The Isolation and Fusion of Pea and Barley Mesophyll Protoplasts (완두와 보리의 엽내세포 원형질체 분이 및 융합)

  • The optimal conditions for the protoplast isolation from the leaves of pea (Pisum sativum L. cv. Sparkle) and barley (Hordeum vulgare L. cv. Baecdong) were determined in order to achieve a somatic hybridization between two species. It was revealed that the use of 0.5M sorbitol as an osmoticum was appropriate for pea. The yield of intact protoplasts was the highest (40%) when pea leaves were incubated in the enzyme solution for 4 hours. In case of barley, the optimal concentrations of cellulase, pectinase and mannitol as the enzyme solution were 2%, 1% and 0.35M, respectively. And the yield of barley protoplasts was the highest(87%) when leaves were incubated in this enzyme solution for 3.5 hours. A fusion of protoplasts from pea and barley was induced by PEG treatment enriched with calcium salts within 60 minutes.

Phytotoxic Effect of Lettuce (Lactuca sativa L.) Leaf Extract on Seedling Growth of Crops and Weeds

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that are biologically active. Aqueous or methanol extracts and residues from leaves of lettuce plants were assayed to determine their allelopathic effects, and the causative allelochemicals from fractions were quantified by means of HPLC analysis and bioassayed. Extracts from oven-dried leaf samples were more phytotoxic than those from freeze-dried samples. Leaf extracts of 40 g L$^{-1}$ were completely inhibitory on root growth of alfalfa (Medicago sativa L.), while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol leaf extracts. The major allelopathic substances analyzed by HPLC were coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid and chlorogenic acid. Of them p-coumaric acid was found as the greatest amount (8.9 mg 100 g$^{-1}$ ) in the EtOAc fraction; only coumarin was found in all the fractions. Hexane and EtOAc fractions of L. sativa reduced alfalfa root growth more than did BuOH and water fractions. These results suggest that lettuce had potent herbicidal activity, and that its activity differed depending on type and amount of causative compounds by fraction.

  • Effect of arsenite, arsenate and dimethylarsinic acid (DMA) on the growth of seedling plants were investigated in order to compare the toxicity of arsenic species in soil environments. Test plants were mung bean (Phaseolus radiatus), wheat (Triticum aestivum), barely (Hordeum vulgare), cucumber (Cucumis sativus L.). Seedling growth in As-contaminated soil were significantly reduced in all test species. Arsenite was more toxic than arsenate and DMA. Among the test plants, mung bean was most sensitive to arsenic, followed by cucumber, wheat, and barely.

A Rapid Screening for Aluminum-tolerant and -sensitive in Barley (Hordeum vulgare L.) and Plasma Membrane H+-ATPase Expression (알루미늄 내성과 민감성 보리의 빠른 screening과 원형질막 H+-ATPase의 발현)

  • Here we report a simple screening system using hematoxylin staining (HS) of the root apex. It allowed rapid classification into different aluminum (Al) tolerance from 65 cultivars within one week. Using this system, we selected the most Al-tolerant (Jayae-2) and-sensitive (Pum-2) barley (Hordeum vulgare L.) The results show that the different responses in Al-induced growth inhibition, Al accumulation and expression of plasma membrane (PM) $H^+$-ATPase in root apices of selected two cultivars. It showed strongly Al-induced growth inhibition in a dosedependant manner only in Pum-2 but not in Jayae-2. Aluminum accumulation in root apices (10 mm) was significantly higher in Pum-2 only. The $H^+$-ATPase expression of PM vesicles by western blotting was decreased in Pum-2 but not in Jayae-2 treated with $20{\mu}M$ Al for 24 h. These finding indicate to screen from our system is rapid and reliable and to sustain the expression of PM $H^+$-ATPase at translational level is an important role in root growth as affected by Al.

Effect of Microbial Flora and Inoculation of Probiotics on Fermenting Characteristics of Naked Barley Grain (Hordeum Vulgare L.) (쌀보리 종실 내 미생물 분포와 생균제 접종이 발효특성에 미치는 영향)

