Environmental Geochemical Characteristics of the Acid Mine Drainage and Stream Sediments in the Abandoned Honam Coal Mine Area, Korea (호남탄전지역 폐탄광의 산성광산배수와 하상퇴적물의 환경지구화학적 특성)

  • The purpose of this study is to investigate environmental geochemical characteristics for the acid mine drainage and stream sediments in the abandoned Honam coal mine area. For the chemical analysis, stream winter samples were collected at 54 sites and for the mineralogical and chemical analysis, stream sediments samples were collected at 34 sites. Physical and chemical characteristics of stream water were measured at the site and chemical compositions were analyzed by ICP-AES, ICP-MS and IC. Mineralogical characteristics of stream sediments samples were determined using XRD, SEM and EDS. In physical and chemical properties, stream water shows thats pH 2.85~8.12, Eh -62~215 mV, EC 0.205~146 ms/m, ER 0.234~255 {$\Omega}{\cdot}$m, DO 0.03~1068 mg/L and TDS 10.96~1420mg/L. In chemical compositions, we obtain that K 0.118~3.184 mg/L, Mg 2.1~114.48 mg/L, Ca 2.59~l25.02 mg/L, Al 0.01~44.72 mg/L, Fe 0.108~89.49 mg/L and Na 5.45~125.41 mg/L. Stream sediments are commonly composed of quartz, illite and goethite in the result of x-ray diffraction. After heat treatment at 98$0^{\circ}C$, the hematite peaks are distinctly shown. In consideration of heavy metals (include Fe) for steam sediments, Fe ranges 22575~34713 ppm, Zn 41.66~970.3 ppm, Cd 0.52~52.07 ppm, Cu 1.25~198.5 ppm and Pb 0.43~77.35 ppm.

Secondary Metabolites with Anti-complementary Activity from the Stem Barks of Juglans mandshurica Maxim

  • Juglans mandshurica is a fast growing hard species, which is a tree in family of Juglandaceae and has a wide distribution in China, Korea and eastern Russia. Plant materials from J. mandshurica have extensively been used in folk medicines to prevent or cure gastric, esophageal, lung and cardiac cancer. As one chain of our searching for anticomplementary agents from natural sources, two epimeric ellagitannins, [2,3-O-4,4',5,5',6,6',-hexahydroxydiphenoyl (HHDP))-(${\alpha},{\beta}$)-D-glucose] (I) and pedunculagin (II) were purified from 70% acetone extracts of the stem barks of J. mandshurica by Thin Layer Chromatography and Sephadex LH-20 column chromatography approaches. The chemical structures of the isolated compounds were characterized by MS, NMR, and a careful comparation with published literatures. The epimeric ellagitannins I and II exhibited inhibitory properties against a classical pathway of complementary system with 50 % inhibitory concentrations ($IC_{50}$) values of 65.3 and $47.7{\mu}M$, respectively, comparing with riliroside ($IC_{50}=104{\mu}M$) and rosmarinic acid ($IC_{50}=182{\mu}M$), which were used as positive controls. Thus, the work indicated both the two secondary metabolites possess excellent inhibitory activity and might be developed as potential anticomplementary chemicals.

Antioxidant Constituents from the Leaves of Cedrela sinensis A. Juss

  • Phytochemical study on the EtOAc fraction from the MeOH extract of the leaves of Cedrela sinensis led to the isolation of five known phenolic compounds (1-5), whose structures were identified as (+)-catechin (1), $kaempferol-3-0-{\alpha}- L-rhamnopyranoside$ (2), quercetin (3), $quercetin-3-O-{\alpha}-L-rhamnopyranoside$ (4), and $quercetin-3-O-{\beta}-D-glucopyranoside$ (5), respectively, by comparing their spectral $(uv,\;JR,\;IH\;and\;^{13}C-NMR,\;and\;ESI-MS)$ and physicochemical data with those reported in the literature. Among the isolated compounds (1-5), compounds 1 and 3-5 exhibited significant DPPH radical scavenging effects with $IC{_50}$ values ranging from $21.3{\pm}1.4\;to\;38.1{\pm}3.2 {\mu}M$ as well as superoxide anion radical scavenging effects with $IC{_50}$ values ranging from $9.4{\pm}0.7\;to\;21.2{\pm}3.6 {\mu}M$. Furthermore, compounds 1 and 3-5 also exhibited considerable inhibitory effects on LDL peroxidation induced by either $CU^{2+}$ or AAPH with $IC{_50}$ values ranging from $1.4{\pm}0.4\;to\;11.9{\pm}1.4\;{\mu}M$. These results indicated that flavonoids are the major constituents of C. sinensis and considered to be antioxidant principles of this plant.

