Immunological Studies on Oriental Pharmaceutical Preparations (I) (한약제제(漢藥製劑)의 면역학적(免疫學的) 연구(硏究) (제 1 보)(第 1 報))

  • To examine effects of oriental pharmaceutical preparations on immunity, four of such preparations which are currently being used were examined. After their fluid extracts were injected intraperitoneally to ICR mice, the counts of total cell number, PMN and macrophage in the peritoneal cavity were made by using non-specific esterase staining method, and plaque-forming cells (PFC) were measured by using the modified method of Cunningham and Jerne. The results showed that 'Bo-Jung-Ik-Ki-Tang', 'Ih-Jung-Tang', 'Ohn-Kyeong-Tang' and 'Ke-Ji-Bok-Ryeong-Hwan-Ka-Dae-Hwang' increased both the macrophages and the total cells of the peritoneal cavity and that these preparations increased the number of PFC in the spleen. Therefore the data indicate that they potentiate both of the cell­mediated and humoral immunities in the mice.

Immunopathological studies in mice exposed to bisphenol A (마우스에서 bisphenol A 노출로 인한 면역병리학적 연구)

  • Bisphenol A (BPA) is a monomer widely used in the manufacturing polycarbonate plastics or epoxy resin, and xenobiotics recently known as endocrine disrupting chemical. In this paper, to assess the effects of bisphenol A on immunopathological parameters (body weight, organ weight, hematological parameters, cellularity and surface marker) in mice, ICR female mice were orally exposed to BPA dissolved in olive oil as concentrations of 100, 500, 1000 ㎎/㎏/day b.w. 5 days a week for 30 days (subacute exposure). Liver - and kidney weight was significantly increased as dose-dependent manner, but body- , spleen- and thymus- weight didn't changed. In hematological parameters, WBC and MCHC were lowered but HCT and MCV were siginificantly enhanced. There was no significant differences in peritoneal macrophages number of the mice exposed to BPA. However, number of splenocytes of spleen, CD3/sup +/ and CD4/sup +/ cell in splenocytes, CD4/sup +/ and CD8/sup +/ cell in thymocytes were decreased at the mice subacutely exposed to BPA. In addition, BPA decreased expression of B7-1 and B7-2 on macrophages. Therefore, these results showed BPA may affect hematological parameters, cellularity and surface marker of immunocytes.

Effects of Neasookseul-Tang on Anti-Cancer Action in Mice (내소옥설탕(內消沃雪湯)의 항암효과(抗癌效果))

  • Objective: This Study was to investigate effects of Neasookseul-Tang on the anti-cancer, proliferation of immunocytes and nitric oxide(NO) production of peritoneal macrophages. Methods: We used Neasookseul-Tang extract(NOT) with freeze-dried, 8wks-old male mice(balb/c, ICR) and cancer cell lines(L1210, sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). Results: 1. NOT was significantly showed cytotoxicity on the L1210 and Sarcoma-180 cell lines. 2. NOT was significantly increased proliferation of thymocytes and splenocytes in vitro. 3. NOT was significantly decreased proliferation of L1210 cells in L1210 cells transplanted mice. 4. NOT was significantly increased proliferation of thymocytes and splenocytes in L1210 cells transplanted mice. 5. NOT was significantly increased NO production from peritoneal macrophages in L1210 cells transplanted mice. 6. NOT was significantly inhibited body weight and tumor weight in Sarcoma-180 cells transplanted mice. 7. NOT was significantly increased in the mean survival days in Sarcoma-180 cells transplanted mice. Conclusions: The present author thought that NOT had action of anti-cancer by accelerating immuno-function.

