Monoclonal antibody K312-based depletion of pluripotent cells from differentiated stem cell progeny prevents teratoma formation

  • Human pluripotent stem cells (PSCs) have been utilized as a promising source in regenerative medicine. However, the risk of teratoma formation that comes with residual undifferentiated PSCs in differentiated cell populations is most concerning in the clinical use of PSC derivatives. Here, we report that a monoclonal antibody (mAb) targeting PSCs could distinguish undifferentiated PSCs, with potential teratoma-forming activity, from differentiated PSC progeny. A panel of hybridomas generated from mouse immunization with H9 human embryonic stem cells (hESCs) was screened for ESC-specific binding using flow cytometry. A novel mAb, K312, was selected considering its high stem cell-binding activity, and this mAb could bind to several human induced pluripotent stem cells and PSC lines. Cell-binding activity of K312 was markedly decreased as hESCs were differentiated into embryoid bodies or by retinoic acid treatment. In addition, a cell population negatively isolated from undifferentiated or differentiated H9 hESCs via K312 targeting showed a significantly reduced expression of pluripotency markers, including Oct4 and Nanog. Furthermore, K312-based depletion of pluripotent cells from differentiated PSC progeny completely prevented teratoma formation. Therefore, our findings suggest that K312 is utilizable in improving stem cell transplantation safety by specifically distinguishing residual undifferentiated PSCs.

Generation of a monoclonal anti-human $\beta$2-adrenergic receptor antibody using GST-$\beta$-adrenergic receptor C-terminal fusion proteins expressed in E.Coli.

  • Among the various receptor molecules discovered so far the ${\beta}$2-adrenergic receptors have been regarded as excellent model systems for the so called 7 transmembrane helix receptor and have been the focus of extensive studies. For the analysis of receptor structure and function a monoclonal antibody plays a crucial role, thus providing useful tools for the study of receptor. However, because of the minute quantity of receptor molecules which could be obtained from natural sources, the generation of specific monoclonal antibody against receptor molecules from the purified receptors has been regarded as virtually impractical in consideration of cost and experimental times. The purpose of the present study was to generate and characterize a monoclonal antibody against human ${\beta}$2-adrenergic receptor. For the production of antibody, C-terminal regions of the human ${\beta}$2-adrenergic receptor was produced as a fusion protein with Glutathion S-transferase (GST) in E. Coli. The expression of the fusion protein was identified by SDS-PAGE and Western blot using monoclonal anti-GST antibody. The fusion protein was purified to an apparent homogeniety by affinity chromatography with Glutathion Sepharose CL-4B and used as an antigen for the immunization of BALB/C mice. The Production of monoclonal antibody was achieved by fusion of the immunized spleen cells and SP/2-0 myeloma cells. Positive hybridomas were screened by ELISA and were cloned by two consecutive rounds of limiting dilution. The monoclonal antibody produced in this study (mAb${\beta}$C02) was IgM type and purified by immunoaffinity chromatography using anti-mouse IgM agarose as an affinity matrix. MAb${\beta}$C02 showed strong and specific immunoreactivity against both the fusion protein and human ${\beta}$2-adrenergic receptor in ELISA and Western blot. The molecular weight of immunoreactive band was 64 kDa and exactly coincided with the previously reported molecular weight of ${\beta}$2-adrenergic recepters. The results of the present study suggest that mAb${\beta}$C02 may be used for the study of receptor function and regulation in normal or nonphysiological status.

8-Azaguanine 내성 인형 형질세포종 세포주의 확립 (Establishment of 8-Azaguanine Resistant Human Plasmacytoma Cell Line)

  • This study was performed to establish human plasmacytoma cell line as the partner cells for producing human hybridoma. Bone marrow cells from a multiple myeloma patient from Seoul National University Hospital, Korea were cultured and established as the cell line, named as HMC-BM4. HMC-BM4 cells were cultivated in RPMI 1640 media containing 8-azaguanine(8-AG; gradually increasing concentration from $1\;{\mu}g/ml$ to $20\;{\mu}g/ml$). 8-AG resistant cells were collected and cloned by limiting dilution. Each clone was divided and tested to die in hypoxanthine, aminopterine and thymidine (HAT) selection media. Finally one clone was selected and named as HMC-AR, which was sensitive to HAT selection media. HMC-AR cells showed typical morphology of plamacytoma in Wright staining. No cell formed the rosette with sheep erythrocytes. Surface membrane $\mu$ heavy chain was detected in 20% of HMC-AR cells and cytoplasmic $\mu$ heavy chain in 90% of them by direct immunofluorescent staining. Ia-like antigen was found in 90% of HMC-AR cells by indirect immunofluorescent staining using anti-Ia-like antigen monoclonal antibody, 1BD9-2. And about $1.0\;{\mu}g/ml$ of human $\mu$ heavy chain was detected in the 3-day culture supernatant of HMC-AR cells. 88% of cells contained 46 chromosomes. Mycoplasma was not detected in HMC-AR cells by Hoechst 33258 staining. This cell line would be used for making hybridomas secreting human monoclonal antibody.

