The Inhibitory Effect of Mahwangbokhapbang 2 on the Obesity (마황복합방(麻黃複合方) 2호(號)가 고지방사료(高脂肪飼料) 식이(食餌)로 유발(誘發)된 비만(肥滿) 생쥐의 혈액 및 조직학적 변화에 미치는 영향(影響))

  • Objective : This experimental study was designed to investigate the effects of Mahwangbokhapbang2 (every abbreviation from now on MHBHB2) on obesity care where it is prescribed for high fat diet-induced mice. Also designed to find out effects of MHBHB2 on controlling th obesity clinically) Method : In order to investigate th obese inhibitory effects of MHBHB2, C57BL/6 mice were induced by high fat diet C57BL/6 mice were divided into four group(normal, high fat diet, high fat diet with reductil, high fat diet with MHBHB2 extract) and fed for 15weeks. Result : 1. Mahwangbokhapbang2(MHBHB2) decreased significantly the body weig-ht. 2. MHBHB2 decreased significantly the weight of adipocyte. 3. MHBHB2 decreased significantly the blood level of serum ALT, AST. 4. MHBHB2 decreased significantly the blood level of serum total cholester-l, LDL- cholesterol, friglyceride and NEFA(free fatty acid). 5. MHBHB2 decreased significantly the blood level of HDL-cholesterol. 6. MHBHB2 500mg/kg decreased significantly the blood level of Leptin. Conclusion : Based on these results, it is prove that MHBHB2 is effective on obesity care and has obese-inhibitory effects in obese-mouse induced by high fat diet. So it is espected that the clinical application of MHBHB2 can help the treatment of obesity.

Ginsenoside Contents and Hypocholesterolemic Effects of a By-Product in Ginseng Radix (인삼부산물 추출액의 ginsenosides 함량 및 고지방 식이에 있어 혈청 콜레스테롤 농도 개선에 미치는 효과)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from mixture of leaf and stem of ginseng radix (Panax Ginseng C.A. Meyer). We conducted analysis of the ginsenoside content by HPLC. Also we investigate the effects of the LSE on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. We examined by analyzing the serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride and atherogenic index and hematological datas and serum metabolic variables. Sprague-Dawley rat weigh $150\;g\;{\pm}\;15\;g$, were ramdomly assigned to 4 groups, basal diet only(BDG), high fat diet weithout LSE(FDCG), high fat diet and 10% LSE(FD10G), high fat diet and 20% LSE(FD20G). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. The activities of ALP, GOT and LDH level were significantly different. Total cholesterol, LDL-cholesterol, triglyceride contentrations in serum and atherogenic index were remarkably reduced in LSE supplemented groups as compared high fat control groups. These result imply that LSE could be used as possible for decrease of serum lipid concentration.

Anti-Diabetic Effects of DA-11004, a Synthetic IDPc Inhibitor in High Fat High Sucrose Diet-Fed C57BL/6J Mice

  • DA-11004 is a synthetic, potent NADP-dependent isocitrate dehydrogenase (IDPc) inhibitor where $IC_{50}$ for IDPc is 1.49 $\mu$M. The purpose of this study was to evaluate the effects of DA-11004 on the high fat high sucrose (HF)-induced obesity in male C57BL/6J mice. After completing a 8-week period of experimentation, the mice were sacrificed 1hr after the last DA-11004 treatment and their blood, liver, and adipose tissues (epididymal and retroperitoneal fat)were collected. There was a significant difference in the pattern of increasing body weight between the HF control and the DA-11004 group. In the DA-11004 (100 mg/kg) treated group the increase in body weight significantly declined and a content of epididymal fat and retroperitoneal fat was also significantly decreased as opposed to the HF control. DA-11004 (100 mg/kg) inhibited the IDPc activity, and thus, NADPH levels in plasma and the levels of free fatty acid (FFA) or glucose in plasma were less than the levels of the HF control group. In conclusion, DA-11004 inhibited the fatty acid synthesis in adipose tissues via IDPc inhibition, and it decreased the plasma glucose levels and FFA in HF diet-induced obesity of C57BL/6J mice.

