EST analysis of regenerating newt retina

  • A vertebrate retina is an organ belonging to the central nerve system (CNS), and is usually difficult to regenerate except at an embryonic stage in life. However, certain species of urodele amphibians, such as newts and salamanders, possess the ability to regenerate a functional retina from retinal pigment epithelial (RPE) cells even as adults. After surgical removal of neural retinas from adult newt eyes, the remaining RPE cells lose their pigment granules, transdifferentiate into retinal progenitor cells, which further differentiate into various retinal neurons, and then finally reform a functional neural network. To understand the molecular mechanisms of CNS regeneration, we attempted to investigate the genes expressing in regenerating newt retina. mRNAs were isolated from regenerating retinas at 18-19 days after the surgical removal of the normal retina, and a cDNA library (regenerating retinal cDNA library) were constructed. Our EST analysis of 112 clones in the regenerating cDNA library revealed that about 70% clones are closely related to the genes previously identified. About 40% clones are housekeeping genes, and about 15% clones encode proteins related to the regulation of gene expression and to the proliferation of the cells. Sequences similar to neural retina- and RPE-specific genes were not detected at all. These results led us to suppose that the regenerating retinal cells are in a state considerably different from those of neither neural retina nor RPE cells.

An Analytical Research of Mass Media's Articles about Korean Apartments' Living - Focused on the Articles of Newspapers and Women's Magazines Between 1960~80's - (아파트 생활기사의 주거담론분석 - 1960~80년대 일간지와 여성지 기사를 중심으로 -)

  • As of 2010, apartment occupies more than half of all houses in Korea. It took just half a century since 1960 when apartment was introduced to ordinary people for the first time. The apartment has spread over the nation very swiftly since then. This phenomena needs more inclusive explanations than the investigations into the government's policies or economical situations. Without satisfaction and consensus of people, the universal spread of apartment might be fairly unrealizable. In this vein, the research and analysis has been performed in order to grasp the picture of everyday life's discourse based on the related articles from five major newspapers and five women's magazines during 1960~1989. From the articles' discourses we tried to understand how modern apartment has been accepted and evaluated through people's living life in it. First, the discourse of living life was generated mostly by housewives through the comparison between apartment and traditional house. Second, the most influential aspect of apartment's popularization was the convenience of efficient housekeeping system by modern equipment and home appliance installed in apartment. This efficiency could allow housewives to perform various urban activities such as work, shopping, and simple going out. Third, the satisfaction from efficiency and privacy brought about a sense of alienation at the same time. As a result, the rapid proliferation of apartment in Korea could be attained not only by the institutional driving forces but through the fulfillments of Korean people's housing needs from modern everyday life.

Factors Associated with the Time Use in Leisure Activity and Social Gathering of the Youth - Focused on the Effects of Engaging Role Types of the Youth - (청년의 여가시간과 교제시간에 대한 영향요인 탐색 -청년 역할유형의 영향을 중심으로-)

  • This study explores the factors associated with the amount of time use in leisure and social gathering activities. The analysis shows that the engaging role types of the youth, for example housekeeping and care giving, or working on the paid job are associated with the amount of time use in leisure activities. However, the factors related to time use in social gathering activities are the engaging role types of youth as well as the socializing opportunity. Compared to the other role types of the youth, the youth who are preparing for a job and is not engaged in employment, education, and training(NEET) spend more time in media leisure, travel and cultural activities, sports and reports activities, however spend less time in social gathering compared to students. The youth who have a job spend the least time in all of the leisure activities except travel and cultural activities for a weekend. Policy should remove the barriers in leisure activities and increase the opportunities for socializing.

An Analysis of Rural Families' Time-Use (농가 가족원의 생활시간 분석)

  • The purpose of this study was to analyze time-use of rural families. The data for 9l4 persons or 400 farm households in eight provinces were collected using time diary. The results were as follows. 1) The time-use of the farming season were different from that of the off-filming season in rural area. 2) In the case of farm wort the working hours of husband were significantly longer than those of wife during the farming and of-filming season. In contrast the working hours of wife for housekeeping were significantly longer than those of husband during both seasons. 3) The time spent of work by rural adults is much longer than average work hours of adults in general during the farming season. And the rural adults spend less time in leisure compared to the adults in general during the farming season.

