Effects of Protein Hydrolysates on Blood and Liver Lipids in Rats fed Fat-enriched Diet (고지방식을 섭취한 흰쥐의 체내지질함량에 대한 단백질 가수분해물의 섭취 효과)

  • The experiments were performed to investigate the effects of protein and protein hydrolysates on lipid metabolism in the hyperlipidemic/hypercholesterolemic rat model induced by feeding fat-enriched diet. In Except 1 male rats were fed four semi-purified high fat and cholesterol diets that contained different nitrogen source, casein(C), casein hydrolysate(CH), corn gluten(G) and corn gluten hydrolysate(GH), for 6 weeks. In Expt. 2 rats were fed high fat and cholesterol diet for 4 weeks to induce hyperlipidemia and hypercholesterolemia. Then the rats were divided into 4 groups and were fed the four kinds of above experimental diets for 4 weeks consecutively. The contents of total lipid , cholesterol and triglyceride in blood, liver and feces were determined. Serum lipid concentrations of CH, G and GH were significantly lower than that of C. Serum cholesterol concentrations of hydrolysate groups(CH and GH) were significantly lower than those of intact protein groups(C and G). Serum HDL -cholesterol concentration tended to increase by hydrolysate intake. The total lipid, cholesterol contents in liver showed similarity results as above. Fecal lipid excretions of CH, G, and GH groups were significantly higher than that of C group. These results indicate that hypolipidemic and /or hypocholesterolemic effect of corn gluten or protein hydrolysates were detected in the process of inducing hyperlipidemia by high-fat and cholesterol diet or after inducing hyperlipidemia.

The Effects of Gamijihwang-tang(GJT) on Hyperlipidemia in Rats (가미지황탕(加味地黃湯)이 고지혈증(高脂血症) 관련(關聯) 인자(因子)에 미치는 영향)

  • Objective : Gamijihwang-tang(GJT) has been used as a therapeutic agent for hyperlipidemia in oriental medicine for several years. This study was performed to investigate the effects of GJT on hyperlipidemia in rats using diverse biological methods. Method : Hyperlipidemia was induced by a hyper-lipidemic diet fed for 4 weeks. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, and glucose were measured in the serum after an oral administration of GJT. Lipid peroxidase, SOD, catalase, ACAT, and HMG-CoA were measured in liver after oral administration of GJT. Result: 1. GJT showed safety against cytotoxicity and toxicity in liver. 2. GJT significantly decreased rat's body and liver weight. 3. GJT significantly decreased serum total cholesterol and LDL-cholesterol, but increased serum HDL-cholesterol. 4. GJT significantly decreased serum triglyceride and glucose. 5. GJT significantly decreased lipid peroxidation and increased SOD and catalase in liver. 6. GJT significantly decreased ACAT and HMG-CoA reductase of cholesterol manifestation in liver. Conclusions : These results suggest that GJT might be effective in treatment and prevention of hyperlipidemia.

The Effect of Herbal Formula KH-204 on Erectile Dysfunction in Hyperlipidemic Rat (고지혈증 흰쥐에서 생약제제 KH-204의 발기부전 치료연구)

  • This study was designed to investigate the effects KH-204 on erectile dysfunction in hyperlipidemic rat. KH-204 has been evaluated antihyperlipidemic and antierectile dysfunction effects on experimental hyperlipidemic rats induced by high fat diet. After oral administration of the water extract KH-204 (50, 100, 200, 300 mg/kg) to hyperlipidemic rats for 8 weeks, the variables including body weight, total cholesterol, HDL and LDL levels in serum, the expression of eNOS and nNOS in penis were measured. And erectile function was determined by measurement of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve. Oral administration of KH-204 significantly inhibited the increases of serum total cholesterol and LDL-cholesterol levels and the decreased of serum HDL-cholesterol levels in hyperlipidemic rats induced by high fat diet. The penile expression level of the two enzyme (eNOS, nNOS) were increased significantly after oral administration of the KH-204 50 mg/kg. Erectile function after 10 volts stimulation was significantly decreased in the hyperlipidemic rat compared with the normal rat, but increased in KH-204 group compared with hyperlipidemic group. These results suggest that KH-204 is effective for erectile dysfunction in hyperlipidemia.

