Inhibition of the Calcineurin Pathway by Two Flavonoids Isolated from Miliusa sinensis Finet & Gagnep.

  • In order to discover plant-derived signaling pathway inhibitors with antifungal properties, a two-component screening system utilizing the calcineurin and Hog1 mitogen-activated protein kinase pathways responsible for the virulence networks of Cryptococcus neoformans was employed, owing to the counter-regulatory actions of these pathways. Of the 1,000 plant extracts tested, two bioactive compounds from Miliusa sinensis were found to act specifically on the calcineurin pathway of C. neoformans. These compounds, identified as pashanone and 5-hydroxy-6,7-dimethoxyflavanone, exhibited potent antifungal activities against various human pathogenic fungi with minimum inhibitory concentration values ranging from 4.0 to >128 μg/ml.

  • To identify plant-derived cell signaling inhibitors with antifungal properties, a twocomponent screening system using both wild-type Cryptococcus neoformans and a calcineurin mutant was employed owing to their counter-regulatory actions on the Hog1 mitogenactivated protein kinase and calcineurin pathways. Of the 2,000 plant extracts evaluated, a single bioactive compound from M. obovata Thunb. was found to act specifically on the calcineurin pathway of C. neoformans. This compound was identified as magnoloside A, and had potent antifungal activities against various Cryptococcus strains with minimum inhibitory concentration values ranging from 1.0 to 4.0 μg/ml.

  • In this paper, we design and implement a automatic control system of wireless sensor network based sprayer for hog barns. The proposed control system is driven by events from sensor nodes. It gathers various sensor readings such as temperature, humid, water level and water temperature, and controls the sprayer in real time by analyzing the sensor readings. Through experiments, we show that the proposed control system manages temperature and humidity steadily. Our proposed system enhances the existing system about 33% for temperature management and 37.3% for humidity management.

  • Past decade systemic mycoses caused by opportunistic human fungal pathogens, including Candida, Aspergillus, and Cryptococcus, have been a growing problem for both immunocompromised and immunocompetent individuals. Particularly, Cryptococcus neoformans has recently emerged as a major fungal pathogen, which can cause fungal pneumonia and meningitis that are lethal if not timely medicated. However, treatment for cryptococcosis has been difficult due to a lack of proper anti-cryptococcal drugs with fungicidal activity and less toxicity. In this review we introduced novel therapeutic methods for treating cryptococcosis by exploring pathogenomic signa1ing networks of C. neoformans with genome-wide transcriptome approaches as well as diverse molecular/genetic tools.

  • The frequency distribution and morphological characteristics of tissue mast cells in the various tissues of normal domestic animals (bovine, swine, dog and chicken) and systematically infected (Hog cholera, Canine distemper and Newcastle disease) animals were studied. The results were as follows: 1 In cattle, density of mast cells was higher in foetus (bovine) and young animals than in adults Differences in frequency distribution among individual animals were also observed. 2. In chicken, the highest number of mast cells was found in age group of 15 to 40 days, the moderate number in age group of one to 10 days, and the lowest number in age group of 40 days or older. 3. The morphologically, mast cells were usually round, ovoid, spindle, oval and irregular, and particularly in ovary of bovine, it was usually round and ovoid forms. 4. The largest numbers of mast cells were shown in ovary of bovine, intestine of swine and dog, and proventriculum of chicken. 5. In the systemic infections, the number of mart cells usually tends to increase.

  • Trypsin inhibitor produced by Streptomyces sp. was investigated its reactive characteristics against trypsin. The mode of inhibition against trypsin was mixed type of non-competitive and competitive with casein, and enzyme-inhibitor complex was formed rapidly. The inhibitory activity was increased by the addition of isoleucine and depressed by silver, mercuric or cupric ion. And when egg albumin or hemoglobin was used as substrate for trypsin, the inhibition ratio was changed. The inhibitor inhibited coagulation of blood of bovine.