The Gene Expression Profiling in Murine Cortical Cells Undergoing Programmed Cell Death (PCD) Induced by Serum Deprivation

  • PCD (programmed cell death) is important mechanism for development, homeostasis and disease. To analyze the gene expression pattern in brain cells undergoing PCD in response to serum deprivation, we analyzed the cDNA microarray consisting of 2,300 genes and 7 housekeeping genes of cortical cells derived from mouse embryonic brain. Cortical cells were induced apoptosis by serum deprivation for 8 hours. We identified 69 up-regulated genes and 21 down-regulated genes in apoptotic cells. Based on the cDNA microarray data four genes were selected and analyzed by RT-PCR and northern blotting. To characterize the role of UNC-51-like kinase (ULK2) gene in PCD, we investigated cell death effect by ULK2. And we examined expression of several genes that related with PCD. Especially GAPDH was increased by ULK2. Theses findings indicated that ULK2 is involved in apoptosis through p53 pathway.

Ideal Image and Fashion of Korean Women in the 1970s (1970년대 한국의 이상적 여성상과 패션)

  • This study examined the ideal image and fashion of Korean women in the 1970s from a socio-cultural context. This study used information on the 1970s politics, economy, and culture provided by "Chosun Ilbo" and "Yosungjungang" as well as their presentation of the ideal image and fashion for Korean women in the 1970s. The ideal image and fashion of women were considered from the viewpoint of Hamilton's Meta-theory. The ideal image of women in the 1970s is divided into two aspects. The image from the traditional Confucian perspective was prevalent and restricted the lives of women to housekeeping chores. On the contrary, women have increasingly participated in society vis-$\grave{a}$-vis education and employment opportunities to present a progressive image of women. These aspects coexisted during the turmoil of social change. Progressive women had money to buy clothes because they were economically independent. These women embraced styles that included mini, midi, maxi, and bell-bottom pants. Further, pants were developed into different styles such as pant suits. T-shirts and blue jeans as casual wear were very popular among the youth. At the end of the 1970s, the tailored look and the big look (which copied men's clothing) were in fashion. Masculine styles such as wide shoulders with pads and neckties strengthened gender equality. Other fashions were dominated by feminine styles described as beautiful, sweet, and elegant that reflected Korean society's tendency to regard women as sex objects. Clothing that exposed the body highlights this sexual objectification aspect. Women wore miniskirts, hot pants, and bikinis because they wanted to enhance their sex appeal, propagating the view of women as sex objects. In conclusion, all aspects of society and culture were closely interrelated with a fashion style that reflected the values of those aspects.

The effects of Ki on the Elderly with Chronic Illness (노인 만성질환에 대한 기(氣) 시술의 효과)

  • The purpose of current study was to investigate the effects of Ki-therapy which improves mood state and relieves pain on the elderly with chronic illness. The subjects were 42 elderly and convenience sampling(incidental sampling) was used to prevent contamination to the control group. Frequency, %, $X^2$, t-test, and repeated measure ANOVA by SPSS PC + program were used to analyze the data. The mean age of subjects was 72.95(Control group), 73.10(experimental group). Number of complains was 2.45(control group), 2.65 (experimental group). All of demographic characteristics of subjects. that is, age, weight. number of complains. educational background. family type. economic state, perceived health state were homogeneous. There are many difficulties in ADL & IADL. 14.2% in shopping, mode of transportation, ability to handle finances. 11.9% in ability to use telephone. continence, 7.1% in bathing, food preparation, transfer, and housekeeping. As the result of this study. slight improvement of mood state and pain relief were found. Although 3 improved items of 'clear-headed', 'lively', and 'nervous' were statistically significant. the pain relief was not statistically significant. It requires repeated treatment and measurement. And suggested case study or qualitative study in further studies because manufacturing of environment(i. e. rain) is very difficult in community people.

