Extraction and Characterization of an Anti-hyperglycemic α-Glucosidase Inhibitor from Edible Mushroom, Pleurotus cornucopiae (식용버섯인 노랑느타리버섯으로부터 혈당상승억제성 α-glucosidase 저해제의 추출 및 특성)

  • The extraction and purification of the anti-hyperglycemic α-glucosidase inhibitor from an edible mushroom, Pleurotus cornucopiae, were investigated. The inhibitor was maximally extracted when the P. cornucopiae fruiting body was treated with distilled water at 30℃ for 12 h. Purification was achieved using Sephadex G-100 and G-50 filtration chromatography, pepsin hydrolysis, and reverse-phase HPLC. The compound’s solid yield and inhibitory activity were 12.2% and 9.10 mg/ml of IC50, respectively. The purified inhibitor contained two hexapeptides with Thr-Ile-Ala-Phe-Ile-Asp (A) and Tyr-Tyr-Ala-Ile-Gly-Asp (B) sequences and molecular weights of 678.79 Da (A) and 643.7 Da (B). The purified inhibitor showed a mixed inhibition pattern to α-glucosidase and a dose-dependent anti-hyperglycemic effect in a streptozotocininduced diabetic Sprague-Dawley rat model, exhibited by decreased blood glucose levels at doses of 50 and 300 mg/kg.

The Mechanism of Nephrotoxicity Formation of Methotrexate in STZ-Induced Hyperglycemic Rats (Streptozotocin 유도 당뇨성 흰쥐에서 methotrexate의 신독성 생성기전에 관한 연구)

  • This study is investigated the effect on mechanism of nephrotoxicity formation of methotrexate(MTX) by hyperglycemic by streptozotocin(STZ). MTX was injected daily at two doses of 3, 6 mg/kg for 1 week in STZ-induced hyperglycemic rats. Activities of BUN, creatinine and LDH were significantly increased by treatment with MTX in STZ-induced diabetic group when compared to MTX treatment group in normal rats' Renal lipid peroxide content and activities of cytosolic enzyme were significantly increased in the treatment of MTX in diabetic group. The concentration of glutathione and glutathione biosynthesis enzymes were decreased by treatment with MTX in STZ-induced diabetic group. These results suggest that nephrotoxicity of MTX in STZ-induced hyperglycemic rat was caused by activation of renal metabolizing enzymes in cytosol and decrease of glutathione concentration.

The Effects of Saengjingamrotang and Saengjingamrotang plus Radix Trichosanthis in Streptozotocin induced Hyperglycemic Rats (생진감노탕(生津甘露湯)과 생진감로탕가천화분(生津甘露湯加天花粉)이 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響))

  • This present study was carried out to investigate the effect of Saengjingamrotang and Saengjingamrotang plus Radix Trichosanthis in streptozotocin induced hyperglycemic rats. Body weigh, serum levels of glucose, insulin, total cholesterol, triglyceride and urine levels of volume, glucose, protein were measured in streptozotocin induced hyperglycemic rats orally receiving extracts of Sanengjingamrotang and Saengjingamrotang plus Radix Trichosanthis for 4 days. The results were as follows: 1. The change of body weight, the medication group of Saengjingamrotang plus Radix Trichosanthis concentration has the notable increase, the medication group of Saengjingamrotang concentration has no notable change, as compared to control group. 2. The change of the content on serum glucose, the medication group of Saengjingamrotang conctntration have notable decrease, as compared to control group. 3. The change of the content on serum insulin, the medication group of Saengjingamrotang plus Radix Trichosanthis concentration has the notable increase, the medication group of Saengjingamrotang concentration has no notable change, as compared to control group. 4. The change of the content on serum total cholesterol, the medication group of saengjingamrotang concentration and the medication group of Saengjingamrotang plus Radix Trichosanthis concentration have notable decrease, as compared to control group. 5. The change of the content on serum triglyceride, the medication group of Saengjingamrotang concentration and the medication group of Saengjingamrotang plus Radix Trichosanthis Concentration have notable decrease, as compared to control group. 6. The change of urine volume, the medication group of Saengjingamrotang plus Radix Trichosanthis concentration has the notable decrease, the medication group of Saengjingamrotang concentration has no notable change, as compared to control group. 7. The change of the content on urine glucose, the medication group of Saengjingamrotang concentration and the medication group of Saengjingamrotang plus Radix Trichosanthis concentration have notable decrease, as compared to control group. 8. The Change of the content of serum protein, the medication group of Saengjingamrotang concentration and the medication group of Saengjingamrotang plus Radix Trichosanthis concentration have notable decrease, s compared to control group. 9. The change of body weight, serum insulin and urine volume, the medication group of saenfjingamrotang plus Radix Trichosanthis concentration has more notable change than the medication group of Saengjingamrotang concentration, as compared to control group. According to above mentioned results, Saengjingamrotang and Saenfjingamrotang plus Radix Trichosanthis have the effect of decreased blood sugar, serum lipid levels and urine volume, protein, glucose in streptozotocin induced hyperglycemic rats, and so they were expected to be appled to the treatment of diabetes mellitus.

