생약재 추출물의 아토피 완화효과 (Effect of Anti-atopic Allergic Reaction in Response to Oriental Herb Extracts)

  • 결명자(Cassia obtusifolia) 및 포공영(Taraxacum platycarpum), 유근피(Ulmus macrocarpa)로 제조된 생약재 추출혼합물이 in vitro와 in vivo상에서 아토피 피부염에 미치는 영향을 실험하였다. HMC-1 cell을 이용하여 비만세포의 $\beta$-hexosaminidase 방출량을 측정한 결과, 단일 추출물보다 추출복합물이 탈과립을 억제하는 효과가 우수한 것으로 측정되었다. 그리고 HMC-1 cell에서 PMA와 A23187로 자극하였을 때 생성되는 염증성 cytokine의 분비에서도 추출혼합물의 저해 효과가 가장 높게 나타났다. 아토피 질환모델인 NC/Nga 마우스를 사용하여 생약재 추출혼합물의 아토피 저감효과를 측정한 결과, 진드기 추출물에 의해 발진된 마우스의 귀두께 및 조직학적인 부종 관찰을 통해 생약재 추출혼합물 농도가 증가함에 따라 대조군에 비해 귀 두께 및 부종의 감소가 관찰되었다. 또한 마우스의 혈중 IgE 농도와 이와 관련된 cytokine의 농도를 측정한 결과, 생약재 추출혼합물 투여농도가 증가함에 따라 IgE의 농도가 유의적으로 감소하였고, IgE의 과발현을 유도하는 IL-4의 분비 억제 및 IFN-$\gamma$의 분비가 증가되는 것으로 나타나 생약재 혼합추출물의 섭취가 아토피 피부염 증세 완화 효과에 도움이 되리라 사료된다.

  • Food-dependent exercise-induced anaphylaxis (FDEIAn) is induced by different types and various intensities of physical activity, and is distinct from food allergies. It has been shown that consumption of allergenic food followed by exercise causes FDEIAn symptoms. Intake of allergenic food or medication before exercise is a major predisposing factor for FDEIAn. Urticaria and severe allergic reactions are general symptoms of FDEIAn. Dermatological tests and serum IgE assays are the typical prescreening methods, and have been used for several decades. However, these screening tests are not sufficient for detecting or preventing FDEIAn. It has been found that exercise may stimulate the release of mediators from IgE-dependent mast cells that can result in FDEIAn when a certain threshold level has been exceeded. Mast cell degradation might be a major factor to induce FDEIAn but this has not been determined. A number of foods have been reported to be involved in the onset of FDEIAn including wheat, eggs, chicken, shrimp, shellfish, nuts, fruits, and vegetables. It is also known that aspirin increases the occurrence of type I allergy symptoms when combined with specific foods. Moreover, high intensity and frequent exercise are more likely to provoke an attack than low intensity and less frequent exercise. In this paper, we present the current views of the pathophysiological mechanisms underlying FDEIAn within the context of exercise immunology. We also present a detailed FDEIAn definition along with etiologic factors and medical treatment for cholinergic urticaria (UC) and exercise-induced anaphylaxis (EIA).

고삼(苦蔘)에탄올 추출물이 $NF{\kappa}B$ 및 JNK, p38 조절을 통한 알레르기성 염증에 미치는 영향 (The Effect of Allergic Inflamation by Sophora Flavescens Aiton Extract Ion Through Inhibition of the $NF{\kappa}B$, JNK and p38 Pathway)

  • 1. Objectives The roots of Sophora flavescens Aiton (SFA) are widely used as a herbal remedy for allergic inflammation. In this study, we invested the effect of SFA on passive cutaneous anaphylaxis reaction and histamin releas and we demonstrated that SFA suppressed the production of pro-inflammatory cytokines, such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin- 6 (IL-6), and interleukin -8 (IL-8), through inhibition of the $NF{\kappa}B$, JNK and p38 pathway in the human mast cell line, HMC-1. 2. Methods To accomplish this, we invested passive cutaneous anaphylaxis reaction and histamin release at an animal experiment. In addition, we investigated the effect of SFA on the production of inflammation-related cytokines in HMC-1 cells that were co-treated with PMA and A23187, which can induce production of pro-inflammatory cytokines. 3. Results and Conclusions SFA induced passive cutaneous anaphylaxis reaction and histamin releas and supressed the expression of TNF-${\alpha}$, IL-6, and IL-8. In addition, the protein levels of TNF-${\alpha}$ were also decreased by SFA treatment. Furthermore, SFA inhibited the nuclear translocation of nuclear factor $NF{\kappa}B$ through inhibition of the phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which is an inhibitor of $NF{\kappa}B$. Moreover, SFA also inhibited induction of MAPKs (JNK, p38) and $NF{\kappa}B$ promoter-mediated luciferase activity. Taken together, these results suggest that SFA could be used as a treatment for mast cell-derived allergic inflammatory diseases.

