Morphometric Characterization of Honey Bee, Apis mellifera Linnaeus, Inbred Lines in Korea (국내 서양종꿀벌 순계의 형태적 특징)

  • The A, C, F colonies of Apis mellifera ligustica Spin. and D, V colony of Apis mellifera caucasia Gorb. bees were collected from 2005-2007. Consequently, inbred lines were derived from the bees of original colonies by matting in the isolated island with due regard for pure breeding. This project helps in the selection of colonies with higher production capacity, aiming to improve honey and royal jelly production and breeding programs. Twenty-three standard morphological traits of honeybee were evaluated, and samples were compared with the data of the two original subspecies. The result suggested that 8 traits partly preserved in bees of inbred lines, and the bees from A. m. ligustica preserved more traits than bees from A. m. caucasia. Among the studied inbred lines, the F line is distinguished by an increase in leg parameters, considered as a favorable phenotypic trait of inbred lines. Importantly, bred of beelines in the same area can be classified as remote and isolated areas. Therefore, we observed differences of inbred lines with the origin subspecies in description acquired with morphometric characteristics as a result of adaptation, breeding, purebred individual lines used as an important resource for breeding novel cross-breeding colonies.

Identification of a Single Nucleotide Polymorphism (SNP) Marker for the Detection of Enhanced Honey Production in Hoenybee (수밀력 우수 꿀벌 계통 판별을 위한 계통 특이 분자마커 개발)

  • Honeybees (Apis mellifera) are common pollinators and important insects studied in agriculture, ecology and basic research. Recently, RDA (Rural Development Administration) and YIRI (Yecheon-gun Industrial Insect Research Institute) have been breeding a triple crossbred honey bee named Jangwon, which have the ability to produce superior quality honey. In this study, we identified a single nucleotide polymorphism (SNP) marker in the genome of Jangwon honeybee, particularly, in the paternal line (D line). Initially, we performed Sequence-Based Genotyping (SBG) using the Illumina Hiseq 2500 in 5 honeybee inbred lines; A, C, D, E, and F; and obtained 1,029 SNPs. Seventeen SNPs for each inbred line were generated and selected after further filtering of the SNP dataset. The 17 SNP markers validated by performing TaqMan probe-based real-time PCR and genotyping analysis was conducted. Genotyping analysis of the 5 honeybee inbred lines and one hybrid line, $D{\times}F$, revealed that one set of SNP marker, AmD9, precisely discriminated the inbred line D from the others. Our results suggest that the identified SNP marker, AmD9, is successful in distinguishing the inbred honeybee lines D, and can be directly used for genotyping and breeding applications.

The Therapeutic Effect of Natural Honeybee (Apis mellifera) Venom in Adjuvant-induced Arthritic Rat (관절염 유발 랫드에 대한 생봉독의 치료 효과)

  • This study was performed to assess that clinco-therapeutic effect of natural Italian honeybee (Apis mellifera) venom in adjuvant-induced arthritic rat. Ninety Sprague- Dawley rats of male were injected with complete Freund's adjuvant (CFA). Adjuvant arthritis was produced by a single subcutaneous injection of 1 mg Mycobacterium butyricum suspended in 0.1 ml paraffin oil into the right hindpaw. Righting reflex was uniformly lost and considered to be the point of arthritis development on day 14 after CFA injection. Experimental groups were divided into three groups. When arthritis was developed in the rat hind-paw, tested groups were administrated with prednisolone (10 mg/kg, p.o) and honeybee venom (one bee, s.c) at an interval of two days. Control group was subcutaneously injected with 0.1 ml of physiological saline solution in the rat at an interval of two days. Clinical findings, hematological values and histopathological findings were observed during or after the drugs administration. In tested groups, the development of inflammatory edema and polyarthritis on day 14 after treatment was suppressed. No significant differences of hindpaw edema volume and lameness score between prednisolone and honeybee venom groups were observed during or after therapeutic drugs treatment. WBC counts of prednisolone and honeybee venom treatment groups as compared with the control group were getting remarkably decreased during or after the therapeutic drugs administration(p<0.01). Erosions of articular cartilage and inflammatory cell infiltrations during or after the therapeutic drugs treatment was effectively suppressed in natural honey venom.

