Effects of Salvia plebeia R. Br. on Antioxidative Enzyme Activities and Oxidative Damage in Rats Fed High-Fat and High-Cholesterol Diets (곰보배추섭취가 고지방과 고콜레스테롤 식이 랫드에서의 항산화 효소활성 및 산화적 손상에 미치는 영향)

  • The purpose of the present study was to investigate the effect of Salvia Plebia R. Br. (SP) powder on the antioxidative defense system and oxidative stress in rats which were fed a high fat high cholesterol diet. Accordingly, the rats were divided into four experimental groups which were composed of a high fat high cholesterol diet group (HF), HF diet with 5% SP powder supplemented group (PA), a HF diet with 10% SP powder supplemented group (PB), and a normal group (N). Consequently, the hepatic catalase activity of the HF group was decreased compared to the normal group (N), but it is recorded that of the PA and PB groups were significantly increased. With this in mind, the PA and PB groups resulted in the case of significantly increased activities of hepatic GSH-px and SOD. The hepatic superoxide radical and hydrogen peroxide contents of the PA and PB groups were significantly decreased, as compared to the HF group. The GOT and GPT activities of the PB group were also significantly decreased when thus compared to the HF group. Notably, the carbonyl values contents of the PA and PB groups were significantly reduced compared to the HF group. The hepatic TBARS values in the liver were significantly reduced as measured in the PA and PB groups. These results suggest that the SP powder may reduce the incidence of oxidative damage, by the activation of an antioxidative enzyme in rats fed with high fat high cholesterol diets.

Immobilization of the Hyperthermophilic Archaeon Thermococcus onnurineus Using Amine-coated Silica Material for H2 Production (아민기가 코팅된 규조토 담체를 이용한 초고온성 고세균 Thermococcus onnurineus의 세포 고정화 및 수소생산 연구)

  • Previously we reported that the hyperthermophilic archaeon, Thermococcus onnurineus NA1 is capable of producing hydrogen (H2) from formate, CO or starch. In this study, we describe the immobilization of T. onnurineus NA1 as an alternative means of H2 production. Amine-coated silica particles were effective in immobilizing T. onnurineus NA1 by electrostatic interaction, showing a maximum cell adsorption capacity of 71.7 mg-dried cells per g of particle. In three cycles of repeated-batch cultivation using sodium formate as the sole energy source, immobilized cells showed reproducible H2 production with a considerable increase in the initial production rate from 2.3 to 4.0 mmol l−1 h−1, mainly due to the increase in the immobilized cell concentration as the batch culture was repeated. Thus, the immobilized-cell system of T. onnurineus NA1 was demonstrated to be feasible for H2 production. This study is the first example of immobilized cells of hyperthermophilic archaea being used for the production of H2.

Determination of Fe(II) ion and Fe(III) ion by Chemiluminescence Method (화학발광법을 이용한 Fe(Ⅱ)이온과 Fe(Ⅲ)이온의 정량)

  • A method to determin Fe(II) and Fe(III) ion in aqueous solution by chemiluminescence method using a stopped flow system has been studied. The method is based on the increased chemiluminescence intensity with the addition of Fe(III) ion to a solution of lucigenin and hydrogen peroxide. The effects of KOH concentration, flow rate of reagents, $H_2O_2$ concentration and citric acid concentration used for the masking of Fe(II) ion on the chemilu-minescence intensity have been investigated. The calibration curve for total Fe was linear over the range from 1.0${\times}$$10^{-6}$ M to 1.0${\times}$$10^{-4}$M, coefficient of correlation was 0.996 and the detection limit was 1.0${\times}$$10^{-7}$M under the optimal exper-imental conditions of 4.0 M, 2.0 M, 3.5 mL/min for the concentration of $H_2O_2,$ KOH and flow rate of reagents, respec-tively. The calibration curve for Fe(Ⅲ) was linear over the range from 1.0${\times}$$10^{-6}$M to 1.0${\times}10^{-4}$ M, the coefficient of correlation was 0.997 and the detection limit was 5.0${\times}$$10^{-7}$M under the optimal experimental conditions.

Investigation on Optimal Aeration Rate for Minimizing Odor Emission during Composting of Poultry Manure with Sawdust (계분톱밥 퇴비화시 악취발생의 최소화를 위한 적정 공기주입을 구명)

  • This study was conducted to find out the optimal aeration rates for minimizing odor emission and for increasing biological activities during composting of livestock manure in the enclosed bench-scale reactor system. It was treated with the mixture of poultry manure and sawdust controlled the initial water content of 60%, then aerated continuously at four different aeration rates (0.1, 0.2, 0.4 and 0.6 L/min/kg dry-solids). The average emitted concentration of ammonia in 0.6 L/min/kg dry-solids during composting reached the level of 40% in comparison with that of 0.2 L/min/kg dry-solids. In cases of sulfur compounds such as hydrogen sulfide, methylmercaptan and ethylmercaptan, their concentrations decreased with increasing aeration rates and the emission time was shortened. But they didn't detect in the treatment of 0.6 L/min/kg dry-solids. The biological activity for composting showed a trend of increasing as aeration rates increased. The treatment of 0.6 L/min/kg dry-solids gave the highest biological activity and the best compost quality.

