Mori Folium(MF) methanol extract and its water soluble fraction showed significant blood glucose lowering effects alloxan-induced hyperglycemic mice. Their hypoglycemic activities seemed to nothing to do with the stimulation of insulin release or insulin-like action, according to our experiments. On the other hand, MF prevents the hyperglycemic responses from an oral load of starch and glucose in vivo. Since complex carbohydrates present in a diet must be degraded to monosaccharides by $\alpha$-glucohydrolase before being absorbed in the gastrointestinal tract, it is thought that blood glucose lowering effects of MF may be related to the inhibition of $\alpha$-glucohydrolase catalyzed enzymatic reaction. In addition, experiments that examined an effect of MF water soluble fraction on gastrointestinal movement showed no significant GI movement inhibitory effect. In conclusion, MF water soluble fraction may possess active component which is a potential candidate as an orally active agent for the treatment of diabetes mellitus.
This study designed to investigate the blood glusose lowering effect of sericultural products using allozan-induced byperglycemic mice. The administration of sericultural products did not affect body weight between normal mice and alloxan induced hyperglycemic mice. High blood glucose level in alloxan-induced mice group was maintained, during all the experimental period. In the 2-3 weeks, a significant decrease in the blood glucose level was observed in the group treated with sericultural products. Up to 5 weeks, blood glucose level of those group was maintained. Sericultural products treatment group have an activity for glucose tolerance test. Sericultural products showed the blood glucose lowering acivity for loading maltose and sucrose.
The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, Anti-fatigue, Decrease of body weight activities in mouse and anti-gastric ulcer activities in stress-induced rats by Mixed Extracts of Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses (100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 4. The swimming time levels in mouse were dose-dependently extended by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 5. Anti-hyperglycemic effects of 17 medicinal plants that have been used for ameliorating diabetes in oriental medicine were evaluated using glucose transport assay in 3T3-L1 adipocytes. Higher activities were obtained by treating water or alcohol extract of Phellodendri Cortex (PC), which showed enhancing effects both on basal and insulin-stimulated glucose uptake. The latter effect of PC was completely inhibited by wortmannin, a specific inhibitor for phosphatidyl inositol 3-kinase (PI 3-kinase), but not affected by SB203580, A specific inhibitor for p38 mitogen-activatedprotein kinase(MAPK). Genistein, an inhibitor for tyrosine kinases, abolished the PC effects completely. Treatment of vanadate, an inhibitor for tyrosine phosphatases, together with PC showed no significant synergic enhancement in glucose uptake. The results of inhibitors associated with insulin signaling pathway indicated that extracts of PC enhance glucose uptake by PI-3 kinase activation which is an upstream event for GLUT4 translocation. Antidiabetic effects of PC extract might be also due to enhanced tyrosine phosphorylation and reduced tyrosine dephosphorylation. In addition, PC accelerated insulin-stimulated glucose uptake in insulin-resistant cells, recovering the uptake level close to that of normal cells. These findings may offer a new way to utilize extracts of PC as novel anti-hyperglycemic agents.
This study was designed to observe the effects of the feeding Gentiana scabra Bunge var.buergeri Max. extract on the improvement of the blood glucose, lipids in the serum of streptozotcin(55mg/kg of body weight, I.P. injection) induced hyperglycemic rats (S.D. strain, ) fed the experimental diets for 4 weeks. Concentrations of blood glucose were significantly higher in the streptozotocin administration(I.P.) groups (group 2(streptozotcin+water), 3(streptozocin+Gentiana scabra Bunge extract), 4(streptozotocin+cholesterol+water), 5(streptozotocin+cholesterol+Gentiana scabra Bunge extract) than those in the control group(group 1(water)). Bolld glucose concentrations were rather lower in the group 3(streptozotocin+Gentiana scabra Bunge extract)than in the group 2(streptozotocin+water), and then those of the group 5 were rather lower than in the group 4. There was almost little significance among the groups. concentrations of total cholesterol in serum were lower in the group 5 than those in the group 4, and the group 3 were rather lower than in the group 2. Concentrations of LDL-cholesterol in serum were significantly lower in the group 3 than those 2. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, Gentiana scabra Bunge extract administration groups were higher percentage than the other goups. Concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the other groups, and then those of the group were higher percentage than the other groups. concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the groups, and then those of the group 3 were significantly lower than in the group 4. Concentrations of triglyceride and phospholipid in serum were decreased in the group 3 than in the other groups. The activities of aspartate aminotransferase and alanine aminotransferase in serum were increased in the all experimental groups than in the control group. From the above research, the Gentiana scabra Bunge var.buergeri Max. extracts were effective on the improvement of the lipid compositions in serum of streptozotcin induced hyperglycemic and hypercholosterolemic rats.