Insect Hormones and Their Actions (곤충의 호르몬과 작용)

  • Basically insect hormones include ecdysteroids (molting hormone), juvenile hormones, and neurohormones comprising neuropeptides and biogenic amines. This article reviewed their chemical structures and biological functions. The active molting hormone is 20-hydroxyecdysone in most insects but makisterone A in some other insects including the honey bee and several phytophagous hemipterans. Most insects use JH III, but lepidopterans JH I and II. Dipterans also use a different JH, so-called JH $B_3$(JH III bisepoxide) and we still do not know the exact chemical structure of JH utilized in hemipterans. Some other insects use methyl farnesoate or hydroxylated JH III analogues as their juvenile hormone. Most diverse pictures can be found in neurohormones (NH), especially in neuropeptides, in terms of their number and structure. There are more than 200 neuropeptides (NP), classified into more than 30 families, which structures have been identified, and more of them are expected to be reported in the near future, partly due to rapid development in molecular biological techniques and in analytical techniques. More than half of them are involved in controlling activity of visceral muscles. But function (s) of many NPs are not clarified yet, even though their amino acid sequences have been identified. It is partly due to the fact that a single NP may have multiple functions. Another interesting point is their gene structure, having many number of independent, active peptides in one gene, apparently working for similar or totally different functions. NH also includes amines, such as octopamine, dopamine, serotonin, etc. From now on, investigation will be concentrated on identifying their function (s) and receptors, and on possibilities of their utilization as control agents against pest insects.

Rearing of a Green Lacewing, Chrysopa pallens Ramber, on Artificial Diets (인공사료를 이용한 칠성풀잠자리붙이의 사육)

  • An aphid predator, Chrysopa pallens Ramber, was reared on the artificial diets containing chicken egg yolk, yeast hydrolysate, brewer's yeast or Vanderzant's vitamin mixture, sucrose andlor bee honey, casein hydrolysate, and cholesterol. On these diets, 20.0 to 70.0% of the 1st instar larvae developed to apparently normal adults depending on diets used. The adults fed on one of these diets which was the most effective laid 230 fertile eggs for her 36 days of adult life span. The nonlipid part of the aphid, Myzus persicae Sulzer was thought to be nutritionally more important than the lipid part for the development of the green lacewing.

Chemical Composition and Bioactivity of Korean Green Tea (Camellia sinensis) Pollen collected by Honey Bee (한국 녹차 화분의 화학적 조성과 기능성 연구)

  • In this study, we investigated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of green tea (Camellia sinensis) pollen grains collected by Apis mellifera bees, for use as a health food. The crude protein and fat content was estimated at 26.14% and 3.49%, respectively. Eighteen amino acids were identified in green tea pollen, including 8 essential amino acids and 10 non-essential amino acids. The predominant amino acids were glutamic acid, proline and aspartic acid accounting for about 33.3% of total free amino acids. The concentration of vitamin C was the highest value of 35.7%, followed by $B_3$ and $B_2$ among the detected vitamins. The predominant minerals were potassium (790.32 mg/100g), followed by phosphorus (707.52 mg/100g) and sulfur (302.67 mg/100g), whereas copper, zinc and sodium were detected as minor elements. The antioxidant activity and phenolic content accounted for 33.8% at $500{\mu}g/mL$ extract and $2.55 {\mu}g/mg$, respectively.

A Study on the System for measuring the Activity of Honeybees inside and outside the Beehive

  • Recently, due to rapid climate change, the population of honey bees has decreased, posing a great threat to the existence of the Earth's ecosystem. In particular, the colony collapse phenomenon in which bees disappeared nationwide in early 2022 had devastating consequences for beekeepers. In order to solve the problems of beekeeping due to climate change, it is urgent to develop a system that can monitor the situation inside the hive through various IoT sensors. This paper develops a system that can measure the activity of bees inside the hive and uses it to measure the number of times of entry and exit of the hive. The data measured by the developed system can be monitored in real time on a smartphone through the cloud server. The system developed in this paper can monitor the ecology of bees according to climate change and measure internal and external bee activities. Using this method, it is possible to check in advance for the colony collapse phenomenon in which bees disappeared in early 2022. This is very meaningful in that it presents an alternative that can identify the cause of the problem through early detection.