Characteristics of ATPases Present in Everted Membrane Vesicles of Helicobacter pylori

  • Everted membrane vesicles of Helicobacter pylori were prepared and the membrane-resided ATPases were characterized. For comparison, Escherichia coli membrane ATPases and hog gastric mucosal H,K-ATPase were employed. ATPase assay revealed that the composite enzyme pool was relatively low in specific activities, below 1/10 times than that found in E. coli. According to their inhibitory specificities, most of the ATPase pool appeared to belong to the P-type ATPase, sensitive to vanadate but not to azide. The enzyme pool was extraordinarily resistant against treatment by N,N'-dicyclohexylcarbodiimide (DCCD). Certain monovalent cations, e.g., $K^+$ or $NH_4^{+}$ stimulated the whole enzyme pool only in the presence of $Mg^{2+}$. On the contrary, $Ni^{2+}$ and $Zn^{2+}$ increased enzyme activity rather effectively without the aid of $Mg^{2+}$. Under a defined condition employed, H. pylori cells could retain the membrane ATPase pool to the extent of $17{\%}$ at pH 3.2. Moreover, its activity was most stable in acidic conditions (pH 5.4-6.4). However, cytoplasmic or peripheral ATPase pools were hardly detected under acidity (below pH 4.6).

Inhibition of the Calcineurin Pathway by Two Tannins, Chebulagic Acid and Chebulanin, Isolated from Harrisonia abyssinica Oliv.

  • In order to discover and develop novel signaling inhibitors from plants, a screening system was established targeting the two-component system of Cryptococcus neoformans by using the wild type and a calcineurin mutant of C. neoformans, based on the counter-regulatory action of high-osmolarity glycerol (Hog1) mitogen-activated protein kinase and the calcineurin pathways in C. neoformans. Among 10,000 plant extracts, that from Harrisonia abyssinica Oliv. exhibited the most potent inhibitory activity against C. neoformans var. grubii H99 with fludioxonil. Bioassay-guided fractionation was used to isolate two bioactive compounds from H. abyssinica, and these compounds were identified as chebulagic acid and chebulanin using spectroscopic methods. These compounds specifically inhibited the calcineurin pathway in C. neoformans. Moreover, they exhibited potent antifungal activities against various human pathogenic fungi with minimum inhibitory concentrations ranging from 0.25 to over $64{\mu}g/ml$.

  • In this paper, we study The Effects of PV Cell's Electrical Characteristics for PV Module Application. Photovoltaic module consists of serially connected solar cell which has low open circuit voltage and high short circuit current characteristics. The whole current flow of PV module is restricted by lowest current of one solar cell. For the experiment, we make PV module composing the solar cells that have short circuit current difference of 0%, 1%, 3% and Random. The PV module exposed about 35days, its the maximum power drop ratio was 4.282% minimum and 6.657% maximum. And PV module of low current characteristics has electrical stress from other modules. The solar cell temperature of PV module was higher compared to PV cell. To prevent early degradation, it is need to have attention to PV cell selection.

A SENSITIVITY STUDY ON NEUTRONIC PROPERTIES OF DUPIC FUEL

  • A sensitivity study has been done to determine the composition of DUPIC fuel from the viewpoint of neutronics fuel design. The spent PWR fuel compositions were generated and fissile contents adjusted by blending fresh uranium after mixing two spent PWR fuel assemblies. The $^{239}$ Pu and $^{235}$ U enrichments of DUPIC fuel were adjusted by controlling the amount of fresh uranium feed and the ratio of slightly enriched and depleted uranium in the fled uranium. Based on the material balance calculation, it is recommended that DUPIC fuel composition be such that spent PWR fuel utilization is more than 90%.. A sensitivity study on the temperature reactivity coefficient of DUPIC fuel has shown that it is desirable to increase the $^{239}$ Pu and $^{235}$ U contents to reduce both the fuel and coolant temperature coefficients. On the other hand, refueling simulations of the DUPIC core have shown that the channel power peaking factor, which is a measure of the reactor trip margin, increases with the total fissile content. Considering these neutronic characteristics of the DUPIC fuel, il is recommended to have enrichments of 0.45 and 1.00 wt% for $^{239}$ Pu and $^{235}$ U, respectively.
