Purpose. To develop a new heterodisperse 650mg acetaminophen HPMC matrix tablet with biphasic sustained release profiles. Methods. Hydroxypropylmethylcellulose(HPMC) matrix tablets were prepared by wet-granulating drug with other excipients, followed by direct compression of the dried granule mixtures into tablet using a rotary tablet machine. A novel oral controlled release tablet which may offer more uniform drug level in the body than simple zero-order was developed. The tablet is composed of three layers; outer film layer, middle part compression-coated hydroxypropylmethylcellulose (HPMC) matrix layer, and inner core layer. Each layer contains nicardipine HCl as a model drug. In vitro dissolution test showed that the tablet released the drug in clear three steps; a rapid initial release, followed by a constant rate of release, and then a second phase of fast release of drug. The dissolution characteristics could be modified easily by changing the grade of HPMC, thickness of matrix layer, content of methylcellulose in matrix layer, content of active ingredient in each layer. The pH of dissolution medium did not affect the release profile. This three-step release system is expected to raise the blood concentration rapidly to effective level and to maintain effective blood level longer than simple slow-release systems.

Acetyl-L-carnitine (ALC), an endogenous component of L-Carnitine, is the acetyl ester of carnitine that has been reported to be beneficial in depressive disorders and Alzheimer's disease.ALC is so hygroscopic that deliquescence took place when it absorbed moisture by 15%(w/w) in a week and then reached steady-state at 45%(w/w) in 40$^{\circ}C$, 75% RH storage condition. Therefore it is necessary to prevent ALC from absorbing atmospheric moisture. For this purpose, we chose hydroxypropylmethylcellulose phthalate (HPMCP) an enteric polymer, as a film former. (omitted)
Tamsulosin or a salt thereof such as its hydrochloride salt has been known to have an adrenaline ${\alpha}$ receptor blocking action for urethra and prostate areas. It has been widely used as a drug which lowers the prostate pressure and improves urinary disturbance accompanied by prostate-grand enlargement, thus for the treatment of prostatic hyperplasia. To avoid dose-dependent side effects of tamsulosin upon oral administration, the development of sustained-release delivery system is essentially required, that can maintain therapeutic drug levels for a longer period of time. The aim of this study was therefore to formulate sustained-release tamsulosin granules and assess their formulation variables. We designed entric coated sustained-release tamsulosin granules for this purpose. Nano-pores in the outer controlled release membrane were needed in order to obtain initial tamsulosin release even in an acidic environment such as gastric region. In our sustained release osmotic granule system, hydroxypropylmethylcellulose in a drug-containing layer was used as a rate controller. The drug-containing granules were coated with hydroxypropylmethylcellulose phthalate (HPMCP) and Eudragit, along with glycerol triacetate as an aqueous nano-pore former. The release of tamsulosin depended heavily on the type of Eudragit such as RS, RL, NE 30D, used in the formulation of controlled release layer. These results obtained clearly suggest that the sustained-release oral delivery system for tamsulosin could be designed with satisfying drug release profile approved by the Korean Food and Drug Administration.
Hydroxypropylmethylcelluloses (HPMC)는 친수성 cellulose 계통의 중합체로서 독성이 적고 가격이 저렴하여 경구용 서방출성 제제에 널리 이용되고 있다. 본 연구에서는 매트릭스로서 HPMC를 이용하여 새로운 서방출성제제인 HPMC/염산슈도에페드린계를 설계하고, 직타법에 의하여 정제를 제조하였으며, 이 팽윤될 수 있는 친수성 약물전달계를 이용하여 약물의 방출에 영향을 주는 여러 인자들이 약물의 방출속도에 미치는 영향에 대하여 조사하였다. 결과로서, HPMC/염산슈도에페드린계에서 매트릭스인 HPMC의 분자량이 클수록, HPMC의 함량이 많을수록 약물의 방출속도가 더 느려짐을 알 수 있었으며, 정제의 경도나 용출시 pH 변화에는 무관한 것으로 나타났다. 특히, 음이온 계면활성제인 sodium laurylsulfate는 HPMC로부터 염산슈도에페드린의 방출속도를 지연시키는데 큰 영향을 미치는 것으로 나타났으며, 이러한 현상은 음이온 계면활성제인 sodium laurylsulfate가 반대의 이온성을 갖는 약물인 염산슈도에페드린과 착물을 형성함으로써 약물의 용해성이 떨어져서 약물방출이 지연되는 것으로 생각된다.

