Effects of Callophyllis japonica Extracts on the Glucide and Lipid Metabolism in Hypercholesterolemic Rats (고콜레스테롤혈증 흰쥐의 당질 및 지질 대사에 볏 붉은잎(Callophyllis japonica)추출액이 미치는 영향)

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.

Effects of the Cynanchum wilfordii Ethanol Extract on the Serum Lipid Profile in Hypercholesterolemic Rats

  • The purpose of this study was to investigate the effects of the ethanol extract of Cynanchum wilfordii (ECW) on the blood lipid profile of hypercholesterolemic rats. Thirty 7-week-old male Sprague-Dawley rats were allowed free access to either a normal diet (AIN-93 diet), or 1% high-cholesterol diet with or without 0.5% or 1% ECW for 5 weeks. After sacrifice, the rat serum lipid profile was analyzed. The diets containing ECW decreased body weight gains compared to the normal diet. Serum HDL-cholesterol levels of ECW-fed groups were significantly increased in the hypercholesterolemic groups and normal groups (P<0.05). When 1% ECW was fed to the normal group, total cholesterol level was increased. Moreover, treatment of ECW in hypercholesterolemic groups yielded a dose-dependent and highly significant decrease in the atherogenic index as compared to the control. These results suggest that intake of Cynanchum wilfordii may help reduce the risks of hypercholesterolemia by increasing blood HDL-cholesterol and lowering the atherogenic index.

  • Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.

  • This study was carried out to investigate the effects of rutin from buckwheat vegetable on rats fed with hypercholesterolemic diets. Sprague-Dawley male rats were fed the diets providing various level of buckwheat vegetable for five weeks. Body weight increased during the experimental period. Body weight gain decreased by addition of cholesterol group (C-C). Rutin feeding group (T-l) and buckwheat vegetable feeding group (T-2, T-3, and T-4) showed the reduction of weight gain. Total cholesterol level decreased whereas HDL-cholesterol level increased with the increasing amount of buckwheat vegetable added. The value of thiobarbituric acid in serum decreased with increasing buckwheat vegetable added. The histopathological changes of the kidney were different among the experimental groups. Kidney of the rats fed with cholesterol added diet showed severe calcification.

Hypocholesterolemic Effect of Indigenous Dadih Lactic Acid Bacteria by Deconjugation of Bile Salts

  • Administration of milk and fermented milks produced from indigenous dadih lactic acid bacteria on serum lipids and bile acids, fecal bile acids and microflora was estimated in hypercholesterolemic rats. Anaerobic lactic acid bacteria decreased and coliforms increased in the feces of the control group; however, the number of fecal lactic acid bacteria remained unchanged when rats were administered milk and fermented milks. Only fermented milk made from Lc. lactis subsp. lactis IS-10285 significantly reduced serum total cholesterol, LDL cholesterol and total bile acids. Milk and fermented milks did not influence the HDL cholesterol. Triglyceride and phospholipid levels were significantly lower in the rats fed fermented milk of Lc. lactis subsp. lactis IS-10285 than rats fed milk and fermented milk of Lc. lactis subsp. lactis IS-29862, but not significantly different from the control group. Hypocholesterolemic effect of Lc. lactis subsp. lactis IS-10285 was attributed to its ability to suppress the reabsorption of bile acids into the enterohepatic circulation and to enhance the excretion of bile acids in feces of hypercholesterolemic rats.

The Effects of Shiryungtang on Hypertension and Hyperlipidemia (시령탕이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響))

  • Experiments were performed to determine the effects of the Shiryungtang (SRT) liquid extract on the hypertension and the hyperlipidemia induced by cholesterol in Sprague-Dawley rat(SDR) and Spontaneous Hypertensive rats(SHR). The results were obtained as follows : 1. The blood pressures of Sample A, B were decreased significantly compared with control group. First day, in case of Sample B, the blood pressure depressed after 1 and 2 hour. 2. After 11days of feeding with SRT, the blood pressure was decreased significantly in the treated group as compared with the control group. 3. After 11days by fed with SRT, the cholesterol had no significant in all treated group compared with the control group. 4. After 11days by fed with SRT, the triglyceride had no significant in all treated group compared with the control group. 5. The cholesterol of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 6. The triglyceride of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 7. The HDL-cholesterol of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 8. The phospholipid of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 9. The total lipid of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 10. The total protein of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 11. The albumin of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. With the above result, it is thought that Shiryungtang can be applied effectively to the hypertension and hyperlipidemia.

