Evaluation of Useful Biological Activities of Hot-Water Extracts of Raw-Red Bean and Boiled-Red Bean (Phaseolus radiatus L.) (생팥 및 삶은 팥의 열수 추출물의 유용 생리활성 평가)

  • Raw-red bean (RR) should be boiled in hot water, and only boiled-red bean (BR) has been used in the food industry. In the course of development of functional food using red- bean (Phaseolus radiatus L), hot- water extracts (HWEs) of RR and BR were prepared, respectively and their components and various biological activities were compared. The extraction yield at $100^{\circ}C$ of RR (16.2%) was higher than that of BR (14.8%), and contents of total polyphenols, total flavonoids and reducing sugars of HWE of RR were 2.5-fold, 2.1-fold and 1.5-fold higher than those of HWE of BR. In anti-oxidation activity assay, scavenging activities against DPPH anion and ABTS cation as well as reducing power of RR was higher than those of BR. The results suggest that the anti-oxidant compounds in red bean might be heat-liable or discarded during boiling in hot-water as a cooking drip. Unexpectedly, nitrite scavenging activity was stronger in HWE of BR than RR. In anti-microbial activity assay, HWE of RR ($500{\mu}g/disc$) showed growth inhibition activity against gram-positive bacteria, whereas HWE of BR did not show any activity against any tested bacteria and fungi. Assay of in-vitro anti-diabetes and anti-thrombosis activities, which were previously reported in ethanol extract of red-bean, revealed that HWEs of RR and BR did not show significant activities against ${\alpha}$-amylase, ${\alpha}$-glucosidase, thrombin, prothrombin, or blood coagulation factors. Our results suggest that the anti-oxidation, anti-diabetes and anti-thrombosis activities of HWEs of RR and BR were lower than those of ethanol extracts of red bean, and bioactive substances in RR were destroyed during boiling or discarded after boiling. Further research on suitable boiling and re-use of cooking drip of red bean is necessary.

Effect of Heterofermentative Lactic Acid Bacteria on the Quality of Italian Ryegrass and Whole-crop Barley Silage (Hetero 발효 유산균주 첨가가 사일리지용 이탈리안 라이그라스와 총체보리의 품질에 미치는 효과)

  • This study was conducted to determine the quality of italian ryegrass (IRG) and whole- crop barley (WCB) silage combined with heterofermentative lactic acid bacteria (LAB) during fermentation. Six strains of LAB (L. plantarum IMAU 70164, L. acidophilus KACC 12419, L. casei KACC 12413, L. reuteri KCTC 3594, L. buchneri KACC 12416 and L. diolivorans KACC 12385) were used in this study. L. casei and L. reuteri had the highest propionic acid production and were therefore used for fermenting the forages. The forages were fermented using monoculture and co-culture of L. casei and L. reuteri for 30, 45 and 60 days of ensiling. Addition of LAB lowered the pH of the IRG silage (p<0.05). Moisture content, lactic acid and acetic acid contents were higher (p<0.05) after addition of LAB. Water soluble carbohydrate was significantly lower (p<0.05) in WCB with a co-culture containing L. casei and L. reuteri. Propionic acid production was comparatively higher after addition of LAB to WCB on days 30, 45 and 60 while butyric acid was only detected in the IRG control on day 60. Fungi was not detected within 60 days after addition of LAB in IRG and WCB. Through this experiment, improved forage preservation was achieved using a co-culture containing L. casei and L. reuteri. WCB silage had higher propionic acid concentration and thus, it was a better forage for ensiling using co-culture of L. casei and L. reuteri.

