The effects of Brassica juncea L. leaf extract on obesity and lipid profiles of rats fed a high-fat/high-cholesterol diet

  • BACKGROUND/OBJECTIVES: Obesity is a global health problem of significant importance which increases mortality. In place of anti-obesity drugs, natural products are being developed as alternative therapeutic materials. In this study, we investigated the effect of Brassica juncea L. leaf extract (BLE) on fat deposition and lipid profiles in high-fat, high-cholesterol diet (HFC)-induced obese rats. MATERIALS/METHODS: Male Sprague-Dawley rats were divided into four groups (n = 8 per group) according to diet: normal diet group (ND), high-fat/high-cholesterol diet group (HFC), HFC with 3% BLE diet group (HFC-A1), and HFC with 5% BLE diet group (HFC-A2). Each group was fed for 6 weeks. Rat body and adipose tissue weights, serum biochemical parameters, and tissue lipid contents were determined. The expression levels of mRNA and proteins involved in lipid and cholesterol metabolism were determined by reverse transcription polymerase chain reaction and western blot analysis, respectively. RESULTS: The HFC-A2 group showed significantly lower body weight gain and food efficiency ratio than the HFC group. BLE supplementation caused mesenteric, epididymal, and total adipose tissue weights to decrease. The serum levels of triglyceride, total cholesterol, and low-density lipoprotein cholesterol were significantly reduced, and high-density lipoprotein cholesterol was significantly increased in rats fed BLE. These results were related to lower glucose-6-phosphate dehydrogenase, acetyl-coA carboxylase, and fatty acid synthase mRNA expression, and to higher expression of the cholesterol $7{\alpha}$-hydroxylase and low density lipoprotein-receptor, as well as increased protein levels of peroxisome proliferator-activated receptor ${\alpha}$. Histological analysis of the liver revealed decreased lipid droplets in HFC rats treated with BLE. CONCLUSIONS: Supplementation of HFC with 3% or 5% BLE inhibited body fat accumulation, improved lipid profiles, and modulated lipogenesis- and cholesterol metabolism-related gene and protein expression.

홍국고구마가 고지방식이를 급여한 흰쥐의 배변량 및 지질대사에 미치는 영향 (Effect of Diets with Red Yeast Sweet Potato Powder Supplement on Fecal Amount and Lipid Metabolism in Rats Fed a High-fat Diet)

  • 홍국고구마가 고지방식이를 섭취한 흰쥐의 배변량 및 지질대사 개선에 미치는 영향을 조사하고자 SD계 수컷 흰쥐를 대상으로 실험 1기(6주)에 고지방식이를 급여하고, 실험 2기(4주)에 3종의 고구마분말(일반, 홍국, 자색)을 5% 첨가한 식이를 급여했을 때, 배변량은 홍국고구마군(RYSP)이 고지방대조군(CON)에 비해 유의하게 증가하였고(p<0.05), 장 통과시간 역시 RYSP군이 가장 짧았다. 분변 중 총콜레스테롤(TC) 함량은 일반고구마군(WFSP)과 홍국고구마군(RYSP)이 유의하게 높았고(p<0.05), 그중에서도 RYSP군이 더 높은 함량을 나타내었으며, 중성지방(TG) 농도는 RYSP군이 WFSP군과 PFSP(자색고구마)군에 비해 각각 9%, 26% 더 높았으나 유의성은 없었다. 반면 1일 총 지질배설량은 TC와 TG 모두 RYSP군이 CON군과 PFSP군에 비해 유의적으로 높았다(p<0.05). 혈청 중 TC 농도는 RYSP군이 CON군에 비해 유의적 차이 없이 20% 감소하였고, LDL-C 농도는 RYSP, PFSP군이 CON군보다 통계적으로 유의하게 낮았으며(p<0.05), TG 농도는 RYSP군이 실험군중 가장 낮았고 이는 WFSP군과 유의적인 차이가 있었다(p<0.05). 이상의 결과에서 홍국고구마 식이는 배변량을 증가시키고, 분변 중 지질 배설량을 다소 증가시키며, 혈청 지질 농도의 감소 효과를 확인할 수 있었다.

