced chronic liver disease (CLD) patients. Interferon-inducible protein 6 (IFI6) delays type I interferon-induced apoptosis in cells. We hypothesized that single nucleotide polymorphisms (SNPs) in the IFI6 could affect the chronicity of CLD. The present study included a discovery stage, in which 195 CLD patients, including chronic hepatitis B (HEP) and cirrhosis patients and 107 spontaneous recovery (SR) controls, were analyzed. The genotype distributions of rs2808426 (C > T) and rs10902662 (C > T) were significantly different between the SR and HEP groups (odds ratio [OR], 6.60; 95% confidence interval [CI], 1.64 to 26.52, p = 0.008 for both SNPs) and between the SR and CLD groups (OR, 4.38; 95% CI, 1.25 to 15.26; p = 0.021 and OR, 4.12; 95% CI, 1.18 to 14.44; p = 0.027, respectively). The distribution of diplotypes that contained these SNPs was significantly different between the SR and HEP groups (OR, 6.58; 95% CI, 1.63 to 25.59; p = 0.008 and OR, 0.15; 95% CI, 0.04 to 0.61; p = 0.008, respectively) and between the SR and CLD groups (OR, 4.38; 95% CI, 1.25 to 15.26; p = 0.021 and OR, 4.12; 95% CI, 1.18 to 14.44; p = 0.027, respectively). We were unable to replicate the association shown by secondary enrolled samples. A large-scale validation study should be performed to confirm the association between IFI6 and HBV clearance.

Immune Reconstitution of CD4+T Cells after Allogeneic Hematopoietic Stem Cell Transplantation and its Correlation with Invasive Fungal Infection in Patients with Hematological Malignancies

  • Objective: To explore the immune reconstitution of $CD4^+T$ cells after allogeneic hematopoietic stem cell transplantation (Allo-HSCT) and its relationship with invasive fungal infection (IFI) in patients with hematological malignancies. Materials and Methods: Forty-seven patients with hematological malignancies undergoing Allo-HSCT in Binzhou Medical University Hospital from February, 2010 to October, 2014 were selected. At 1, 2 and 3 months after transplantation, the immune subpopulations and concentration of cytokines were assessed respectively using flow cytometry (FCM) and enzyme linked immunosorbent assay (ELISA). The incidence of IFI after transplantation and its correlation with immune reconstitution of $CD4^+T$ cells were investigated. Results: The number of $CD4^+T$ cells and immune subpopulations increased progressively after transplantation as time went on, but the subpopulation cell count 3 months after transplantation was still significantly lower than in the control group (p<0.01). In comparison to the control group, the levels of interleukin-6 (IL-6) and IL-10 after transplantation rose evidently (p<0.01), while that of transforming growth factor-${beta}$ (TGF-${beta}$) was decreased (p<0.01). There was no statistically significant difference level of interferon-${\gamma}$ (IFN-${\gamma}$) (p>0.05). The incidence of IFI was 19.2% (9/47), and multivariate logistic regression revealed that IFI might be related to Th17 cell count (p<0.05), instead of Th1, Th2 and Treg cell counts as well as IL-6, IL-10, TGF-${beta}$ and IFN-${\gamma}$ levels (p>0.05). Conclusions: After Allo-HSCT, the immune reconstitution of $CD4^+T$ cells is delayed and Th17 cell count decreases obviously, which may be related to occurrence of IFI.

