Clinical Comparison between Paclitaxel Liposome (Lipusu®) and Paclitaxel for Treatment of Patients with Metastatic Gastric Cancer

  • Aim: To compare the efficacy and safety of paclitaxel liposome (Lipusu$^{(R)}$) with paclitaxel in combination with tegafur and oxaliplatin in treating patients with advanced gastric cancer. Materials and Methods: Patients with advanced gastric cancer receiving chemotherapy were retrospectively collected, and divided into two groups. Patients in group A received paclitaxel liposomes at a dose of 135 $mg/m^2$ on day 1 of each cycle, and patients in group B were given paclitaxel at the same dose with the same timing. All patients received tegafur at a dose of 500 $mg/m^2$ on days 1-5, and oxaliplatin at a dose of 80-100 $mg/m^2$ on day 1 for 2 cycles (each cycle was 21 d in total). Results: Fifty-eight patients could be evaluated for efficacy. The overall response rate was 47% in group A (14/30), and 46% in group B (13/28). Disease control rate was 73% in group A (22/30), and 71% in group B (20/28) (P>0.05). No significant differences were detected in hematologic and neurologic toxicities between the two groups (P>0.05). However, nausea, vomiting and hypersensitive reactions were significantly lower in group A than in group B (P<0.05). Conclusion: Paclitaxel liposomes are as effective as paclitaxel when combined with tegafur and oxaliplation in treating patients with advanced gastric cancer, but adverse reactions with paclitaxel liposomes are less common.

Poly(ADP-ribose) protects vascular smooth muscle cells from oxidative DNA damage

  • Vascular smooth muscle cells (VSMCs) undergo death during atherosclerosis, a widespread cardiovascular disease. Recent studies suggest that oxidative damage occurs in VSMCs and induces atherosclerosis. Here, we analyzed oxidative damage repair in VSMCs and found that VSMCs are hypersensitive to oxidative damage. Further analysis showed that oxidative damage repair in VSMCs is suppressed by a low level of poly (ADP-ribosyl)ation (PARylation), a key post-translational modification in oxidative damage repair. The low level of PARylation is not caused by the lack of PARP-1, the major poly(ADP-ribose) polymerase activated by oxidative damage. Instead, the expression of poly(ADP-ribose) glycohydrolase, PARG, the enzyme hydrolyzing poly(ADP-ribose), is significantly higher in VSMCs than that in the control cells. Using PARG inhibitor to suppress PARG activity facilitates oxidative damage-induced PARylation as well as DNA damage repair. Thus, our study demonstrates a novel molecular mechanism for oxidative damage-induced VSMCs death. This study also identifies the use of PARG inhibitors as a potential treatment for atherosclerosis. Three transcripts of EDS1-like genes respond differently to Vitis flexuosa infection

  • Enhanced disease susceptibility1 (EDS1) is a regulator of basal defense responses required for resistance mediated by TIR-NBS-LRR containing R proteins. We identified three transcripts of EDS1-like genes encompassing diverse/separate expression patterns, based on the transcriptome analysis by Next Generation Sequencing (NGS) of V. flexuosa inoculated with Elsinoe ampelina. These genes were designated VfEDL1 (Vitis flexuosa Enhanced Disease Susceptibility1-like1), VfEDL2 and VfEDL3, and contained 2464, 1719 and 1599 bp, with 1791, 1227 and 1599 bp open reading frames (ORFs), encoding proteins of 596, 408 and 532 amino acids, respectively. The predicted amino acid sequences of all three genes showed the L-family lipase-like domain (class 3 lipase domain), and exhibited a potential lipase catalytic triad, aspartic acid, histidine and serine in the conserved G-X-S-X-G. All three VfEDL genes were upregulated at 1 hpi against the bacterial and fungal pathogens Rizhobiumvitis and E. ampelina, respectively, except VfEDL1, which was downregulated against E. ampelina at all time points. Against E. ampelina, VfEDL2 and VfEDL3 showed downregulated expression at later time points. When evaluated against R. vitis, VfEDL1 showed downregulated expression at all time points after 1 hpi, while VfEDL3 showed upregulation up to 24 hpi. Based on the expression response, all three genes may be involved in plant resistant responses against R. vitis, and VfEDL2 and VfEDL3 show additional resistant responses against E. ampelina infection.

Isolation and Characterization of Pathogen-Inducible Putative Zinc Finger DNA Binding Protein from Hot Pepper Capsicum annuum L.

