Anti-allergy Action of Soeumin Kwakhyangjeonggisan (소음인(少陰人) 곽향정기산(藿香正氣散)의 항(抗) Allergy 작용(作用))

  • The purpose of this research was to investigate the effects of Kwakhyangjeonggisan (KJS) on the anti-allergic action. In the present study, we examined the effect of KJS on type I and type IV allergic reaction. KJS inhibited the systemic anaphylaxis induced by compound 48/80 and platelet activating factor (PAF), and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KJS dose-dependently inhibited the release of histamine from peritoneal mast cells in rat. Also, KJS inhibited the delayed type hypersensitivity (DTH) induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene (DNFB). KJS inhibited the proliferation of splenocytes, the subpopulation of B220+ cells and CD4+CD8-(Th) cells in splenocytes and the production of γ-interferon in serum and splenocytes. These findings suggest that KJS prevented the type I allergy by the inhibition of histamine release from mast cells and the type IV allergy by the inhibition of γ-interferon production and B lymphocytes subpopulation. These results indicate that KJS may be useful for the prevention and treatment of type I and type IV allergy related disease.

Inhibitory Effects of Chrysanthemum zawadsaki Ethanolic Extract on $Fc{\varepsilon}RI$ ${\alpha}$ Chain Expression (구절초 에탄올 추출물의 $Fc{\varepsilon}RI$ ${\alpha}$ chain 발현 저해효과)

  • Human basophilic KU812F cells express a high affinity for IgE receptor, $Fc{\varepsilon}RI$, which plays an important role in immunoglobulin E (IgE)-mediated allergic reactions. Using the human basophilic KU812F cells, we assessed the inhibitory effects of the ethanolic extract from Chrysanthemum zawadsaki(CZ) on $Fc{\varepsilon}RI$ ${\alpha}$ chain expression. Flow cytometric analysis showed that cell surface $Fc{\varepsilon}RI$ ${\alpha}$ chain expression was suppressed when the cells were cultured with CZ extract. RT-PCR analysis showed that the mRNAs for $Fc{\varepsilon}RI$ ${\alpha}$ chain decreased by the treatment of CZ extract. Moreover, CZ extract was also observed to result in a reduction in the release of histamine from anti-$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells. These results suggest that CZ extract may exert anti-allergic activity through down-regulation of $Fc{\varepsilon}RI$ ${\alpha}$ chain expression and a subsequent decrease in histamine release.

Vaccinium angustifolium Root Extract Suppresses FcεRI Expression in Human Basophilic KU812F Cells.

  • Vaccinium angustifolium, commonly known as the lowbush blueberry, is a rich source of flavonoids, with which various human physiological activities have been associated. The present study focuses on the investigation of the effect of the methanolic extract of V. angustifolium root extract (VAE) on high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$) ${\alpha}$ chain antibody (CRA-1)-induced allergic reaction in human basophilic KU812F cells. The total phenolic content of VAE was found to be $170{\pm}1.9mg$ gallic acid equivalents/g. Flow cytometry analysis revealed that the cell surface expression of $Fc{\varepsilon}RI$ was suppressed in a concentration-dependent manner upon culture with VAE. Reverse-transcriptase polymerase chain reaction analysis showed that the mRNA level of the $Fc{\varepsilon}RI$ ${\alpha}$ chain was reduced in a concentration-dependent manner as a result of VAE treatment. Western blot analysis revealed that the protein expression of $Fc{\varepsilon}RI$ and the phosphorylation of extracellular signal-regulated kinases (ERK) 1/2 were concentration-dependently inhibited by VAE. We determined that VAE inhibited anti-CRA-1-induced histamine release, in addition to the elevation of intracellular calcium concentration ([$Ca^{2+}$]i), in a concentration-dependent manner. These results indicate that VAE may exert an anti-allergic effect via the inhibition of calcium influx and histamine release, which occurs as a result of the down-regulation of $Fc{\varepsilon}RI$ expression through inhibition of ERK 1/2 activation.

Inhibitory Effects of Allergic Reaction of Aqueous Extract of Salviae Radix Root by Anal Therapy (항장요법(肛腸療法)에 의한 단삼추출액(丹蔘抽出液)의 알레르기반응 억제효과(抑制效果))

