국외 도입 열대·아열대 작물의 국내 재배실태 및 과제 (Current Situations and Prospects on the Cultivation Program of Tropical and Subtropical Crops in Korea)

  • 기후변화 적응과정에서 도입 작물의 국내 재배실태를 조사하고 대응과제를 도출코져본 연구를 수행하였으며, 그 결과는 다음과 같다. 도입 열대 아열대 채소 재배현황(2015년 기준)은 18작목으로 재배농가수는 920농가, 재배면적은 321 ha로 조사 추정되었고, 재배면적이 많았던 작물은 울금, 여주, 아스파라거스, 삼채, 허브류 순이었다. 도입 열대 아열대 과수류 재배 현황은 2015년 국내재배 농가수와 재배면적은 264농가 106.5 ha로 추정되며, 전년대비 각각 51.7%와 83.7% 증가하였다. 도입 특약용작물 재배현황(2015년 기준)은 10작물로 753농가에서 재배면적 276.3 ha로 조사 되었으며 재배면적이 많았던 작물은 강황, 감초, 마카, 모링가, 포포나무 순이었다. 도입 열대 아열대 작물의 국내 재배가 안정적으로 정착되기 위해서는 국내 재배적응성 검정에 의거 재배환경면에서의 안전성과 경제성이 있어야 하며, 생산물의 이용과 유통 및 판로의 확보 문제, 외국현지 수입산과의 경쟁력 등을 고려할 필요가 있고, 국가 간 이동에 따른 생물다양성협약 등 국제협약 및 국내 종자산업법에 의하여 도입되어야 할 것이다.

Transient Expression of Homologous Hairpin RNA Interferes with Broad bean wilt virus 2 Infection in Nicotiana benthamiana

  • Broad bean wilt virus 2 (BBWV2), genus Fabavirus, subfamily Comovirinae, family Secoviridae, causes damage in many economically important horticultural and ornamental crops. Sequence alignments showed several conserved sequences in 5' non-coding regions (5' NCRs) of RNA 1 and RNA 2 in all BBWV2 strains characterized so far. Based on this observation, we generated a hpRNA construct (pIR-BBWV2) harboring an inverted repeat containing a 210 bp cDNA fragment homologous to 5' NCR portion of BBWV2 RNA 1 to investigate the silencing potential for its ability to interfere with a rapidly replicating BBWV2. Agrobacterium-mediated transient expression of the IR-BBWV2 had a detrimental effect on BBWV2 infection, showing no distinct symptoms in non-inoculated leaves of the agroinfiltrated Nicotiana benthamiana plants. BBWV2 genomic RNAs were not detected by RT-PCR from tissues of both the inoculated leaves and upper leaves of the agroinfiltrated plants. Accumulation of virus-derived small interfering RNAs was detected in the inoculated leaf tissues of N. benthamiana plants elicited by transient expression of IR-BBWV2 indicating that RNA silencing is responsible for the resistance to BBWV2.

Development of Gene-based Markers for the Allelic Selection of the Restorer-of-fertility Gene, Rfo, in Radish (Raphanus sativus)

  • Cytoplasmic male sterility (CMS) and fertility restoration have been utilized as valuable tools for $F_1$-hybrid seed production in many crops despite laborious breeding processes. Molecular markers for the selection of CMS-related genes help reduce the expenses and breeding times. A previously reported genomic region containing the Ppr-B gene, which is responsible for restoration of fertility and corresponds to the Rfo locus, was used to develop gene-based or so-called "functional" markers for allelic selection of the restorer-of-fertility gene (Rfo) in $F_1$-hybrid breeding of radish (Raphanus sativus L.) Polymorphic sequences among Rfo alleles of diverse breeding lines of radish were examined by sequencing the Ppr-B alleles. However, presence of Ppr-B homolog, designated as Ppr-D, interferes on specific PCR amplification of Ppr-B in certain breeding lines. The organization of Ppr-D, resolved by genome walking, revealed extended homology with Ppr-B even in the promoter region. Interestingly, PCR amplification of Ppr-D was repeatedly unsuccessful in certain breeding lines implying the lack of Ppr-D in these radishes. Ppr-B could only be successfully amplified for analysis through designing primers based on the sequences unique to Ppr-B that exclude interference from Ppr-D gene. Four variants of Rfo alleles were identified from 20 breeding lines. A combination of three molecular markers was developed in order to genotype the Rfo locus based on polymorphisms among four different variants. These markers will be useful in facilitating $F_1$-hybrid cultivar development in radish.

Effect of Basal Medium and Plant Growth Regulator on in vitro Plant Regeneration from Axillary Buds of Walnut New Cultiver "Sinlyeong"

  • The walnut (Juglans regia L.), a member of the Juglandaceae, is native to the mountain ranges of central Asia. This species of walnut is valued commercially for its nuts and in some areas for its timber. The seeds of walnut are recalcitrant and it has strong integument dormancy and their germination is irregular, making its natural propagation difficult. Low percentage of seed germination and long propagation cycle are the main problems of propagation. This study was conducted medium composition on in vitro plantlet regeneration from axillary buds of walnut. It has proved to be the most generally applicable and reliable method of in vitro propagation. Micropropagation culture that axillary buds are excised aseptically enables faster multiplication of plants. The axillary buds of walnut new cultivar "Sinlyeong" were cultured on two basal media which contained the different plant growth regulators depending on the respective shooting and rooting stage. After 12 weeks, the shoot generation rate was 85.3%, the shoot number and its length were 1.9/explant and 2.7 cm in the most favorable medium composition. The percentage of rooting was 25.4%. From these results, it was found the optimum basal medium and plant growth regulator for in vitro plant regeneration from axillary buds of walnut new cultivar "Sinlyeong". However, we have continued to search the other medium additives to enhance the rate of walnut root.