  • To better understand plant defense responses against pathogen attack, we identified the transcription factor-encoding genes in the hot pepper Capsicum annuum that show altered expression patterns during the hypersensitive response raised by challenge with bacterial pathogens. One of these genes, Ca1244, was characterized further. This gene encodes a plant-specific Type IIIA - zinc finger protein that contains two Cys$_2$His$_2$zinc fingers. Ca1244 expression is rapidly and specifically induced when pepper plants are challenged with bacterial pathogens to which they are resistant. In contrast, challenge with a pathogen to which the plants are susceptible only generates weak Ca1244 expression. Ca1244 expression is also strongly induced in pepper leaves by the exogenous application of ethephon, an ethylene releasing compound. Whereas, salicylic acid and methyl jasmonate had moderate effects. Pepper protoplasts expressing a Ca1244-smGFP fusion protein showed Ca1244 localizes in the nucleus. Transgenic tobacco plants overexpressing Ca1244 driven by the CaMV 355 promoter show increased resistance to challenge with a tobacco-specific bacterial pathogen. These plants also showed constitutive upregulation of the expression of multiple defense-related genes. These observations provide the first evidence that an Type IIIA - zinc finger protein, Ca1244, plays a crucial role in the activation of the pathogen defense response in plants.

알러지성 비염 병태 모델에 대한 방풍통성산(防風通聖散)의 항알러지 효과 (The Effect of Bangpungtongsung-San on Model of Allergic Rhinitis)

  • Background and Objective : Allergic rhinitis is a IgE-mediated hypersensitive reaction at nasal mucosa. In oriental medicine, Bangpungtongsung-San is clinically widely used for the treatment of the allergic rhinitis. The objective of this study is to investigate the effects of Bangpungtongsung-San on allergic rhinitis experimentally. Material and Methods : BALB/c mouse were divided into there groups: intact, control, experimental groups. Control and experimental group were induced allergic rhinitis by Ovalbumin as· the method of Levin and Vaz. Experimental group was orally administered the Bangpungtongsung-San for 28days. Total IgE, Interleukin-4, Interleukin-5 and $Interferon-{\gamma}$ were measured at three groups. The statistical significance was examined by Independent Samples T test. Results : 1. The experimental group shows increase of $IFN-{\gamma}$ by 17% compared with control group but there was no statistical significance. 2. The experimental group shows increase of IL-4 and IL-5 compared with control group but there was no statistical significance. 3. The experimental group shows increase of Total IgE and diminution of OVA-specific IgE by 32% compared with control group but there was no statistical significance. Conclusion : The effect of Bangpungtongsung-San on allergic rhinitis is not immunological but seems to be anti-imflammatory and anti-stress effect.

치주질환 치료후 미세 결정형 수산화 인회석 함유치약의 지각과민 억제효과에 관한 임상적 연구 (CLINICAL EVALUATION OF MICROCRYSTALLINE HYDROXYAPATITE CONTAINING TOOTHPASTE IN THE CONTROL OF DENTIN HYPERSENSITIVITY AFTER PERIODONTAL TREATMENT)

  • The purpose of this study was to observe the control effect of hypersensitivity after periodontal treatment in the 19% microcrystalline hydroxyapatite containing toothpaste for the subject of 85 persons of both sexes, who complained hypersensitivity. At 2 weeks and 4 weeks after periodontal treatment, comparison of control effect was performed between the 19% microcrystalline hydroxyapatite containing toothpaste group and control group. The result were as follows, 1. The main causes of dentin hypersensitivity are the root exposure with gingival recession and cervical abrasion. 2. The occurance rate of hypersensitive tooth in the upper jaw was higher than that of the lower jaw, and more or less, the molar area showed more occlurance of hypersensitivity than the premolar and incisor area in both jaw. 3. Patients showed very sensitive response to the thermal stimulus, especially cold stimulus. 4. Exellent control effect of hypersensitivity in 19% microcrystalline hydroxyapatite containing toothpaste group showed 83.02% at 2weeks, 92.45% at 4weeks and these values were higher than the control group. In conclusion, we find that 19% microcrystalline hydroxyapatite containing toothpaste have the control effect of hypersensitivity and the proper toothbrushing method is the key in attaining more effectiveness of the toothpaste.

