A report of 9 unrecorded radiation resistant bacterial species in Korea

  • Five bacterial strains, ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 assigned to the genus Deinococcus were isolated from soil samples collected from Namyangju-si in Gyeonggi-do, Gangnam-gu and Dongdaemun-gu in Seoul, Korea. In addition, four bacterial strains, KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 assigned to the genus Hymenobacter were isolated from soil samples collected from Gangnam-gu and Dongdaemun-gu in Seoul, in South Korea. The five Deinococcus species were Gram-stain positive, pink-pigmented, and short-rod or coccus shaped. The four Hymenobacter species were Gram-stain negative, red-pigmented, and short-rod shaped. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strains ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 were most closely related to Deinococcus citri $NCCP-154^T$ (with 99.8% similarity), Deinococcus grandis DSM $12784^T$ (99.0%), Deinococcus marmoris DSM $12784^T$ (98.8%), Deinococcus claudionis $PO-04-19-125^T$ (98.7%), and Deinococcus radioresistens $8A^T$ (99.8%), respectively. KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 were most closely related to Hymenobacter algoricola $VUG-A23a^T$ (99.1% similarity), Hymenobacter elongatus $VUG-A112^T$ (99.1% similarity), Hymenobacter gelipurpurascens $Txg1^T$ (99.1% similarity), and Hymenobacter psychrotolerans $Tibet-IIU11^T$ (99.3% similarity), respectively. These nine species have never been reported in Korea; thus, five Deinococcus species are reported in the family Deinococcaceae, order Deinococcales, class Deinococci, phylum Deinococcus-Thermus and four Hymenobacter species are reported in the family Cytophagaceae, order Cytophagales, class Cytophagia, phylum Bacteroidetes.

민물환경에서 분리된 novel Hymenobacter sp. B2의 분류학적 특성연구 (Taxonomic characterization of novel Hymenobacter sp. B2 isolated from a freshwater environment)

  • Hymenobacter 속(genus)은 Bacteroidota 문(phylum), Hymenobacteraceae 과(family)의 대표 속(type genus)이다. 이 속에 속하는 세균들은 붉은색 색소를 함유하는 그람 음성 간균으로서, 자연계의 다양한 환경에서 분리되고 있다. 본 연구에서 붉은색 색소를 함유하는 그람 음성 간균이 경남 창원시 소재 창원대학교 교내의 연못에서 분리되었고, 이 세균은 균주 B2로 명명되었다. 균주 B2를 계통분석 및 생화학적으로 분석한 결과, Hymenobacter 속에 속하는 것으로 밝혀졌다. 이 세균의 16S rRNA 유전자 염기서열을 genbank의 BLAST로 분석해 본 결과, 다른 어떠한 세균과도 16S rRNA 유전자 염기서열의 상동성이 새로운 미생물로 인정되는 기준인 98.7%보다 낮은 것으로 나타났다. 균주 B2의 지방산을 분석해 본 결과, 주된 지방산은 summed feature 3(C16:1 ω7c and/or C16:1 ω6c, 22.8%), iso-C15:0(16.2%), anteiso-C15:0(12.9%), C16:1ω5c(12.4%) 및 summed feature 4 (iso-C17:1 I/anteiso-C17:1)(9.5%)인 것으로 밝혀졌는데, 결과적으로 균주 B2의 지방산 함량은 다른 Hymenobacter 종들의 지방산 함량과 뚜렷한 차이가 있는 것을 알 수 있었다. 이 세균의 16S rRNA 유전자 염기서열은 genbank에 accession number OQ318247로 등록되었다.