  • The characteristics of the six different agricultural soil from Michigan were as follows. Colwood and Capac soil were loam, Gilford and Ceresco were sandy clay loam, Sission was clay loam, and Spinks was sandy loam. pH of Gilford sandy clay loam was 6.6 whereas that of the soil ranged $5.4{\sim}5.9$. Gilford sandy loam found to contain a relatively higher amount of organic matters as compared to other soils. Furthermore, the numbers of bacteria in Gilford sandy clay loam were significantly higher than those in other soils. The populations of fungi in Gilford sandy clay loam and Colwood loam soils were significantly greater than those in other soils. On the other hand, the densities of actinomycetes in Gilford sandy clay loam and Ceresco sandy clay loam soils were significantly different from those in other soils. The population of anaerobic bacteria varied depending on the soils; Ceresco sandy clay loam, Capac loam, Colwood loam soils have higher numbers of bacteria, whereas Gilford sandy clay loam was very lesser than the other soils. In the $^{14}C-glucose$ respiration by soil microorganisms after 10 hrs, the respiration rate was decreasing in the order of Ceresco sandy clay loam, Spinks sandy loam, Colwood loam, Sission clay loam, Capac loam and Gilford sandy clay loam. Germination of test propagules on natural soil soil was $0{\sim}5%$, and it was germinated $90{\sim}98%$ on autoclaved soil and PDA. The propagules differed in thier germination response to nutrients added to the soils. In general, more nutrients were required to promote germination on Capac loam and Gilford sandy loam soil than Spinks sandy loam soil. Especially Mortierella n. sp. required more nutrients for germination to obtain the same ratio as Helminthosporium victoriae.

Degradation Ability and Population of Resistant Strains of Chlorothalonil in Upland Soil Distributed in Honam Area (호남지역 밭토양에 분포된 Chlorothalonil 내성균(耐性菌)의 밀도(密度)와 분해능(分解能))

  • This experiment was conducted to obtain the basis of degradation of remaining agricultural chemicals accumulated in upland soils of Honam district in Korea. The population. relative growth rate(RGR). chlorothalonil(TPN)-degradation ability and bacterialogical characteristics of TPN resistant strains were investigated in TPN levels of 0, 25, 50, 100 and $500{\mu}l/ml$ compared with Mancozeb. A number of TPN-resistant bacteria were differ in the area of examined and were decreased with higher levels of TPN. The resistance of bacteria was stronger in TPN than Mancozeb but the resistance of fungi was vise versa. RGR of bacteria in the culture was the highest at the level of $50{\mu}l/ml$ and the lowest in $500{\mu}l/ml$ of TPN. TPN-degradation ability of bacteda isolated in various TPN levels was varied : only 8 percentage of bacteria showed 75 percentage or more degradation ability. The higher the concentration in TPN resistance, the larger the number of strains carried great ability to decompose pesticide residues. The strains having higher decomposition ability was rod-shapes cells and senstive to heat. Analyses of the indol production, methyl red, and V-P test have given similar results, with negative reaction in all these strain, while the other biochemical characteristics were differ in the strains. Based on these, these strains might be classified into Pseudomonas sp., Corynebacterium sp., Acinetobacter sp. and Moraxcella sp.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. Site Characteristics and Vegetation Structure of the Habitat of Cauliflower Mushroom (Sparassis crispa) (꽃송이버섯 자생지의 입지특성 및 식생구조)

  • From 2006 to 2008, natural habitat of Sparassis crispa were surveyed for investigating ecological conditions at sixteen different sites in Korea. The investigated sites showed very wide altitudinal distribution ranged from 240 meters to 1,100 meters above the sea level. In general, S. crispa showed distinct feature of heart-rot fungi as it occurs on soils around the stems of larch (Larix kaempferi) and Korean white pine (Pinus koraiensis). But it also could be found on stems or on the stumps of dead trees, which indicated that the fungus might have several invasion routes and capabilities to grow on various ecological conditions. All of the sixteen sites were pure stands consisted with conifers such as larch or Korean white pine. The dominant tree layer showed $15.3{\sim}38.0$ meters for tree heights, thicker than twenty centimeters for the diameter at breast height (DBH), and all of them were older than thirty years. Since the stands were pure stand, species diversity of trees in the sites was extremely low. While the dominant tree layer showed only pure coniferous stand, the co-dominant tree layer, shrub layer and herbaceous layer showed more diverse features with higher Shannon-Wiener (H') indices. Soil texture of thirteen sites among sixteen investigated sites were loamy soils, and the contents of organic matter in soil were more or less higher than general forest soils in Korea with $3.79{\sim}14.32%$. The cation exchange capacity (CEC) was also relatively higher than general forest soils with $16.1{\sim}27.2$ cmol+/kg. The data indicated that the cauliflower mushroom occurring sites were relatively fertile than general forest soils. The soils were acidic with pH ranged from 4.2 to 5.2, which were typical features for conifer stands in Korea.