  • This study was performed to analyze the resident microbial flora and the effects of probiotic inoculation on the fermentation characteristics of whole grain naked barley (Hordeum Vulgare L.) with the goal of evaluating the possibility of utilization as fermented feedstuff. Naked barley grains were harvested 35 days after heading, and the microbial flora was analyzed using MALDI-TOF mass spectrometer. After inoculation of commercial microbes to the naked barley grain (BT), the pH and number of bacteria, such as aerobic bacteria, lactic acid bacteria, yeast and E. coli, were measured and compared with the non-inoculated control (BC). A total of 122 colonies was isolated from the naked barley grain and the most popular bacteria species was Staphylococcus xylosus (n = 30, 24.59%). The pH value decreased more rapidly in BT than in BC, and was significantly lower after 7 days of fermentation at $4.33{\pm}0.02$ and $4.83{\pm}0.01$, respectively. The number of aerobic bacteria, lactic acid bacteria and yeast showed an increasing trend within the first 7 days of fermentation, however, their numbers decreased at 28 and 42 days of fermentation. The population of lactic acid bacteria in BT was higher than in BC, but there was no significant different at 7 days of fermentation, with respective levels of $9.24{\pm}0.20$ and $9.01{\pm}0.10logCFU/g$ (p>0.05). The initial number of E. coli was very high in the naked barley grain but subsequently decreased significantly. After 7 days of fermentation, E. coli was not detected in either BT or BC samples. From these results, it appears that the fermentation of naked barley grain proceeded adequately after 7 days, and that fermentation contributes to the safety of naked barley grain during storage.

  • A barley(Hordeum vulgare L.) cultivar 'Youjin' with hooded spike type having good silage quality was developed at National Institute of Crop Science, RDA in 2016. 'Youjin' showed both high yielding and cold resistance through the preliminary and advanced yield trials(PYT, AYT) from 2012 to 2013. We conducted regional yield trials(RYT) of 'Youjin' in six locations around Korea for three years from 2014 to 2016. It had erect plant type, growth habit of II, the green leaf and hooded awn type. In the paddy field its heading date was April 24 and the maturing date was May 25. Plant height was 99 cm and the number of spikes per ㎡ was 696. It has high rate of leaf blades, resistance to BaYMV(Barley Yellow Mosiac Virus) and good winter hardiness. The average dry matter of Youjin was about 17.2 MT ha-1 in the field. And feed quality of 'Youjin' was 10.6% of crude protein content, 24.8% of ADF(Acid Detergent Fiber), 43.5 % of NDF(Neutral Detergent Fiber), 69.1% of TDN(Total Digestible Nutrients). And also 'Youjin' had grade I of silage quality.

Effect of Zebularine Soaking on the Early Growth Stage and Mitotic Chromosomes of Barley (Hordeum vulgare L.) (제부라린의 침종처리가 보리 생육초기 생장 및 체세포 염색체에 미치는 영향)

  • The objective of this study was to identify the effect of zebularine soaking on the early growth stage of barley (Hordeum vulgare L.). Hence, root elongation was measured daily according to the different concentrations of zebularine (1.0, 2.5, 5.0, and 10.0 μM) for 4 days. On the first day, root length at 2.5 and 5.0 μM was significantly longer than that in the non-treated control. On the second day, root length was not significantly different among all concentrations and controls. On the third day, root elongation was suppressed by the effect of zebularine, except at 2.5 μM. The treatment time of zebularine accounted for the largest proportion of the variation in root elongation. After transplanting, plant growth velocity was similar to that of the control; however, plants at 2.5 μM showed faster growth velocity than that of the other concentrations and the control. In the metaphase of mitosis, most chromosomes of cells under zebularine treatment were ordinary regardless of the concentration, while some cells with short chromosomes were investigated at around 2%. The short chromosome showed a centromere. In addition, it showed short and long arms based on the centromere. The lengths of the short and long arms were different for each short chromosome. It is necessary to study the effect of the short chromosome as a chromosomal function on plant growth and phenotype through investigation of meiosis and fertilization at the chromosome level.

Effects of Application of Liquid Pig Manure on Yield of Green manure in Single and Companion Cropping (청보리와 헤어리베치 단작 및 혼작 재배지에서 돈분액비 시용이 녹비생산량에 미치는 영향)

  • To evaluate the effects of liquid pig manure (LPM) on green manure crop yields, plant height and biomass of green manure crops were investigated in single and companion cropping. Green manure crops used for this experiment were Hordeum vulgare L. (green barley) and Vicia villosa roth (hairy vetch). Field experiment was designed with LPM 1.65 (LPM of $1.65ton\;10a^{-1}$ + single and companion green crops), LPM 3.3 (LPM of $3.3ton\;10a^{-1}$ + single and companion green crops) and LPM 6.6 (LPM of 6.6$6.6ton\;10a^{-1}$ + single and companion green crops). In single cropping, plant height were high in LPM 3.3. In companion cropping, plant height were high in LPM 6.6. Thus, we found that application of LPM is general enough to reduce use of chemical fertilizer. Also, optimum application level in single cropping would be LPM 3.3 and in companion cropping would be higher than LPM 3.3.
