  • With a view to obtaining basic informations on the morphological, physiological and ecological aspects of hop in Korea, this experiments were conducted in 4 locations (Suwon, Hongchun, Pyungchang and Hoengsung) from 1972 to 1974. The varieties used in this experiments were Hallertau, Saaz, Shinshuwase and Cascade. the results obtained are summarized as follows.

  • Neuropharmacological profile of Humulus lupulus (hop) extract was studied in mice. At doses above 100 mg/kg(i.p.), it decreased spontaneous locomotor activity and raised the nociceptive threshold in the hot-plate test. At doses above 250 mg/kg (i.p.), it increased pentobarbital-induced sleeping time and produced muscle relaxant effect. At the dose of 500 mg/kg, anticonvulsive effect against pentylenetetrazole-induced convulsion and hypothermic effect was observed.

Assessment of Stability and Safety of Maskne Cosmetic

  • Wearing a mask is still advised since COVID-19 continues to spread. However, masks may also irritate the skin and cause mask acne, often known as "maskne", which is a type of acne mechanica caused by friction between the skin and clothing. Therefore, there is a need to develop an effective maskne cosmetic. In this study, we made the maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 and investigated its stability and safety. To measure stability, a centrifugation test and heat-cool cycling were done, and changes in viscosity and pH were measured for 8 weeks. The Cumulative Irritation Test (CIT, WKIRB-202111-HR-096) was performed and positive reactions were determined by the ICDRG criteria. The results indicated that the samples were stable after centrifugation, temperature cycling, viscosity, and pH tests. In addition, cosmetic safety test results revealed that maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 did not cause any skin responses. These findings indicate that prepared maskne cosmetics' stability and safety were comparable to those of currently available commercial cosmetics.

Inhibition of Matrix Metalloproteinases-12 (MMP-12) and Anti-oxidant Effect of Xanthohumol from Hop (Humulus lupulus L.)

  • Xanthohumol was isolated from hops (Humulus lupulus L.), and then investigated anti-oxidant effect by AAPH-induced LLC-PK1 cell and oxygen radical absorbance capacity (ORCA) assays and MMP-12 inhibitory effect by direct MMP-12 inhibition assay. The treatment of xanthohumol protected LLC-PK1 cells from AAPH-induced cell damage such as cell viability, SOD and GSH-px reduction in a dose dependant manner (0.1, 1, and $5{\mu}M$), the SOD value was 2.98, 4.51, and 5.77 U/mg protein, and GSH-px value was 30.12, 49.32, and 60.11 U/mg protein. ORAC value of xanthohumol was showed as 4320, 12004, and $14209{\mu}M$ TE/g at the concentration 0.1, 1, and $5{\mu}M$, respectively. The change of SOD and GSH-px values was significantly correlated with the results of ORAC assay, that is, AAPH-induced cell and ORCA assays. In addition, inhibition of MMP-12 that is known to play an important role in skin aging was 14%, 37%, 46%, and 79% at the concentration of 0.01, 0.1, 1, and $5{\mu}M$, respectively. On the basis of these results, xanthohumol from hops (Humulus lupulus L.) showed interesting biological and pharmacological activity such as anti-oxidant effect and anti-aging.

홉(Humulus lupulus L.) 조직배양 시 재분화율에 미치는 식물생장조절제의 영향 (The Effect of Plant Growth Regulators on Regeneration Rate during Tissue Culture of Hop (Humulus lupulus L.))