Characterization of a Salicylic Acid- and Pathogen-induced Lipase-like Gene in Chinese Cabbage

  • A cDNA clone for a salicylic acid-induced gene in Chinese cabbage (Brassica rapa subsp. pekinensis) was isolated and characterized. The cabbage gene, designated Br-sil1 (for $\underline{B}$rassica $\underline{r}$apa $\underline{s}$alicylate-$\underline{i}$nduced $\underline{l}$lipase-like 1 gene), encodes a putative lipase that has the family II lipase motif GDSxxDxG around the active site serine. A database search showed that plant genomes have a large number of genes that contain the family II lipase motif. The lipase-like proteins include a myrosinase-associated protein, an anther-specific proline-rich protein APG, a pollen coat protein EXL, and an early nodule-specific protein. The Br-sil1 gene is strongly induced by salicylic acid and a non-host pathogen, Pseudomonas syringae pv. tomato, that elicits a hypersensitive response in Chinese cabbage. Treatment of the cabbage leaves with BTH, methyl jasmonate, or ethephon showed that the Br-sil1 gene expression is induced by BTH, but not by methyl jasmonate or ethylene. This indicates that the cabbage gene is activated via a salicylic acid-dependent signaling pathway. An examination of the tissue-specific expression revealed that the induction of the Br-sil1 gene expression by BTH occurs in leaves and stems, but not in roots and flowers. Without the BTH treatment, however, the Br-sil1 gene is not expressed in any of the tissues that were examined.

Molecular Characterization of a thiJ-like Gene in Chinese Cabbage

  • A cDNA clone for a salicylic acid-induced gene in Chinese cabbage (Brassica rapa subsp. pekinensis) was isolated and characterized. The cabbage gene encoding a protein of 392 amino acids contained a tandem array of two thiJ-like sequences. ThiJ is a thiamin biosynthesis enzyme that catalyzes the phosphorylation of hydroxymethylpyrimidine (HMP) to HMP monophosphate. Although the cabbage gene shows a similarity to bacterial thiJ genes, it also shares a similarity with the human DJ-1, a multifunctional protein that is involved in transcription regulation, male fertility, and parkinsonism. The cabbage thiJ-like gene is strongly induced by salicylic acid and a nonhost pathogen, Pseudomonas syringae pv. tomato, which elicits a hypersensitive response in Chinese cabbage. Treatment of the cabbage leaves with BTH, methyl jasmonate, or ethephon showed that the cabbage thiJ-like gene expression is also strongly induced by BTH, but not by methyl jasmonate or ethylene. This indicates that the cabbage gene is activated via a salicylic acid-dependent signaling pathway. Examination of the tissue-specific expression revealed that the induction of the cabbage gene expression by BTH occurs in the leaf, stem, and floral tissues but not in the root.

  • 분생포자는 발아관의 수에 따라 포자내에 격막이 형성된다. 저항성 품종(금장 2호, 홍일품)과 감수성 품종(하늘초, 새고추) 간의 부착기 형성율은 큰 차이가 없었고 부착기의 모양은 엔형, 타엔형 및 성장체 등 다양하며 접종 12시간에서 그 형성율은 최고에 달했다. 접종 24시간에 저항성 품종의 잎과 열매에서는 과민성인 작은 갈색병반이 관찰되면서 표피하 조직세포내에 심한 원형질분리가 일어났으나 감수성 품종에서는 시간의 경과에 따라 큰 병반으로 급속히 진전 확대되었고 시간이 경과함에 따라 균사가 하부조직으로 확산되었고 분생자시 및 강모의 출현은 접종 96시간 후에 관찰되었다. 감염종자의 종피는 심하게 붕괴되었을 뿐만 아니라 침입균사도 관찰되었고 배유와 배는 심한 원형질분리를 일으켰다.

Differential Subcellular Responses in Resistance Soybeans Infected with Soybean Cyst Nematode Races

  • Early nematode development and subcellular responses in resistant soybean lines PI 88788 and PI 437654 infected with races 3 (R3) and 14 (R14) of soybean cyst nematode (SCN), Heterodera glycines Ichinohe, were compared. SCN R14 nematodes penetrated and developed significantly more than R3 at 5-6 days after inoculation. Both races also penetrated and developed more in PI 88788 than in PI 437654. Syncytia, characterized by cell wall dissolution and cellular hypertrophy, were developed more in PI 88788 than in PI 437654 and more by R14 than R3, for which less necrotic responses occurred in the former than the latter. This suggests that the latter two may be more resistant and less virulent than the former two, respectively. A common structural feature found in each of PI 437654 and PI 88788 in relation to SCN-resistance was the formation of prominent cell wall appositions and nuclear degeneration prior to cytoplasmic degradation in syncytial cells, respectively. Necrosis and cell wall apposition are types of hypersensitive responses occurring at early stages of the nematode infection so that these structural modifications indicate the inhibition of initial syncytial development related to the early nematode development. As soybean cultivars and lines with identical or similar genotypes have the same types of structural features related to SCN-resistance, the structural modifications induced by SCN infection may result from the expression of inheritable resistance genes, of which the information can be used for breeding soybean cultivars and lines specifically resistant to SCN races.
