Comparison of Genetic Diversity and Population Structure of Kalopanax pictus (Araliaceae) and its Thornless Variant Using RAPD

  • Kalopanax pictus is a long-lived woody species mostly distributed in East Asia. K. pictus has been regarded as medically and ecologically important species in Korea. Thornless castor aralia variant, local name 'Cheongsong' is an endemic to Cheongsong province in Korea. Random amplified polymorphic DNA (RAPD) was used to investigate the genetic variation and structure of Korean populations of two species. A high level of genetic variation was found in six K. pictus populations. Twelve primers revealed 49 loci, of which 29 were polymorphic (59.2%). Nei's gene diversity for K.pictus and K. pictus variant were 0.119 and 0.098, respectively. Mean of genetic diversity in K. pictus was higher than average values for species with similar life history traits. The asexual and sexual reproduction, perennial habitat, and longevity are proposed as possible factors contributing to high genetic diversity. An indirect estimate of the number of migrants per generation (Nm=0.857) indicated that gene flow was not extensive among Korean populations of K.pictus. It is suggested that the isolation of geographical distance and reproductive isolation between K.pictus and K.pictus variant populations may have played roles in shaping the population structure of this species.

  • Most of the naval shipboard equipments strongly require isolation devices to keep their performance and durability from severe noise and vibration loading. The rubber mount is well-known as one of most effective way to isolate major equipments from noise and vibration. The performance of the rubber mount is determined by characteristics of shock stiffness and vibration stiffness and the frequency range of interest is up to several Kilo Hz in navy vessels. In this study, an vibration experiment method for the rubber mount has been developed as a foundation work for assessing dynamic stiffness up to high frequency. Also experiments for a typical rubber mount has been carried out and the results has been discussed.

  • Isolation of BacilLus sp. producing multi-enzyme and optimization of medium conditions for its production using feedstuffs for probiotics were carried out in this study. A bacterium isolated from natural resources, namely Bacillus subtilis 4-3, has multi-enzyme activity (phytase. cellulase, xylanasc, protease, and amylase. In the culture of B. subtilis 4-3 using soybean meal and rice bran. relatively low phytate degradation was noted using whereas high phytate degradability was observed with wheat bran (80.63%). The optimal composition of medium using feedstuffs was 1.0% (w/v) soybean meal and 2% (w/v) molasses to yield high cell growth.

  • In this paper, we designed resistive MMIC mixer using $0.5{\mu}m$ p-HEMT process. This Mixer is designed to have a similar performance in -4 ~ 4 dBm local oscillator signal power level and to maintain a constant conversion loss and linear performance due to the variation of local signal. In order to have such characteristics, we designed new feedback circuit topology by using FET, and minimized performance change for LO signal power level variation, also obtain MMIC mixer characteristics which is able to apply in wideband. In the design result, When the LO signal power is -4 ~ 4 dBm, there was 6 dB conversion loss and it came up with the excellent result that IIP3 got over 30 dBm in 0.5 ~ 2.6GHz frequency band.

  • Response to nerve stimulation platform for implementing measures to detect finger movement has been functioning as an important factor. This stimulated finger on the nerve and muscle responses would vary. In other words, the finger movement of the muscle response to nerve stimulation and sensing Actuator for the H/W development is needed. In addition, a low power embedded CPU based on the top was used.   • A solid state transformer (SST) that can interface an MVAC of three-phase 13.2 kV and a 1.5 kV DC distribution. SST consists of an AC/DC converter and a DC/DC converter with a high-frequency isolation transformer (HFIT). The AC/DC converter consists of cascaded NPC full-bridge to cope with the MVAC. The DC/DC converter applies a quad active bridge (QAB) topology to reduce the number of the HFIT. Topology analysis and controller design for this specific structure are discussed. In addition, the insulation of HFIT used in DC/DC converters is considered. The discussion is validated using a 300 kVA three-phase SST prototype.

  • Micro-scale magnetic beads are widely used for isolation of proteins, DNA, and cells, leading to the development of in vitro diagnostics. Efficient isolation of target biomolecules is one of the keys to developing a simple and rapid point-of-care diagnostic. A zinc finger protein (ZFP) is a double-stranded (ds) DNA-binding domain, providing a useful scaffold for direct reading of the sequence information. Here, we utilized two engineered ZFPs (Stx2-268 and SEB-435) to detect the Shiga toxin (stx2) gene and the staphylococcal enterotoxin B (seb) gene present in foodborne pathogens, Escherichia coli O157 and Staphylococcus aureus, respectively. Engineered ZFPs are immobilized on a paramagnetic bead as a detection platform to efficiently isolate the target dsDNA-ZFP bound complex. The small paramagnetic beads provide a high surface area to volume ratio, allowing more ZFPs to be immobilized on the beads, which leads to increased target DNA detection. The fluorescence signal was measured upon ZFP binding to fluorophore-labeled target dsDNA. In this study, our system provided a detection limit of ≤ 60 fmol and demonstrated high specificity with multiplexing capability, suggesting a potential for development into a simple and reliable diagnostic for detecting multiple pathogens without target amplification.

  • This paper presents a tri-wideband bandpass filter (TWB-BPF) with compact size, high band-to-band isolation, and multiple transmission zeros (TZs). The proposed TWB-BPF is based on a multiple-mode resonator (MMR), which is interpreted by the method of the even- and odd-mode analysis technique. The MMR can excite 11 resonant modes, where the first two modes comprise the first passband, the next four modes form the second passband, and the last five modes are used to generate the third passband. In addition, 10 TZs are yielded to obtain high band-to-band isolation and wide stopband suppression characteristics up to $14.95f_{c1}$ ($f_{c1}$ is the center frequency of the first passband). To verify the proposed filter, a TWB-BPF with 3-dB fractional bandwidths (FBWs) of 37.4%, 43.5%, and 40.4% is designed, fabricated, and measured.

  • Current codes of practice do not exist in isolation, and rules that have been developed assume certain material properties and minimum workmanship in fabrication and erection. These are either in separate documents or different parts of the code. This paper explains the main requirements for materials and workmanship and how they can be related to design and construction in general. The use of very high strength steels is also considered and the measures that may be needed to allow their use with modern codes are also presented.

  • This paper presents analysis of control characteristics of high power hybrid multilevel PWM rectifier without bulky and heavy isolation transformers. It will be shown that the DC voltages for multilevel output generation may be directly built up from AC utility source. The multilevel PWM rectifier is analyzed by using the circuit DQ transformation whereby the static and dynamic characteristics and some useful design equations are obtained. Finally to confirm the validity of the analysis MATLAB simulation are carried out.