  • 홉은 맥주 생산에서 풍미와 쓴 맛을 제공하며 방부제의 역할을 한다. 최근 수제 맥주 호황에 의해 외국의 홉 품종을 도입하여 국내 재배 및 생산을 도모하고 있으나, 효율적인 무병묘 생산에 있어 어려움을 겪고 있다. 이에 따라 본 연구는 홉(Humulus lupulus L.) Cascade 품종을 대상으로 조직배양 기술을 통한 효율적인 기내 증식 방법과 우량묘 생산기술을 개발하고자 수행되었다. 실험에 사용된 식물 생장조절제는 MS 배지에 auxin 계열 IAA와 cytokinin 계열의 2iP, zeatin, BAP, TDZ을 사용하였다. 식물 재료는 신초를 대상으로 정단에서 3마디를 제거하고 사용하였다. 홉 기내배양 시 재분화율은 IAA만 첨가한 조건에서 가장 높았으며, cytokinin을 함께 사용했을 때 보다 약 21% 높았다. 하지만 IAA 0.1 mg/L + BAP 1 mg/L 조건에서 재분화율이 약 91%로 우세하였다. 초장은 IAA 0.1 mg/L + BAP 1 mg/L를 사용하는 것이 유리하다. TDZ가 첨가된 배지에서 다경 유도가 이루어졌으며, control이나 IAA만 첨가된 조건의 경우 callus 형성이 이루어지지 않고 지상부와 뿌리 생장이 진행되었다. Cytokinin을 첨가한 배지에서 callus가 형성되며 무게가 증가하였다. 홉 조직배양묘의 순화 시 생존율을 높이기 위한 실험 결과, 배양병에 멸균된 상토를 넣고 상대습도를 100%로 맞추어 준 다음점진적으로 기내의 상대습도를 낮추어 가며 21일 이후 야외로 나갔을 때 90%의 생존율을 확인하였다. 이러한 연구 결과는 다른 홉 품종에도 적용하여 각 품종별로 적합한 배지 조성을 찾거나 callus 및 다경 유도를 통한 무병묘 증식이나 육종 연구 등 사용자의 필요에 따라 적합한 조직배양 배지 조성을 찾는데 도움이 될 것으로 판단된다.

  • Hop cones (Humuli Strobili) are the female inflorescences of hop plants (Humulus lupulus L.) belonging to the family Cannabaceae. They have been used as herbal remedies to treat mood and sleep disturbances, and mainly to add as a bittering ingredient in brewing process. Considerable interests on pharmacological and biological activities of hop cones have been focused on their major constituents, namely, phloroglucinols (humulone, lupulone), terpenes (myrcene, humulene), and prenylated flavonoids (xanthohumol, isoxanthohumol, 6-prenylnaringenin, and 8-prenylnaringenin). The present review describes and discusses biological activity profiles of these major compounds in the hop cones.

  • Nutrition influences cancer incidence and offers a variety of preventive dietary factors including non-nutritive plant metabolites. To identify novel potential chemopreventive agents, we have set up cell- and enzyme-based in vitro marker systems relevant for prevention of carcinogenesis in vivo. This experimental approach led to the identification of Xanthohumol (Xn), a prenylated chalcone from hop (Humulus lupulus L.) as a most promising broad-spectrum chemopreventive agent.(omitted)

Optimized mixture of hops rho iso-alpha acids-rich extract and acacia proanthocyanidins-rich extract reduces insulin resistance in 3T3-L1 adipocytes and improves glucose and insulin control in db/db mice

  • Rho iso-alpha acids-rich extract (RIAA) from Humulus lupulus (hops) and proanthocyanidins-rich extracts (PAC) from Acacia nilotica exert anti-inflammatory and anti-diabetic activity in vitro and in vivo. We hypothesized that a combination of these two extracts would exert enhanced effects in vitro on inflammatory markers and insulin signaling, and on nonfasting glucose and insulin in db/db mice. Over 49 tested combinations, RIAA:PAC at 5:1 ($6.25{\mu}g/mL$) exhibited the greatest reductions in $TNF{\alpha}$-stimulated lipolysis and IL-6 release in 3T3-L1 adipocytes, comparable to $5{\mu}g/mL$ troglitazone. Pretreatment of 3T3-L1 adipocytes with this combination ($5{\mu}g/mL$) also led to a 3-fold increase in insulin-stimulated glucose uptake that was comparable to $5{\mu}g/mL$ pioglitazone or $901{\mu}g/mL$ aspirin. Finally, db/db mice fed with RIAA:PAC at 5:1 (100 mg/kg) for 7 days resulted in 22% decrease in nonfasting glucose and 19% decrease in insulin that was comparable to 0.5 mg/kg rosiglitazone and better than 100 mg/kg metformin. RIAA:PAC mixture may have the potential to be an alternative when conventional therapy is undesirable or ineffective, and future research exploring its long-term clinical application is warranted.