Study on Antitumor Effect of Kamicheungyeolhaedogtang(KCHT)(I) (가미청열해독탕(加味淸熱解毒湯)의 항암활성(抗癌活性)에 관(關)한 연구(硏究)( I ))

  • To evaluate the antitumor activity and antimetastatic effects of Kamicheungyeolhaedog tang(KCHT), studies were done experimentally. The results were obtained as follows: 1. KCHT extracts exhibited a significant cytotoxicity against A549, SK-MEL-2, SK-OV-3, and B16-BL6 cell lines. 2. KCHT extracts showed significant inhibitoty effect on DNA topoisomerase I 3. The T/C% was 145.8% in KCHT treated group in S-180 bearing ICR mice. 4. KCHT extracts exhibited efficient affect adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 5. In vitro neovascularization assays, angiogenesis was insignificantly inhibited in KCHT treated group as compared with control group. These results suggested that KCHT extracts might be usefully applied for prevention and treatement of cancer.

Study on Antitumor Activity of Kamibojungikgi-tang (가미보중익기탕이 항암활성 및 항전이효과에 관한 연구)

  • To explore the possible cancer agent from oriental prescriptions, we have examined its antitumor and anti metastatic activities of Kamibojungikgi-tang(KBIT). KBIT extracts exhibited cytotoxicity against P388, A549 and B16-F10 cell lines in a dose-dependent manner and showed antiadhesive effect of A549 cell to complex extracellular matrix at 1 ㎎/㎖ in vitro. In DNA topoisomerase I assay, KBIT extracts showed strong inhibitoty effect in a dose-dependent manner. In pulmonary colonization assay with B16BL6, a number of colonies in the lungs were decreased effectively in KBIT treated group as compared with control group. Moreover, in CAM assay, KBIT extracts significantly inhibited angiogenesis at 15㎍/egg as compared with control. The T/C% was 141% in KBIT treated group in S-180 bearing ICR mice. From the above results it was concluded that KBIT had antitumor and anti metastatic activities. So it is expected to be clinically helpful on the prevention and treatment of cancer, although it is still necessary to study its mechanism on molecular biology and immunology.

Effects of Oenanthe javanica Extracts on Mercury Accumulation in Organs of the Mouse (미나리 추출물이 마우스의 장기내 수은 축적에 미치는 영향)

  • This study was performed to investigate the antitoxic effect of Oenanthe javanica extracts on orally administered mercury compound. Adult male ICR mice were exposed to methylmercuric chloride (CH3HgCl)through drinking water. The control, mercury treated and Oenanthe javanica treated groups not showed significant differences in mean body and organ weights of mice. The distribution of mercury in the cerebellum, kidney, liver and spleen of the mouse were examined according to a histochemical mathod. Grains of mercury traces were located in the purkinje cell and granular layers of the cerebellum and cortex of kidney respectively. Lesser staining of the grains was seen in the collecting tubules of medulla. in the liver, mercury accumulations were present primarily in the hepatocytes around portal area containing interlobular bile duct, artery and portal vein. Also grains of mercury traces were accumulated in the white pulp of the spleen. In the group of Oenanthe javanica extracts, staining intensity of mercury was decreased in the Purkinje cell layer of cerebellum and in the portal area of liver respectively. Staining patterns in kidney and spleen of extracts group were similar to that of only mercury treated group.

In Vitro Development and Conception Rate after Transfer of Mouse Embryos Bisected at Morula or Blastocyst Stage (상실배 및 포기배에 분할한 생쥐 수정란의 체외발달 속도 및 이식 후 수태율)

