The Influence of the Dissolved Oxygen of Bottom Water on the Temporal Variation of the Benthic Polychaetous Community Structure in Dangdong Bay (저층수 용존산소가 당동만 저서다모류군집 구조의 시간적 변동에 미친 영향)

  • This study was carried out at 5 sites 11 times over two years to identify the variation of benthic environments and benthic polychaetous community and analyze the benthic healthiness in Dangdong Bay, a small semi-enclosed inner bay of Jinhae Bay. The temperature of bottom water showed the typical temporal fluctuation of a temperate zone and was in the range of 5.94 ~ 23.94℃. The salinity did not change significantly during the study period and was in the range of 32.93 ~ 35.72 psu. The concentration of dissolved oxygen of bottom water fluctuated a great deal and was in the range of 0.31 ~ 10.20 mg/L. The lowest DO value was recorded in July 2015, as 0.31±0.04 mg/L corresponding to the hypoxic water mass. The hypoxic water mass was formed continuously at some sites also in July and August 2016. The mean grain size was in the range of 7.57 ~ 9.81Ø and the average was 8.89±0.20Ø. The surface sediments were mainly composed of fine sediment (mud) above 85%. The mean of TOC was 3.09±0.22% and LOI was 13.30±0.47%, showing very high levels in Korean coastal waters. The concentration of AVS was in the range of 0.33 ~ 1.28 mgS/g-dry. The high values of organic contents and AVS indicated that there had been the serious organic enrichment in Dangdong Bay. The number of species and the density of the benthic polychaetous community in Dangdong Bay were in the range of 2 ~ 38 species and 2 ~ 2,185 ind./㎡ during the study period. The number of species and density were highly sustained in winter and spring, and then decreased gradually with the formation of a hypoxic water mass in summer, and the lowest number of species and density were recorded in autumn. In September and November 2015, the dead zone expanded to almost the whole study area. Dominant polychaetous species were Capitella capitata, Lumbrineris longifolia, Paraprionospio patiens and Sigambra tentaculata, each known as opportunistic species and potential organic pollutant indicator species. In particular, Paraprionospio patiens showed a very high population density of 2,019 ind./㎡ in December 2016. Polychaetous communities at each sampling time were classified into 4 temporal groups according to dominant species in each period by cluster analysis and nMDS. 'Period Group AI' was formed in winter and spring of 2015, dominated by Capitella capitata, 'Period AII' in summer dominated by Lumbrineris longifolia, 'Period B' in autumn with no fauna in the dead zone, and particularly 'Period C' in winter of 2016 dominated by Paraprionospio patiens. As a result of analysis of benthic healthiness, the study area was estimated to be in a Fair~Very Poor condition by AMBI and in a Poor~Very Poor condition by BPI during the study period. Both AMBI and BPI showed that the study area was in a Very Poor condition in September and November 2015, and when the dead zone occurred. In Dongdong Bay, the fact that the formation of a hypoxic water mass occurred in summer and a dead zone in autumn were confirmed. In addition, the dominance of opportunistic and organic pollutant indicator species was also observed clearly. The benthic healthiness indexes such as AMBI and BPI showed that organic enrichment was serious in Dangdong Bay.

  • Hypoxic ischemic encephalopathy(HIE) is a condition that occurs when the entire brain is deprived of an adequate oxygen supply and associated in most cases with oxygen deprivation in the neonate due to birth asphyxia. Children with HIE develop problems such as cerebral palsy, mental retardation, learning difficulties, and other disabilities. We hereby report the case of treating a child patient with HIE whose chief complaint of severe lingual ulceration. A 12-month-old boy referred to our hospital to treat ulcers of ventral surface of tongue. He has been suffering from lingual ulceration due to persistent tongue thrusting. He has bitten his own tongue involuntary. The ulceration was apparently seemed to be not healed because of the constant irritation by lower incisors. So we designed tongue protector to prevent lingual ulcers. The appliance was proved to be effective to reduce tongue trauma by lower incisors and periodic follow-up is needed.