Melanin Synthesis Inhibition and Radical Scavenging Activities of Compounds Isolated from the Aerial Part of Lespedeza cyrtobotrya

  • The EtOAc fraction of Lespedeza cyrtobotrya showed mushroom tyrosinase inhibitory and radical scavenging activities. Four active compounds were isolated based on Sephadex LH-20 chromatography and HPLC, and the structures were elucidated, on the basis of their LC-MS and NMR spectral data, as 2-(2,4-dihydroxyphenyl)-6-hydroxybenzofuran (1), eriodictyol-7-O-glucopyranoside (2), haginin A (3), and dalbergioidin (4), respectively. Compound (1) showed mushroom tyrosinase inhibitory activity with an $IC_{50}$ value of $5.2\;{\mu}M$ and acted as a competitive inhibitor. Furthermore, $37.3\;{\mu}M$ of compound 1 reduced 50% of the melanin content on human melanoma (MNT-1) cells. The radical scavenging activities of compounds 1, 2, 3, and 4 were shown to have $IC_{50}$ values of 11.0, 24.5, 9.0, and $36.5\;{\mu}M$, respectively, in an ABTS system and $IC_{50}$ values of 42.7, 36.0, 37.7, and $61.7\;{\mu}M$, respectively, in a DPPH system. The mushroom tyrosinase inhibitory activity of the EtOAc fraction of Lespedeza cyrtobotrya was contributed by compounds 1, 3, and 4, and its radical scavenging activity was contributed by compounds 1-4.

  • Activity-guided isolation from an ethylacetate-soluble fraction of a 70% (v/v) ethanolic extract from the roots of Taraxacum ohwianum, using a pancreatic lipase inhibition assay, resulted in isolation and identification of five phenolic metabolites of previously known structure; these were 3,5-di-O-caffeoylquinic acid, chicoric acid, caffeic acid, protocatechuic aldehyde, and luteolin. All structures were confirmed by NMR and MS scpectroscopic data. Of these compounds 3,5-di-O-caffeoylquinic acid exhibited the most potent inhibitory activity, with $IC_{50}$ of $65.1{\pm}0.7\;{\mu}M$ against pancreatic lipase.

Antioxidant and Anti-Inflammatory Activities of Eugenol and Its Derivatives from Clove (Eugenia caryophyllata Thunb.) (정향(Eugenia caryophyllata Thunb.) Eugenol 및 그 유도체의 항산화 및 항염증활성)

  • Antioxidant and anti-inflammatory activities of eugenol and its derivatives from clove (Eugenia caryophyllata Thunb.) were evaluated using in vitro assay systems by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, cyclooxygenase-2 (COX-2), and 15-lipoxygenase (15-LOX). Among eight different crude medicinal drugs tested, volatile extracts of clove extracted by steam distillation extraction (SDE) showed potent DPPH radical scavenging activity ($IC_{50}$=8.85 ${\mu}g/mL$) as well as strong inhibitory activity against COX-2 (58.15%) and 15-LOX (86.15%) at 10 ${\mu}g/mL$ and 25 ${\mu}g/mL$, respectively. Major volatile components of clove were identified as eugenol, trans-caryophyllene, and acetyleugenol by GC-MS analysis. Out of three eugenol derivatives, eugenol, methyl eugenol, and acetyl eugenol, eugenol showed the strongest DPPH radical scavenging activity and COX-2 inhibitory activity, whereas methyl eugenol exhibited the strongest 15-LOX inhibitory activity. Finally, the contents of the three eugenol derivatives in clove were quantified by analytical HPLC. Contents of eugenol and acetyl eugenol in clove were 6.95% and 1.85% per dry weight, respectively. These results suggest that eugenol and its derivatives in steam distilled extract of clove may be useful as potential antioxidant and anti-inflammatory agents.

  • In order to find anti-dementia drugs from natural products, prolyl endopeptidase inhibitors were purified from Puerariae Flos by consecutive solvent partition, followed by silica gel, Sephadex LH-20, and HPLC. Four isoflavonoid inhibitors were isolated and identified as tectorigenin, genistein, 5,7-dihydroxy-4',6-dimethoxyisoflavone, and 5-hydroxy-6,7,4'-trimethoxyisoflavone by means of instrumental analyses including $^{1}H-$, $^{13}C-$, $^{2}D-NMR$ and MS and $IC_{50}$ values against PEP were 5.30 ppm$(17.7\;{\mu}M)$, 10.39 ppm$(38.5\;{\mu}M)$, 13.92 ppm$(44.3\;{\mu}M)$, and 20.61 ppm$(62.8\;{\mu}M)$, respectively. Some previous mistakes in $^{13}C-NMR$ assignment were revised by careful investigation of HMBC and HMQC data.