Effect of thinning ratio on the forest environment and fruiting of ectomycorrhizal mushrooms in a Pinus densiflora stand (소나무림에서 간벌률이 산림 내 환경과 외생균근성 버섯 발생에 미치는 영향)

  • To investigate the effect of thinning intensity on environmental factors and ectomycorrhizal mushroom fruiting in forest ecosystems, we studied canopy closure, throughfall, soil temperature, soil moisture, light response of understory vegetation, and ectomycorrhizal mushroom fruiting in a 10-year-old pine forest after 34%, 45%, and 60% thinning. Canopy closure was significantly higher in the 34% treatment and control plots, ranging from 80-85% in April. However, in November, all thinning treatment plots showed a decrease of approximately 5-10% compared with the control plot. The 60% treatment plot had over 200 mm of additional throughfall compared with the control plot, and monthly throughfall was significantly higher by more than 100 mm in October. The soil temperature in each treatment plot increased significantly by up to 1℃ or more compared with the control plot as the thinning rate increased. The soil moisture increased by more than 5% in the thinning treatment plots during rainfall, particularly in the 34% treatment plot, where the rate of moisture decrease was slower. The photosynthetic rate of major tree species (excluding Pinus densiflora)was highest in Quercus mongolica, with a rate of 7 µmolCO2·m-2·s-1. At a lightintensity of 800 μmol·m-2·s-1, Q. mongolica showed the highest photosynthetic level of 6 ± 0.3 μmolCO2·m-2·s-1 in the 45% treatment. The photosynthetic rate of Fraxinus sieboldiana and Styrax japonicus increased as the thinning intensity increased. The Shannon-Wiener index of mycorrhizal mushrooms did not significantly differ among treatments, but the fresh weight of mushrooms was approximately 360-840 g higher in the 34% and 45% treatments than in the control. Additionally, the fresh weight of fungi in the 60% treatment was 860 g less than that in the control. There were more individuals of Amanita citrina in the control than in the thinning treatment, while Suillus bovinus numbers increased by more than 10 times in the 34% thinning treatment compared with the control.

Bioactivities and Isolation of Functional Compounds from Decay-Resistant Hardwood Species (고내후성 활엽수종의 추출성분을 이용한 신기능성 물질의 분리 및 생리활성)

  • Wood of Robinia pseudoacacia and bark of Populus alba$\times$P. glandulosa, Fraxinus rhynchophylla and Ulmus davidiana var. japonica were collected and extracted with acetone-water(7:3, v/v) in glass jar to examine whether its bioactive compounds exist. The concentrated extracts were fractionated with hexane, chloroform, ethylacetate and water, and then freeze-dried for column chromatography and bioactive tests. The isolated compounds were sakuranetin-5-O-$\beta$-D-glucopyranoside from Populus alba $\times$Pl glandulosa, 4--ethyoxy-(+)-leucorobinetinidin frm R. pseudoacacia and fraxetion from F. rhynchophylla and were characterized by $^1H$ and$^{13}C $ NMR and positive FAB-MS. Decay-resistant activity was expressed by weight loss ratio and hyphae growth inhibition in the wood dust agar medium inoculated wood rot fungi. R. pseudoacacia showed best anti-decaying property in both test and its methanol untreated samples, indicating higher activity than methanol treated samples in hyphae grwoth test. In antioxidative test, $\alpha$-tocopherol, one of natural antioxidants, and BHT, one of synthetic antioxidants, were used as references to cmpare with the antioxidant activities of the extacted fractions. Ethylacetate fraction of F. rhynchophylla bark indicated the hightest activity in this test and all fractions of R. pseudiacacia extractives also indicated higher activities compared with the other fractions. In the isolated compounds, aesculetin isolated from F. rhynchophylla bark showed best activity and followed by robonetinidin from R. pseudoacaica.