  • Mast cell is a cell that functions mainly in our body with a respect to inflammation and allergic response. Bee venom has been progressed in a study as a model related to mechanism in alleviation of pain until now, but it is being progressed in a study relevant to immunocyte in anti-inflammation or anti-allergic response. The present study is aimed to present the basis related to a future study of gene, by researching the influence of melittin and MCD Peptide, which are major ingredients in Bee venom, upon the expression of gene in the mast cell strain. In this study, it dealt with melittin and MCD Peptide respectively, in the effective concentration after passing though the experiment of cytotoxicity by using human mast cell strain. Also, with the respect in the aspect of expression in gene that changes at this time, information was obtained through the technique of analyzing microarray. Through experimental statistics, when regarding a case that global M is significant in more than 1 or -1, in melittin, all 7 genes were accelerated, and 8 inhibited. In MCDP, 7 genes were accelerated and 17 genes inhibited. The function in the body to which these genes are related, was associated with the protein binding within a cell, the activation in the function of lymphocyte, the acceptor related to macrophage antigen. In cell nucleus, substance related to GABA A receptor, protein associated with cAMP reactive element, substance related to complement system No.8 and to B-cell, protein substance related to polycystic kidney disease, substance related to inflammation, and the protein substance of influencing coagulation of blood. Through these results of analysis, it could obtain more useful materials in clarifying the mechanism of action in melittin and MCD peptide, which are in charge of mainly medical action in the abdomen. Also, it is thought that an in-depth study on the influence of main ingredients in Bee venom, the wholly honey bee venom aqua upon anti-allergic response or anti-inflammation are further required.

Acute toxicity of ethyl formate to nontarget organisms and reduction effect of sodium silicate on ethyl formate-induced phytotoxicity (에틸포메이트의 비표적생물에 대한 급성독성 및 sodium silicate의 약해저감 효과)

  • Ethyl formate (EF) is a naturally occurring insecticidal compound and is used to control pests introduced from abroad, in quarantine, by a fumigation method. In particular, it is mainly used as a substitute for methyl bromide and is less toxic to humans and less harmful to plants. This study aimed to investigate the possible acute toxicity of EF to useful organisms, and how to reduce phytotoxicity in watermelon, zucchini, and oriental melon. After fumigation with EF for 2 h, the LC50 values for earthworms, honey bees, and silkworms were 39.9, 7.09, and 17.9g m-3, respectively. The degree of susceptibility to EF was in the order of earthworms, silkworms, and honey bees based on the LC50 value, and EF fumigation induced stronger acute toxicity to honey bees. Phytotoxicity was observed in watermelon leaves treated with a concentration of 7.5 g m-3 EF, and when treated with a concentration of 10.0g m-3, it was confirmed that the edges of watermelon leaves were charred and seemed to be damaged by acids. Zucchini and melon, and other cucurbits, showed strong damage to the leaves when treated with a concentration of 10 g m-3, and sodium silicate, at concentrations of 10% and 20%, was used to reduce phytotoxicity. Therefore, acute toxicity towards nontarget organisms and phytotoxicity during the fumigation of EF should be reduced for efficient agricultural pest control.

Evaluation of Toxicity of Several Environmental Friendly Agricultural Materials on Honeybee (Apis mellifera) and Asian Multicolored Ladybird Beetle (Harmonia axyridis) (꿀벌과 무당벌레에 미치는 몇가지 친환경유기농자재에 대한 독성평가)

  • Toxicity of seven environment friendly agricultural materials (EFAM), which have been used in the domestic market were evaluated on honeybee (Apis mellifera) and asian multicolored ladybird beetle (Harmonia axyridis). Three EFAMs made from plant extract agents (Wangjoongwang Eco, Bogum Eco and Bestop Eco) and four EFAMs made from microbial utilizing agents (Worldstar Eco, Goodmorning, Bluechip and Cameleon) were investigated as EFAMs. In evaluation of toxicity on honeybee, the $RT_{25}$ values of 3 EFAMs made from plant extract agents ranged from 1 to 3 days. Therefore, honeybee should be released 1-3 days after application of these EFAMs. Meanwhile, the four agricultural materials made from microbial utilizing agents did not show any mortality against honeybee. In evaluating the toxicity to adult and larva ladybird beetles, all seven EFAMs made from plant extract agents and microbial utilizing agents to show any mortality.