Cobalt Chloride-induced Hypoxia Ameliorates NLRP3-Mediated Caspase-1 Activation in Mixed Glial Cultures

  • Hypoxia has been shown to promote inflammation, including the release of proinflammatory cytokines, but it is poorly investigated how hypoxia directly affects inflammasome signaling pathways. To explore whether hypoxic stress modulates inflammasome activity, we examined the effect of cobalt chloride ($CoCl_2$)-induced hypoxia on caspase-1 activation in primary mixed glial cultures of the neonatal mouse brain. Unexpectedly, hypoxia induced by oxygen-glucose deprivation or $CoCl_2$ treatment failed to activate caspase-1 in microglial BV-2 cells and primary mixed glial cultures. Of particular interest, $CoCl_2$-induced hypoxic condition considerably inhibited NLRP3-dependent caspase-1 activation in mixed glial cells, but not in bone marrow-derived macrophages. $CoCl_2$-mediated inhibition of NLRP3 inflammasome activity was also observed in the isolated brain microglial cells, but $CoCl_2$ did not affect poly dA:dT-triggered AIM2 inflammasome activity in mixed glial cells. Our results collectively demonstrate that $CoCl_2$-induced hypoxia may negatively regulate NLRP3 inflammasome signaling in brain glial cells, but its physiological significance remains to be determined.

Characterization of human cardiac mesenchymal stromal cells and their extracellular vesicles comparing with human bone marrow derived mesenchymal stem cells

  • Cardiac regeneration with adult stem-cell (ASC) therapy is a promising field to address advanced cardiovascular diseases. In addition, extracellular vesicles (EVs) from ASCs have been implicated in acting as paracrine factors to improve cardiac functions in ASC therapy. In our work, we isolated human cardiac mesenchymal stromal cells (h-CMSCs) by means of three-dimensional organ culture (3D culture) during ex vivo expansion of cardiac tissue, to compare the functional efficacy with human bone-marrow derived mesenchymal stem cells (h-BM-MSCs), one of the actively studied ASCs. We characterized the h-CMSCs as CD90low, c-kitnegative, CD105positive phenotype and these cells express NANOG, SOX2, and GATA4. To identify the more effective type of EVs for angiogenesis among the different sources of ASCs, we isolated EVs which were derived from CMSCs with either normoxic or hypoxic condition and BM-MSCs. Our in vitro tube-formation results demonstrated that the angiogenic effects of EVs from hypoxia-treated CMSCs (CMSC-Hpx EVs) were greater than the well-known effects of EVs from BM-MSCs (BM-MSC EVs), and these were even comparable to human vascular endothelial growth factor (hVEGF), a potent angiogenic factor. Therefore, we present here that CD90lowc-kitnegativeCD105positive CMSCs under hypoxic conditions secrete functionally superior EVs for in vitro angiogenesis. Our findings will allow more insights on understanding myocardial repair.

Hypoxia Inducible Factor-1α Directly Induces the Expression of Receptor Activator of Nuclear Factor-κB Ligand in Chondrocytes

  • Receptor activator of nuclear factor-${\kappa}B$ ligand (RANKL) is an osteoblast/stromal cell-derived essential factor for osteoclastogenesis. During endochondral bone formation, hypertrophic chondrocytes calcify cartilage matrix that is subsequently resorbed by osteoclasts in order to be replaced by new bone. Hypoxia-induced upregulation of RANKL expression has been previously demonstrated in an in vitro system using osteoblasts; however, the involved mechanism remains unclear in chondrocytes. In the present study, we investigated whether hypoxia regulates RANKL expression in ATDC5 cells, a murine chondrogenic cell line, and hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) mediates hypoxia-induced RANKL expression by transactivating the RANKL promoter. The expression levels of RANKL mRNA and protein, as well as HIF-$1{\alpha}$ protein, were significantly increased in ATDC5 cells under hypoxic condition. Constitutively active HIF-$1{\alpha}$ alone significantly increased the levels of RANKL expression under normoxic conditions, whereas dominant negative HIF-$1{\alpha}$ reduced hypoxia-induced RANKL expression. HIF-$1{\alpha}$ increased RANKL promoter reporter activity in a HIF-$1{\alpha}$ binding element-dependent manner in ATDC5 cells. Hypoxia-induced RANKL levels were much higher in differentiated ATDC5 cells, as compared to proliferating ATDC5 cells. These results suggested that under hypoxic conditions, HIF-$1{\alpha}$ mediates induction of RANKL expression in chondrocytes; in addition, hypoxia plays a role in osteoclastogenesis during endochondral bone formation, at least in part, through the induction of RANKL expression in hypertrophic chondrocytes.