Effects of Yam on Lowering Cholesterol Level and Its Mechanism (마(Dioscorea)의 콜레스테롤 저하작용 및 그 작용기전)

  • The effects of yam(Dioscorea) on lowering cholesterol level and its mechanism were investigated. The concentrations of plasma and liver lipids, and the excretions of fecal neutral sterol and bile acid were measured in male Sprague-Dawley rats. Five Groups of 8 rats were fed hypercholesterolemic diet(1% cholesterol, 10% lard ; control), hypercholesterolemic diet plus 15% or 30% dried yam powder prepared by either hot-air(15HY, 30HY) or freeze dry(15FY, 30FY) for 4wk. Plasma total lipid, total cholesterol and cholesterol level was also significantly lower(28%, p<0.05), buy HMG-CoA reductase activity was higher in 30FY(230%, p<0.05) than in control. Although no significant differences in fecal neutral sterols were observed among groups, the yam-fed rats apparently had less bacterial degradation of cholesterol as indicated by a significantly greater of fecal cholesterol to coprostanol than in controls. Total fecal bile acids were significantly greater in rats fed yam(15HY : 5 folds, 15FY ; 12,30HY ; 12, 20FY ; 22) than in controls. The ratio of secondary to primary bile acids was almost 8 times lower in 30FY than in control. These data indicate that yam lowers cholesterol both in plasma and in liver through increasing fecal bile acid excretion as well as HMG-CoA reductase activity. Freeze-dried yam, which possesses viscosity, was more effective in cholesterol-lowering action than hot-air dried one.

Antioxidant Effects of Cranberry Powder in Lipopolysaccharide Treated Hypercholesterolemic Rats

  • This study was conducted to investigate the effects of cranberry power on antioxidant defense system in rats fed an atherogenic diet and injected with lipopolysaccharide (LPS). Sprague-Dawley rats were divided into the following 5 groups: normal diet+saline (NS), atherogenic diet+saline (AS), atherogenic diet+LPS (AL), atherogenic diet with 5% cranberry powder+LPS (AL-C5), and atherogenic diet with 10% cranberry powder+LPS (AL-C10). Total antioxidant status measured by ferric reducing ability of plasma (FRAP) was significantly reduced by LPS injection (24%) and was restored by the cranberry powder treatment (P<0.05). In addition, the mean level of plasma total phenolics was significantly decreased by LPS injection (P<0.05) and tended to be increased when cranberry powder was incorporated in to the diet. Activity of serum superoxide dismutase (SOD) tended to be lowered by LPS injection and declined further in cranberry powder fortified groups. Overall results indicate that dietary cranberry powder may provide appropriate antioxidants to counter the diminished antioxidant status induced by exposing hypercholesterolemic rats to LPS.

Effects of Dietary Calcium and Sodium Levels on Lipid Metabolism in Hyperlipidemic/Hypercholesterolemic Rats (고지혈증 모델 흰쥐에서 칼슘과 소디움 섭취수준이 체내 지질대사에 미치는 영향)

  • The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.

Effects of the Chitosan Oligosaccharide Intake on the Improvement of Serum Lipid Level in Hypercholesterolemic Rats (키토산 올리고당이 고콜레스테롤혈증 흰쥐의 혈청 지질농도 개선에 미치는 영향)

  • This study was performed to investigate the effects of the chitosan oligosaccharide administration on the improvement of the lipid compositions, blood glucose and enzyme activities in the serum of hypercholesterolemic rats fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglyceride (TG), phospholipid (PL) and blood glucose in serum were higher in the hypercholesterolemic group (group CW) and cholesterol+chitosan oligosaccharide administration group (group CCW) than those in the control group (group BW, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG, PL and blood glucose in serum were lower in the group CCW than those in the CW, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the group CCW were higher than in the group CW. The activities of alkaline phosphatase (ALP) and aminotransferase (AST, ALT) in serum were lower in the group CCW than in the hypercholesterolemic group CW. From the above results, it was suggested that chitosan oligosaccharide administration was effective in the prevention and improvement of the lipid level, blood glucose and enzyme activities in serum of hypercholesterolemic rats.
