A Study on the Composition and Enzymatic Hydrolysis of Some Agricultural Waste Products (농산폐기물(農産廢棄物)의 성분분석(成分分析) 및 효소분해(酵素分解)에 관(關)한 연구(硏究))

  • For the purpose of utilizing agricultural waste products for animal feeds, chemical analysis of some important agricultural wastes in Korea and enzymatic hydrolysis of rice hull were undertaken and summarized as follows: 1) Rice hull and straws of rice, barley and wheat were analyzed for their proximate principles and carbohydrate composition. 2) A strain of Aspergillus niger was selected as having the highest productivity of rice hull decomposing activity among many species of molds. The optimum conditions for the culture on washed wheat bran were. 3 days of culture period, 55% moisture content and neutral pH and one hour was proper for the extraction of enzymes. 3) The extent of hydrolysis of xylan and rice hull by a crude enzyme preparation from this strain was 63% and 7%, respectively, and the optimum pH for its activity was 4.0 and 4.5, respectively. 4) Enzymatic hydrolysis of rice hull was generally suppressed by gamma-ray irradiation ($0.5{\sim}5\;Mrad$, in dry state), but the effect was less by irradiation in wet state.

  • For the production of microbial cells from cellulosie materials by cellulore-assimilating bacteria, Cellulomonas flavigena GFB 24-1, isolated by authors, utilization of this organism on some microbiological properties was investigated. The results of these studies were summarized at follows; 1. When the organism was incubated in the growth medium at pH 7.0 for 50 hours, its growth was the most effective and the level of excreted total protein in the menstruum increased continuously during the stationary phase of cell growth. 2. The optimal enzyme activity was observed in the pH region of 5 to 7 and culture period of 40 to 50 hours. 3. The microbial degestibility of cellulosic wastes such as sawdust, rice hull, rice straw, peanut hull and used newspaper was less than 30%, whereas that of cellulose powder was 47.1% and rice straw was digested 77% by NaOH treatment. 4. Bacterial cells incubated in the growth medium were increased up to 8% of sustrate concentration and showed a decrease on further concentration. 5. The production of microbial cells from NaOH treated rice straw was obtained 10.6mg/ml of culture medium.

Effect of KOH Concentrations and Pyrolysis Temperatures for Enhancing NH4-N Adsorption Capacity of Rice Hull Activated Biochar (KOH 농도 및 탄화온도가 왕겨 활성 바이오차의 NH4-N 흡착능 향상에 미치는 영향)

  • BACKGROUND: Recently, biomass conversion from agricultural wastes to carbon-rich materials such as biochar has been recognized as a promising option to maintain or increase soil productivity, reduce nutrient losses, and mitigate greenhouse gas emissions from the agro-ecosystem. This experiment was conducted to select an optimum conditions for enhancing the NH4-N adsorption capacity of rice hull activated biochar. METHODS AND RESULTS: For deciding the proper molarity of KOH for enhancing its porosity, biochars treated with different molarity of KOH (0, 1, 2, 4, 6, 8) were carbonized at 600℃ in the reactor. The maximum adsorption capacity was 1.464 mg g-1, and an optimum molarity was selected to be 6 M KOH. For the effect of adsorption capacity to different carbonized temperatures, 6 M KOH-treated biochar was carbonized at 600℃ and 800℃ under the pyrolysis system. The result has shown that the maximum adsorption capacity was 1.76 mg g-1 in the rice hull activated biochar treated with 6 M KOH at 600℃ of pyrolysis temperature, while its non-treated biochar was 1.17 mg g-1. The adsorption rate in the rice hull activated biochar treated with 6 M KOH at 600℃ was increased at 62.18% compared to that of the control. Adsorption of NH4-N in the rice hull activated biochar was well suited for the Langmuir model because it was observed that dimensionless constant (RL) was 0.97 and 0.66 at 600℃ and 800℃ of pyrolysis temperatures, respectively. The maximum adsorption amount (qm) and the bond strength constants (b) were 0.092 mg g-1 and 0.001 mg L-1, respectively, for the rice hull activated biochar treated with 6 M KOH at 600℃ of pyrolysis. CONCLUSION: Optimum condition of rice hull activated biochar was 6M KOH at 600℃ of pyrolysis temperature.