고지방식이 병행섭취 시 연잎, 연 줄기, 연자방 분말가루가 흰쥐의 혈중 생화학적 인자에 미치는 영향 (The Effect of Lotus (Nelumbo nucifera) Leaf, Stem, and Yeonjabang Powder Extract on the Biochemical Factors in Serum in Mice Fed a High-Fat Diet)

  • In this study, we analyzed the biochemical factors in lotus (Nelumbo nucifera) leaf, stem, and yeonjabang and their effects on serum factor levels in mice fed a high-fat (HF) diet. The loutus leaf showed $9.47{\pm}0.30%$ moisture content, $8.25{\pm}0.39%$ ash, $21.45{\pm}1.25%$ crude protein, and $2.21{\pm}0.13%$ crude fat content; the lotus stem showed $11.84{\pm}0.43%$ moisture, $10.21{\pm}0.64%$ ash, $17.55{\pm}0.92%$ crude protein, and $4.16{\pm}0.23%$ crude fat content; and the lotus yeonjabang showed $11.86{\pm}0.50%$ moisture, $6.81{\pm}0.51%$ ash, $18.71{\pm}1.02%$ crude protein, and $3.95{\pm}0.15%$ crude fat. Blood triglyceride levels were higher in the HF group ($146.43{\pm}38.81mg/dL$), and lower in the HF+yeonjabang groups ($98.00{\pm}17.18mg/dL$). In particular, blood triglyceride levels were significantly lower in the groups that had 10% dry yeonjabang powder added to the high-fat diet. The inclusion of excessive high-fat diet increased concentrations of serum insulin and leptin. Serum leptin concentrations were highest in the HF group mice ($3.00{\pm}1.35ng/dL$), whereas they were significantly lower in the HF+yeonjabang groups by $1.34{\pm}0.52ng/dL$ (p<0.05). Thus, addition of dry yeonjabang powder to the high-fat diet was more effective in regulating the levels of serum triglycerides and leptin in mice. Additional studies would help in the development of yeonjabang as a functional food.

고지방식이 비만모델에서 방기황기탕(防己黃芪湯) 합(合) 영계출감탕(苓桂朮甘湯)의 항비만 효과 (Anti-obesity Effects of Banggihwnggi-tang-hap-yeonggyechulgam-tang in High Fat Diet Induced Obese Mice Model)

  • Objectives This study is to investigate anti-obesity effects of Banggihwanggi-tang-hap-yeonggyechulgam-tang (BY), an herbal formula, in high fat diet induced obese mice model. Methods Fourty five male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (Nor, n=9), high fat diet control group treated with water (Veh, n=9), high fat diet group treated with orlistat (Oris; n=9, Orlistat 40 mg/kg), high fat diet group treated with low concentraion BY (BYL; n=6, BY 0.87 g/kg) and high fat diet group treated with high concentration BY (BYH; n=6, BY 1.74 g/kg). Results Seven weeks later, antioxidative capacity, body weight, epididymal fat pad and liver weight, reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL), superoxide dismutase (SOD), catalase, glutathione peroxidase (Gpx), heme oxygenase (HO)-1 and histology of liver were evaluated. In the BYH group, 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis (3 ethybenzothiazoline-6-sulfonic acid) radical scavenging activity were more than L-ascorbic acid. Body weight gain were significantly less than Veh group. Epididymal fat pad and liver weight gain were significantly less than Veh group. ROS and $ONOO^-$ were significantly less than with Veh group. ALT and AST were significantly less than with Veh group. Total cholesterol, triglyceride and LDL were significantly less, HDL were significantly more than Veh group. SOD, catalase, Gpx, HO-1 significantly increased compared with Veh group. Injury on liver was lesser than Veh group. Conclusions It can be suggested that BY has anti-obesity effects in high fat diet induced obese mice model.

수풍순기환 분할처방 투여가 고지방, 고탄수화물 식이로 유발된 비만형 제2형 당뇨병 동물모델에 미치는 영향 (The Effects of Supungsunki-hwan Partitioned Prescriptions on Obese Type 2 Diabetes Mouse Model Induced by High Fat, High Carbohydrate Diet)

  • Objectives : Recently a lot of research is being done for find antidiabetic medicine which has no side effects. This study aimed to investigate the antidiabetic and antiobesity effects of Supungsunki-hwan partitioned prescriptions on obese type 2 diabetes mouse. Methods : Type 2 diabetes mellitus and obesity were induced by Surwit's high fat, high sucrose diet for 8 weeks. Mice were divided into 3 groups of ND (normal diet, n=10) HFD (high fat and high sucrose diet, n=10) and SPP (high fat and high sucrose diet with Supungsunki-hwan partitioned prescriptions, n=10) groups. Body weights were measured every week. After 7 weeks, fasting blood sugar and oral glucose tolerance tests were conducted. After 8 weeks, blood samples of all mice were taken from their heart and analyzed biochemically. At the same time, epididymal fat pad and liver weights were measured. Histological size of white adipocyte were measured as well. Results : Compared with a HFD group, body weight, fructosamine, epididymal fat pad weight and white adipocyte size decreased. High-density lipoprotein cholesterol levels increased in the SPP group. Conclusions : These results suggest that SPP has antidiabetic and antiobesity effects in high fat, high sucrose diet induced obese mice.