한강물로부터 분리된 방사선 내성 세균들의 계통학적 다양성 및 UV 내성 분석 (Phylogenetic diversity and UV resistance analysis of radiation-resistant bacteria isolated from the water in Han River)

  • 이 논문은 서울 한강물에서 분리한 방사선 내성 세균군집과 분리된 신종 세균의 UV 내성 특성에 관한 내용이다. 세균은 R2A agar와 1/10 R2A agar를 사용하여 3 kGy가 조사된 한강물에서 분리되었다. 그 결과 방사선에 내성을 가지는 것으로 추측되는 균주를 60주 분리하였고, 본 연구 분석 자료로 사용하였다. 16S rRNA 유전자 염기서열 분석을 통해 분리균주 60개의 계통수를 파악한 결과, 3개의 문(4개의 속)이 확인되었고, Deinococcus-Thermus (Deinococcus)가 61.7%, Firmicutes(Exiguobacterium)는 15%, Bacteroidetes (Hymenobacter, Spirosoma)는 23.4%의 비중을 나타냈다. 분리균주 중 29개 균주가 Deinococcus, Hymenobacter, Spirosoma에 속하는 신종 또는 다른 신속으로 분류될 가능성을 보였으며, 앞으로 추가적인 신종 실험이 진행될 예정이다. 그리고 신종 예상균주를 9개 선정하여 UV 내성 실험을 진행한 결과, 9개 균주 모두 D. radiodurans $R1^T$ 균주 만큼 높은 UV 내성을 가지는 것으로 나타났다. 또한 분리된 Firmicutes (Exiguobacterium) 균주는 아직까지 방사선 내성 연구 보고가 되어 있지 않아서 추가적인 방사선 내성 연구가 필요하다.

  • Fourteen bacterial strains, low10-4-1, J11015, 17J27-22, 17G22-9, 17G9-4, 17Bio_15, 17gy_33, 17SD1_21, Strain8, 17Sr1_17, J21014T, H31021, 17J49-9, and 17J80-6 assigned to the phylum Actinobacteria, Bacteroidetes, Deinococcus-Thermus, and Firmicutes were isolated from soil samples. Phylogenetic analysis based on 16S rRNA gene sequence revealed that strains low10-4-1, J11015, 17J27-22, 17G22-9, 17G9-4, 17Bio_15, 17gy_33, 17SD1_21, Strain8, 17Sr1_17, J21014T, H31021, 17J49-9, and 17J80-6 were most closely related to Marmoricola aurantiacus (98.9%), Calidifontibacter indicus (99.8%), Gordonia soli (98.8%), Rhodococcus globerulus (99.5%), Pseudarthrobacter siccitolerans (99.1%), Hymenobacter qilianensis (98.7%), Hymenobacter terrae (99.0%), Deinococcus yunweiensis (99.2%), Deinococcus proteolyticus (99.7%), Domibacillus indicus (99.2%), Exiguobacterium mexicanum (100.0%), Kurthia senegalensis (99.1%), Lysinibacillus composti (99.6%), and Bacillus loiseleuriae (99.3%). These fourteen species have never been reported in Korea, therefore we report them here for the first time.

군산지역 초등학교 정수기 물의 미생물학적 수질 (Microbiological Water Quality of Water Purifiers at Elementary Schools in Gunsan Area)

  • 2007년 7월부터 12월까지 총 5회 전라북도 군산에 위치한 초등학교의 정수기 물을 대상으로 미생물학적 수질에 대하여 조사하였다. 조사기간 중 정수기 물의 종속영양세균의 분포는 $0{\sim}1.2{\pm}0.2{\times}10^4CFU/ml$의 범주에서 나타났는데, 이 중 최대값은 우리나라 먹는 물 수질기준을 120배 초과하는 값을 나타냈다. 월별 수질기준을 초과하는 정수기 물의 백분율은 7월과 9월에 90%, 10월과 11월에 87.2%, 12월에 93.7%로 조사되었다. 한편 총대장균군과 병원성 세균인 Salmonella와 Shigella는 정수기 물과 수돗물에서 모두 검출되지 않았다. 총 5회의 실험 중 정수기 물에서 분리한 세균을 분자생물학적인 방법으로 동정한 결과, Sphingomonas, Methylobacterium, Caulobacter, Novosphingobium, Bosea, Brevundimonas, Aminobacter, Ralstonia, Mitsuaria, Variovorax, Acidovorax, Massilia, Pseudomonas, Acinetobacter, Aeromonas, Bacillus, Staphylococcus, Brevibacillus, Microbacterium, Lapillicoccus, Micrococcus, Arthrobacter, Janibacter, Flavobacterium, Chryseobacterium, Hymenobacter 등 26개 속이 동정되었다. 검출된 세균의 대부분은 일반 환경에서 다량 존재하는 세균들로서 병원성이 없으나, Pseudomonas fluorescens, Staphylococcus epidermidis, Flavobacterium johnsoniae, Acinetobacter johnsonii 등은 기회감염균으로 면역력이 낮은 초등학교 학생들의 경우 감염의 가능성이 있다.

