Crlz1 promoter의 상위에 인접한 DNase I hypersensitive site 8의 enhancer 기능 (DNase I Hypersensitive Site 8 as an Enhancer is Mapped in the Upstream Vicinity of the Crlz1 Promoter)

  • Crlz1 유전자는 B 세포 분화 과정 중 pre-B 세포 분화 단계에서 특이적으로 발현됨이 밝혀져 있다. Crlz1 유전자의 발현과 연관되는 3개의 DNase I hypersensitive site (HSS8, 9, 10)가 Crlz1 유전자의 전사 시작점으로부터 바로 위쪽의 chromatin 상에서 발견되었고, 그 중에서 HSS9/10은 Crlz1 promoter 지역에 해당하며 매우 높은 전사 활성을 가지는 것으로 이미 보고 되었다. 본 연구에서는 앞의 연구에서 더 나아가 HSS8이 Crlz1 promoter의 전사활성을 약 2 배 증가시키는 enhancer의 역할을 하고 있는 현상을 밝혔으며, 또한 HSS8의 위치가 Crlz1 유전자의 전사 시작점을 기준으로 하였을 때 -1055와 -1159 사이, 즉 104 bp 길이의 chromatin DNA 상에 존재하며, 이러한 HSS8의 위치는 luciferase reporter의 활성 측정으로 비교 분석되어진 deletion construct들의 전사 활성에 대한 결과와도 잘 부합되었다.

양독백호탕(陽毒白虎湯)이 이식(移植) 피부(皮膚) 거부반응(拒否反應) 지연(遲延)에 미치는 영향(影響) (Inhibitory effects of Yangdokbackhotang on the Immune Hypersensitive Reaction in skin transplant mice)

  • Objective: This study was carried out to evaluate the inhibitory effect on immme hypersensitive reaction by Yangdokbackhotang extract. methods: To study, We divided 4 weeks old rats into three groups of 20, which are the constrasive group with nothing treated, the skin transplated only, and the Yangdokbackhotang extract fed before skin transplantation. And did the experiment. Results: 1. The ear swelling in mice treated with YB noticeably decreased than only DNCB treated mice. 2. As shown in Table3, also results in a significant decrease of YB group than ST group in the number of positive cells of $NF-{\kappa}B$, COX-2, IL-12B, and ICAM-1. From the above results, it could be concluded that Yangdokbackhotang showed benificial immunosupressive effect on the hypersensitive reaction.

The Hypersensitive Response. A Cell Death during Disease Resistance

  • Host cell death occurs during many, but not all, interactions between plants and the pathogens that infect them. This cell death can be associated with disease resistance or susceptibility, depending on the nature of the pathogen. The most well-known cell death response in plants is the hypersensitive response (HR) associated with a resistance response. HR is commonly regulated by direct or indirect interactions between avirulence proteins from pathogen and resistance proteins from plant and it can be the result of multiple signaling pathways. Ion fluxes and the generation of reactive oxygen species commonly precede cell death, but a direct involvement of the latter seems to vary with the plant-pathogen combination. Exciting advances have been made in the identification of cellular protective components and cell death suppressors that might operate in HR. In this review, recent progress in the mechanisms by which plant programmed cell death (PCD) occurs during disease resistance will be discussed.

Systemic Immediate Hypersensitive Reactions after Treatment with Sweet Bee Venom: A Case Report