To find the antidiabetic activity of the tripterpenoid-rich fractions of Rubus coreanus (TRF-cor) and R. crataegifolius (TRF-cra) leaves or its main component niga-ichigoside $F_1$ (Niga-$F_1$), anti-hyperglycemic and antihyperlipidemic effects were investigated in the diabetic rat model induced by streptozotocin (STZ). Treatments of rats with 200 mg/kg of the TRF-cor, TRF-cra (each, p.o.) or 20 mg/kg of Niga-$F_1$ significantly inhibited the increase of blood glucose concentration about 44.8%, 28.7% or 20.6%, respectively, in the diabetic rats. In addition, treatments with those fractions inhibited the increase of serum concentrations of triglyceride, total cholesterol or LDL-cholesterol caused by STZ. The inhibitory rate on atherogenic index (AI) values of the TRFcor (200 mg/kg), TRF-cra (200 mg/kg) or Niga-$F_1$ (20 mg/kg)-treated groups were decreased about 55.7%, 36.3% or 22.6%, respectively, comparable to STZ-treated group. In the oral glucose tolerance test, treatment of TRF-cor or TRF-cra inhibited the increase of blood glucose concentration in the STZ-induced rats. Administration of 20 mg/kg of Niga-$F_1$ (p.o.) also exhibited similar effects with the effects of both TRFs at 200 mg/kg dose (p.o.). These results support that the triterpenoids, in particular Niga-$F_1$, are contributed to the antidiabetic effects of R. coreanus or R. crataegifolius.
Hyperglycemia is one of the major risk factors for stroke. Hyperglycemia can lead to a more extensive infarct volume, aggravate neuronal damage after cerebral ischemia. ${\alpha}$-Synuclein is especially abundant in neuronal tissue, where it underlies the etiopathology of several neurodegenerative diseases. This study investigated whether hyperglycemic conditions regulate the expression of ${\alpha}$-synuclein in middle cerebral artery occlusion (MCAO)-induced cerebral ischemic injury. Male Sprague-Dawley rats were treated with streptozotocin (40 mg/kg) via intraperitoneal injection to induce hyperglycemic conditions. MCAO were performed four weeks after streptozotocin injection to induce focal cerebral ischemia, and cerebral cortex tissues were obtained 24 hours after MCAO. We confirmed that MCAO induced neurological functional deficits and cerebral infarction, and these changes were more extensive in diabetic animals compared to non-diabetic animals. Moreover, we identified a decrease in ${\alpha}$-synuclein after MCAO injury. Diabetic animals showed a more serious decrease in ${\alpha}$-synuclein than non-diabetic animals. Western blot and reverse-transcription PCR analyses confirmed more extensive decreases in ${\alpha}$-synuclein expression in MCAO-injured animals with diabetic condition than these of non-diabetic animals. It is accepted that ${\alpha}$-synuclein modulates neuronal cell death and exerts a neuroprotective effect. Thus, the results of this study suggest that hyperglycemic conditions cause more serious brain damage in ischemic brain injuries by decreasing ${\alpha}$-synuclein expression.
본 연구에서는 예로부터 전해 내려오는 당뇨처방전에 사용되고 있는 문헌고찰을 토대로 6종의 약용식물(당귀, 백복령, 상백피, 상엽, 총목피 및 홍삼)의 항산화 및 항당뇨 활성을 비교 검토하고 이들의 상관관계를 검토하여 가장 효과적인 추출방법을 조사하였다. 당귀의 열수와 에탄올 추출물에서 각각 39.43와 37.75%로 다른 시료에 비하여 추출수율이 높았다. 폴리페놀 함량은 상백피 에탄올추출물에서 가장 높았고, 홍삼 초음파추출물에서 가장 낮았다. 플라보노이드 함량과 DPPH $RC_{50}$ 값은 상엽에탄올 추출물에서 가장 우수하였다. ${\alpha}$-Glucosidase저해활성은 상엽과 상백피 초음파추출물에서 높았고 추출방법 따른 저해율은 차이를 보였으나 세가지 추출물 모두 acarbose보다 높은 활성을 보였다. ${\alpha}$-Amylase저해활성은 상엽 에탄올추출물에서 가장 높은 저해활성을 보였고, 상백피와 백복령 에탄올추출물에서도 acarbose와 비슷한 결과를 보였다. 항산화 활성과 항당뇨 저해활성의 상관성은 에탄올 추출물과 초음파 추출물에서 유의적인 상관관계를 보였다. 에탄올 추출물에서 ${\alpha}$-glucosidase저해활성에 대한 폴리페놀 함량 또는 플라보노이드 함량과는 양의 상관관계($r^2$=0.792, 0.588)를 보였고, ${\alpha}$-amylase저해활성에 대한 플라보노이드 함량 또는 DPPH 소거능의 $RC_{50}$ 값에서도 음의 상관관계($r^2$=0.918, -0.594)를 나타내었다. 또한 ${\alpha}$-amylase 저해활성과 ${\alpha}$-glucosidase저해활성에서는 양의 상관관계($r^2$=0.746)를 보였다. 초음파 추출물에서는 ${\alpha}$-glucosidase저해활성에 대해 폴리페놀 함량과 플라보노이드 함량에서만 양의 상관관계($r^2$=0.763, 0.618)를 보였다. 항산화능에서의 상관관계를 살펴 본 결과 열수, 에탄올 및 초음파추출물에서 모두 폴리페놀 함량과 플라보노이드 함량, DPPH $RC_{50}$ 값과 유의적으로 높은 양의 상관관계가 있는 것으로 나타났다. 결론적으로 약용식물이 추출방법에 따른 항산화, 항당뇨 활성과 상관성에 차이를 보이고 있음을 알 수 있었다.
