A gene for a putative glucoamylase, stg, of a hyperthermophilic archae on Sulfolobus tokodaii was cloned and expressed in Escherichia coli. The recombinant glucoamylase (STGA) had an optimal temperature of $80^{\circ}C$ and was extremely thermostable with a D-value of 17 hr. The pH optimum of the enzyme was 4.5. Being different from fungal glucoamylases, STGA hydrolyzed maltotriose (G3) most efficiently. Gel permeation chromatography and sedimentation equilibrium analytical ultracentrifugation analysis showed that the enzyme existed as a dimer. STGA was stable enough to hydrolyze liquefied com starch to glucose in 4 hr at $90^{\circ}C$ with a yield of95%. Comparison of the $k_{cat}$ values for the hydrolysis and the reverse reaction at $75^{\circ}C$ and $90^{\circ}C$ indicated that glucose production by STGA was more efficient at $90^{\circ}C$ than $75^{\circ}C$. Therefore, STGA showed great potential for application to the industrial glucose production process due to its high thermostability.
We report the existence of new type of phosphatidylcholine-hydrolyzing phospholipase D (PLD), which has been characterized and partially purified in the scuticociliate, Uronema marinum. The enzyme from partial purification showed that it was existed in membrane fraction and was a neutral PLD, which catalyzed both transphosphatidylation and hydrolysis reaction. The activity of partially purified membrane-bound PLD was also found to be optimal at pH 7.0-7.5 for 2 hours at 37℃ and depended strictly on the presence of Ca2+ (2.5 mM) and Mg2+ (1.6 mM). Immunoblot analysis indicated that the enzyme was distinct from hPLD1 (human PLD1) and hPLD2 (human PLD2) because it was not recognized by a polyclonal antibody raised to the 12 terminal amino acid of these enzymes. We also found that the membrane-bound PLD is a PIP2-dependent PLD and that GTP-binding proteins are not implicated in the regulation of this enzyme: This enzyme activity is markedly stimulated by phosphatidylinositol 4,5-bisphosphate (PIP2) but not by the small G-protein Arf and GTPrS. In addition, this enzyme was capable of hydrolyzing phosphatidylcholine (PC) but not phosphatidylethanolamine (PE), implying that PC was a preferred substrate.
Recently, graphene based solution-gated field-effect transistors (SGFETs) have been received a great attention in biochemical sensing applications. Graphene and reduced graphene oxide (RGO) possess various advantages such as high sensitivity, low detection limit, label-free electrical detection, and ease of fabrication due to their 2D nature and large sensing area compared to 1D nanomaterials- based nanobiosensors. Therefore, graphene or RGO -based SGFET is a good potential candidate for sensitive detection of protons (H+ ions) which can be applied as the transducer in various enzymatic or cell-based biosensing applications. However, reports on detection of H+ ions using graphene or RGO based SGFETs have been still limited. According to recent reports, clean graphene grown by CVD or exfoliation is electrochemically insensitive to changes of H+ concentration in solution because its surface does not have terminal functional groups that can sense the chemical potential change induced by varying surface charges of H+ on CVD graphene surface. In this work, we used RGO -SGFETs having oxygen-containing functional groups such as hydroxyl (OH) groups that effectively interact with H+ ions for expectation of increasing pH sensitivity. Additionally, we also investigate RGO based SGFETs for bio-sensing applications. Hydroloytic enzymes were introduced for sensing of biomolecular interaction on the surface of RGO -SGFET in which enzyme and substrate are acetylcholinesterase (AchE) and acetylcholine (Ach), respectively. The increase in H+ generated through enzymatic reaction of hydrolysis of Ach by AchE immobilized on RGO channel in SGFET could be monitored by the change in the drain-source current (Ids).
ZnO, CdS, rutile-$TiO_2$ 및 혼합 rutile-$TiO_2$/ZnO와 같은 여러 반도체를 이용하여 Congo Red를 광 촉매 분해시켰다. 연구 결과 ZnO, CdS, rutile-$TiO_2$ 중에서는 CdS의 광 촉매 효과가 제일 컸는데 이것은 CdS가 제일 작은 band gap 에너지를 가지고 있기 때문이었다. 또한 혼합 촉매에서는 ZnO의 함량이 rutile-$TiO_2$에 비하여 상대적으로 높을수록 분해 반응을 촉진하였다. 이것은 $Zn^{2+}$ 가수분해 생성물이 구조적으로 안정한 화합물인 rutile-$TiO_2$의 표면을 덮음으로서 자외선 흡수를 차단하기 때문에 라디칼 생성을 저해하기 때문이었다.