Effect of Hovenia dulcis Thunb Extract on the Hyperglycemic Mice Induced with Streptozotocin (자구자(Hovenia dulcis Thunb) 추출물이 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향)

  • This study has been carried out to understand the effect of Hovenia dulcis on the hyperglycemic mice induced with streptozotocin (STZ). Mice in control group were administered with $0.9\%$ saline (2 mL/kg), and experimental groups were administered Hovenia dulcis extract (H1 group, 0.01 g/kg: H2 group, 0.04 g/kg) after hyperglycemic state was induced. Blood glucose concentrations of tile H1 and H2 groups administered with Hovenia dulcis extract for 6 weeks were significantly (p<0.01), compared to control group. Blood glucose tolerance was more favorable in H1 and H2 groups than control group. The Langerhan's islet of pancreas was destructed by treatment of STZ in the control group, but pancreatic islet of the experimental groups was partially recovered from damage, and a number of insulin-positive cells were observed. A number of insulin-like growth factor- I and II (IGF-I and IGF-II) positive cells occurred in the acinar cells of H1 and H2 groups. These results suggest that administration of Hovenia dulcis extract help mice recover from the damage induced with STZ.

Effect of Persistant Mild Hyperglycemic Hyperinsulinemia on Development of Insulin Resistance in Rats (지속적인 경한 고혈당과 고인슐린증이 인슐린 저항성의 발생에 미치는 영향)

  • The effect of persistant mild hyperglycemic hyperinsulinemia on the development of the insulin resistance in rats was studied in vivo. Also, the characteristics of the insulin resistance compared with the insulin resistance of STZ diabetic rats. Persistant mild hyperglycemic hyperinsulinemic rat model was produced by ingestion of glucose polymer for 8 days. The glucose disappearance and infusion rate was measured by hyperinsulinemic euglycemic clamp technique at steady state of blood glucose and insulin levels. The clamped level of blood glucose was 100 mg/dl, and the clamped levels of insulin were $70{\mu}U/ml$ (physiologic condition) and $3000{\mu}U/ml$ (supramaximal condition). Hepatic glucose producticon rate was calculated using measured data. And the glycogen synthetic capacity of skeletal muscle(soleus) and liver was measured after 2 hours of hyperinsulinemic euglycemic clamp study. The glucose disappearance and glucose infusion rate in glucose polymer group was decreased in the both physiological and supramaximal insulin level compared to the rate of the normal control group. The rate of STZ diabetic group wase lowest at supramaximal insulin level among two another experimental groups. The hepatic glucose production rate of glucose polymer group was decreased compared to normal control but increased in STZ diabetic group. The glycogen synthetic capacity of skeletal muscle and liver of glucose polymer group was not significantly different from normal control group, but it was markdly decreased in STZ diabetic group. These results suggest that persistant mild hyperglycemic hyperinsulinemia may induce insulin resistance, but glycogen synthetic capacity is intact.