Inhibition of Chitinase-3-like-1 by K284-6111 Reduces Atopic Skin Inflammation via Repressing Lactoferrin

  • Chitinase-3-like-1 (CHI3L1) is known to induce inflammation in the progression of allergic diseases. Previous our studies revealed that 2-({3-[2-(1-cyclohexen-1-yl)ethyl]-6,7-dimethoxy-4-oxo-3,4-dihydro-2-quinazolinyl}sulfanyl)-N-(4-ethylphenyl)butanamide (K284-6111; K284), the CHI3L1 inhibiting compound, has the anti-inflammatory effect on neuroinflammation. In this study, we investigated that K284 treatment could inhibit the development of atopic dermatitis (AD). To identify the effect of K284, we used phthalic anhydride (5% PA)-induced AD animal model and in vitro reconstructed human skin model. We analyzed the expression of AD-related cytokine mediators and NF-κB signaling by Western blotting, ELISA and quantitative real-time PCR. Histological analysis showed that K284 treatment suppressed PA-induced epidermal thickening and infiltration of mast cells. K284 treatment also reduced PA-induced release of inflammatory cytokines. In addition, K284 treatment inhibited the expression of NF-κB activity in PA-treated skin tissues and TNF-α and IFN-γ-treated HaCaT cells. Protein-association network analysis indicated that CHI3L1 is associated with lactoferrin (LTF). LTF was elevated in PA-treated skin tissues and TNF-α and IFN-γ-induced HaCaT cells. However, this expression was reduced by K284 treatment. Knockdown of LTF decreased the expression of inflammatory cytokines in TNF-α and IFN-γ-induced HaCaT cells. Moreover, anti-LTF antibody treatment alleviated AD development in PA-induced AD model. Our data demonstrate that CHI3L1 targeting K284 reduces AD-like skin inflammation and K284 could be a promising therapeutic agent for AD by inhibition of LTF expression.

장류 및 젓갈 분리 균주 추출물의 비만세포 매개 항염증효과 (Anti-Allergic Inflammatory Effect of Bacteria Isolated from Fermented Soybean and Jeotgal on Human Mast Cell Line (HMC-1))

  • 본 연구에서 장류 및 젓갈 유래 균체를 각각 sonication, $100^{\circ}C$에서 30 min, $125^{\circ}C$에서 15 min의 조건으로 추출하여 HMC-1에 처리하였을 때, 이 추출방법에 따른 HMC-1매개 염증반응 억제효과를 조사하였다. 추출방법을 달리하여 추출한 장류 및 젓갈 유래 균체 J80, G147이 HMC-1생존율에 미치는 영향은 대조구 B (Not treated)와 비교하였을 때 86~96%로 유의적 차이를 보이지 않아 장류 및 젓갈 유래 균체에 의한 세포 독성은 나타나지 않았다. 장류 및 젓갈 유래 균체의 모든 전처리구에서 histamine 유리 억제효과를 나타내었으며 특히, $100^{\circ}C$에서 30 min 동안 가열하였을 때 histamine 억제율이 J80에서 28.86%, G147에서 41.14%으로 가장 높게 나타냈다. IL-8 분비량은 J80, G147의 모든 전처리구에서 IL-8 분비억제효과가 나타났으며, J80의 경우 $100^{\circ}C$, 30 min에서 처리한 구에서 분비량이 0.78 ng/ml로 대조구 B (Not treated)에 비하여 약 28.45%의 분비억제효과로 가장 높게 나타내었다. G147의 경우 $125^{\circ}C$, 15 min에서 처리한 구에서 분비량이 0.73 ng/ml로 약 36.52%의 분비억제효과로 가장 높게 나타내었다. IL-6 분비량을 측정한 결과 J80, G147의 모든 실험구에서 IL-6 분비억제효과가 나타났으며, J80에서는 $100^{\circ}C$, 30 min한 D구의 분비량이 0.40 ng/ml으로 대조구 B (Not treated)에 비하여 약 38.46%의 분비억제효과로 가장 높았으며, G147 역시 $100^{\circ}C$, 30 min 동안 처리한 D구의 분비량이 0.27 ng/ml으로 대조구 B (Not treated)에 비하여 56.45%의 분비억제효과로 가장 높게 나타냈다. TNF-${\alpha}$ 분비량 역시 IL-6와 마찬가지로 J80, G147에서 대조구 B보다 낮은 분비량으로 TNF-${\alpha}$ 분비 억제효과가 나타났으며 J80의 경우 $100^{\circ}C$, 30 min 처리한 D의 TNF-${\alpha}$ 분비량이 0.052 ng/ml으로 66.67%의 분비억제효과로 가장 높았으며, G147의 경우 sonication을 처리한 C구의 TNF-${\alpha}$ 분비량이 0.086 ng/ml으로 41.1%의 분비억제효과로 다른 구에 비해 높은 억제효과를 나타내었다. 위의 결과를 통해서 장류 및 젓갈 유래 균체의 3가지 전처리 방법 중 $100^{\circ}C$, 30 min 처리 시 염증 반응 억제 효과가 가장 높을 것으로 예상된다. 결과를 미루어보아 장류를 섭취하는 대표적인 방법인 가열하여 찌개로 조리과정 중 장류 및 젓갈 속의 균체가 열에 가열되면서 염증억제효과 높일 수 있는 가장 좋은 조리 방법으로 사료되며, 이는 대중들이 가장 가깝게 염증반응 억제효과를 볼 수 있는 식품일 것이다. 또한 염장처리과정만을 거친 젓갈을 섭취하는 것보다 열처리과정을 거친 젓갈을 섭취할 경우 염증억제효과를 더 높일 수 있을 것으로 예상된다.