  • These experiments were carried out to determine the effect of pregnancy in bisected embryo. The embryos of ICR mouse were microsurgically bisected at morula and blastocyst stage using microsurgical blade attached a micromanipulator. These bisected embryos without zona pellucida were cultured up to blastocyst stage and cell count and diameter of stained blastomere, and transferred pseudopregnant mice. And the development of these bisected embryos was compared with the results of production of young of the corresponding intact embryos or cell stage. When the bisected mouse embryos were cultured in vitro for 20 to 24 hours in morula stage(77.2%) or 3 to 6 hours in blastocyst stage(84.1%), them were developed to the expanded blastocyst stage. There were no significant(P<0.05) differences in the development rate of bisected embryos between in morula and blastocyst stages. The embryo size of blastocyst developed in vitro from bisected embryo was small(P<0.05)than intact embryo. However, the number of blastomeres with bisected embryo (24.7+1.3and 21.5+1.2 respectively) were significantly(P<0.05) reduced, compared with that of intacted embryos(36.3+1.1 and 41.4+1.2 respectively). When compared with the result of pregnancy rate(63.6%) after surgical transfer of bisected morulae, a similar result(65.4%) was obtained with bisected blastocyst stage(P< 0.05). However, production of youngs (38.8%) after transfer of bisected morula, a similar result (38.1%) was obtained with bisected blastocyst stage (P<0.05).

  • With an aim to disclose causal factors scably grains intoxications, the screening of toxic Fusaria and the detection of toxic priniciples were performed in respect of cultured cells bioassay with HeLa cells, skin-necrotizing effect, histopathological investigation and also chromatographic analysis sith following results ; 1. Among the fungi, Fusarium sp. F-27, F-63 and F-61 were highly toxic to mice, causing liver injury characterized necrosis and inflammation. 2. HeLa cell culture bioassay demonstrated that the cell of the isolated strains of Fusaria were suspected to produce toxic material (Fusarenon-X). 3. The culture filtrates of Fusarium nivale Fn-2B, F-27, and F-63, were injected subcutaneously, and caused inflammation followed by crust on the skin ICR-mice. 4. The observation method of skin-necrotizing effect to the mice can be used to the screening to the toxin-producing fungi isolated from many fusarial contaminations.

Anti-cancer Effects of Kamiboa-tang and some other Traditional Medical Prescriptions (가미보아탕(加味保我湯) 및 수종(數種) 한방처방의 항암효과에 대한 연구)

  • Objectives : Anticancer and immune-modulating effects of several Korean medical prescriptions including Yukgunja-tang, Bohwa-tang, Sogam-Won, and Kamiboa-tang were investigated. Methods : In vitro anti-cancer effects were measured by cytotoxicity MTT assay using SNU-1 gastric cancer cell lines, In vivo anti-cancer effects were measured by increased life span of S-180 sarcoma-injected ICR mouse. Immune-modulating effects were analyzed by measuring hemagglutinin titer, appearance of rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice. Results : In vitro assay showed that only Sogam-won showed cytotoxic effect with $IC_{50}$ of 87.9 ${\mu}g/ml$. All other prescriptions showed no cytotoxic effects against SNU-1 gastric cancer cell line. However, in vivo assay showed that Sogam-won showed lowest anti-cancer effects in contrast to its highest cytotoxic effects, Kamiboa-tang, which showed no cytotoxic effect, showed the highest in vivo anticancer effects, with increased life span of 140%. Kamiboa-tang showed significant immune-enhancing activities by significantly increasing rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice (P<0.05). Conclusion : The anticancer effect of Kamiboa-tang is not mediated by direct inhibition of cancer cells but is mediated by improving immune reactions against cancer cells.

  • The purpose of this study was to investigate the effect of Kamigedang-tang(KGDT) water extract on the antitumor and anti metastatic activity. The results were summarized as follows: KGDT extracts exhibited a significant cytotoxicity against P388, SK-MEL-2, SK-OV-3, and B16-F10 cell lines and showed significant inhibitoty effect on DNA topoisomerase I from calf thymus. The T/C% was 122.9% in KGDT treated group in S-180 bearing ICR mice. Also, KGDT extracts exhibited efficient affect adhesive effect of A549 cell to complex extracellular matrix. In CAM assay, KGDT extracts inhibited angiogenesis at 15㎍/egg concentration insignificantly as compared with control. These results suggested that KGDT extracts might be usefully applied for prevention and treatement of cancer.