Unique Cartilage Matrix-Associated Protein Alleviates Hyperglycemic Stress in MC3T3-E1 Osteoblasts (Unique cartilage matrix-associated proteins에 의한 MC3T3-E1 조골세포에서의 고혈당 스트레스 완화 효과)

  • Unique cartilage matrix-associated protein (UCMA) is an extrahepatic vitamin K-dependent protein rich in γ-carboxylated (Gla) residues. UCMA has been recognized for its ability to promote osteoblast differentiation and enhance bone formation; however, its impact on osteoblasts under hyperglycemic stress remains unknown. In this paper, we investigated the effect of UCMA on MC3T3-E1 osteoblastic cells under hyperglycemic conditions. After exposure to high glucose, the MC3T3-E1 cells were treated with recombinant UCMA proteins. CellROX and MitoSOX staining showed that the production of reactive oxygen species (ROS), which initially increased under high-glucose conditions in MC3T3-E1 cells, decreased after UCMA treatment. Additionally, quantitative polymerase chain reaction revealed increased expression of antioxidant genes, nuclear factor erythroid 2-related factor 2 and superoxide dismutase 1, in the MC3T3-E1 cells exposed to both high glucose and UCMA. UCMA treatment downregulated the expression of heme oxygenase-1, which reduced its translocation from the cytosol to the nucleus. Moreover, the expression of dynamin-related protein 1, a mitochondrial fission marker, was upregulated, and AKT signaling was inhibited after UCMA treatment. Overall, UCMA appears to mitigate ROS production, increase antioxidant gene expression, impact mitochondrial dynamics, and modulate AKT signaling in osteoblasts exposed to high-glucose conditions. This study advances our understanding of the cellular mechanism of UCMA and suggests its potential use as a novel therapeutic agent for bone complications related to metabolic disorders.

  • E. creticum has been used in folk medicine in Jordan as a remedy for scorpion stings in the rural areas and as a hypoglycemic agent. The hypoglycemic activity of an aquous decoction of plant arial parts of E. creticum was tested in normoglycemic and streptozocin-hyperglycemic rats. Results indicate that those extract caused significant reductions in blood glucose concentration when given orally.

Optimal Dextrose Concentration for Pain Control in Healthy Newborns during Hepatitis B Vaccination

  • Purpose: Glucose has been recommended as an analgesic for mild to moderately painful procedures in neonates. The goal of this study was to assess the optimal dextrose concentration for pain control in newborns. Methods: This prospective, randomized, blinded clinical trial included 116 healthy full-term newborns. The neonates were randomly assigned to the following four groups by drawing straws: groups receiving sterile water or a 10%, 20%, or 40% dextrose solution orally. Each group was treated with the assigned solution prior to hepatitis B vaccination. The Neonatal Facial Coding System (NFCS) and the Neonatal Infant Pain Scale (NIPS) scores were evaluated before, immediately after, and 2 minutes after the injection in all neonates. Premature Infant Pain Profile (PIPP) scores were evaluated during the injection. All procedures were video-recorded, and pain scores were assessed by two independent observers who were not involved in the care of the newborns studied. The pain scores were compared among the four groups. Results: The 40% dextrose solution significantly reduced the NFCS (P=0.002) and the PIPP scores (P=0.001) compared with sterile water. No hyperglycemic events were noted in the study subjects 2 hours after the injection. Conclusion: The 40% dextrose solution effectively relieved pain due to intramuscular injection in full-term newborns without causing hyperglycemic events. However, the 10% and 20% dextrose solutions did not affect neonatal pain scores.

Effect of Rosa rugosa, Thunbergii to the Streptozotocin induced Hyperglycemic Rats (매괴화(玫瑰花) 잎, 줄기, 뿌리가 streptozotocin으로 유발(誘發)된 당뇨(糖尿)흰쥐에 미치는 영향(影響))

  • This paper aims to probe the effect of Rosa rugosa, Thunbergii which streptozotocin induced hyperglycemic rats was examined by glucose contents, body weight, kidney weight and histological observation. The results obtained were as follows: 1. The glucose level is decreased in the adminstered group of the Rosa rugosa compared with control group. These effects are in order named root group, stem group and leaf group group. BUN is increased fold of 1.52 in the control group compared with normal group, but the administered group of Rosa rugosa are decreased., in order named leaf group, root group and stem group. 2. The kidney hypertrophy to body weight is the higest in the control group compared with the other group in rats, and glucose contents are in order named stem group, leaf group and root group. 3. The loss of body weight is the highest in the control group, and the loss is in order named stem group, leaf group and root group in rats. 4. In histological observations, pancreatic pancreatic islets of control group shows disrupted or condensed nuclei, and the intercellular space between acini are enlarged. In the experimental group, histological damages of the acini and pancreatic islet are recovered compared with control group. Thes results suggest that the extracts of Rosa rugosa are effected to diabetes mellitus, root extract correlated with the function of kidney, and leaf extract is decreased plasma glucose.