  • Dansam, the root of Salvia miltiorrhiza BGE. (Labiatae), has a bitter and a slightly 'cold' property. and is nontoxic. It has been used for the treatment of diseases such as tumors, bruises, gynecologic diseases, menoxenia, anemia and so forth. As an oriental medicine pathway, anal therapy has many kinds of treatments, Retention enema is one the most useful, advantageous methods of anal therapy. This experiment was performed in order to study the effect of an aqueous extract of Salviae radix root(SRRAE) on Immediate type allergic reaction by Retention enema. The results were obtained as follows, 1. SRRAE inhibited compound 48/80-induced Immediate type allergic reaction 100% with the dose of 0.1g/kg by anal treatment. However, SRRAE showed no significant inhibitory effect on the same reaction by oral treatment. When mice were pretreated with SRRAE at a concentration ranging from 0.001 to 1.0g/kg, by intra-anal treatment, the serum histamine levels were reduced in a dose-dependent manner. 2. SRRAE also inhibited by 79.8% local cutaneous allergic reaction activated by anti-dinitrophenyl (DNP) IgE. 3. SRRAE close-dependently inhibited the histamine release from rat peritoneal mast cells(RPMC) by anti-DNP IgE., but SRRAE not inhibited compound 48/80-induced histamine release. 4. SRRAE dose-dependently inhibited tumor necrosis $factor-\;{\alpha}$ $(TNF-\;{\alpha}$) production from RPMC by anti-DNP IgE. Moreover, the level of cAMP in RPMC, when SRRAE was added, significantly increased compared with that of a normal control. According to the above results, anal therapy(Retention enema) of SRRAE may be beneficial in the treatment of systemic and local Immediate type allergic reactions.

The Effects of Gyejijakyakjimo-Tang on the Allergic Contact Dermatitis induced by DNCB (계지작약지모탕(桂枝芍藥知母湯)이 DNCB로 유발된 알레르기성 접촉피부염에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effects of Gyejijakyakjimo-Tang on the Allergic Contact Dermatitis caused by 2,4-dinitro-chlorobezene(DNCB). Methods : Twenty eight mice were divided into four groups ; normal, control, experimental group A and B. Control and experimental groups were induced allergic contact dermatitis by DNCB. Experimental group A was orally administered the Gyejijakyakjimo-Tang and experimental group B was orally administered the prednisolone. In this study, ear thickness measurement, auricle microphotograph observation, MPO(Myeloperoxidase) activity measurement, Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA level of TNF-$\alpha$, IL-$1{\beta}$ were performed on these four groups. In addition, the effect of Gyejijakyakjimo-Tang on cell viability and the effect of Gyejijakyakjimo-Tang on the compound 48/80-induced histamine release from HMC and RPMC were measured. Results: 1. Both experimental group A and B had decreased ear thickness compared with control group In contact hypersensitivity assay. 2. In experimental group A, inflammatory edema was similarly observed comparing to control group. Nevertheless, inflammatory edema was obviously reduced in experimental group B. In both experimental group A and B, pathological lesion of dermatitis were alleviated. In addition, the numbers of infiltrated inflammatory cells were decreased compared with control group. 3. Compared to the normal group, there was a noticeable increase in MPO activity in control group. However, in experimental group A and B, it showed remarkable inhibition of the increase in MPO activity comparing with control group. 4. The level of expression of TNF-$\alpha$, IL-$1{\beta}$ in experimental group A and B were meaningfully lower than those in control group. 5. In MTT assay, the concentrations of Gyejijakyakjimo-Tang that were used on the test had no cytotoxicity. 6. Gyejijakyakjimo-Tang dose-dependently inhibited the compound 48/80-induced histamine release from both HMC and RPMC. Conclusions : According to above experiments, Gyejijakyakjimo-Tang was effective on allergic contact dermatitis.

Inhibitory Effects of Histamine Production in Mackerel Muscle by Medicinal Herbs and Seaweed Extracts (한약재 및 해조류 추출물에 의한 고등어 육중의 Histamine 생성 억제 효과)

  • This study was conducted in order to investigate the inhibitory effects of natural materials on histamine production in mackerel. Antimicrobial activities on Photobacterium phosphoreum of medicinal herbs and seaweeds were investigated using the paper disc assay and MIC (minimum inhibitory concentration) test. According to the results, Sargassum sagamianum and Ecklonia cava ethanol extracts exhibited antibacterial activity. In particular, Sargassum sagamianum ethanol extract showed excellent antibacterial activity at 0.015625 mg/mL by the MIC test. Anti-histamine release activities of natural materials were further investigated by examining their inhibitory effects on histidine decarboxylase (HDC) activity in the crude enzyme preparation from Photobacterium phosphoreum. The ethanol extracts of Ecklonia cava and Eisenia bicyclis exhibited the strongest HDC inhibitory activity, with 32% and 22%, at a concentration of 1 mg/mL, respectively. Therefore, natural materials may reduce histamine poisoning through decrease of histamine production in mackerel.