  • Objectives: A previous study showed that bee venom (BV) could cause anaphylaxis or other hypersensitivity reactions. Although hypersensitivity reactions due to sweet bee venom (SBV) have been reported, SBV has been reported to be associated with significantly reduced sensitization compared to BV. Although no systemic immediate hypersensitive response accompanied by abnormal vital signs has been reported with respect to SBV, we report a systemic immediate hypersensitive response that we experienced while trying to use SBV clinically. Methods: The patient had undergone BV treatment several times at other Oriental medicine clinics and had experienced no adverse reactions. She came to acupuncture & moxibustion department at Semyung university hospital of Oriental medicine (Je-cheon, Korea) complaining of facial hypoesthesia and was treated using SBV injections, her first SBV treatment. SBV, 0.05 cc, was injected at each of 8 acupoints, for a total of 0.40 cc: Jichang (ST4), Daeyeong (ST5), Hyeopgeo (ST6), Hagwan (ST7), Yepung (TE17), Imun (TE21), Cheonghoe (GB2), and Gwallyeo (SI18). Results: The patient showed systemic immediate hypersensitive reactions. The main symptoms were abdominal pain, nausea and perspiration, but common symptoms associated with hypersensitivity, such as edema, were mild. Abdominal pain was the most long-lasting symptom and was accompanied by nausea. Her body temperature decreased due to sweating. Her diastolic blood pressure could not be measured on three occasions. She remained alert, though the symptoms persisted. The following treatments were conducted in sequence; intramuscular epinephrine, 1 mg/mL, injection, intramuscular dexamethasone, 5 mg/mL, injection, intramuscular buscopan, 20 mg/mL, injection, oxygen ($O_2$) inhalation therapy, 1 L/minutes, via a nasal prong, and intravascular injection of normal saline, 1 L. After 12 hours of treatment, the symptoms had completely disappeared. Conclusion: This case shows that the use of SBV does not completely eliminate the possibility of hypersensitivity and that patients who received BV treatment before may also be sensitized to SBV. Thus, a skin test should be given prior to using SBV.

과민성치근에 대한 Argon laser조사의 치료효과에 관한 실험적 연구 (AN IN VITRO STUDY OF THE THERAPEUTIC EFFECT OF ARGON LASER IRRADIATION ON HYPERSENSITIVE ROOTS)

  • In order to evaluate the therapeutic effect of argon laser irradiation on hypersensitive root, 60 mandibular premolars that had been extracted due to periodontal disease were treated by thorough scaling, root planing and root conditioning with tetracycline HCI(100mg/ml, 5 min.) for induction of hypersensitive root. Within middle one third of root mesial surface(30) or distal surface(30) was randomly irradiated by 0.5W-, 0.75W- and 1.0W-power for 2 minutes argon laser(HGM Inc., USA) with or without topical application of 1.23% acidulated phosphate fluoride gel(Pascal, USA). After all teeth were maintained in 2% methylene blue solution for 48 hours and cross-sectioned through irradiated surface of dried roots, methylene blue stained area was compared between irradiated side and nonirradiated side. Following results were obtained : 1. Methylene blue stained area was significantly less in the laser irradiated group than control group(p<0.05). 2. There was no significant difference in methylene blue stained area between 0.5W-power and 0.75W-power group(p>0.1) and between 0.75W-power and 1.0W-power group(p>0.1), but there was significant difference between 0.5W-power and 1.0W-power group(p<0.05). 3. There was no significant difference in methylene blue stained area between two groups with and without topical application of 0.12% acidulated phosphate fluoride gel(p>0.1). The results suggest that argon laser irradiation on hypersensitive root may reduce the hypersensitivity by obstruction of dentinal tubules.

Similarities of Tobacco Mosaic Virus-Induced Hypersensitive Cell Death and Copper-Induced Abiotic Cell Death in Tobacco

  • Hypersensitive cell death of plants during incompatible plant-pathogen interactions is one of the efficient defense mechanisms of plants against pathogen infections. For better understanding of the molecular mechanisms involved in the plant hypersensitive response (HR), TMV-induced biotic plant cell death and CuSO4-induced abiotic plant cell death were compared in terms of expression patterns of ten different defense-related genes as molecular markers. The genes include five pathogenesis-related protein genes, two plant secondary metabolite-associated genes, two oxidative stress-related genes and one wound-inducible gene isolated from tobacco. Northern blot analyses revealed that a same set of defense-related genes was induced during both biotic and abiotic cell death but with different time and magnitude. The expression of defense-related genes in tobacco plants was temporarily coincided with the time of cell death. However, when suspension cell cultures was used to monitor the expression of defense-related genes, different patterns of the gene expression were detected. This result implies that three are common and, in addition, also different branches of signaling pathways leading to the induced expression of defense-related genes in tobacco during the pathogen- and heavy metal-induced cell death.

