Carbon nanotubes (CNTs) stand at the frontier of nanotechnology and are destined to stimulate the next industrial revolution. Rapid increase in their production and use in the technology industry have led to concerns over the effects of CNT on human health and the environment. The prominent use of CNTs in biomedical applications also increases the possibility of human exposure, while properties such as their high aspect ratio (fiber-like shape) and large surface area raise safety concerns for human health if exposure does occur. It is crucial to develop viable alternatives to in vivo tests in order to evaluate the toxicity of engineered CNTs and develop validated experimental models capable of identifying CNTs' toxic effects and predicting their level of toxicity in the human respiratory system. Human lung epithelial cells serve as a barrier at the interface between the surrounding air and lung tissues in response to exogenous particles such as air-pollutants, including CNTs. Monolayer culture of the key individual cell types has provided abundant fundamental information on the response of these cells to external perturbations. However, such systems are limited by the absence of cell-cell interactions and their dynamic nature, which are both present in vivo. In this review, we suggested two viable alternatives to in vivo tests to evaluate the health risk of human exposure to CNTs.
Purpose: Measuring viability of a three-dimensional in vitro organotypic human oral tissue model has been suggested as an alternative test method to the oral mucosa irritation test of oral care products. The aim of this study was to investigate the production of two different cytokines using organotypic human oral tissue model following exposure to chemicals that are commonly used in oral care products. Materials and Methods: The organotypic human oral tissues were exposed to ethanol, sodium lauryl sulphate or hydrogen peroxide for 90 minutes. Following exposure, interleukin (IL)-1α and IL-8 productions were assessed and correlated with cell viability testing as well as histology of the organotypic human oral tissues. Result: High levels of IL-8 were released from organotypic human oral tissues in all of the test and control groups without any significant differences between them. In contrast, differences were found in IL-1α release between the test and control groups. Additionally, the trend of IL-1α release corresponded to the phenotypes observed in histological analysis while different trend existed between IL-1α release and cell viability. Conclusion: The study concluded the non-specific release of IL-8 for the assessment of oral care product chemicals' toxicity, while potential of measuring IL-1α cytokine level as the possible alternative test method.
Background: Arsenic is a metalloid of public health significance due to its unique material properties and toxicity and the widespread pollution in the environment. Arsenic exists as inorganic arsenic and organic arsenic with distinct chemical properties. Its toxicity varies depending on the properties. Objectives: Although the carcinogenicity of arsenic has been identified, the various diseases that occur after acute and chronic exposure to arsenic are not yet clearly known. Methods: Research on the effects of chronic exposure to arsenic on human health was searched and the results were summarized. Results: It has been found that cancer occurs due to exposure to high concentrations of arsenic in areas with elevated exposure to arsenic, but research results have recently been presented on health effects caused by chronic exposure to low concentrations of arsenic. Cancers have also been identified to be related to inorganic arsenic, including skin cancer, lung cancer, and bladder cancer. Significant relationships with neurological diseases, cardiovascular diseases, and diabetes mellitus have been suggested as well. Conclusions: Our results suggest that it is necessary to evaluate the health impact on residents around abandoned metal mines and industrial complexes in South Korea.
Cytokines are hormone-like proteins which mediate and regulast inflammatory and immune responses. Transforming growth factor -$\beta$1(TGF-$\beta$) plays an important role in the control of the immune response and wound healing, and in the development o various tissues and organs, Nitric oxide(NO) is major messenger molecule regulating immune function and blood vessel dilation and serving as a neurotransmitter in the brain and peripheral nervous system. Also, NO is to be a potent mutagen that cause mutation in the p53 tumor suppressor gene in early phases of human gastric carcinogenesis. The purpose of this study was to investigate the effect of Helicobacter phlori lystes, lipopolysaccharide (LPS), and Staphylococcus enterotoxin B(SEB) on production of TGF-$\beta$1 and NO by human fibroblasts. Primary cultured human fibroblasts were incubated with H. pylori lysates(Hp), LPs, SEB, Hp+LPS, Hp+SEB, Hp+LPS+SEB. Cultured supernatants that were collected at 24, 48 and 72 hr were assessed for TGF-$\beta$1 by enzyme-linked immunosorbent assay and NO production by quantification of nitrite ion. TGF-$\beta$1 production in fibroblasts exposed with Hp, LPS or SEB for 48 hrs was enhanced, but for 72 hrs inhibited. Its production by doble exposure such as Hp+LPS, Hp+SEB, Hp+LPS+SEB was lowered in comparison with single exposure of Hp in cases of 24 and 48 hrs incubation, but for 72 hrs decreased in Hp vaculoating toxin(+), increased in Hp vacuolating toxin(-). No production in fibroblasts increaed at all doses of LPS. But its production by exposure of SEB increased or decreased according to dose and incubation time. Also, NO production by Hp vacuolating toxin(+) increased at all doses, but its production by Hp vacuolating toxin(-) decreased. Its production by doble exposure such as Hp+LPS, Hp+SEB, Hp+LPS+SEB decreased in comparison with single exposure Hp Therefore, quantities pf TGB-$\beta$1 and NO released by human fibroblasts shows differences according to kinds of stimulants. Also, in care stimulated with same kinds of stimulants, its productions exhibit quantitative differences according to exposure times. These results suggest that the decreased of TGF-$\beta$1 in fibroblasts by mixed exposure with Hp producing vacuolating toxin and bacterial toxins such as LPS and SEB may effect negatively in healing of host tissue and increased of NO by infection oh H. pylori may related to the increased susceptibility for human gastric carcinogenesis.