Induction of antiviral mechanisms by interferon-related genes in rock bream (Oplegnathus fasciatus) infected with rock bream iridovirus (RBIV)

  • We evaluated the transcriptional response of interferon (IFN)-related genes in rock bream iridovirus (RBIV)-infected rock bream under high-, low-, or no-mortality conditions induced by different stocking water temperatures. Under the high susceptibility condition (group A, water temperature 26℃, 100% mortality), only the Mx gene was expressed early, with prolonged expression, and with heavy viral loads of approximately 106~107 major capsid protein gene copies/μL from 4 to 10 days post infection (dpi). However, IRF1, IRF3, IRF8, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56 were activated at later time points (8 dpi) and then quickly decreased (10 dpi). For the low susceptibility condition, the water temperature was set at 23℃ for 7 days (group B) and then reduced to 17℃. Group B exhibited a 28% mortality rate, in which persistent and effective antiviral responses were observed for long periods of time. In particular, at 20 and 22 dpi, when virus replication was peaked at approximately 107/μL, the expressions of most of the IFN-related genes (IRF1, IRF3, IRF8, Mx, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56) were significantly higher in group B than in the control group. Moreover, prolonged and higher levels of IRF3 (at least 30 dpi), IRF8 (at least 30 dpi), ISG15 (at least 30 dpi), PKR (at least 28 dpi), Viperin (at least 30 dpi), and IFI44 (at least 30 dpi) were also observed in the recovery stage of infection. Under the no-susceptibility condition at 17℃ (0% mortality), significantly elevated levels of IRF3, Mx, ISG15, and PKR were observed mostly until 20 dpi. The findings indicate that RBIV infection can induce an efficient IFN-mediated antiviral immune response in low- and no-susceptibility conditions. The findings could be valuable for effective control of viral pathogens in fish.

Echinacea 추출물이 단구와 단구유래 수지상세포의 유전자발현에 미치는 효과 (The Effects of Echinacea Extract on the Gene Expression of Monocytes and Monocyte-derived Dendritic Cells)

  • 목 적 : Echinacea는 면역증강제로 이미 사용되고 있는 재래 식물로서 최근에 Echinacea의 추출물로 단구를 중심으로 면역세포들에 의한 면역증강효과에 대한 연구가 이루어지고 있다. 본 연구는 단구와 수지상세포에서 Echinacea에 의해 유전자의 발현이 증가되는 면역관련 유전자들을 cDNA microarray chip을 사용하여 선별하고 이들을 토대로 Echinacea의 면역증강 효과에 대한 연구를 할 때 기초 자료가 되고자 하였다. 방 법 : 실험 1과 2는 3명의 공여자의 말초혈 단구로 실험하였는데 실험 1은 단구에 최종 농도가 $50{\mu}g/mL$ 되게 Echinacea를 첨가하여 1일간 배양하였고, 실험 2는 실험 1의 대조군으로서 Echinacea를 첨가하지 않고 배양하였다. 실험 3과 4는 2명의 공여자의 단구로 실험하였는데 실험 3은 GM-CSF와 IL-4를 첨가하여 5일간 배양시켜 수지상세포로 분화시킨 뒤 Echinacea를 첨가하여 1일간 더 배양시켰고, 실험 4는 실험 3의 대조군으로서 수지상세포로 분화시킨 뒤 Echinacea를 첨가하지 않고 1 일간 더 배양하였다. Echinacea에 의한 단구와 수지상세포의 유전자발현 효과를 알아보기 위해서 cDNA microarray chip을 이용하여 대조군에 대한 실험군의 각 유전자의 발현비를 구하였다. Echinacea를 첨가하지 않은 단구(실험 2의 단구)에 대한 Echinacea를 첨가한 단구(실험 1의 단구)의 각 유전자들의 발현 비를 구하였고, Echinacea를 첨가하지 않은 수지상세포(실험 4의 수지상세포)에 대한 Echinacea를 첨가한 수지상세포(실험 3의 수지상세포)의 각 유전자들의 발현비를 구하였다. 여기서 실험 1과 2에서는 세 공여자의 단구에서 나온 유전자 발현비의 결과를, 실험 3과 4에서는 두 공여자의 수지상세포에서 나온 유전자 발현비의 결과를 평균하여 그 발현비가 2.5 이상 되는 것을 의미있게 발현된 유전자로 보았다. 결 과 : Echinacea를 첨가하지 않은 단구를 대조군으로 하여 Echinacea를 첨가한 단구의 유전자 발현비가 2.5 이상으로 증가한 것들 중 면역과 관계된 유전자들은 17개였다. Echinacea를 첨가하지 않은 수지상세포를 대조군으로 하여 Echinacea를 첨가한 수지상세포의 유전자 발현비가 2.5 이상으로 증가한 것들 중 면역과 관계된 유전자들은 24개였고, 실험에 사용한 수지상세포들은 모두 미성숙 수지상세포의 특징적인 표면항원들을 가지고 있음을 유세포 분석으로 확인하였다. Echinacea가 단구와 수지상세포 둘 다에서 의미있게 유전자발현비가 증가된 것들이 7개 있었는데, 이들은 CD44, IFI 30, MRC 1, CCR 7, CLK 2, syntenin, cytochrome C oxidase subunit VIII 등의 유전자들이었다. 특히 발현비가 3.5 이상으로 높은 유전자들을 그 발현비 순으로 나열하면 단구에서는 IFI 30, CLK 2, syntenin, superoxide dismutase 2 등 4개의 유전자들이 있었고, 수지상세포에서는 somatomedin A, methyl-CpG binding domain protein 3, IFI 30, small inducible cytokine subfamily A(Cys-Cys), member 22, ubiquitin-conjugating enzyme E2L 6, hexosaminidase B, nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor epsilon, CCR 7 등 8개의 유전자들이 있었다. 결 론 : 본 연구는 Echinacea가 $CD14^+$ 단구 및 수지상세포에서 발현을 증가시키는 면역관련 유전자들을 cDNA microarray chip을 이용하여 검색하였고, 향후 이 유전자들을 기초로 정량적이고 기능적으로 분석할 수 있는 토대를 마련하였다.