일반적으로 고내구성 재료로 알려져 있는 FRP 재료가 유해한 환경요소에 노출될 경우 가수분해와 같은 화학적 반응으로 인하여 재료적 열화와 재료의 성질이 감소될 수 있다. 따라서, FRP 재료를 건설 주부재로 사용하기 위해서는 사용 환경에 따른 역학적 특성을 정확하게 파악하는 것이 중요하다. 이에 본 연구에서는 화학약품에 의한 급성 또는 만성적인 극심한 변화들을 가상적인 시뮬레이션을 실시하고자 강산과 강알칼리화합물 용액에 FRP 부재를 일정기간 보관한 후 압축강도, 인장강도, 전단강도 그리고 휨강도 실험을 실시하여 화학약품의 종류 및 보관일수에 따른 강도 변화를 분석하였다. 결론적으로 본 연구를 통하여 FRP 부재의 우수한 내화학약품성을 확인할 수 있었다.
본 실험에서는 MMA와 tBMA의 공중합체를 합성하고, tert-butyl 그룹의 가수분해를 선택적으로 유도하여 poly(MMA-co-MA)를 제조하였다. 또한 말단에 에폭시기를 함유한 다양한 분자량의 PEO와 MA와의 grafting-onto 커플링 반응을 통해 같은 주사슬을 가지지만 부사슬의 길이가 다른 poly(MMA-co-PEGMA)를 합성하여 조성이 이온전도도에 미치는 영향을 평가하였다. AC-impedance로 측정한 상온 이온전도도는 MMA의 몰분율이 82%에서 $5.11{\times}10^{-8}Scm^{-1}$의 값이 얻어진 반면, 48%에서는 $1.88{\times}10^{-6}Scm^{-1}$로서 많은 PEGMA에서 높게 관찰되었다. 또한 에폭시기를 함유한 PEO의 분자량에 따라 이온전도도의 차이가 발생하는데, grafting-onto 법의 입체적 장애가 원인으로 고려되었다. 한편, 합성된 poly(MMA-co-PEGMA) 고분자 전해질은 상온에서 6V까지 우수한 전기화학적 안정성을 보여주었다.
A xylanase (xylanase I) was purified 11.9-fold from the culture filtrate of Trichoderma koningii ATCC 26113 by the column chromatography on Sephadex G-75, SP-Sephadex C-50, DEAE-Sephadex A-50 and Sephadex G-50 with an overall yield of 8.2%. The molecular mass determined by gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis was found to be a monomeric polypeptide of ca. 35 kDa. The isoelectric point of the enzyme was estimated to be 9.3. The optimal reaction pH and temperature are 5.8 and 55.deg.C, respectively. The enzyme is stable up to 60.deg.C, while 78% of its activity is lost after the incubation for 10 min at 70.deg.C. The enzyme hydrolyzes sylan with relatively high activity, as well as carboxymethyl cellulose and avicel. The $K_{m}$ values of the enzyme for oat-spelf sylan, larchwood xylan and Avicel were 3.5, 1.6 and 10. 1 mg/ml, respectively. The enzyme hydrolyzed oat-spelt sylan to sylose, sylobiose, sylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose, xylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose and xylotriose as the major products. The hydrolysis patterns indicate that xylanase I is endo-enzyme.e.
벼 도열병에 대하여 선택적으로 방제효과가 있는 2-이미노-1,3-티아졸린 유도체 1의 최적화 과정 중의 하나로서, 선도물질의 C-5 위치에 메틸기가 도입된 새로운 화합물 교를 합성하고 생물활성을 시험하였다. $\beta$-케토 에스터 7을 브롬화한 다음 thiourea와 반응시키고 가수분해하여 2-아미노-5-메틸-1,3-티아졸린 카르복실산 3을 얻었다. 이것을 아닐린 유도체와 각각 반응시켜 17종의 상응하는 2-이미노-5-메틸-1,3-티아졸린 카르복스 아닐리드 유도체 2를 합성하였다. 벼 도열병에 대한 화합물 2의 방제효과는 화합물 1보다 미약하였다. 2-이미노-1,3-티아졸린 유도체의 벼 도열병에 대한 방제효과는 C-5 위치의 치환체에 의하여 매우 큰 영향을 받았으며, 이것은 이 계열 화합물의 선도물질 최적화 과정에서 분자설계를 위한 중요한 자료가 된다.
Protox 저해제인 cyclic imide type 화합물 중에서 EK 5439와 S-275 같은 화합물들의 필수 중간체인 3-chloro-2-(4-chloro-2-fluoro-5-hydroxyphenyl)-4,5,6,7-tetrahydro-2H-indazole은 2-(2-fluoro-4-chloro-5-hydroxyphenyl)-2,3a,4,5,6,7-hexahyoindazol-3-one을 포스겐과 반응시켜서 얻을 수 있다고 알려져 있으나, 수율이 매우 낮고 재현성이 없다. 따라서 이 반응의 부산물들을 분리 확인하여, 이 부산물들이 반응물과 목적물의 이량체임을 밝히고, 이들을 가수분해함으로서 목적물의 합성수율을 높이고 반응물을 거의 대부분 회수할 수 있는 효과적인 방법을 개발하였다.