Effect of solvents on physical characteristics and release characteristics of monolithic acetaminophen (APAP) hydroxypropylmethylcellulose (HPMC) matrix granules and tablets were examined. Various types and amounts of solvents were employed for granulation and coating. APAP and other excipients were mixed and were then wet-granulated in a high-speed mixer. The dried granules were then directly compressed and film-coated with low viscosity grade HPMC. As the amount of water increased, the size of granules also increased, showing more spherical and regular shape. However, manufacturing problems such as capping and lamination in tableting occurred when water was used alone as a granulating solvent. The physical properties of HPMC matrix granules were not affected by the batch size. The initial release rate as well as the amount of APAP dissolved had a tendency to decrease as the water level increased. Addition of nonaqueous solvent like ethanol to water resulted in good physical properties of granules. When compared to water/ethanol as a coating solvent, the release rate of film-coated HPMC matrix tablets was more sensitive to the conditions of coating and drying in methylene chloride/ethanol. Most of all, monolithic HPMC matrix tablet when granulated in ethanol/water showed dual release with about $50\%$ drug release immediately within few minutes followed by extended release. It was evident that the type and amount of solvents (mainly water and ethanol) were very important for wet granulation and film-coating of monolithic HPMC matrix tablet, because the plastic deforming and fragmenting properties of material were changed by the different strengths of the different solvents.
Tamsulosin has been frequently used for the treatment of benign prostatic hyperplasia. To avoid dose-dependent side effects of tamsulosin upon oral administration, the development of sustained-release delivery system is required, that can maintain therapeutic drug levels for a longer period of time. The aim of this study was therefore to formulate sustained-release tamsulosin matrix tablets and assess their formulation variables. We designed enteric coated sustained-release tamsulosin matrices to fulfill above statement. Aqueous microchannels in the enteric film need to be formed in order to obtain tamsulosin release even in an acidic environment such as gastric region. In the sustained-release tamsulosin matrix, low viscosity hydroxypropylmethylcellulose was used as a rate controller. Povidone K30 was also added to the matrices to facilitate water uptake so that a decrease in the release rate of tamsulosin as time elapses was prevented, possibly leading to pseudo zero-order release of the drug. The matrices were enteric-coated with hydroxypropylmethylcellulose phthalate (HPMCP), along with povidone K30 as an aqueous microchannel former. With the aqueous microchannels formed within the enteric film, tamsulosin could be released in an acidic condition. The release of tamsulosin decreased with increasing thickness of HPMCP membrane while the release rates of tamsulosin from those having different HPMCP thickness in pH 7.2 aqueous media were not considerably different, indicating that the enteric film was promptly dissolved at pH 7.2. These results clearly suggest that the sustained-release oral delivery system for tamsulosin could be designed with satisfying drug release profile approved by the KFDA.
To develop a sustained-release tablet which had the similar dissolution to commercial ibudilast-loaded sustained-release capsule, the tablets were prepared using hydroxypropylmethylcellulose phthalate (HPMCP), ethylcellulose (EC) and hydroxypropylcellulose (HPC), and dissolution test were carried out with paddle method in KP. The tablet prepared only with HPMCP and EC showed sno similar dissolution pattern to the commercial product. As the ratio of HPMCP/HPC in tablet decreased, the dissolution rate of drug decreased in pH 1.2 but increased in pH 6.8. Furthermore, an ibudilast-loaded sustained-release tablet composed of [ibudilast/EC/HPMCP/HPC (10/10/170/10 mg/tab)] gave similar dissolution to commercial product in pH 1.2 for 3 h and in pH 6.8 for 10 h. Thus, it could be a potential candidate for the substitute of commercial capsule.

Sustained release microspheres containing phenylpropanolamine HCI (PPA) were prepared with acrylic polymer (Eudragit RL/RS) sand hydroxypropylmethylcellulose phthalate (HPMCP) using a emulsion-solvent evaporation method. Magnesium strate was used a smoothing agent for preparation of microspheres. The microspheres obtained were very spherical and free-flowing particles. Scanning electron microscopy showed that microspheres have a smooth surface and a sponage-like internal structure. The dissolution rate of PPA from the microspheres was dependent on the pH of dissolution media. PPA showed faster relase in hP 1. 2 solution than in pH 7.4 solution due to the solubility of PPA. Therefore we prepared new microspheres containing 5% (w/v) HPMCP in order to control the release of PPA. The release rate of PPA from these new microspheres was similar in pH 1.2 and pH 7.4 solution.