HpaXm from Xanthomonas citri subsp. malvacearum is a Novel Harpin with Two Heptads for Hypersensitive Response

  • A novel harpin-like protein, HpaXm, was described from cotton leaf blight bacteria, Xanthomonas citri subsp. malvacearum. The hpaXm was found to be localized between hrp2 and hrcC. A phylogenetic analysis of the complete amino acid sequence or solely the 13 highly conserved residues $H_2N$-SEKQLDQLLTQLI-COOH in the N-terminal $\alpha$-helix indicates that HpaXm is evolutionarily closer to HpaGXag and HpaXac than to Hpa1Xoo and Hpa1Xoc. A synthesized peptide containing two heptads, 39-LDQLLTQLIMALLQ-52, from the N-terminal a-helical region of HpaXm displayed comparable activity in inducing a hypersensitive response, but two other synthesized derivatives, $HpaXm{\Delta}T44C$ and $HpaXm{\Delta}M48Q$, showed reduced HR-triggering activity. The data from a GST trap test revealed that HpaXm was released into the extracellular medium, hpaXm mutant deficient for the leader peptide (1-MNSLNTQIGANSSFL-15) was unable to be secreted outside cells but still induced HR in tobacco leaves.

A Marine Bacterium with Animal-Pathogen-Like Type III Secretion Elicits the Nonhost Hypersensitive Response in a Land Plant

  • Active plant immune response involving programmed cell death called the hypersensitive response (HR) is elicited by microbial effectors delivered through the type III secretion system (T3SS). The marine bacterium Hahella chejuensis contains two T3SSs that are similar to those of animal pathogens, but it was able to elicit HR-like cell death in the land plant Nicotiana benthamiana. The cell death was comparable with the transcriptional patterns of H. chejuensis T3SS-1 genes, was mediated by SGT1, a general regulator of plant resistance, and was suppressed by AvrPto1, a type III-secreted effector of a plant pathogen that inhibits HR. Thus, type III-secreted effectors of a marine bacterium are capable of inducing the nonhost HR in a land plant it has never encountered before. This suggests that plants may have evolved to cope with a potential threat posed by alien pathogen effectors. Our work documents an exceptional case of nonhost HR and provides an expanded perspective for studying plant nonhost resistance.

Light- and Relative Humidity-Regulated Hypersensitive Cell Death and Plant Immunity in Chinese Cabbage Leaves by a Non-adapted Bacteria Xanthomonas campestris pv. vesicatoria

  • Inoculation of Chinese cabbage leaves with high titer (107 cfu/ml) of the non-adapted bacteria Xanthomonas campestris pv. vesicatoria (Xcv) strain Bv5-4a.1 triggered rapid leaf tissue collapses and hypersensitive cell death (HCD) at 24 h. Electrolyte leakage and lipid peroxidation markedly increased in the Xcv-inoculated leaves. Defence-related gene expressions (BrPR1, BrPR4, BrChi1, BrGST1 and BrAPX1) were preferentially activated in the Xcv-inoculated leaves. The Xcv-triggered HCD was attenuated by continuous light but accelerated by a dark environment, and the prolonged high relative humidity also alleviated the HCD. Constant dark and increased relative humidity provided favorable conditions for the Xcv bacterial growth in the leaves. Pretreated fluridone (biosynthetic inhibitor of endogenous abscisic acid [ABA]) increased the HCD in the Xcv-inoculated leaves, but exogenous ABA attenuated the HCD. The pretreated ABA also reduced the Xcv bacterial growth in the leaves. These results highlight that the onset of HCD in Chinese cabbage leaves initiated by non-adapted pathogen Xcv Bv5-4a.1 and in planta bacterial growth was differently modulated by internal and external conditional changes.

Ubiquitous Technology를 이용한 주의력결핍 과잉행동장애 증상 중 과잉활동증 아동 예진 서비스 (Preliminary ADHD Symptom of the Hyperactivity Diagnosis Service Using Ubiquitous Technology)

  • The purpose of this study was to detect early children with hyperactivity which is one of the symptoms of Attention Deficit-Hyperactivity Disorder (ADHD). This study used two methods: K-CBCL and observation of children's behavior. K-CBCL was done online by parents at home. For observation of children's behavior, the school asked children to wear a 3-axis accelerometer on their wrists. The data from K-CBCL and 3-axis accelerometer were analyzed and clustered to separate hypersensitive children from ordinary children. This experiment confirmed that 3-axis accelerometer which is one of Ubiquitous techniques and the K-CBCL questionnaire were helpful for detection of hypersensitive children.