  • In 2018, as a subset study to discover indigenous prokaryotic species in Korea, a total of 18 unreported bacterial strains were discovered. From the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade, it was determined that each strain belonged an independent and predefined bacterial species. There were no official report that these 18 species were previously described in Korea; therefore, one strain of Williamsia, one strain of Rhodococcus, three strains of Microbacterium, three strains of Agromyces, one strain of Arthrobacter, one strain of Paeniglutamicibacter, one strain of Pseudarthrobacter, one strain of Nocardioides, one strain of Fibrella, one strain of Hymenobacter, one strain of Deinococcus, two strains of Fictibacillus, and one strain of Paenibacillus are described as unreported bacterial species in Korea. Gram reaction, basic biochemical characteristics, and colony and cell morphologies are described in the species description section.

Twelve unrecorded UV-resistant bacterial species isolated in 2020

  • In 2020, a total of 12 bacterial strains were isolated from soil after a comprehensive investigation of indigenous prokaryotic species in Korea. It was determined that each strain belonged to independent and predefined bacterial species, with high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species. This study identified four families in the phylum Actinobacteria, two families in the phylum Proteobacteria, one family in the phylum Bacteroidetes one family in the phylum Firmicutes; and four species in the family Nocardiaceae, two species in the family Nocardioidaceae, one species in the family Cellulomonadaceae, one species in the family Hymenobacter, one species in the family Methylobacteriaceae, one species in the family Microbacteriaceae, one species in the family Bacillaceae and one species in the family Sphingomonadaceae. There is no official report of these 12 species in Korea, so they are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are included in the species description section.

A report of 31 unrecorded bacterial species isolated from freshwater

  • A total of 31 bacterial strains were isolated from the Geum River basin in the Republic of Korea during our investigation of indigenous prokaryotic species. The isolated bacterial strains had high 16S rRNA gene sequence similarity (>98.7%) with those of validly published bacterial species, which have not been reported in Republic of Korea. The 31 bacterial strains were phylogenetically diverse and assigned to 4 phyla, 8 classes, 18 orders, 21 families, and 27 genera. At the genus level, the unreported species were affiliated with Kineococcus, Pedococcus, Rhodoluna, Salinibacterium, Rhodoluna, Arthrobacter, Williamsia, Nakamurella, Nocardioides of the class Actinobacteria, Patulibacter of the class Thermoleophilia, Pontibacter, Hymenobacter of the class Cytophagia, Flavobacterium of the class Flavobacteriia, Geomicrobium of the class Bacilli, Brevundimonas, Gellertiella, Rhizobium, Paracoccus, Taonella, Sphingomonas of the class Alphaproteobacteria, Burkholderia, Polaromonas, Hydrogenophaga, Chitinilyticum, Azospira, Zoogloea of the class Betaproteobacteria, and Pseudomonas of the class Gammaproteobacteria. The unreported bacterial species were further characterized by examining their morphological, cultural, physiological, and biochemical properties. The detailed descriptions of the 31 bacterial strains were provided.