Effect of hydrogen-rich water on the lactic acid level in metformin-treated diabetic rats under hypoxia

  • The present study aims to investigate the impact of hydrogen-rich water on the lactic acid level in metformin-treated diabetic rats under hypoxia. Thirty Sprague-Dawley rats were randomly divided into five groups, including normal diet group, and diabetes model (DM) group, DM + metformin treatment (DMM) group, DMM + hypoxia treatment (DMMH) group and DMMH + hydrogen-rich water (DMMHR) group. We found that the levels of lactic acid, pyruvate and lactate dehydrogenase were significantly lower in the blood of DMMHR group than DMMH group. Superoxide dismutase and glutathione levels in liver and heart were significantly higher in DMMH group after hydrogen-rich water treatment, while malondialdehyde and oxidized glutathione levels were decreased in DMMHR group when compared with DMMH group, which indicates that hydrogen-rich water could reduce oxidative stress. qPCR analysis demonstrated that that pro-apoptotic genes Bax/Caspase-3 were upregulated in DM group and metformin treatment suppressed their upregulation (DMM group). However, hypoxic condition reversed the effect of metformin on apoptotic gene expression, and hydrogen-rich water showed little effect on these genes under hypoxia. HE staining showed that hydrogen-rich water prevented myocardial fiber damages under hypoxia. In summary, we conclude that hydrogen-rich water could prevent lactate accumulation and reduce oxidant stress in diabetic rat model to prevent hypoxia-induced damages. It could be served as a potential agent for diabetes patients with metformin treatment to prevent lactic acidosis and reduce myocardial damages under hypoxic conditions.

  • Hypoxia indicates the condition of low oxygen levels in tissues. In oncology, hypoxia can induce cancer progression and metastasis, as well as cause resistance to cancer therapies. The detection of hypoxia by using molecular imaging, particularly, positron emission tomography (PET) has been extensively studied due to many advantages. Nitroimidazoles, the moieties that can be trapped in hypoxic tissues due to selective reduction, have been used to design and synthesize of hypoxia-targeting radiopharmaceuticals. This review provides a summary of synthetic routes towards 18F-labeled-nitroimidazole radiotracers for PET imaging of hypoxia.

Effects of epigallocatechin gallate on $CoCl_2-induced$ apoptosis in PC12 cells (PC12 세포에서 $CoCl_2$ 유발 세포자멸사에 대한 epigallocatechin-gallate의 역할)

  • Neuronal apoptotic events, consequently resulting in neuronal cell death, are occurred in hypoxic/ischemic condition. This cell death has been shown to be accompanied with the production of reactive oxygen species (ROS), which can attack cellular components such as nucleic acids, proteins and phospholipid. However, the underlying mechanisms of apoptosis induced in hypoxic/ischemic condition and its treatment methods are unsettled. Cobalt chloride $(CoCl_2)$ has been known to mimic hypoxic condition including the production of ROS. Epigallocatechin gallate (EGCG), a green tea polyphenol, has diverse pharmacologial activities in cell growth and death. This study was aimed to investigate the apoptotic mechanism by $CoCL_2$ and effects of EGCG on $CoCl_2-induced$ apoptosis in PC12 cells. Administration of $CoCl_2$ decreased cell survival in dose- and time-dependent manners and induced genomic DNA fragmentation. Treatment with $100{\mu}M$ EGCG for 30 min before PC12 cells were exposed to $150{\mu}M$ $CoCl_2$, being resulted in the cell viability and DNA fragmentation being rescued. $CoCl_2$ caused morphologic changes such as cell swelling and condensed nuclei whereas EGCG attenuated morphologic changes by $CoCl_2$. EGCG suppressed the apoptotic peak and a loss of ${\Delta}{\psi}_m$ induced by $CoCl_2$. $CoCl_2$ decreased Bcl-2 expression but Bax expression was not changed in $CoCl_2$- treated cells. EGCG attenuated the Bcl-2 underexpression by $CoCl_2$. $CoCl_2$ augumented the cytochrome c release from mitochondria into cytoplasm and increased caspase-8, -9 and caspase-3 activity a marker of the apoptotic executing stage. EGCG ameliorated the incruement in caspase-8, -9 and -3 activity, and cytochrome c release by $CoCl_2$ NAC (N-acetyl-cysteine), a scavenger of ROS, attenuated $CoCl_2-induced$ apoptosis in consistent with those of EGCG. These results suggest that $CoCl_2$ induces apoptotic cell death through both mitochondria- and death receptor-dependent pathway and EGCG has neuroprotective effects against $CoCl_2-induced$ apoptosis in PC12 cells.