Background: Chemical emissions in the environment have rapidly increased with the accelerated industrialization taking place in recent decades. Residents of industrial complexes are concerned about the health risks posed by chemical exposure. Objectives: This study was performed to suggest modeling methods that take into account multimedia and multi-pathways in human exposure and risk assessment. Methods: The concentration of benzene emitted at industrial complexes in Daesan, South Korea and the exposure of local residents was estimated using the Caltox model. The amount of human exposure based on inhalation rate was stochastically predicted for various activity stages such as resting, normal walking, and fast walking. Results: The coefficient of determination (R2) for the CalTOX model efficiency was 0.9676 and the root-mean-square error (RMSE) was 0.0035, indicating good agreement between predictions and measurements. However, the efficiency index (EI) appeared to be a negative value at -1094.4997. This can be explained as the atmospheric concentration being calculated only from the emissions from industrial facilities in the study area. In the human exposure assessment, the higher the inhalation rate percentile value, the higher the inhalation rate and lifetime average daily dose (LADD) at each activity step. Conclusions: Prediction using the Caltox model might be appropriate for comparing with actual measurements. The LADD of females was higher ratio with an increase in inhalation rate than those of males. This finding would imply that females may be more susceptible to benzene as their inhalation rate increases.
There are numerous and evidential findings that TCDD (2, 3, 7, 8-tetrachlorodibenzo-Pdioxin, or dioxin) is a potential carcinogen and general toxin in rodents. flowever, human risk assessment for dioxin exposure has been a topic of debate, owing in part to the large animal interspecies differences in its toxicity. We review dioxin-related reports indicating its toxicity, toxic effects in animal, and human epidemiologic findings. The intent of this paper does not provide a causal inference about chronic human diseases related to dioxin exposure. This summary would give a valuable clue for a researcher to conduct or design a further dioxin-related study.
The purpose of this study was to investigate consumer competencies of urban wives an variables on it. The framework of this study was consumer socialization theory. The results were as follows; 1. There were significant differences in competencies of urban wives according to employment status, interaction with family members, interaction with peers and frequency of media exposure. 2. Employment status, interaction with family members and frequency of media exposure were independent influencing factors on consumer competencies of urban wives. 3. Reviewing a causal model, the results were as follows. Benzene and ethylbenzene (BE), the volatile organic compounds (VOCs) are common constituents of cleaning and degreasing agents, paints, pesticides, personal care products, gasoline and solvents. VOCs are evaporated at room temperature and most of them exhibit acute and chronic toxicity to human. Chronic exposure of benzene is responsible for myeloid leukemia and also ethylbenzene is also recognized as a possible carcinogen. To evaluate the BE effect on human, whole human genome 35 K oligonucleotide microarray were screened for the identification of the differential expression profiling. We identified 280 up-regulated and 201 down-regulated genes changed by more than 1.5 fold by BE exposure. Functional analysis was carried out by using DAVID bioinformatics software. Clustering of these differentially expressed genes were associated with immune response, cytokine-cytokine receptor interaction, toll-like signaling pathway, small cell lung cancer, immune response, apoptosis, p53 signaling pathway and MAPKKK cascade possibly constituting alternative or subordinate pathways of hematotoxicity and immune toxicity. Gene ontology analysis methods including biological process, cellular components, molecular function and KEGG pathway thus provide a fundamental basis of the molecular pathways through BEs exposure in human lymphoma cells. This may provides a valuable information to do further analysis to explore the mechanism of BE induced hematotoxicity.
This study assessed the stabilization of fluorine (F)-contaminated soil using calcium hydroxide (Ca(OH)2) and the consequent changes in human health risk. The bioavailable F decreased to 3.5%, (i.e., 57.9 ± 1.27 mg/kg in 6% Ca(OH)2-treated soil sample) from 43.0%, (i.e., 711 ± 23.4 mg/kg in control soil sample). This resulted from the conversion of water-soluble F to stable calcium fluoride, which was confirmed by XRD spectrometry. Soil ingestion, inhalation of fugitive dust from soil, and water ingestion were selected as exposure pathways for human health risk assessment. Non-carcinogenic risks of F in soils reduced to less than 1.0 after stabilization, ranging from 4.2 to 0.34 for child and from 3.0 to 0.25 for adult. Contaminated water ingestion owing to the leaching of F from soil to groundwater was considered as a major exposure pathway. The risks through soil ingestion and inhalation of fugitive dust from soil were insignificant both before and after stabilization, although F concentration exceeded the Korean soil regulatory level before stabilization. Our data suggested that substantial risk to human health owing to various potential exposure pathways could be addressed by managing F present in soil.
There has been an increase in the public concern about possible health risks by electromagnetic exposure from mobile phones. Recently, increase in the use among young children of portable telephones public concern regarding potential health hazards due to a hot spot appearing inside the infant head, has been growing. Since the biological hazards due to RF exposure are caused mainly by a temperature-rise in tissue, the effect of localized SAR for portable telephones should also be related to the temperature-rise in the human head. In this paper, it was measured that in the actual use of portable telephone the temperature of the local tissue in the human-head change. As a result, it should be noted that the mean temperature of human-body and localized tissue is rising from beginning call. However the temperature variation of localized tissue is recovered rapidly as normal temperature, although the mean temperature of human-body rising continuously at ending call of portable telephone.