Anti-Obesity Effects of Fermented Samjung-hwan in Hign Fat Diet Rats (고지방 식이를 섭취한 흰쥐에서 발효 삼정환의 항비만 효과)

  • Objectives: This study was performed to evaluate the effects of fermented Samjung-hwan (SJH) extracts on weight, serum lipids and blood glucose. Methods: SJH was fermented using three different probiotic bacterial strains (Lactobacillus plantarum [LP], Leuconostoc mesenteroides [LM], Bifidobacterium longum [BL]) separately. Thirty-six rats were divided into normal, control (high fat diet), SJH-UF (high fat diet+unfermented SJH 200 mg/kg), SJH-LP (high fat diet+LP fermented SJH 200 mg/kg), SJH-LM (high fat diet+LM fermented SJH 200 mg/kg) and SJH-BL (high fat diet+BL fermented SJH 200 mg/kg). For 8 weeks later, we examined body weight, total cholesterol, high-density lipoprotein (HDL)-cholesterol and blood glucose. Results: The control group showed significantly increased weight gain compared with normal group and SJH-LP and BL groups had less weight gain than control group, significantly. In the lipid serum tests, control group showed significantly increased total cholesterol levels compared with normal group and only SJH-LP represented decreased total cholesterol levels compared with control group. However there was no significant change in the HDL-cholesteol levels. In the blood glucose tests, that of control group significantly incereased more than that of normal group, SJH-BL showed significantly decreased blood glucose levels compared with control group. Conclusions: SJH-LP, SJH-BL showed weight control effect, SJH-LP decreased TC and SJH-BL reduced blood glucose.

The Effects of Salvia miltiorrhiza on High Fat Diet-induced Obese Diabetic Mouse Model (단삼투여가 고지방식이로 유발된 비만형 당뇨병 동물모델에 미치는 영향)

  • Objectives : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. In this study, we investigated the effects of Salvia miltiorrhiza on high fat diet-induced obese type 2 diabetic mouse models. Methods : Diabetes was induced in ICR male mouse (23~25 g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups (n=10) of normal, control, Salvia miltiorrhiza, and metformin. After 8 weeks, body weight, OGTT, fructosamine, lipid profile, serum level of adiponectin and leptin, epididymal fat pad, liver weight and epididymal adipocyte size were measured. Results : Salvia miltiorrhiza significantly reduced oral glucose tolerance levels, fructosamine serum level, epididymal fat weight, and epididymal adipocyte size. Salvia miltiorrhiza also increased HDL-cholesterol, adiponectin and leptin serum levels. Conclusions : These results show that Salvia miltiorrhiza improves insulin resistance. Therefore we suggest that Salvia miltiorrhiza would be an effective treatment for obese type 2 diabetic patients.

A study on the analysis of artificial sweeteners in processed foods including emulsified foods by HPLC-DAD

  • This study has been carried out to develop a standard method for quantifying of 3 permitted artificial sweeteners (including sodium saccharine, aspartame, acesulfame potassium) contained in foods by HPLC-DAD. A simple and rapid sample pretreatment method was used to remove fat and protein from the test solution with Carrez clearing regent precipitant know to be effective for protein and fat removal. The artificial sweeteners in the test solution purified through sample pretreatment were detected by high performance liquid chromatograph using a Reverse phase C18 column (5 ㎛, 4.6 × 250 mm). The simultaneous quantitative test of 3 kinds of artificial sweeteners can be effectively performed on the high fat emulsified foods containing a large amount of fat. Using the established simultaneous quantitative test method, artificial sweeteners were tested in foods such as dairy products, snacks and chocolate. The results calibration curve showed good linearity with high regression coefficients and the result of recovery test showed satisfactory recoveries within 80~110 %.

The Effects of Mori folium on Insulin Resistance and Adipose Tissue Inflammation in an Experimental Mouse Model of Obesity (상엽(桑葉)이 비만 유발 생쥐의 인슐린 저항성 및 지방세포 염증에 미치는 영향)

  • Objective: This study was undertaken to investigate the effects of Mori folium on insulin resistance and adipose tissue inflammation in an experimental mouse model of obesity.Methods: Obesity was induced in C57BL/6 mice by feeding them a high-fat diet. The mice were divided into four groups (n=6): a normal diet, high-fat diet, high-fat diet with 40 mg of Mori folium, and high-fat diet with 800 mg of Mori folium groups. After 13 wk, the body weights, fasting blood glucose and fasting serum insulin levels, insulin resistance (homeostatic model assessment) levels, oral glucose tolerance test levels, epididymal fat and liver weights, and gene expression of tumor necrosis factor-α, interleukin-6, and interferon-γ were measured. In addition, adipose tissue macrophages were analyzed by fluorescence-activated cell sorting.Results: Mori folium significantly reduced blood glucose levels, oral glucose tolerance levels, and liver weights. It also reduced adipose tissue macrophage numbers and tumor necrosis factor receptor-α gene expression.Conclusions: These results show that Mori folium has insulin resistance reduction and anti-inflammatory effects in an experimental mouse model of obesity.

