Salivary secretion and salivary stress hormone level changes induced by tongue rotation exercise

  • PURPOSE. Prevention of xerostomia and stress is important to prolong healthy life expectancy and improve the quality of life. We aimed to investigate the effects of tongue rotation exercise for increasing salivary secretions and stabilizing salivary stress hormone levels. MATERIALS AND METHODS. Twenty four participants without subjective oral dryness were enrolled. The exercises comprised tongue rotation exercise and empty chewing. The salivary stress hormone level was measured using a Salivary Amylase Monitor. Unstimulated whole saliva volume and salivary amylase activity were measured before tongue rotation exercise or empty chewing and subsequently 5, 10, and 15 minutes after these exercises. Differences in the rates of change of unstimulated whole saliva volume and salivary amylase activity were analyzed by repeated measure analysis of variance. RESULTS. Statistically significant differences among the rates of change were not observed after empty chewing for unstimulated whole saliva volume and salivary amylase activity at the four measurement times. However, the rate of change of unstimulated whole saliva volume and salivary amylase activity were statistically significantly different among the four time points: before the tongue rotation exercise and 5, 10, and 15 minutes post-exercise (P<.05 and P<.01, respectively). CONCLUSION. Tongue rotation is effective in increasing saliva secretion, reducing stress, improving oral function, and extending healthy life expectancy.

  • The relation of endocrine in the western medicine and zangfu-organ functions in Korean medicine, related to growth disturbance, was studied and derived the following results. 1. The hormone most related to growth disturbance is, directly secreted from the anterior pituitary or is stimulated and secreted in the target grands, growth hormone, thyroid, adrenocortical hormone, gonadial hormone and is insulin secreted from $\beta$ cell of langerhans' slands of pancreas. 2. the pituitary has the most close relation with the kidney in the five zang-organ. Because the kidney is innate origin(先天之本) and promotes qi and blood(生化氣血), stores the essence of life(藏精), dominates the bones(主骨) and promotes the marrow(生骨髓). Especially it is connected with brain(通於腦). 3. In the children growth, the endocrine action in the pituitary has the most close relation with the kidney, As in the reports of the brain and spinal cord, bone, store essence of life, sexual maturation and decline(kidney-qi, sexual functions of both sexes(天癸)) etc, and cause of cretinism, dwarf in the main subject. 4. Somatomedin is the most important factor of the growth factors, IGF in another word. The unification of IGF and secretion is controlled firstly according to growth hormone, however is very closely related to the nutrition status in the non-hormonic causes. Also, it is affected very much by the insulin. 5. Insulin is one of the important hormone related to the growth and is secreted from the pancreas. Pancreas belongs to the functional system of spleen in oriental medicine, thus the growth disturbance, occurred due to error in insulin secretion and nutrition(in another words, the lack of postnatal essential substance from food-stuff(水穀精氣)), is closely related to the spleen. 6. From the results driven above, the hormone action of endocrine and problem in secretion, related to the growth disturbance, must be focused on the differentiation of symptoms and signs of the kidney and the spleen in oriental medicine.

Circadian rhythm of melatonin secretion and growth-related gene expression in the tiger puffer Takifugu rubripes

  • Somatostatin (SS) and growth hormone-releasing hormone (GHRH) are primary factors regulating growth hormone (GH) secretion in the pituitary. To date, it remains unknown how this rhythm is controlled endogenously, although there must be coordination of circadian manners. Melatonin was the main regulator in biological rhythms, and its secretion has fluctuation by photic information. But relationship between melatonin and growth-related genes (ghrh and ss) is unclear. We investigated circadian rhythms of melatonin secretion, ghrh and ss expressions, and correlation between melatonin with growth-related genes in tiger puffer Takifugu rubripes. The melatonin secretion showed nocturnal rhythms under light and dark (LD) conditions. In constant light (LL) condition, melatonin secretion has similar patterns with LD conditions. ss1 mRNA was high during scotophase under LD conditions. But ss1 rhythms disappeared in LL conditions. Ghrh appeared opposite expression compared with melatonin levels or ss1 expression under LD and LL. In the results of the melatonin injection, ghrh and ss1 showed no significant expression compared with control groups. These results suggested that melatonin and growth-related genes have daily or circadian rhythms in the tiger puffer. Further, we need to know mechanisms of each ss and ghrh gene regulation.

Secretory Expression of Human Growth Hormone in Saccharomyces cerevisiae Using Three Different Leader Sequences

  • A recombinant human growth hormone(hGH) was expressed as a secretory product in the yeast Saccharomyuces cerevisiae. There different leader sequences derived from the mating fac-tor $\alpha$1(MF$\alpha$1) inulinase and invertase were used to direct the secretion of hGH into the extracel-lular medium. Among three leader sequences tested, the inulinase leader sequence was found to be the most efficient in the secretory expression of hGH. In contrast, no hGH was detected in the ex-tracellular medium with the invertase leader sequence. After 48 h shake-flask culture, the yields of hGH secreted into th emedium by the invertase. MF$\alpha$1 inulinase and invertase leader sequences were approximately 0, 0.3 and 0.9 mg/L, respectively. The secretion efficiencies were also found to be 0, 3.8 and 13% for the invertase , MG$\alpha$1 and inulinase leader sequences, respectively.