Convergent research on how golf affects the subjective happiness of mothers who have children with disabilities (골프운동이 장애자녀를 둔 어머니의 주관적 행복감에 미치는 융합 연구)

  • This study was conducted an convergence research on 210 mothers in order to examine the effect of golf on subjective happiness of mothers who have children with disabilities. The factors were conducted surveys using SPSS WIN 18.0 program. Firstly, were mostly 40s, college graduates, Buddhists or had no religion, had housekeeping as occupation, had average household income, and most of them were healthy. In addition, descriptive statistics on their years of experience in golf, golf strokes, field, practice frequency, and practice hours were all above average. Secondly, significant correlations were suggested between experience in golf, golf strokes, field, practice frequency, silence during practice hours, and in the subjective happiness of negative correlation as a result of analysing the correlations between the factors. Thirdly, the field, practice frequency, and practice hours affected the subjective happiness.

Significant Variables Influencing on Adolescents' Self-Efficacy (청소년의 자기효능감에 영향을 미치는 관련변인)

  • Since self-efficacy was suggested by Bandura(1977) as the most important factor that affects human behavior, preceding studies about self-efficacy are frequently related with children, and it is a main stream that only personal factors are chosen as related factors of these studies. Considering we don't have enough studies about family factors that reflect qualitative and relational factors of adolescents' home background, this study was aimed to seize the actual condition of self-efficacy meaning perceived and emotional faith about their own ability. These are our major concerns: 1. What kind of tendency does self-efficacy and appearance stress of adolescents, family functioning show? 2. How strong are relative influences of variables on self-efficacy of adolescents? This study was operated on July and August, 2002, aimed 378 middle school and high school students in Jeon-Nam, Mokpo area. 378 question sheets were used for analysis. Followings are results of this study. First, aimed students' average mark of self-efficacy was 2.09, which meant a normal level. And in low-ranked part, self-control-efficacy was the highest, and confidence task difficulty preference was following. Furthermore, whole self-efficacy showed a little difference from the demographic factor of adolescents. In low-ranked part, however, confidence and self-efficacy showed a somewhat difference. As students grew up, confidence decreased, and male students showed higher self-control-efficacy than female ones. Second, appearance stress and family functioning degree had a strong relation with self-efficacy. Family functioning, had important interrelation not only with whole self-efficacy but also with low-ranked part of self-efficacy. In low-ranked part, self-control-efficacy and task difficulty preference were proportional to the family functioning degree. From these results, we can find that the appearance stress was an actual factor that affected the self-efficacy of adolescents, and their self-efficacy was strongly related with family functioning degree. Accordingly, affirmative emotion in family, harmonic communication among family members and sharing housekeeping works had an immediate influence on elevation of self-efficacy.

Molecular typing of uropathogenic Escherichia coli isolated from Korean children with urinary tract infection

  • Purpose: We investigated the molecular types of uropathogenic Escherichia coli (UPEC) by using conventional phylogrouping, multilocus sequence typing (MLST), and fimH genotyping. Methods: Samples of patients younger than 18 years of age were collected from the Chung-Ang University Hospital over 2 years. Conventional phylogenetic grouping for UPEC strains was performed by polymerase chain reaction (PCR). Bacterial strain sequence types (STs) were classified on the basis of the results of partial sequencing of seven housekeeping genes. In addition, we analyzed nucleotide variations in a 424-base pair fragment of fimH, a major virulence factor in UPEC. Results: Sixty-four UPEC isolates were analyzed in this study. Phylogenetic grouping revealed that group B2 was the most common type (n=54, 84%). We identified 16 distinctive STs using MLST. The most common STs were ST95 (35.9%), ST73 (15.6%), ST131 (12.5%), ST69 (7.8%), and ST14 (6.3%). Fourteen fimH allele types were identified, of which 11 had been previously reported, and the remaining three were identified in this study. f1 (n=28, 45.2%) was found to be the most common allele type, followed by f6 and f9 (n=7, 11.3% each). Comparative analysis of the results from the three different molecular typing techniques revealed that both MLST and fimH typing generated more discriminatory UPEC types than did PCR-based phylogrouping. Conclusion: We characterized UPEC molecular types isolated from Korean children by MLST and fimH genotyping. fimH genotyping might serve as a useful molecular test for large epidemiologic studies of UPEC isolates.