Influence of Periodicity of Eating on Body Fat Accumulation and Lipases in Rat Adipose Tissue (급식횟수가 체내의 지방축적과 지방조직의 lipase에 미치는 영향)

  • Activities of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) in adipose tissue, accumulation of carcass fat, and serum triglyceride have been determined in meal-fed (MF) and ad libitum-fed (AD) rats. At each feeding frequency, the animals received diets providing total fat as 15% or 30% of calories and polyunsaturated fatty acids (PUFA) as 2.5% or 11% of calories. The food intake of the MF rats was 75% of that consumed by the AD rats but MF rat utilized their food more efficiently, as evidenced by weight gain per 100 Kcal consumed. Meal feeding, as contrasted to ad libitum feeding, resulted in greater activities of both LPL and HSL. This suggested a higher turnover of fat in the adipose tissue of MF rats. In AD rats, body fat was significantly correlated with LPL and the ratio of LPL/HSL. Meal feeding significantly increased the ratio of LPL/HSL, indicating a greater capacity for energy storage and fat deposition in the MF rat. However, at the limited caloric intake, MF rats failed to realize this potential; there was no significant difference in percentage of body fat at the two feeding frequencies. Body fat deposition was greater in rats fed the 30% fat diet, as compared with the 15% diet, regardless of the rate of food ingestion. This was coupled with a higher ratio of LPL/HSL. The significant correlation of serum triglycerides with body fat and with the ratio of LPL/HSL in AD rats suggests that LPL activity and fat deposition may be controlled by the concentration of circulating triglycerides. Both serum triglycerides and adipose LPL activity were significantly reduced when the diet contained high levels of PUFA. The percentage of body fat was also lower in animals whose intake of PUFA was high.

Development of Low Fat Mayonnaise Made with Modified Starch (변성전분을 이용한 저지방 마요네즈의 개발)

  • This study was examined closely physiological activation and intended to present the possibility of developing food low-fat food. Developing carbohydrate fat replacer as materials for low-fat food made of Korean potato starch, it is expected that the new demand of fat replacer will be created. Potato starch was modified by chemical modification. The calorie of starch of GPS was measured to be 3.0 kcal/g, those of chemically modified starch, HPR showed 2.5 kcal/g respectively, suggesting that calorie is decreased by modified treatment. The appropriateness of processing food was experimented by substituting the existing oil and fat containing food with gel of starch and modified starch in constant rate through utilization of modified starch. When producing mayonnaise by substituting edible oil with gel of modified starch in 10-50%, calorie was reduced by 44${\sim}$45% when substituted by 50%, suggesting the potential of low-fat food. Measuring viscosity of mayonnaise by Brookfield viscometer, the mayonnaise with HPR showed high viscosity and the chemical modified starch group of EZ also showed high viscosity. Generally, the material property of mayonnaise tended to reduce in all measured items when oil and fat are substituted by starch substituting materials and the substituting materials increase. When it comes to the emulsification stability of mayonnaise with starch substituting materials, emulsification stability of all mayonnaise with starch substituting materials is lower than that of compared group. While the group with NL as commercial fat replacer showed emulsification stability which was slightly higher than group with modified starch and the substitution group of HPR showed higher emulsification stability. Sensory evaluation for low-fat mayonnaise by substituting oil the products substituted by modified starch was more preferred than general starch substituting products such as GPS. While NL as commercially fat replacer showed the hight preference, products with H40, EZ were also highly preferred.

Effects of Duchesnea chrysantha on Regulation of Antioxidative defense System in Rats Fed a High-fat·High-cholesterol Diet (뱀딸기풀의 항산화 활성 및 고지방·고콜레스테롤 식이 흰쥐의 항산화 방어계 조절에 미치는 영향)

  • This study was attempted to investigate the effects of Duchesnea chrysantha (DC) on antioxidative activities by in vivo. Rats were divided into four experimental groups which are composed of normal diet group (N group), high fat high cholesterol diet group (HF group), high fat high cholesterol diet with 5% DC powder supplemented group (DA group) and high fat high cholesterol diet with 10% DC powder supplemented group (DB group). Supplementation of DC powder groups resulted in increased activities of hepatic glutathione peroxidase and catalase. The microsomal superoxide radical contents of the DA and DB groups were significantly reduced compared to the high fat high cholesterol diet group. The mitochondrial superoxide radical contents of the DB group were significantly reduced compared to the high fat high cholesterol diet group. Hepatic hydrogen peroxide contents in cytosol were significantly reduced 5% and 10% DC powder supplemented group. The carbonyl values contents in mitochondria and microsome of the DA and DB groups were significantly reduced compared to the HF group. Thiobarbituric acid reaction substance (TBARS) values in liver were reduced in 10% DC powder supplemented group compared to the HF group. These results suggest that DC powder may have a strong regulatory effect in the activation of the antioxidative defense system.