Designing Signal Peptides for Efficient Periplasmic Expression of Human Growth Hormone in Escherichia coli

  • The secretion efficiency of a protein in a Sec-type secretion system is mainly determined by an N-terminal signal peptide and its combination with its cognate protein. Five signal peptides, namely, two synthetic Sec-type and three Bacillus licheniformis alpha-amylase-derived signal peptides, were compared for periplasmic expression of the human growth hormone (hGH) in E. coli. Based on in silico predictions on the signal peptides' cleavage efficiencies and their corresponding mRNA secondary structures, a number of amino acid substitutions and silent mutations were considered in the modified signal sequences. The two synthetic signal peptides, specifically designed for hGH secretion in E. coli, differ in their N-terminal positively charged residues and hydrophobic region lengths. According to the mRNA secondary structure predictions, combinations of the protein and each of the five signal sequences could lead to different outcomes, especially when accessibility of the initiator ATG and ribosome binding sites were considered. In the experimental stage, the two synthetic signal peptides displayed complete processing and resulted in efficient secretion of the mature hGH in periplasmic regions, as was demonstrated by protein analysis. The three alpha-amylase-derived signal peptides, however, were processed partially from their precursors. Therefore, to achieve efficient secretion of a protein in a heterologous system, designing a specific signal peptide by using a combined approach of optimizations of the mRNA secondary structure and the signal peptide H-domain and cleavage site is recommended.

Resistance to Thyroid Hormone Syndrome Mutation in THRB and THRA: A Review

  • Resistance to thyroid hormone syndrome (RTH) is a genetic disease caused by the mutation of either the thyroid hormone receptor-β (THRB) gene or the thyroid hormone receptor-α (THRA) gene. RTH caused by THRB mutations (RTH-β) is characterized by the target tissue's response to thyroid hormone, high levels of triiodothyronine and/or thyroxine, and inappropriate secretion of thyroid-stimulating hormone (TSH). THRA mutation is characterized by hypothyroidism that affects gastrointestinal, neurological, skeletal, and myocardial functions. Most patients do not require treatment, and some patients may benefit from medication therapy. These syndromes are characterized by decreased tissue sensitivity to thyroid hormones, generating various clinical manifestations. Thus, clinical changes of resistance to thyroid hormones must be recognized and differentiated, and an approach to the practice of personalized medicine through an interdisciplinary approach is needed.

Effects of Massage on Stress, Depression and Level of Milk Secretion Hormones in Mothers (신생아 어머니의 혈중 코티졸과 우울 및 혈중 프로락틴에 대한 신생아 마사지 적용 효과)

  • Purpose: To investigate changes in mother's stress, degree of depression and level of milk secretion hormones when the mothers provided massage to their new born babies. Method: The study was conducted with 65 women who had delivered at a convalescence institute in Incheon between Sep 25, 2003 and Feb 10, 2004. The mothers were assigned to a control group or an experimental group. Infants of mothers in the experimental group received massage from their mothers twice a day for 10 days beginning on the 4th day after delivery. The data were processed using $x^2-test$, paired t-test and repeated measures ANOVA using the SAS program. Results: The level of cortisol, a stress hormone and depression were significantly different between the two groups (p=0.01 ;p=0.02). While fur plasma prolactin level, a milk secretion hormone, there was no significant difference(p=0.14). Conclusions: When massage was provided to new-born babies the cortisol level of the mothers decreased significantly compared to the control group which did not receive the massage. As the level of depression of the control group declined as time passed while the experiment group showed no difference and there was no difference in the plasma prolactin level, further study is suggested.

Effect of The Herb medicines to Growth and secretion of Growth Hormone in Pigs (한약(韓藥) 복합제제(複合製劑)의 투여(投與)가 돼지의 성장(成長) 및 성장(成長)호르몬 분비(分泌)에 미치는 영향(影響))

  • The present study is designed that the herb medicines which are returned to kidney, pancreas and are known by efficacy in growth are supplied to pigs, thereafter the effects to growth and secretion of growth hormone in serum is investigated. For this purpose the 24 days old pigs, which are crossbreeding, castrated are used. The conclusions are that after 4 weeks' supply of herb medicinal compounds, pigs' weight gain has increased in low nutritional group with statistical meaning(p〈0.05). And one day's weight gain has increased with statistical meaning(p〈0.05) in all groups. Regarding food take, the second nutritional group has more significant increase than control group. Utilization rate of nutrition doesn't have any statistical meaning in all groups, but it has the tendency that as herb medicinal compounds are added, utilization rate of nutrition is increased. Increase of vertebra lengths and level of growth hormone in serum according to supply of herb medicinal compound don't have any statistical meaning because of small sample size, but 10 - 20 % increase of vertebra lengths and 10 - 30 % increase of level of growth hormone are observed.