Socioeconomic impact of traditional Korean medicine, Pyeongwee-San (KMP6) as an anti-allergic inflammatory drug

  • The prevalence of allergic disease has been increasing over the past few decades in the majority of Western industrialized nations. There are some socioeconomic disparities regarding allergic disease status and management. Pyeongwee-San (KMP6) is Korean medicine for the treatment of gastrointestinal tract disease. It is known that KMP6 has an improving effect on the spleen and stomach functions in traditional Korean medical theory. Here, we hypothesized that KMP6 could be used to regulate the inflammatory reaction. We show the molecular mechanisms of Pyeongwee-San (KMP6) on inflammatory reactions. A molecular docking simulation showed that hesperidin, component of KMP6, regulate the enzymatic activity by interaction in the active site of caspase-1. KMP6 control the activity of caspase-1 in activated human mast cell line (HMC-1 cells). KMP6 reduced the expression of receptor interacting protein (RIP)-2 in HMC-1 cells. Thymic stromal lymphopoietin protein production and mRNA expression were inhibited by KMP6. In the activated HMC-1 cells, KMP6 suppressed the activation of mitogen-ativated protein kinase and nuclear factor-kappaB. In addition, KMP6 significantly inhibited the expression of inflammatory cytokines. Our findings indicate that KMP6 may attenuate allergic reactions via the regulation of caspase-1/RIP-2 signaling pathway. These studies will help advance the social welfare system.

구풍제습탕(驅風除濕湯)이 DNCB로 유도된 NC/Nga mice의 아토피 피부염에 미치는 영향 (Effect of Gupoongjeseuptang (GPJST) on DNCB (dinitrochlorobenzene)-induced Atopic Dermatitis-like Model NC/Nga Mice)

  • Objectives : The purpose of this study is to investigate the effect of Gupoongjeseuptang (GPJST) on atopic dermatitis by in vivo experiment using NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to the atopic dermatitis of human. Methods : To investigate the effect of GPJST on atopic dermatifis, we evaluated atopic dermatitis-like skin lesions by clinical skin index and analyzed immunological parameters in peripheral blood mononuclear cells (PBMCs), splenocytes, draining lymph node (DLN) and performed skin histology in ears and dorsal skin of atopic dermatitis-like skin NC/Nga mouse in vivo. Results : In vivo, clinical skin severity score were significantly lower in GPJST group than control group. IgE, IL-6, $TNF-{\alpha}$, IgG1, IgM, IgG2a and IgG2b levels in serum decreased remarkably in GPJST group than control group. Also, total absolute number of $CD3^+CD69^+$, and $CCR3^+$ cells recovered as normal in PBMCs and $CD3^+$, $CD3^+CD69^+$ decreased significantly compared with control group in isolated DLN from NC/Nga mouse and total absolute number of $CD11b^+Gr-1^+$, $CCR3^+CD3^+$ in dorsal skin of NC/Nga mouse decreased by GPJST. We analyzed ear and neck-back skin after biopsy and dyeing by hematoxyline/eosin (H&E) and toluidine staining (mast cells marker) and obtained results that GPJST are very effective to histological symptoms (dermal and epidermal thickening, hyperkeratosis and inflammatory cell (CD4, $CCR3^+$) infiltration). Conclusions : This study demonstrates immunological activity of GPJST on atopic dermatitis-like model mice.