마황(麻黃), 마황합록다(麻黃合綠茶) 약침(藥鍼)이 고지혈증(高脂血症) 유발(誘發) 백서(白鼠)에 미치는 영향(影響) (Effects of herbal acupuncture (Ephedra sinica and Green tea) at Umnungchon(SP9) and Pungnyung(ST40) in obese Rats induced by high fat diet)

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Ephedra sinica and Green tea) at Umnungchon(SP9) and Pungnyung(ST40) on weight gain, food intakes, food efficiency ratios, HDL-cholesterol, triglyceride, free fatty acid on rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into normal group(Sham), high fat diet group(Con), high fat diet and Ephedra sinica-herbal acupuncture in SP9 ST40 treated group(ES), high fat diet and Green tea-herbal acupuncture in SP9 ST40 treated group (GT), high fat diet and Ephedra sinica+Green tea-herbal acupuncture in SP9 ST40 treated group(ES+GT). Herbal acupuncture was bilaterally treated at the level of 19.125mg/ml per 2 days during 5 weeks. Results : Body weight, feed intake and feed efficiency ratio were decreased in Sham, ES, ES+GT, compared with control group. The level of serum HDL-cholesterol was increased in sham, tender to increase in ES, ES+GT. The change on that of serum triglyceride was decreased in sham, ES, ES+GT and free fatty acid was decrease in sham, ES+GT. Conclusions : Ephedra sinica with Green tea-herbal acupuncture in SP9 ST40 can control the Body weight, feed intake and feed efficiency ratio, HDL-cholesterol, triglyceride, free fatty acid.

고지방 식이와 일반사료를 섭취한 랫드에서 미생탕의 항비만 효과 (Anti-obesity Effects of Misaengtang in Rats Fed on a High-Fat Diet or Normal Diet)

  • Misaengtang (MST), a formula of Korean herbal medicines, has been used as a weight-controlling recipe. We have investigated two experiment of body weight regulation by MST In rats. i) The anti-obesity effect of MTS on a high fat diet-induced obesity, male Sprague-Dawley rats were fed with a high-fat diet containing 30% in the absence or presence of MST (0.3, 1 or 3%) or a reference orlistat (0.05%) for 6 weeks. ii) weight-decrease effect of MST on normal diet, same animal were fed with a normal diet in the absence or presence of MST (3%) for 6 weeks. And the body weights, daily feed and water consumptions, organ weights, fat weights serum biochemistry were measured. In both experiments, MST and orlistat did not affect the body weight gain. But orlistat significantly increased the feed and water consumptions, leading to low-feed efficiency, and orlistat markedly reduced abdominal, paratesticular and perirenal fat weights, although increased the kidney weights. In MST, low dose (0.3%) of MST decreased the perirenal fat and increased the kidney weights in rats fed HFD, and MST 3% decreased the abdominal fat weights in rats fed normal diet. In addition, Orlistat caused changes in parameters of hepatotoxicity (AST and glucose), nephrotoxicity (BUN and B/C ratio) and lipid metabolism (HDL and triglycerides). In comparison, MST decreased AST, ALP and ALT, the hepatotoxicity markers, and somewhat improved the hepatic fatty degeneration. Taken together, it is suggested that MST does not exert anti-obesity activity as well as remarkably direct effects, but MST may be potentially benefit for dietary cure and exercise-cure of obesity.