Analysis of Quality Properties and Fermentative Microbial Profiles of Takju and Yakju Brewed With or Without Steaming Process (증자 혹은 무증자 탁주 및 약주의 품질특성 및 발효관련 미생물 분석)

  • Steamed rice is usually used as an essential ingredient when Takju or Yakju is brewed in Korea. Alternatively, non-steamed rice can be used to keep thermolabile nutrients and fresh tastes richer in Takju or Yakju. In this study, therefore, the physicochemical properties (ethanol and sugar contents, pH, total acidities, and turbidities) and the fermentative microbial profiles (aerobic mesophillic bacteria (AMB), fungi, lactic acid bacteria (LAB), acetic acid bacteria (AAB), and Escherichia coli and coliforms) have been compared among 4 Takju and 1 Yakju samples brewed using steamed or non-steamed rice. Yakju brewed using non-steamed rice has approximately 2-3 times higher ethanol and sugar contents than other 4 Takjus brewed using steamed or non-steamed rice. The pH and total acidity values of all the 5 samples ranged 3.77-4.30 and 0.12-0.35, respectively. As for turbidities, Yakju brewed using nons-teamed rice was transparent, but other 4 Takjus were not. The AMB and fungal counts for Yakju brewed using nons-teamed rice were approximately $10^4$-fold less than those for 4 Takjus. The LAB counts for Takju and Yakju brewed using non-steamed rice were $10^3$-fold less than those for Takjus brewed using steamed rice. The AAB counts ranged $2-6\;log_{10}\;CFU/mL$ for all the 5 samples. E.coli and coliforms were not detected. Overall, there was no significant difference in microbial counts among 4 Takjus brewed using steamed or non-steamed rice, but Takju has higher microbial counts than Yakju. All the 5 samples were conclusively considered to be hygienically brewed and processed containing plenty of beneficial microorganisms.

Characteristics of Soil Electrical Conductivity in Plastic Film House Located in Southern Part of Korea (남부지방(南部地方) 시설재배지(施設栽培地) 토양(土壤)의 염농도(鹽濃度) 특성(特性))

  • Characteristics of electrical conductivity(EC) of seventy one-greenhouse soil located in southern part of Korea were surveyed to obtain the basic information for sustainable management of plastic film house soils. The averaged soil EC were 5.84 and 2.49dS/m for top-and sub-soil, respectively, showing distribution of 11.3% for top-soils and 50.7% for sub-soils within the optimum level of 2.0 dS/m. On the whole, the soils with flowering plants revealed higher EC than those with fruiting vegetables showing 2.75~6.34dS/m for the former and 2.29(sub-soils)~5.13dS/m(top-soils) for the latter. In a view of soil sampling time, the soils at Feb., 1996 marked 4.3~37.7% and 10.3(top-soils)~11.3%(sub-soils) higher EC compared with the soils at Sept. and Dec., 1995, respectively. The linear regression correlation coefficients of soil minerals to EC were in oder of $Mg^{2+}$ > $NO_3{^-}$ > $Cl^-$ > $Ca^{2+}$ > $SO_4{^{2-}}$ > $K^+$ for top-soils and were $Cl^-$ > $SO_4{^{2-}}$ > $NO_3{^-}$ > $Ca^{2+}$ > $Mg^{2+}$ > $K^+$ for sub-soils. In especial, the concentration of soil EC accompanied the increasing population ratio of soil fungi to bacteria and/or actinomycetes.
