삼릉추출물의 멜라닌 생성 저해 효과 (The Inhibitiory Effects of the Scirpi rhizoma on Melanogenesis)

  • 본 연구에서는 한방 천연 미백 소제를 개발하기 위하여 삼릉추출물의 미백 효과에 대하여 연구하였다. 삼릉추출물은 프리 라디칼 소거활성(DPPH)에서 $IC_{50}=638{\mu}g/mL$, superoxide radicals 소거활성은 $IC_{50}=21.7{\mu}g/mL$을 나타냈다. 삼릉추출물의 B16 melanoma 세포에서의 멜라닌 생합성 저해효과는 $100{\mu}g/mL 삼릉추출물을 48 h 처리한 세포에서는 멜라닌 합성량이 27% 감소되었다. 삼릉 추출물은 농도에 비례하여 B16 melanoma 세포내의 타이로시네이즈 활성을 저해하였으며, western blot을 이용하여 타이로시네이즈와 타이로시나아제 관련 단백질(TRP-1)의 발현감소를 확인하였다. 또한, RT-PCR을 이용하여 삼릉추출물이 멜라닌 생성 과정에 관련하는 유전자 발현을 조사한 결과 타이로시네이즈와 TRP-1의 mRNA발현을 억제하는 것을 확인하였다. 그러므로 삼릉추풀물은 항산화 효과와 함께 멜라닌 저해 효과가 우수하여 새로운 미백 화장품으로 응용할 수 있을 것으로 생각된다.

Antioxidant Effect of Berberine and its Phenolic Derivatives Against Human Fibrosarcoma Cells

  • Berberine (B1), isolated from stems of Coscinium fenestratum (Goetgh.) Colebr, was used as a principle structure to synthesize three phenolic derivatives: berberrubine (B2) with a single phenolic group, berberrubine chloride (B3) as a chloride counter ion derivative, and 2,3,9,10-tetra-hydroxyberberine chloride (B4) with four phenolic groups, to investigate their direct and indirect antioxidant activities. For DPPH assay, compounds B4, B3, and B2 showed good direct antioxidant activity ($IC_{50}$ values=$10.7{\pm}1.76$, $55.2{\pm}2.24$, and $87.4{\pm}6.65{\mu}M$, respectively) whereas the $IC_{50}$ value of berberine was higher than $500{\mu}M$. Moreover, compound B4 exhibited a better DPPH scavenging activity than BHT as a standard antioxidant ($IC_{50}=72.7{\pm}7.22{\mu}M$) due to the ortho position of hydroxyl groups and its capacity to undergo intramolecular hydrogen bonding. For cytotoxicity assay against human fibrosarcoma cells (HT1080) using MTT reagent, the sequence of $IC_{50}$ value at 7-day treatment stated that B1 < B4 < B2 ($0.44{\pm}0.03$, $2.88{\pm}0.23$, and $6.05{\pm}0.64{\mu}M$, respectively). Berberine derivatives, B2 and B4, showed approximately the same level of CAT expression and significant up-regulation of SOD expression in a dose-dependent manner compared to berberine treatment for 7-day exposure using reverse transcription-polymerase chain reaction (RT-PCR) assays. Our findings show a better direct-antioxidant activity of the derivatives containing phenolic groups than berberine in a cell-free system. For cell-based system, berberine was able to exert better cytotoxic activity than its derivatives. Berberine derivatives containing a single and four phenolic groups showed improved up-regulation of SOD gene expression. Cytotoxic action might not be the main effect of berberine derivatives. Other pharmacological targets of these derivatives should be further investigated to confirm the medical benefit of phenolic groups introduced into the berberine molecule.