Effect of Soil Textures on Fruit Yield, Nitrogen and Water Use Efficiencies of Cucumber Plant as Affected by Subsurface Drip Fertigation in the Greenhouse

  • Growing crops under different soil textures may affect crop growth and yield because of soil N availability, soil N leaching, and plant N uptake. The objective of this study was to evaluate effects of three different soils (sandy loam, loam, and clay loam) on cucumber (Cucumis sativus L.) yield, nitrogen (N) use efficiency (NUE), and water use efficiency (WUE) by subsurface drip fertigation in the greenhouse. Three different soil textures are sandy loam, loam, and clay loam with 3 replications. The dimension of each lysimeter was $1.0m(W){\times}1.5m(L){\times}1.0m(H)$. Cucumber was transplanted on April $8^{th}$ and Aug $16^{th}$ in 2011. The subsurface drip line and tensiometer was installed at 30 and 20 cm soil depth, respectively. An irrigation with $100mg\;NL^{-1}$ concentration was automatically applied when the tensiometer reading was 10 kPa. Volumetric soil water content for cucumber cultivation was the highest in 30 cm soil depth regardless of soil texture and was lowered when soil depth was deeper. The volumetric soil water contents at soil depths of 10, 30, 50, and 70 cm were the highest at clay loam, followed by loam, and sandy loam. The growth of cucumber at the $50^{th}$ day after transplanting was the lowest at sandy loam. Cucumber fruit yields were similar for all three soil textures. The highest amount of water use at sandy loam was observed. Nitrogen and water use efficiencies for cucumber were higher for clay loam, followed by loam and sandy loam, while the amount of N leaching was the greatest under sandy loam, followed by loam, and clay loam. Overall, growing cucumber on either loam or clay loam is better than sandy loam if subsurface drip fertigation is used in the greenhouse.

배지내 천연유기물 첨가가 딸기 "설향" 배양묘 기내 생육에 미치는 영향 (Effect of Natural Additives on In Vitro Growth Medium of Strawberry 'Seolhyang')

  • Strawberry which is the genus Fragaria under family Rosaceae is one of the most important fruit plants for both fresh consumption and food processing in the temperate and subtropical countries. Propagation of strawberry is achieved either through runners or by in vitro micropropagation. Meristem tips, generally obtained from runners of virus-free plants, are commonly used to establish in vitro cultures, which are employed for mass propagation or as a source of plant material for regeneration and transformation experiments. This study was conducted to determine the optimal natural additives strength to improve sprouting shoot rate of apical meristem of strawberry 'Seolhyang'. Strawberry apical meristem at size (0.2 mm to 0.3 mm) with leaf primordials were cultured on the 1/3MS(Murashige & Skoog) medium supplemented with five natural additives such as coconut milk, maple sap, banana powder and peptone. The sprouting ratio and growth characteristics were evaluated after eight weeks after in vitro culture. Shoot ratio of 'Seolhyang' apical meristem was 72.9% in 1/3MS medium supplemented with maple sap. On the other hand, the low shoot ratio was observed 47.7% in 1/3MS medium supplemented with banana powder. Shoot length was different as natural additives but numbers of leaf was not significantiy different among the natural additives. As a result, the sprouting ratio and plant growth were enhanced effectively in 1/3MS medium with maple sap compared to the others.

Effect of Medium Composition on in vitro Propagation and Plantlet Regeneration from Nodal Explants of Cassava (Manihot esculenta Crantz)

  • The Cassava (Manihot esculenta Crantz) is a perennial woody shrub cultivated mainly in the tropics for its starchy tuberous roots. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. Thus, a study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with single node after sterilization, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, single-node stem segments, approximately 1 cm in length, were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate was 55.6%, the shoot number and its length were 1.0/explant and 2.3 cm in the most favorable medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

  • The Cassava(Manihot esculenta Crantz) is a tropical root crop, originally from Amazonia, that provides the staple food of an estimated 800 million people worldwide. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. So we tried to optimize protocols for mass production of somatic embryo amenable to large-scale vegetative propagation of Cassava. After in vitro eight-week culture of leaves of Cassava, the medium which contained the 2,4-D, BAP and IBA showed the highest callus induction rate, embryogenesis callus formation rate and somatic embryo formation in Cassava culture. In the medium with GA3 and myo-inositol, shoots were most vigorously regenerated from somatic embryos of Cassava. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

  • Where temperature permits plant growth, the existence of plants is controlled chiefly by the water availability. Without water, plants cannot grow, and man and animals cannot survive. With too much water, plant growth is also rather limited due partly to oxygen deficit in root zone (Ro et al., 1995).