단세포군항체를 이용한 효소면역억제측정 법에 의한 유구낭미충증의 혈청학적 진단 (Serodiagnosis of cysticercosis by ELISA-inhibition test using monoclonal antibodies)

  • 유구낭미충 항원에 대한 단세포군항체를 제조하여 그 특성을 분석하고, 이를 효소면역억제측정법 (ELISA-inhibition test)에 응용함으로써 통상적인 효소면역측정법(ELISA)의 특이도를 더욱 높이고자 하였다. 유구낭미충의 두절항원으로 면역한 BALB/c 마우스의 비장세포와 형질세포종세포를 융합하여 림프잡종세포를 만들고, 이 중에서 유구낭미충 항원에 대한 특이 단세포군 항체를 생성하는 4개의 림프잡종세포군을 선택하였다. EITB상 Cya-1 단세포군항체는 25.5 kDa, Cya-7은 28 kDa, Cya-28은 87.5 kDa, Cya-31은 12.5 kDa 항원대에 각각 반응하는 것을 알 수 있었다. 간접면역 형광항체법으로 Cya-1는 칼슘소체에, Cya-7은 실질조직에, Cya- 28과 Cya-31은 충체 표피에 주된 반응을 나타내고 있었다. 유구낭미충 항원을 사용한 효소면 역측정법으로는 유구낭미충 감염자 28명의 혈청 중 23명 (82. l%)이 양성 반응을 보였으며 폐흡충 감염자 31명 중 6명(19.4%), 스파르가눔 감염자 9명 중 1명(11. l%)이 교차 반응을 보였다 반면, 특이 단세포군항체 Cya-7를 이용한 효소면역 억제측정법으로는 유구낭미충 감염자는 28명 중 19명 (67.9%)이 양성반응을 보였고, 다른 기생충 감염자 혈청과의 교차 반응은 관찰되지 않았다.

DEAE-Trisacryl 크로마토그래피법에 의한 IgG1 Type 쥐 단일클론 항체의 분리정제 (Purification of IgG1 Type Mouse Monoclonal Antibodies with DEAE-Trisacryl Chromatography)

  • 하이브리도마를 쥐의 복강이나 in-vitro에서 배양한 뒤 생산된 IgG1 type의 쥐 단일클론 항체를 정제하기 위하여 음이온 교환 크로마토그래피를 이용하였다. 배양이 끝난 배지를 원심분리하여 세포를 제거하고 50-60% ammonium sulfate로 침전물을 만든 다음 0.025M Tris-HCI(pH8.2)용액으로 투석하여 salt가 제거된 sample을 DEAE-Trisacryl M에 부하하였다. Column에 결합된 항체는 30-40mM NaCl 을 포함하는 0.025M Tris-HCI(pH8.2)용액으로 용출하였다. 혈청농도가 높은 배지 (10% FBS)에서는 50% ammonium sulfate 처리로 90% 이상의 항체가 회수되었으나 저혈청 배지 (2% FBS)에서는 60% ammonium sulfate 처리에도 회수율이 84%에 그쳤다. 후자의 경우 한외여과법 (ultrafiltration)을 이용하여 항체 회수율을 91%까지 증가시킬 수 있으나 농축된 항체를 크로마토그래피로 정제하였을 때 그순도가 ammonium sulfate 침전법에 비해 낮아졌다. 하이브리도마 Alps 25-3, HCGK, A4W, KW를 여러가지 배양조건에서 배양한 뒤 생산된 항체를 DEAE-Trisacryl M chromatography를 이용하여 정제해 본 결과 대체로 순도 70-80%의 항체를 얻을 수 있었고 이때 항체 회수율은 65% 선이었다. 항체의 순도를 높이기 위해서는 affinity chromatography 혹은 gel filtration과 같은 2차적인 방법이 필요 할 것으로 보이며 한 예로 affinity chromatography를 이용하여 순도 95%의 항체를 얻었다.