Study on distribution and extermination of scuticociliatids parasitizing to japanese flounder, Paralichthys olivaceus in southern Korea (남해 양식산 넙치(Paralichthys olivaceus) 치어에 기생한 스쿠티카 섬모충(scuticociliatids) 동태 및 구제에 관하여)

  • We investigated on the prevalence and extermination of scuticociliatids parasitic on cultured japanese flounder, Paralichthys olivaceus in land-marine tank system of southern Korea from January to February in 1997. The gills and the skin showed the highest infection rate(60%), and the brain showed the lowest(22%). Also, fish secreted large quantity of mucus with a bleeding and ulcerated lesions on the infected sites. The number of the parasites in inflowing sea water, surface water and bottom water of farming tank ranged 0~1 individuals/$100m\ell$, 0~413 individuals/$100m\ell$ and $7\sim7.3{\times}10^4$ individuals/$100m\ell$, respectively. This parasite was died within 2 hours in 50~500 ppm, 48 hours of 10 ppm formalin or hydrogen peroxide, 1 hour in 50~500 ppm, 80 minutes of 10 ppm oligo chitosan and 10 minute in 100% but did not died until 48 hours in 10~70% fresh water.

Effects of Fermented Scutellaria Baicalensis Extract on H2O2 - Induced Impairment of Long-term Potentiation in Hippocampal CA1 Area of Rats (흰쥐 해마 CA1 영역에서 H2O2에 의한 장기강화 억제에 대한 발효황금 추출물의 효과)

  • Scutellaria baicalensis (SB) has widely used in the treatment for various brain diseases in the field of Oriental medicine. Biofermantation of SB can make major chemical constituents of SB to pass blood-brain barrier easily and to have more potent anti-oxidant ability. There is a little information about the contribution of fermented SB (FSB) to the formation or maintenance of the neural plasticity in the hippocampus. The purpose of this study was to evaluate effects of FSB extract on hydrogen peroxide (H2O2) - induced impairments of the induction and maintenance of long-term potentiation (LTP), an electrophysiological marker for the neural plasticity in the hippocampus. From hippocampal slices of rats, the field excitatory postsynaptic potentials (fEPSPs) were evoked by the electrical stimulation to the Schaffer collaterals - commissural fibers in the CA1 areas and LTP by theta-burst stimulation by using 64 - channels in vitro multi-extracellular recording system. In order to induce oxidative stress to hippocampal slices two different concentrations (200, 400 μM) of H2O2 were given to the perfused aCSF before and after the LTP induction, respectively. The ethanol extract of FBS with concentration of 25 ㎍/ml, 50 ㎍/ml was diluted in perfused aCSF that had 200 μM H2O2, respectively. Oxidative stress by the treatment of H2O2 resulted in decrease of the induction rate of LTP in the CA1 area with a dose - dependent manner. However, the ethanol extract of FSB prevented the reduction of the induction rate of LTP caused by H2O2 - induced oxidative stress with a dose - dependent manner. These results may support a potential application of FSB to ameliorate impairments of hippocampal dependent neural plasticity or memory caused by oxidative stress.

A Study on Occupational Hydrofluoric Acid Burns in a Hydrofluoric Acid Manufacturing Factory (불산제조업체에서 발생한 불산화상에 관한 조사연구)

  • Hydrofluoric acid is one of the strongest irritating, corrosive and poisonous inorganic chemicals. Hydrofluoric acid burns are occurring with ever-increasing frequency due to the wide use of this acid in industries. Hydrofluoric acid burns are characterized by severe progressive tissue destruction and excruciating pain due to the unique properties of the freely dissolvable fluoride ion. The authors reviewed medical records of 32 cases (36 spells) of hydrofluoric acid burns which occurred in a hydrofluoric acid manufacturing factory from Sep. 1, 1990 to June 30, 1993. The results are as follows; 1. Eleven measurements of air concentrations of hydrofluoric acid by detection tube method from 1990 to 1992 were all below TLV (Department of Labor, R.O. K). 2. There were 19 cases (22 spells) of hydrofluoric acid burns which occurred during the study period among regular employees. The overall incidence density of hydrofluoric acid was 17.8 cases (20.6 spells) per 100 person-year. Incidence density was 19.0 cases (22.0 spells) per 100 person-year among male workers and there were no female cases. Incidence density was 32.9 cases (38.3 spells) per 100 person-year among production workers and 1.9 cases (1.9 spells) per 100 person-years among management workers with the difference being statistically significant (P<0.01). 3. Of 32 cases (36 spells) of hydrofluoric acid burns among workers who were regularly employed or temporarily employed, 26 spells (81.2%) were between age 20 to 39. In 15 spells(41.7%) burns occurred between 12:00 and 17:59 with 16 spells(44.3%) having arrived at hospital within 2 hours after the accident. 4. Of 36 spells, the main cause of hydrofluoric acid burns were by splashes (8 spells, 22.2 %). The most frequent site of burns were fingers and pain was the most frequent symptom. Thirty spells (83.3%) of the hydrofluoric acid burns were treated with local injection of antidote (calcium gluconate). Complete recovery without scarring were observed in most of the cases (34 out of 36 cases, 94.4%). The study results suggest that to prevent hydrofluoric acid burns, environmental control and the wearing of hydrofluoric acid resistant protective clothes and gloves are important. It is also stressed that establishment of an emergency management and a transfer system for hydrofluoric acid burn victims is necessary.