양파껍질 열수 추출물이 혈중지질 성분과 혈당에 미치는 영향 (Effects of Onion Peel Water Extract on the Blood Lipid Profiles and Glucose)

  • Objectives : Onion (AlliumcepaL.) is one of the richest sources of flavonoids in human diet. In this study, we studied the effects of onion peel water extract (OPE) on the blood lipid profiles and glucose in rats. Methods : The experimental groups were divided with 5 groups (n = 6) of SD rats: normal diet + distilled water (NC), high-fat diet + Quercetin (PC), high-fat diet + onion peel water extract 4 mg/kg (OPE-4), high-fat diet + onion peel water extract 20 mg/kg (OPE-20), high-fat diet + onion peel water extract 100 mg/kg (OPE-100). Results : The liver fat showed significantly lower weights and size in the OPE-100 group as compared with NC group (p<0.05), The epididymal fat and retroperitoneal fat showed significantly lower weights and sizes in the OPE-4 and OPE-20 group as compared with NC group (p<0.05). The serum levels of total cholesterol, LDL cholesterol and triglyceride were significantly lower in the OPE-4 and OPE-20 group as compared with NC group (p<0.05). The OPE-4 and OPE-20 group showed higher HDL cholesterol concentration than NC group (p<0.05). Atherogenic index of OPE-4 and OPE-20 group was significantly lower in as compared with NC group (p<0.05). The serum levels of glucose significantly lower in the OPE-20 group as compared with NC group (p<0.05). Conclusions : In these results, we suggests that onion peel water extracts supplementation can reduces the serum lipid components and improves the lipid metabolism in hyperlipidemic SD rat induced with a high-fat diet.

Effects of corn gluten hydrolyzates, branched chain amino acids, and leucine on body weight reduction in obese rats induced by a high fat diet

  • In this study, we compared corn gluten hydrolyzates, BCAAs, and leucine for their effects on body weight reduction in high fat-induced obese rats in order to determine the major active components in the corn gluten hydrolyzates. After obesity was induced for 13 weeks with high fat diet, the overweight-induced SD rats (n = 64) were stratified according to body weight, randomly blocked into eight treatments, and raised for 8 weeks. Four groups were changed to a normal diet and the other groups remained on the high fat diet. Each of the groups within both diets was fed either casein, corn gluten hydrolyzates, leucine, or branched chain amino acids, respectively. Daily food intake, body weight gain, and food efficiency ratio were significantly lower in the corn gluten hydrolyzate groups compared to the other groups, regardless of the high fat diet or normal fat diet. The rats fed the corn gluten hydrolyzates diet had the lowest perirenal fat pad weights whereas muscle weight was significantly increased in the corn gluten hydrolyzates groups. Plasma triglyceride, hepatic total lipid, and total cholesterol contents were significantly reduced in the corn gluten hydrolyzates groups. Other lipid profile measurements were not significantly changed. Plasma triglyceride and hepatic total lipid were also significantly reduced in the BCAA and leucine groups. Leptin levels were significantly lower and adiponectin was significantly higher in the corn gluten hydrolyzates groups. Fasting blood glucose, insulin, C-peptide, and HOMA-IR levels were also significantly reduced in the corn gluten hydrozylates groups, regardless of fat level.

Influence of high fat and different types of carbohydrate diet on energy metabolism in growing mice

  • [Purpose] The purpose of this study was to determine whether different types of carbohydrate diets with or without exercise changes energy metabolism at rest and during exercise. [Methods] To minimize differences in food and energy intake between experimental groups, mice were pairfed. After 1 week of adaptation, 40 male ICR mice (6 weeks old) were randomly divided into four groups: Sta. (high fat + high starch), Scu. (high fat + high sucrose), StaEX. (high fat + high starch + exercise), and SucEX. (high fat + high sucrose + exercise). StaEX. and ScuEX. groups underwent training by running on a treadmill five times a week. After 10 weeks of training, energy metabolism was measured for 24 h and during a 1 h exercise period. [Results] The final body weight showed no significant difference between the groups. However, the weight of abdominal tissues (epididymal, perirenal, and mesenteric adipose tissue) in training groups was markedly decreased following 10 weeks of training. Results of all energy metabolism (24 h at rest and during 1 h of exercise) showed no significant interactions between diet and exercise. A brief summary of the results of the energy metabolism is that the metabolism related indicators over 24 h were more affected by the dietary pattern than the exercise but during the 1 h of exercise, training had more effect on energy metabolism than diet. [Conclusion] Our findings confirm that: (a) the type of carbohydrates included in the diet influence the metabolic responses over 24 h, (b) training had more effect on energy metabolism than diet during 1 h of exercise, (c) both results; abdominal adipose tissue weight and fat oxidation during exercise are suggestive for a beneficial effect of moderate physical activity on weight maintenance.