A genome-wide association study of social genetic effects in Landrace pigs

  • Objective: The genetic effects of an individual on the phenotypes of its social partners, such as its pen mates, are known as social genetic effects. This study aims to identify the candidate genes for social (pen-mates') average daily gain (ADG) in pigs by using the genome-wide association approach. Methods: Social ADG (sADG) was the average ADG of unrelated pen-mates (strangers). We used the phenotype data (16,802 records) after correcting for batch (week), sex, pen, number of strangers (1 to 7 pigs) in the pen, full-sib rate (0% to 80%) within pen, and age at the end of the test. A total of 1,041 pigs from Landrace breeds were genotyped using the Illumina PorcineSNP60 v2 BeadChip panel, which comprised 61,565 single nucleotide polymorphism (SNP) markers. After quality control, 909 individuals and 39,837 markers remained for sADG in genome-wide association study. Results: We detected five new SNPs, all on chromosome 6, which have not been associated with social ADG or other growth traits to date. One SNP was inside the prostaglandin $F2{\alpha}$ receptor (PTGFR) gene, another SNP was located 22 kb upstream of gene interferon-induced protein 44 (IFI44), and the last three SNPs were between 161 kb and 191 kb upstream of the EGF latrophilin and seven transmembrane domain-containing protein 1 (ELTD1) gene. PTGFR, IFI44, and ELTD1 were never associated with social interaction and social genetic effects in any of the previous studies. Conclusion: The identification of several genomic regions, and candidate genes associated with social genetic effects reported here, could contribute to a better understanding of the genetic basis of interaction traits for ADG. In conclusion, we suggest that the PTGFR, IFI44, and ELTD1 may be used as a molecular marker for sADG, although their functional effect was not defined yet. Thus, it will be of interest to execute association studies in those genes.