Characterization of Bio-oils Produced by Fluidized Bed Type Fast Pyrolysis of Woody Biomass (목질바이오매스의 급속열분해에 의해 생성된 바이오오일의 특성 분석)

  • Using fluidized bed type fast pyrolysis system (capacity 400 g/h) bio-oils were produced from beech (Fagus sylvatica) and softwood mixture (spruce and larch, 50:50). The pyrolysis was performed for 1~2 s at the temperature of $470{\pm}5^{\circ}C$. Pyrolysis products consisted of liquid form of bio-oil, char and gases. In beech wood bio-oil was formed to ca. 60% based on dry biomass weight and the yield of bio-oil was 49% in soft wood mixture. The moisture contents in both bio-oils were ranged between 17% and 22% and the bio-oil's density was measured to $1.2kg/{\ell}$. Bio-oils were composed of 45% carbon, 47% oxygen, 7% hydrogen and lower than 1% nitrogen,which was very similar to those of original biomass. In comparison with oils from fossil resources, oxygen content was very high in bio-oils, while no sulfur was found. More than 90 low molecular weight components, classified to aromatic and non aromatic compounds, were identified in bio-oils by gas chromatographic analysis, which amounted to 31~33% based on the dry weight of bio-oils.

Syngas Concentration and Efficiency in Heavy Residual Oil Gasification with 1 Ton/Day-Class Entrained-Bed Reactor (1톤/일급 분류층 가스화기에서 중질잔사유의 가스화 합성가스 조성 및 효율 변화)

  • With the 1 ton/day-class entrained-bed gasification system, heavy residual oil from local refinery was gasified at the operating conditions of 1,000~1,20$0^{\circ}C$ and 3 $kg_f$/$\textrm{cm}^2$ in order to determine the variation of syngas composition, carbon conversion, and cold gas efficiency. Produced syngas consists of mainly CO, H$_2$, $CO_2$, and the methane concentrations. Results yielded a maximum syngas composition of 45% H$_2$ and 26%, CO at the 31 kg/hr feeding condition. The maximum carbon conversion and cold gas efficiency were 87% and 68%, respectively at the feeding conditions of 20 kg/hr and oxygen/feed ratio of 1.2. When oxygen feeding amount that is one of the most important operating parameter in gasification was increased, concentration of hydrogen in the syngas is greatly increased comparing to the concentration of CO and $CO_2$. The temperature exhibited about 11$0^{\circ}C$ raise while oxygen/feed ratio changed from 0.6 to 1.2. Methane concentration showed enhanced dropping rate with increase in gasifier temperature and the useful relationship between the gasifier temperature and methane concentration existed such that it can be employed as an indirect measure of inside gasifier temperature.

Study on Torrefaction Characteristics of Solid Biomass Fuel and Its Combustion Behavior (바이오매스 고형연료의 반탄화 특성 및 반탄화물의 연소특성에 관한 연구)

  • Torrefaction is a thermochemical process proceeded at the temperature around $250^{\circ}C$ in an inert gas condition. By torrefaction, the hemicellulose portions contained in biomass are broken down to change into the volatile gas which is removed from biomass eventually. The main purpose of biomass torrefaction is to improve the energy density of the biomass to minimize the transport energy consumption, though the flammability can be elevated for transportation. In this study two types of solid biomass fuel, waste wood and rice straw, were torrefied at various temperature range from $200^{\circ}C$ to $300^{\circ}C$ to evaluate the torrefied biomass characteristics. In addition torrefied biomass were tested to evaluate the combustion characteristics using TGA (Thermogravimetric Analysis). After the torrefaction of biomass, the C/H (carbon to hydrogen ratio) and C/O (carbon to oxygen ratio) were measured for aquisition of bio-stability as well as combustion pattern. Generally C/H ratio implies the soot formation during combustion, and the C/O ratio for bio-stability. By torrefaction temperature at $300^{\circ}C$, C/H ratio and C/O ratio were increased by two times for C/H and three times for C/O. The torrefied biomass showed similar TGA pattern to coal compared to pure biomass; that is, less mass decrease at lower temperature range for torrefied biomass than the pure biomass.