  • The buffer capacity of food waste lowers during the collecting and transportation period. Food waste usually shows deficiency of micro nutrients especially molybdenum(Mo) and cobalt(Co). Therefore, food waste can be considered as a good mixture of livestock waste to enhance methane production. The objective of this study was to investigate the correlation between properties of substrates (local food waste and livestock manure) and methane yields for successive anaerobic fermentation process and its stable management. Food wastes were taken at an intermediate storage or treatment system provided by eight local authorities (Gangnam, Gangdong, Gwanak, Guro, Dongjak, Songpa, Yeongdeungpo, and Younsan) in Seoul. The solid content and potential methane yield of food wastes were average of 16% and $446.6STP-m{\ell}/g-VS$ (range from 334.8 to $567.5STP-m{\ell}/g-VS$) respectively. As for the beef cattle manure, the solid content and potential methane yield had an average of 26% and $280.6STP-m{\ell}/g-VS$ respectively. Potential methane yield had a positive correlation with fat content, and hydrogen content and a negative correlation with carbohydrate content ($r^2>0.8$). Therefore, the potential methane yield can be predicted based on the substrate characterization results with reasonable accuracy. Further research may be needed to investigate the relation of the properties of the mixture substrate and methane production rate. The mixtures may include food waste, livestock waste, and bulking agents (saw dust, rice hull, or agricultural byproducts etc.) to determine best combination of these substrates for maximum methane production rate.

Development of Substrates for the Production of Basidiocarps of Flammulina velutipes (팽나무버섯 자실체 생산을 위한 기질개발)

  • Agricultural and forest wastes were tested as substrate for the production of Flammulina velutipes. Among the meranti, pine tree, mixed sawdust, coffee waste and peanut hull tested, coffee waste was the best basal substrate for the production of basidiocarps. When various supplements, such as ricebran, brewer's grain, defatted corn flour, defatted soybean flour and defatted rapeseed flour were tested, defatted corn flour was chosen for the production of basidiocarps. Maximum yield of basidiocarps(129.38 g/bottle) was obtained from the mixed medium at 4 : 1 ratio of coffee waste and defatted corn flour.

Study on the Activation Plan for Utilization of Agri-food by-products as Raw Materials for TMR (TMR 원료로 이용하는 농식품 부산물 사료 이용 활성화 방안에 관한 연구)

  • This study was conducted to survey and analyze the quantity of various organic wastes and to vitalize the utilization of agri-food by-products as raw materials for Total mixed ration (TMR), to improve feed cost savings and the quality of animal products. On-the-spot obstacles for animal farmers, along with legal and institutional alternatives are presented. The results are as follows. First, organic wastes in Korea are managed by the Allbaro system created in the Wastes Control Act, which processes 10,488 tons of cooking oil waste, 832,493 tons of animal and plant residues, 5,740 tons of animal carcasses, 1,171,892 tons of animal residues, and 2,172,415 tons of plant residues including 12,905 tons of rice hull and bran, for a total of 4,205,931 tons. Raw materials for TMR, namely rice hulls and bran as well as plant residues, accounted for 51.7% of the total national organic waste. The top 10 municipalities process 76~100% of all organic wastes and a supply management system is needed for the waste. Second, the 10 major agri-food by-products used as raw materials for TMR are bean curd by-product, rice bran, oil-cake, brewers dried grain, Distiller's Dried Grains with Solubles (DDGS), barley bran, soy sauce by-product, citrus fruit by-product, mushroom by-product and other food by-product (bread, noodles, snacks, etc.). Third, the biggest difficulties in using agri-food by-products are legal obstacles. Because agri-food by-products are regulated as industrial wastes by the Waste Control Act, animal farmers that wish to use them have legal reporting obligations including the installation of recycling facilities. To enable the use of agri-food by-products as raw materials for TMR, waste management system improvements such as 'the end of waste status' and the establishment of more than 10 public distribution centers nationwide are deemed essential.

Effect of AlF3 on Zr Electrorefining Process in Chloride-Fluoride Mixed Salts for the Treatment of Cladding Hull Wastes (폐 피복관 처리를 위한 염소계-불소계 혼합용융염 내 지르코늄 전해정련공정에서 삼불화알루미늄의 효과 연구)

  • Zr electrorefining is demonstrated herein using Zirlo tubes in a chloride-fluoride mixed molten salt in the presence of $AlF_3$. Cyclic voltammetry reveals a monotonic shift in the onset of metal reduction kinetics towards positive potential and an increase in intensity of the additional peaks associated with Zr-Al alloy formation with increasing $AlF_3$ concentration. Unlike the galvanostatic deposition mode, a radial plate-type Zr growth is evident at the top surface of the salt during Zr electrorefining at a constant potential of -1.2 V. The diameter of the plate-type Zr deposit gradually increases with increasing $AlF_3$ concentration. Scanning electron microscopy-energy-dispersive X-ray spectroscopy (SEM-EDX) and X-ray photoelectron spectroscopy (XPS) analyses for the plate-type Zr deposit show that trace amount of Al is incorporated as Zr-Al alloys with different chemical compositions between the top and bottom surface of the deposit. Addition of $AlF_3$ is effective in lowering the residual salt content in the deposit and in improving the current efficiency for Zr recovery.