한국노인의 연구참여를 위한 동의능력평가척도의 타당성 연구 (The Validation of a Measurement for Assessing the Capacity of Korean Older Adults to Consent to Research)

  • 본 연구의 목적은 국내에서 노인복지연구 참여를 위한 고지된 동의과정에 동의능력평가가 활발하게 적용될 수 있도록 한국형 동의능력척도를 실증적으로 검증하는 것이다. 본 연구를 위해 재가복지서비스를 이용하거나 노인복지시설에서 보호를 받고 있는 404명의 노인을 조사하였다. 연구 1(n=202)에서는 동의능력검사척도의 신뢰도, 탐색적 요인분석, 수렴 및 기준 타당도 분석을 하였다. 연구 2(n=202)는 연구 1을 통해 수정된 척도의 신뢰도와 타당도는 내적 신뢰도, 확증적 요인분석, 수렴 및 기준 타당도 분석 등을 통해 검증하였다. 연구결과 본 척도의 내적신뢰도 계수가 .90로 높은 수준이었고, 모델 적합도 지수(RMSEA=.057, NNFI=.94, IFI=.96, CFI=.96)도 양호하게 나타났다. 또한 관련변수와의 높은 상관관계 계수를 통해 수렴 및 기준타당도가 검증되었다. 한국형 동의능력평가척도가 우리나라 노인을 대상으로 임상(clinical) 및 조사(survey)연구 참여를 위해 동의능력을 평가하는 척도로서 적합함을 통계적으로 확인하였다. 본 척도는 노인을 대상으로 수행될 연구와 프로그램에서 고지된 동의과정을 위해 활용될 것으로 기대된다.

융복합 사회에서 민간 항공사 운항승무원의 조직몰입 결정요인: 안전, 근무조건, 조직공정성을 중심으로 (Factors Affecting on Organizational Commitment of Airline Flight of Crew in Convergency Society: Focus to Safety, Working Condition and Organizational Justice)

  • 본 연구는 운항 승무원의 삶의 질을 높이기 위한 방안마련을 위해 안전과 근무조건 및 조직공정성, 조직몰입 정도를 측정하고, 조직몰입에 영향을 미치는 관련요인을 파악하기 위해 경로분석을 시행하였다. 2014년 11월 2일부터 3일까지 인천국제공항 및 김포국제공항을 운항하는 국내 7개 민간 항공사의 운항승무원 230명을 대상으로 설문조사를 실시하였다. 안전, 근무조건, 조직공정성, 조직몰입 등을 리커트 5점 만점으로 측정하여 신뢰도와 타당성 검정을 수행하였다. 경로모형의 적합도는 $X^2=88.424$, p<0.001, df=48, $X^2/df=1.842$, RMR=0.044, GFI=0.940, NFI=0.926, CFI=0.964, IFI=0.965로 나타났으며, 최종 연구모형에서 근무조건과 안전은 조직공정성과 조직몰입의 유의미한 영향요인으로 나타났다. 따라서 민간 항공사의 조직공정성과 조직몰입을 위해서는 안전을 강화하고 근무조건을 향상시키고 안전에 대한 노력을 강화해야 한다고 제시된다.

축제 앱 사용자를 위한 축제 검색 요인분석 (Festival search factor analysis for festival application users)

  • 본 논문에서는 단조롭고 일률적으로 보여주는 축제 앱의 단점을 보완하기 위하여 문화관광축제 종합 평가 보고서의 만족도 평가항목과 국내 외 학자들의 축제 품질요인을 이용하여 요인분석을 실시하였다. 요인분석 결과 탐색적 요인분석에서 KMO 값은 0.821로서 나타났다. 요인적재치가 기준 이하인 항목은 제거를 하여 축제 검색요인은 접근성, 홍보안내, 행사내용, 축제상품, 먹을거리, 편의시설을 도출하였다. 신뢰도 분석에서는 Cronbach's ${\alpha}$(Alpha)값이 0.816으로 나타났다. 확인적 요인분석에서는 카이스케어 검정결과 p값이 0.289로 나타났으며, CMIN/DF 값도 1.100로 나타났다. GFI, AGFI, CFI, NFI, IFI 모두 적합도 기준치인 0.9 이상으로 나타났고 RMSEA 역시 0.018로 나타났다.