수종의 청열약물의 미백 및 항알러지 효과에 대한 실험적 연구 (Experimental studies on the Whitening and Anti-allergic effect of various Herbal medicines that clear heat)

  • Objective : In this study, herbal medicine(GJE, Gardenia jasminoides Ellis; HCT, Houttuynia cordata Thunb.; CIL, Chrysanthemum indicum Linne; PMS,Paeonia moutan Sims, P. subfruticosa Makino; APL, Agrimonia pilosa Ledebour) were screened for their inhibitory activities against Tyrosinase and PMA plus A23187-induced $TNF-{\alpha}$, IL-6, IL-8 productions in HMC-1 cells to reveal their skin -whitening and anti-allergic effect. Method : To investigate Tyrosinase inhibition we treated Mushroom Tyrosinase(Fluka, 93898) $10{\mu}{\ell}$ and 7.5mM Tyrosine (Sigma, T3754) $20{\mu}{\ell}$ with 80% ethanol medicine extracts. Then we observed 96well micro plate extinction at 490nm. In the next experiment, to investigate Anti-allergic effect we blended cultured Human Mast Cells(HMC-1) with medicine extracts. We treated the blended solution with Phorbol 12-myristate 13-acetate(PMA) and A23187, then observed $TNF-{\alpha}$, IL-6, IL-8 by ELISA (enzyme-linked immunosorbent assay) at 450nm. Results : In inhibiting Tyrosinase the results are as follows. 1. We observed 22% inhibition of Mushroom Tyrosinase at $500{\mu}g/m{\ell}$ concentration of GJE extracts. 2. We also could observe that the decreased Mushroom Tyrosinase activities in HCT, CIL extracts. In inhibiing $TNF-{\alpha}$, IL-6, IL-8 productions in HMC-1 cells the results are as follows. 1. Of the extracts examined, HCT, PMS, APL extracts showed over 50% inhibitions of Cytokines at $200{\mu}g/m{\ell}$ concentration. 2. In particular, APL extracts showed the best inhibitory effect on Cytokine productions in a dose-dependent manner. Conclusion : These results suggest that GJE extracts contributes to the anti melanin activities and represent a potential source of whitening agent. Thus these herbal medicines suggest novel drugs on anti-allergic effects.

Anti-allergic effects of Perilla frutescens var. acuta Kudo 30% ethanol extract powder

  • Perilla frutescens var. acuta Kudo (PF) is a traditional Korean medicinal herb for allergic reaction regulation. In the present study, we investigated the effect of 30% ethanol extract powder of PF (EPPF) and rosmarinic acid (RA), the active compound of EPPF on various allergic reactions using in vivo and in vitro models. EPPF and RA significantly inhibited compound 48/48-induced systemic anaphylactic reaction and histamine release (P < 0.05). In addition, EPPF and RA significantly inhibited passive cutaneous anaphylaxis (PCA) in a dose-dependent manner (P < 0.05). These effects were stronger than those of disodium cromoglycate, the reference drug tested. EPPF and RA also significantly inhibited production of inflammatory cytokines, tumor necrosis factor-a interleukin (IL)-6, and vascular endothelial growth factor on the PCA reaction and phorbol 12-myristate 13-acetate and calcium ionophore A23187-stimulated human mast cell line, HMC-1 cells (P < 0.05). Moreover, EPPF and RA showed an inhibitory effect on lipopolysaccharide (LPS)-induced IL-4 production from whole spleen cells. Finally, EPPF and RA significantly decreased IL-4-dependent IgE production by LPS-stimulated whole spleen cells (P < 0.05). In conclusion, these results indicate that EPPF has potent anti-allergic activities.

Safety Evaluation of Topical Valproate Application

  • The potential role of topical valproate (VPA) in hair regrowth has been recently suggested. However, safety reports of VPA as a topical formulation are lacking. Therefore, in the present study, we investigated whether VPA causes skin irritation in humans. We first performed a cell viability test and showed that VPA did not exhibit toxicity toward HaCaT keratinocytes, fibroblasts, and RBL-3H mast cells. We then performed clinical patch test and skin irritation test through transdermal drug delivery with the help of microneedle rollers. No significant findings were obtained in the clinical patch test. In the skin irritation test, only 1 patient showed erythema at 1 hr, but the irritation reaction faded away within a few hours. Erythema and edema were not observed at 24 hr. We concluded that VPA has minimal potential to elicit skin irritation. Therefore, we consider that VPA can safely be applied to human skin.