Efficacy of 20 Select Herbs Against Common Cold (감기 치료 빈용 처방 구성 약물의 효능 연구)

  • Objectives: To investigate the anti-inflammatory, analgesic, anti-pyretic and anti-histamine activities of 20 herbal medicines to test their efficacy in common cold treatment. Methods: For all experiments, the herbal medicines were extracted with 80% ethanol and freeze-dried. To determine the anti-oxidative properties, we tested DPPH-free radical-scavenging activity and xanthine oxidase inhibitory activity. To determine anti-inflammatory and analgesic potential, we investigated acetic acid-induced vascular permeability and writhing test in ICR mice. For anti-pyretic activities, an LPS-induced pyrexia study was conducted in rabbits. To evaluate the anti-histamine activity, we examined compound 48/80-induced systemic anaphylaxis in ICR mice and the release of β-hexosaminidase on rat basophilic leukemia (RBL-2H3) cells. Results: Ephedrae herba, Forsythiae fructus, Cinnamomi ramulus, and Cimicifugae rhizome showed potent free-radical scavenging activities. Gentianae macrophyllae radix inhibited acetic acid-induced vascular permeability. Schizonepetae spica and Cimicifugae rhizome inhibited acetic acid. Cinnamomi ramulus and Angelicae decursivae radix inhibited LPS-induced pyrexia. Angeliace dahuricae radix and Asari radix inhibited compound 48/80. Scutellariae radix, Cinnamomi ramulus, Ephedrae herba, and Zingiberis rhizoma crudus potently inhibited the release of β-hexosaminidase. Conclusions: We examined the anti-inflammatory, analgesic, anti-pyretic and anti-histamine activities of 20 herbal medicines;We examined the anti-inflammatory, analgesic, anti-pyretic and anti-histamine activities of 20 herbal medicines Codonopsis pilosulae radix, Zingiberis rhizoma crudus, and Cinnamomi ramulus showed novel efficacy. These results suggest that some of herbal medicines may be very effective in treating common cold.

Effect of Panax Ginseng Alcohol Extract on the Contractility of Isolated Guinea Pig Tracheal Muscle (인삼(Panax Ginseng)주정추출액이 기관지 평활근의 수축력에 미치는 영향)

  • It has been reported that administration of Ginseng powder to the Guinea pig reduces anaphylactic shook induced by horse serum (Lee, 1939). However, Lee et al. (1960) and Paik et al. (1976) have demonstrated that Ginseng increases capillary permeabilites and histamine release from the mast cell. These facts suggest that Ginseng acts directly on the bronchial muscle causing it to dilate. Recently, a number of investigators(Kidakawa & Iwasiro 1963; Takagi et al. 1973) have reported that Ginseng reverses acetylcholine- or histamine- induced contraction in the isolated Guinea pig ileum. We, therefore, undertook the present study to examine if Ginseng relaxes the spasm of bronchial muscle induced by acetylcholine or histamine. We have also attempted to identify the mechanism of the Ginseng effect. Male Guinea Pig was sacrificed by a blow on the head, The trachea was removed and sectioned with scissors into about 12 rings. After the 'C' shaped ring of cartilage was sectioned the one end of ring was tied to the bottom of the incubation bath and the other end was connected to a force transducer (FTO 3C) to record tension on a Polygraph. When the antispasmodic action of Ginseng effect was first examined in the normal trachea which was not treated by the drug. And then the Ginseng effect was tested in the muscle treated by histamine hydrochloride, acetylcholine hydrochloride or barium chloride. The results indicate that Ginseng alcohol extract relaxes the contraction of isolated tracheal muscle induced by histamine $(1{\mu}g/ml{\sim}10{\mu}g/ml)$, acetylcholine $(1{\mu}g/ml{\sim}5{\mu}g/ml)$ and barium chloride (1.5 mg/ml). The mechanism of this action is in Pa.1 due to nonspecific antimuscarinic and antihistaminic effect and in part by predominant action in the adrenergic ${\beta}-receptor$ although the ${\alpha}-receptor$ is also involved. We, therefore, conclude that Ginseng can be act as a bronchodilator.

Inhibitory Effect of Saururus chinensis (Lour.) Baill Extracts on Allergy in Mouse Models (Mouse 모델 알레르기 반응에서 삼백초(三白草)가 미치는 영향)

  • We investigated the effect of Saururus chinensis (Lour.) Baill (SCB) on allergy in mice. We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.

Inhibitory Effect of Salvia plebeia on Compound 48/80-Induced Immediate Hypersensitivity Reaction (Compound 48/80 유도 즉시형 과민반응에 대한 뱀차즈기의 억제효과)

  • We studied the inhibitory effect of the aqueous extract of Salvia plebeia (SPAE) on immediate hypersensitivity reactions. SPAE inhibited immediate hypersensitivity reaction induced by compound 48/80 in mice. When SPAE was employed in an immediate hypersensitivity reaction test, the plasma histamine levels were reduced in a dose-dependent manner. SPAE also inhibited the histamine release from rat peritoneal mast cells (RPMC) by compound 48/80. The level of cAMP in RPMC, when SPAE was added, significantly increased compared with that of a normal control. These results indicate that SPAE may be beneficial in the treatment of immediate hypersensitivity reactions.
