  • The genus Hypoxylon is a member of the Xylariaceae which has brown to dark brown and pha- seoliform single cell ascospores with a conspicuous full germ slit. The isolates of Hypoxylon collected from England, Hong Kong and Korea are compared with similar taxa and described. These isolations under Hypoxylon section are detailed and illustrated, using light microscopy and scanning electron microscopy. Hypoxylon howeianum and H. rubiginosum are described and compared with former collection from Korea. A key to the species is also included.

These isolations under Annulata section are illustrated with a light microscopy and a scanning electron microscopy. Hypoxylon archeri is newly recorded from Korea. A key to the species is also included.

  • The genus Hypoxylon is a member of the Xylariaceae that has brown to dark brown and phaseoliform single cell ascospores with a conspicuous full germ slit. Hypoxylon truncatum differs from H. annulatum, it has conspicuous perithecial mounds, dark brown stromatal surface, and absence of enriched ostiola disc. The isolate of Hypoxylon, collected from Mt. Deuk-yu in Korea, is compared with similar taxa and described. The isolation of H. truncatum is compared with similar taxa, H. annulatum, with a precise description in the SEM level in Korea.

  • The tissue types of stromata were observed intensively in four species of Hypoxylon under a scanning electron microscope (SEM). These stromata were sectioned with a freezing fracture method for observation. Several tissue types were recognized and stable in each species. This study presents the most intensive observation of tissue types of each layer of stromata. It will be useful for taxonomic criteria for the species level. However, the tissue types can not be major taxonomical criteria for the genus Hypoxylon.

  • Latent infections of healthy-appearing oaks of Hypoxylon atropunctatum complicates field studies by interfering with inoculation experiments to follow pathogenesis, fungal development and reproduction of this canker rot fungus. Mutants with unique and easily scorable phenotypes would be useful for inoculation studies. There is a broad range in the capacity of wild-type isolates to utilize nitrate as a sole nitrogen sources. Several types of nitrate-nonutilization mutants (nit1, Nit3, NitM) were selected from nitrate-utilizing wild-type isolates. Also, a few mutants of Hypoxylon atropunctatum were selected that could only grow poorly on basal medium supplemented with various nitrogen sources and even on yeast extract agar. These unknown mutants need to be characterized further. Nit mutants of Hypoxylon atropunctatum were readily selected, grew well and were recovered after inoculation into oak stems. These results suggest that nit mutants could be useful for inoculation studies in trees that contain latent infections.

  • The genus of Hypoxylon, well known wood degrading fungi, is a member of the Xylariales, which has woody to carbonaceous, brown to dark brown stromata. Hypoxylon moriforme, which was isolated from heavily decayed hardwood, is fairly described by electron microscope. The isolation of H. moriforme will be the first record and the first description in Korea. The species collected, having small size of ascospores $7.5\sim9.0\times2.5\sim4.0{\mu}m$ and asci $75\sim130\times3\sim5{\mu}m$, are taxonomically compared with H. truncatum, which is well known as a popular species in Northeast Asia. And H. bovei and H. annulatum, which are the tropic to subtropic species and also compared with other temperate species of H. moriforme.

  • Tremella fuciformis produces white jelly fruitbody which is used as a special food in the orient. Symbiotic relationship between T. fuciformis and Hypoxylon sp. is important for mass production of fruitbody in T. fuciformis. T. fuciformis showed the peak of 24mg/2mL on the 9th day, after that mycelial growth maintained a gentle curve. Protein content increased into 0.69㎍/mL in rapid, T. fuciformis fruiting body maintained high galactose, mycelia of T. fuciformis showed 42.6% trehalose.

  • An unrecorded Hypoxylon sp. was observed on the bed-log of oak-mushroom (Lentinula edodes) in Korea. The stroma of the fungus was formed on the bed-log (Quercus spp.) damaged by Korean squirrel (Sciurus vulgaris coreae). Perithecia were $200{\sim}400{\mu}m$, and asci containing 8 ascospores were $7{\sim}8{\times}150{\sim}230{\mu}m$ in size. Paraphysis was cylindrical shape, Ascospores were $5{\sim}9{\times}12{\sim}17.5{\mu}m$ in size, pale brown to dark brown, elliptical, and smooth. They have germination cleft and $1{\sim}2$ oil droplets.

  • Two strains, each of Tremella fuciformis and Hypoxylon sp. were isolated and their mycelial properties were investigated. T. fuciformis produced yeast-like conidia and dikaryotic mycelia from white to yellow color. The diameter of the hyphae was $1.5\;to\;3.0\;{\mu}m$, and septa with clamp connections were present. Secondary mycelia with clamp connections could produce fruitbodies on sawdust medium when the environmental conditions were suitable for fructification. The symbiotic fungus, Hypoxylon sp. produced white feather-like mycelia. But the color of old mycelia was changed to light yellow or light brown and pigmented the culture medium from light brown to brown or very dark green. Generally it did not produce conidia, but in a special case it produced conidia of which color was yellow green to grass green and of which shape was sub-elliptical with the size of approximately $3\;to\;5\;{\mu}m$.

  • The fungal flora of Korean wood-rotting fungi were studied for two years from March of 1990 to February of 1992. Fresh fungi were collected from national parks, some local areas, and several islands throughout the country. Fleshy ascomycetes and jelly fungi were examined through identification and literature studies. They were counted 52 species, 1 subspecies, and 1 forma among which, Hypoxylon and Femsjonia were confirmed as unrecorded genera and Hypoxylon punctulatum, Exidia recisa, and Femsjonia pezizaeformis as unrecorded species to Korea and are registered here with descriptions.