Antiproliferative Effects of Crocin in HepG2 Cells by Telomerase Inhibition and hTERT Down-Regulation

  • Crocin, the main pigment of Crocus sativus L., has been shown to have antiproliferative effects on cancer cells, but the involved mechanisms are only poor understood. This study focused on probable effect of crocin on the immortality of hepatic cancer cells. Cytotoxicity of crocin ($IC_{50}$ 3 mg/ml) in hepatocarcinoma HepG2 cells was determined after 48 h by neutral red uptake assay and MTT test. Immortality was investigated through quantification of relative telomerase activity with a quantitative real-time PCR-based telomerase repeat amplification protocol (qTRAP). Telomerase activity in 0.5 ${\mu}g$ protein extract of HepG2 cells treated with 3 mg/ml crocin was reduced to about 51% as compared to untreated control cells. Two mechanisms of inhibition, i.e. interaction of crocin with telomeric quadruplex sequences and down regulation of hTERT expression, were examined using FRET analysis to measure melting temperature of a synthetic telomeric oligonucleotide in the presence of crocin and quantitative real-time RT-PCR, respectively. No significant changes were observed in the $T_m$ telomeric oligonucleotides, while the relative expression level of the catalytic subunit of telomerase (hTERT) gene showed a 60% decrease as compared to untreated control cells. In conclusion, telomerase activity of HepG2 cells decreases after treatment with crocin, which is probably caused by down-regulation of the expression of the catalytic subunit of the enzyme.

Metastatic Inhibitory and Radical Scavenging Efficacies of Saponins Extracted from the Brittle Star (Ophiocoma erinaceus)

  • Echinodermata use saponins in chemical defense against pathogens and predators. The molecular mechanisms of antimetastatic effects of brittle star saponins are still unknown. The present study examined antioxidant capacity and invasive ability in HeLa carcinoma cells exposed to brittle star crude saponins. Discolorating methods with DPPH and ABTS and expression of SOD-2 with RT-PCR were used to estimate the antioxidant activity. The anti-invasive activity of extracted saponins was examined through adhesion of HeLa cells to extracellular matrix, wound healing and evaluation of the mRNA levels of MMP-2 and MMP-9 by real time-PCR. The results showed that extracted saponins had cytotoxicity against cervical cancer cells and ABTS and DPPH scavenging properties with $IC_{50}$ values of 604.5, $1012{\mu}g/ml$, respectively. Further, we found that, in wound healing assay, brittle star saponins could prevent invasion of HeLa cells in a concentration dependent manner. Furthermore, cell adhesion assay demonstrated blockage of cell attachment to extracellular matrix with an $IC_{50}$ concentration of $16.1{\mu}g/ml$. The significant dose dependent down regulation of MMP-2 and MMP-9 in treated cells demonstrated that isolated saponins can decline tumor metastasis in vitro. The brittle star saponins remarkably prevented cervical cancer invasion and migration associated with down regulation of matrix metalloproteinase expression. Therefore, saponins could be suggested as an anti-invasive candidate against cervical cancer and an antioxidant as well.

백서 간손상에 의한 구기자의 유리자유기 소거능에 관한 연구 (The Study of Free Radical Scavenging Effect of Lycii Fructus by Liver Injury of Rats)

  • In the present study, we investigated the protective effect of the Lycii Fructus water extracts (LFE) against CCl4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of LFE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation, In addition, the pretreatment of LFE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CC1₄-injected rats. The LFE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 μg/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of LFE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the LFE may be related to antioxidant effects and regulation of CYP2E1 gene expression.

$CC1_4$로 유발된 백서의 간손상에 대한 갈근의 간보호작용 연구 (The Study of Protective Effect of Puerariae Radix against $CC1_4$-induced Hepatotoxicity)

고량강으로부터 분리된 galangin의 RAW 264.7 세포주에서 LPS로 유도된 nitric oxide 생성 저해활성 (Inhibitory Effect of Galangin from Alpinia officinarum on Lipopolysaccharide-induced Nitric Oxide Synthesis in RAW 264.7 macrophages)