PPARα-Target Gene Expression Requires TIS21/BTG2 Gene in Liver of the C57BL/6 Mice under Fasting Condition

  • The $TIS21^{/BTG2/PC3}$ gene belongs to the antiproliferative gene (APRO) family and exhibits tumor suppressive activity. However, here we report that TIS21 controls lipid metabolism, rather than cell proliferation, under fasting condition. Using microarray analysis, whole gene expression changes were investigated in liver of TIS21 knockout (TIS21-KO) mice after 20 h fasting and compared with wild type (WT). Peroxisome proliferator-activated receptor alpha ($PPAR{\alpha}$) target gene expression was almost absent in contrast to increased lipid synthesis in the TIS21-KO mice compared to WT mice. Immunohistochemistry with hematoxylin and eosin staining revealed that lipid deposition was focal in the TIS21-KO liver as opposed to the diffuse and homogeneous pattern in the WT liver after 24 h starvation. In addition, cathepsin E expression was over 10 times higher in the TIS21-KO liver than that in the WT, as opposed to the significant reduction of thioltransferase in both adult and fetal livers. At present, we cannot account for the role of cathepsin E. However, downregulation of glutaredoxin 2 thioltransferase expression might affect hypoxic damage in the TIS21-KO liver. We suggest that the $TIS21^{/BTG2}$ gene might be essential to maintain energy metabolism and reducing power in the liver under fasting condition.

SOD and Inorganic Nutrient Fluxes from Sediment in the Downstream of the Nagdong River (낙동강 하류 수계에서 저질퇴적층의 SOD와 영양염 용출)

  • Nutrient fluxes and sediment oxygen demands (SOD) were measured with intact sediment cores collected from three stations in the downstream of Nagdong River. The sediments were subjected to controlled oxic and hypoxic conditions and temperature gradients (from $10^{\circ}C$ to $30^{\circ}C$) of the overlying waters in laboratory batch system. The effect of temperature and labile layer thickness of the sediment on SOD were examined. $PO_4\;^{3-}$ and $NH_4\;^+$ fluxes were elevated above $20^{\circ}C$ and large mobilities were observed when they were coupled with a hypoxic and high-temperature condition. In the well oxygenated conditions, $PO_4\;^{3-}$ fluxes were negative or negligible but $NH_4\;^+$ fluxes ranged from 1.3 mg N $m^{-2}\;hr^{-1}$ to 2.3 $m^{-2}\;hr^{-1}$. Temperature quotients($Q_{10}$) of $PO_4\;^{3-}$ fluxes were 3.7 ${\sim}$ 7.3 ranges to have the most high values. $PO_4\;^{3-}$ and $NH_4\;^+$ fluxes had the logarithmic increase with temperature, while $NO_3\;^-$ was negatively absorbed to the sediment and linearly correlated with the temperature. $SiO_2$ fluxes showed no difference among oxic and hypoxic conditions and sediment texture. The nutrient fluxes would be closely correlated with pore water chemistry of sediments and activated by the top sediment layer composition such as labile organic matters or algal detritus. The ecological implications of the nutrient fluxes were discussed in terms of sources and sinks of nutrients coupled to algal productions in the Nagdong River.