Effects of Different Substrate on the Growth and Microstructure of Fruit Body in the Basidiomycetes, Pleurotus ostreatus. (느타리버섯 자실체 생육 및 미세구조에 미치는 배지의 영향)

  • Comparison of fruit bodies of Pleurotus ostreatus cultivar chunchu No .2 grown on the sawdust, rice straw and cotton waste substrates revealed differences in the pattern of differentiation of hyphal compartments. Required period for primordium induction of fruit bodies grown on sawdust substrate was 13 days. Physical structure shown as hardness of stipes grown on the sawdust substrate, fruit bodies were harden than control. Pileocystidia were well developed on the surface of pileus in the fruit body cultivated on rice straw. Microstructures of fruit body grown on the sawdust and cotton wastes substrates shown fast-discharge of basidiospore and sytoms ageing. Hyphae of fruit bodies formed on sawdust substrate had less stainable cytoplasmic material and many more vacuoles than hyphae of fruit bodies formed on synthetic substrate with 50% of pine sawdust, 30% of cotton seed hull and 20 of beet pulp(control).

Effect of Artificial Zeolite on Fermentation and Emission of Ammonia and Methane during Animal Waste Composting (인공제올라이트 처리가 가축분 퇴비의 발효 및 암모니아, 메탄가스 발생에 비치는 영향)

  • This study was carried out to investigate the influence of artificial zeolite on the change of temperature, gas emission, water content and chemical properties during the composting process with the mixture of animal feces, broken bark and extruded rice hull. Artificial zeolite was added 0, 0.5, 1, 3 and 5% volume of the raw composting material, and proceeded 1.2m every day with mobile stacking escalator. Temperature was increased, and water content was decreased in the composting pile by addition of artificial zeolite. This caused to accelerate decomposition of organic matter during composting. $NH_3$ was emitted the highest at 6th day after stacking, then decreased gradually. And addition of artificial zeolite caused to decrease greatly in $NH_3$ emission from composting pile. As result of this, content of nitrogen in the compost was increased by addition of artificial zeolite. Emission of $CH_4$ was the highest at early stacking stage, and that was decreased drastically at 8th day. Emission of $CH_4$ was also decreased greatly by addition of artificial zeolite at 5th days after stacking. It may be resulted from adsorption of $CH_4$ into the molecular sieve structure of artificial zeolite and low water content by high temperature fermentation.

  • Biogas production from agricultural wastes were summarized as follows: 1. Biogas Generation Characteristics of Various Manures and Residues a. Gas yield from crop residues like rice straw, rice hull, corn stalk and coconut husk can be improved by addition of animal manures. b. Gas yield from coconut husk can be improved through aerobic fermentation for at least one week before loading in the digester. c. Gas yield from fresh rice straw is better than from pre-fermented one, whether alone or in combination with animal manures. d. Initial study has shown that fresh azolla can be substituted for animal manures in manurerice straw combinations and gas yield derived based on unit volatile solids loaded is actually better than for manure-residue combinations. e. Gas production is highly sensitive to substrate pH and becomes almost nil at a pH of below 6. 2. Effect of ambient conditions and other factors on biogas production in a house hold-size digester. a. Results showed that compaction of rice straw in straw-manure combination can reduce gas yield compared with loosely mixed straw. b. The effective gas production period extended to 70 days using freshly threshed rice straw and fresh cattle manure as feed material. c. Underground and above ground digesters with shade have relatively more stable substrate temperature than aboveground exposed digesters. This relative temperature instability may likely be the reason for lower gas yield for the exposed aboveground digester loaded with loose straw-cattle manure substrate, compared with the underground digester with the same substrate. 3. Economic Analysis a. Based on prevailing costs of fuel, materials, and labor in the Philippines, biogas produced from the household size system is cheaper than either LPG or kerosene. b. If other benefits like organic fertilizer, pollution control and convenience are considered, biogas will surely be the best alternative fuel source.