Antioxidant Activity of The Residue Generated During Pervaporation of Bioethanol Produced from Lignocellulosic Biomass (목질계 바이오매스로부터 생산된 바이오에탄올 투과증발 과정에서 발생한 투과증발 잔류물의 항산화 활성)

  • In this study, we produced bioethanol from the original hydrolysate obtained during oxalic acid pretreatment of lignocellulosic biomass. The bioethanol was separated and concentrated by pervaporation and the residue after pervaporation was evaluated for its antioxidant activity. Xylose ($37.28g/{\ell}$) was the major product in the original hydrolysate. The original hydrolysate contained acetic acid, furfural and total phenolic compounds (TPC) as fermentation inhibitors. Acetic acid was removed by electrodialysis (ED), and $12.21g/{\ell}$ of bioethanol was produced from ED-treated hydrolysate. The TPC of ethyl acetate extracts from the residue obtained (OA-E) during pervaporation was 86.81 mg/100 g (extract). The $IC_{50}$ values of DPPH and ABTS radical scavenging activities, and reducing power of OA-E were $0.87mg/m{\ell}$, $0.85mg/m{\ell}$, and $0.59mg/m{\ell}$, respectively. Sugar degradation products and the phenolic compounds in OA-E were determined by GC-MS.

Responses of Tobacco Photomixotrophic Cultured Cells to Various Herbicides (다양한 제초제에 대한 담배 Photomixotrophic 배양세포의 반응)

  • To establish an efficient screening system for new herbicides using plant cultured cells, responses of tobacco photomixotrophic cultured (PH) cells to various herbicides with different modes of action were surveyed by measuring the cell growth and ion conductivity in medium. The cells were cultured in Murashige and Skoog (MS) medium containing 0.7mg/L 2,4-D, 0.3mg/L kinetin and 30 g/L sucrose at $25^{\circ}C$ in the light (100 rpm). Chemicals were treated to suspension cultures of tobacco PH cells at the time of subculture. The cell growth and ion conductivity in the medium were investigated on 12 days after chemical treatment. The ion conductivity assay gave well correlated results to the cell growth inhibition data. The responses of tobacco PM cells were dependent on the modes of action of chemicals tested. Atrazine, an inhibitor of photosynthetic electron transport (PET), strongly inhibited both the cell membrane and cell growth ($IC_{50}$/, about 1 $\mu$M). Butachlor (an inhibitor of cell division), glufosinate (an inhibitor of amino acid biosynthesis), and fluridone (an inhibitor of carotenoid biosynthesis) showed a dose-dependent inhibition. However, Quinclorac, a herbicide with an auxin activity, did not affect the cell growth and ion leakage. These results suggested that tobacco PM cells is suitable materials for the simple screening of new herbicides such as PET, amino acid biosynthesis, ceil division inhibitors by measuring the cell growth and ion conductivity.

Performance Evaluation of Hazardous Substances using Measurement Vehicle of Field Mode through Emergency Response of Chemical Incidents

  • BACKGROUND: Chemical accidents have increased owing to chemical usage, human error and technical failures during the last decades. Many countries have organized supervisory authorities in charge of enforcing related rules and regulations to prevent chemical accidents. A very important part in chemical accidents has been coping with comprehensive first aid tool. Therefore, the present research has provided information with the initial applications concern to the rapid analysis of hazardous material using instruments in vehicle of field mode after chemical accidents. METHODS AND RESULTS: Mobile measurement vehicle was manufactured to obtain information regarding field assessments of chemical accidents. This vehicle was equipped with four instruments including gas chromatography with mass spectrometry (GC/MS), Fourier Transform Infrared Spectroscopy (FT-IR), Ion Chromatography (IC), and UV/Vis spectrometer (UV) to analyses of accident preparedness substances, volatile compounds, and organic gases. Moreover, this work was the first examined the evaluation of applicability for analysis instruments using 20 chemicals in various accident preparedness substances (GC/MS; 6 chemicals, FT-IR; 2 chemicals, IC; 11 chemicals, and UV; 1 chemical) and their calibration curves were obtained with high linearity ( r 2 > 0.991). Our results were observed the advantage of the high chromatographic peak capacity, fast analysis, and good sensitivity as well as resolution. CONCLUSION: When chemical accidents are occurred, the posted measurement vehicle may be utilized as tool an effective for qualitative and quantitative information in the scene of an accident owing to the rapid analysis of hazardous material.