  • 각종 염증성 질환 및 패혈증으로 인한 치명적인 저혈압을 예방치료하는 약물 개발을 위한 기초 연구로서 유도성 NOS (inducible nitric oxide synthase, iNOS) 에 의한 NO의 과다 생성을 저해하는 성분을 천연물로부터 찾아내고자 본 연구를 수행하였다. NO 생성 저해활성의 검정은 대식세포주인 RAW 264.7 세포를 LPS로 활성화한 후, 유도되는 iNOS에 의해 생성되는 NO를 Griess 시약을 이용해 $NO_2{^-}$의 형태로 정량하였다. 또한 Western blot 실험 및 RT-PCR 실험을 시행하여 iNOS의 mRNA의 발현 및 단백 합성에 대한 영향을 조사하였다. 고량강(Alpinia officinarum Hance, Zingiberaceae)의 메탄올 추출물로부터 극성에 따른 용매 분획을 시행하여 활성성분을 분리하고 분광학적 분석법을 이용하여 분리한 단일성분이 flavonol 구조인 3,5,7-trihydroxy-2-phenylchromen-4-one (galangin, GLG)임을 확인하였다. 작용기전을 알아보기 위해, Western blot 및 RT-PCR 실험결과, 분리한 flavonol 성분(GLG)의 NO 생성저해 활성은 iNOS mRNA발현을 저해하여 iNOS 효소 단백질의 생성이 억제됨에 기인하는 것으로 확인하였다. 따라서, 고량강 추출물로부터 분리한 flavonol 화합물(GLG)이 iNOS 발현의 억제를 통해 다량의 NO 생산을 저해함으로써, 고량강(Alpinia officinarum)의 NO 과량생성과 관련된 염증성 질환에 대한 응용 가능성이 클 것으로 기대된다.

정천탕의 알러지에 대한 실험적 연구 (The Experimental study of Jungchun-tang on Allergies)

  • 목적 : 정천탕(定喘湯, JCT)이 IgE/mast cell로 유포되는 천식(喘息)등의 알러지성 질환에 대한 효과를 실험적으로 검증하고자 하였다. 방법 : IgE의 혈중농도와(in vivo) 분비 량을 측정하였으며(in vitro), 알러지 관련 유전자의 발현을 RT-PCR로 검색하였고, JCT의 수용성 복합 추출물이 compound 48/80로 유발되는 mast cell line(IC-2)의 histamine의 방출에 미치는 직접적인 영향을 측정하였다(in vitro). 결과 : 정천탕은 ovalbumin으로 유발시킨 마우스의 혈중 IgE량과 IL-4와 CD-40로 유도시킨 spleen B 임파구의 IgE 생성량도 현저하게 감소시켰다. 또한 INF-r의 발현은 증가시킨 반면, IL-4와 IL-6의 발현과 mast cell의 histamine 방출은 억제하는 것으로 나타났다. 결론 : 정천탕의 항알러지 효과를 실험적으로 확인 할 수 있었으며 임상적으로는 천식등 알러지성 질환에 사용 할 수 있을 것으로 사료된다.

Synthesis of Heterocycle-linked Thioureas and Their Inhibitory Activities of NO Production in LPS Activated Macrophages

  • A series of thioureas were synthesized as inhibitors of NO production in lipopolysaccharide-activated macrophages. We investigated the effect of lipophilic moiety and N-substituents of the thioureas on the activity. Phenoxazine and carbazole-containing derivatives revealed higher activity than indole-containing thioureas. The appropriate spacer between lipophilic tail and thiourea head and methyl substituent at N3 position of thiourea brought beneficial effect on the inhibition of NO production. Among prepared compounds, phenoxazine-containing derivative 2a was the most potent with $2.32 {\mu}M$ of $IC_{50}$ value. RT-PCR analysis suggested that the prepared thioureas inhibited NO production through the suppression of iNOS mRNA expression.

Inhibition of Nitric Oxide Production, iNOS and COX-2 Expression of Ergosterol Derivatives from Phellinus pini

  • Ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were screened for their ability to inhibit nitric oxide (NO) production in LPS-activated RAW 264.7 cells. Compounds 1, 2, and 3 reduced NO production in the assay with $IC_50$ values of 29.7 ${\mu}M$ (1), 15.1 ${\mu}M$ (2), and 18.4 ${\mu}M$ (3) respectively. They also suppressed the expression of protein and m-RNA of iNOS and COX-2 in a dose dependent manner by western blot analysis and RT-PCR experiment in LPS-activated microglial cells.