Inhibitory Effect of Euscaphic Acid and Tormentic Acid from the Roots of Rosa rugosa on High Fat Diet-Induced Obesity in the Rat (고지방 식이로 유도된 비만 흰쥐에서 해당근에서 분리된 Euscaphic Acid 및 Tormentic Acid의 효과)

  • The roots of Rosa rugosa have been used to treat diabetes mellitus in the folkloric society of Korea. To demonstrate the active component for the rat obesity induced by high fat diet for 6 weeks, the phytochemical fractionation and the pharmacological activity test were performed on this crude drug. It was shown that the methanolic extract and its EtOAc fraction inhibited the weight increase of the rat body, abdominal fat pad and hyperlipidemia at 200 mg/kg dose. Further, the triterpenoids, euscaphic acid and tormentic acid, isolated from R. rugosa roots were active at 30 mg/kg in the same assay. The two components shifted serum total-, HDL, and LDL-cholesterol levels toward the values of the unteated group, suggesting that the active compounds has hypolipidemic effects. The rats fad euscaphic acid and tormentic acid also reduced thiobarbituric acid-reactive substance (TBARS) and hydroxyl radical in the rat blood and increased superoxide dismutase activity compared to the control. TBARS values and carbonyl contest of the hepatic protein were reduced by treatment with the two triterpenoids. Antioxidative enzyme (SOD, glutathione peroxidase, and catalase) activities in hepatic were increased by treatment of rats with the triterpenoids, which suggests that triterpenoids inhibited the reduction of hepatic antioxidative activity caused by high fat diet. Taken together, these results support that euscaphic acid and tormentic acid improve a high fat diet-induced hyperlipidemia via the activation of antioxidative mechanism.

Lipid-lowering and Antioxidant Effects of Curcuma Radix in Poloxamer 407-induced Hyperlipidemia Model Rat Models

  • Objectives : This study investigated the hypolipidemic and antioxidant effects of Curcuma radix using a rat model induced by poloxamer 407 injection. Methods : Serum lipid parameters and oxidative stress-associated biomarkers were determined. Additionally, hepatic cholesterol and triglyceride as well as lipid metabolism-associated gene expressions were observed in hepatic tissue. Results : 1. Curcuma radix ameliorated elevation of serum cholesterol, triglyceride, LDL-cholesterol, MDA, hepatic cholesterol level, and reduction of serum TAC, SOD, GSH, GSH-reductase level. 2. Curcuma radix augmented up-regulated ACAT gene expression. 3. Curcuma radix almost completely ameliorated down-regulated CYP-7A1 but up-regulated HMG-CoA gene expression. Conclusions : The hypolipidemic and antioxidant properties of Curcuma radix were evidenced. This study provides a scientific basis for the clinical application of Curcuma radix and development of hypolipidemics using this herb in the future.

Hypolipidemic and Antioxidative Effects of Bangpoongtongsungsankamibang on Diet-Induced Obesity Rats (비만유도 흰쥐에 대한 방풍통성산가미방 (防風通聖散加味方)의 항고지혈 효과 및 항산화 효과)

  • Obesity is associated with a number of pathological disorders such as non-insulin-dependent diabetes, hypertension, hyperlipidemia, and cardiovascular diseases. Bangpoongtongsungsankamibang (BTSK) has been widely used in the oriental medicine for the treatment of several diseases associated with inflammatory abnormalities in cardiovascular and nervous system. The BTSK is the modified prescription of Bangpoongtongsungsankamibang in which sea tangle (Laminaria japonica) were added. This study was carried out to detemine the anti-obestic effects of BTSK. Pretreatment with the BTSK at daily dose of 100 or 200 mg/kg (p.o.) far 4 weeks reduced serum triglyceride, total cholesterol contents in rat induced by Poloxamer-407 or Triton WR-1339, respectively. Furthermore, post-treatment with BTSK far four weeks also inhibited body weight gain, adipose tissue mass and hyperlipidemia induced by the high fat diet for six weeks. The BTSK shifted serum total-, HDL- and LDL-cholesterol levels toward the values of normal group, suggesting that BTSK has hypolipldemlc effects. The rats fed BTSK reduced lipid peroxide and hydroxy radical in the rat blood and increased superoxide dismutase (SOD) activity compared to the control group. Taken together, these results superoxide that BTSK improve hyperlidemia and obesity via the upregulation of anti-oxidative mechanism.

Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats (산마늘 기능성 활성물질 탐색 및 자원화)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

Preventive effects of Saponariae Vaccariae Semen and Melandryi Firmi Herba on hyperlipidemia and liver damage induced by alcohol (왕부류행(王不留行)과 경엽여루채(硬葉女婁菜)가 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : This study was performed to compare the preventive effect of water extracts from Saponariae Vaccariae Semen(SVS) and Melandryi Firmi Herba(MFH) on hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, I fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. I measured the serum component in rat's blood, body weight and weight of liver. Results : At first, I observed effects of SVS and MFH on hyperlipidemia induced by alcohol. SVS and MFH group didn't show the significant decrease of total cholesterol in comparison with those of the control group. SVS and MFH group showed the significant increase of HDL-cholesterol in comparison with those of the control group. SVS group showed the significant decrease of triglyceride in comparison with those of the control group, but MFH group didn't show the significant decrease of triglyceride in comparison with those of the control group. SVS group showed the significant increase of body weight in comparison with those of the control group at 4weeks and 8weeks. But, MFH group didn't show the significant changes of body weight in comparison with those of the control group at 4weeks and 8weeks. By the way, SVS group showed the significant decrease of triglyceride in comparison with those of MFH group. SVS group showed the significant increase of body weight in comparison with those of MFH group at 8weeks. At second, I observed effects of SVS and MFH on liver damage induced by alcohol. SVS and MFH group showed the significant decrease of AST(aspartate aminotransferase), ALT(alanine aminotransferase), ALP(alkaline phosphatase) and LDH(lactate dehydrogenase) in comparison with those of the control group. SVS group showed the significant increase of liver weight in comparison with those of the control group, but MFH group didn't show the significant increase of liver weight in comparison with those of the control group. By the way, SVS group showed the significant increase of liver weight in comparison with those of MFH group. Conclusions : Reviewing these experimental results, it suggests that water extracts from SVS and MFH have pharmaceutical preventive efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate the pharmaceutical efficacy of these in depth.

The Effect of Herbal-acupuncture with Moutan Cortex Radicis Extract (목단피 약침이 고지혈증과 동맥경화에 미치는 영향에 대한 실험적 연구)

  • Objective & Methods : The present study was to investigate the effect of Moutan Cortex Radicis Herbal-acupuncture (MCR-HA) at GB34 (Yangleungchean) on hyperlipidemia and arteriosclerosis induced by high fat diet in rats. Results : 1. Moutan Cortex Radicis herbal-acupuncture infusion solution (MCR-HAS) increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. MCR-HA at GB34 decreased serum total cholesterol level, and increased serum phospholipid/total cholesterol in rats with high fat diet. 3. MCR-HA at GB34 decreased atherogenic index in rats with high fat diet. 4. MCR-HA at GB34 increased hepatic catalase activity in rats with high fat diet. Conclusion : These results suggested that Moutan Cortex Radicis herbal-acupuncture at GB34 has therapeutic effects on hyperlipidemia and related complications in rats with high fat diet.

Effect of Cheunggansoyosan-gamibang on Hyperlipidemic mice (청간소요산가미방(淸肝逍遙散加味方)이 생쥐의 고지혈증에 미치는 영향)

  • Hyperlipidemia is one of dangerous factors causing the arteriosclerosis. For the study of Cheunggansoyosan-gamibang( CGSYS) on antihyperlipidemic effect, we had fed mice divided to three groups (basal diet, hyperlipidemic diet, hyperlipidemic diet + CGSYS), and observed the change of weight, total cholesterol. triglyceride, HDL-cholesterol. LDL-cholesterol. sGOT, sGPT on the serum per every 7days for 6weeks. Also to compensate for the results above, we had tested rat liver. The results of this Study were obtained as fallows ; 1. Cholesterol change on blood was decreased by CGSYS. 2. Total cholesterol. triglyceride and LDL-cholesterol were significantly decreased by CGSYS. 3. HDL-cholesterol was significantly increasd by CGSYS. 4. sGPT was significantly decreased by CGSYS. 5. sGOT was not changed by CGSYS. 6. In case of supplying CGSYS. fatty degeneration was not observed in liver portal area. This study suggests that Cheunggansoyosan-gamibang was significant effect with hyperlipidemia.
