Purification and Characteristics of Pullulanase from Bacillus cereus subsp. mycoides (Bacillus cereus subsp. mycoides가 생산하는 Pullulanase의 정제와 특성)

  • The optimum cultural temperature and time for the pullulanase production by Bacillus cereus subsp. mycoides were 35$\circ $C and 48 hrs, respectively. The addition of egg albumin and casein to the basal medium increased the enzyme production. The enzyme was purified by ammonium sulfate fractionation and DEAE-cellulose column chromatography. specific activity of the purified enzyme was 82.37 U/mg protein and yield of theenzyume activity was 62.1%. The purified enzuyme showed a single band on ployacrylamide disc gel electrophoresis and its molecular weight was estimated to be 66.,000 by SDS-polyacrylamide disc gel electrophoresis. The isoelcular point for the purified enzyme was pH 5.0. The optimum temperature and pH were 50$\circ $C and pH 6.5, respectively. The purified enzyme was stable below 40$\circ $C and in the pH range of 6.5~10.0 The pullulanase activity was greatly inhited by Ag$^{+}$, Hg$^{2+}$ and EDTA, and its heat stability was increased by the addition of Ca$^{2+}$. The tydrolysis product with the enzyme on pullulan was maltotriose.

  • Generally, it is known that the composition of the cation of the dog's RBCs is high in potassium(K) and low in sodium(Na). However, it is reported that certain kinds of dogs have HK, HG phenotype which contains a large amount of reduced glutathione(GSH) by the effect of Na-K pump on the cell membrane of RBC with high concentration of K and low concentration of Na. Although this HK phenotype is not regarded as a disease, it is supposed to be an important assignment to examine the distribution and the occurrence rate of the dogs that contain HK cell in their RBCs for the proper clinical treatments as these HK dogs are very sensitive to aromatic disul-fide or onions and have a tendency to cause hemolysis. Accordingly, present study was performed to measure the concentration of K, Na and GSH in the RBCs of Jindo dogs and that of Dosa dogs at the same time.

Optimization of Conditions for In Vitro Germination of Seeds of Couroupita guianensis Aubl. - A Threatened Tree with Recalcitrant Seeds

  • An efficient and reproducible protocol for in vitro germination of seeds of Couroupita guianensis has been developed in the present study. Couroupita guianensis is a medicinally important tree and categorized as threatened plant species by the IUCN. The seeds were surface sterilized using 0.1% $HgCl_2$ and inoculated on MS (Murashige and Skoog) and Woody Plants (Llyod and McCown) medium. The seeds with partially removed seed coat responded better (100%) on half strength MS medium as compared to the full (51%), one-fourth strength MS (79%) and the WP medium. Half strength MS medium conjunct with $2.0mg{\cdot}L^{-1}$ indole-3 butyric acid (IBA) was found best among the different types of auxins and cytokinins tested for seed germination, as all the seeds germinated on this medium combination. The seedlings were successfully hardened in the greenhouse and restored in the field with 83% survival rate. The protocol can be used as conservation measure for large scale multiplication of this threatened forest tree species.

Preparation of the Internal Mammary Artery Graft in Coronary Artery Bypass Surgery - Comparison of Free Mammary Artery Flows - (관상동맥 우회로술에서 내유동맥 이식편의 처치방법에 따른 문합전 내유동맥 혈류량의 비교)

  • To compare two methods of mammary pedicle graft preparations with free internal mammary artery flow, we studied 31 patients who had the left internal mammary artery harvested for coronary artery bypass grafting. The free flow was measured at the transected opening of 2 to 3 cm distal to the point of bifurcation on mean arterial pressure of 50 to 55 mmHg during cardiopulmonary bypass. Group I comprised 14 patients, whose grafts were sprayed and wrapped in sponges soaked in diluted papaverine solution (60 mg in 40 ml Hartmann's solution). An average 80 minutes after the preparations, free flow of the internal mammary artery ranged from 20 to 80 ml/min (mean 37.7 ml/min). Group II comprised 17 patients, who had internal mammary artery takedown under the exact conditions used in group I. The grafts were sprayed and wrapped in sponges soaked in the diluted papaverine solution as in group I. After an average of 28 minutes, free flow ranged from 8 to 28 ml/min (mean 17.6 ml/min). Intraluminal papaverine of the same dilution was then injected without any hydrostatic dilatation and flows increased upto 37 to 150 ml/min (mean 74.7 ml/min). This study shows that intraluminal papaverine preparation method markedly increases free mammary artery flow which is inadequate with external papaverine preparation.

Stroke Risk and Knowledge in Women with Prehypertension (고혈압 전 단계 성인여성의 뇌졸중 위험과 뇌졸중 지식)

  • Purpose: This study was undertaken to identify stroke risk with risk factors and knowledge of stork in women with prehypertension. Methods: A total of 46 pre-hypertensive women in urban area aged 44.8 (SD 11.4) yr old in average were selected by a convenient sampling. Demographic data, risk factors, and knowledge of stroke were assessed through face to face interview. Stroke risk scores were calculated based on points given to age, systolic blood pressure, smoking, DM, and heart disease in Framingham stroke risk score. Data were analyzed by SPSS WIN 12.0 using descriptive statistics, two sample t-test, and Pearson's correlation coefficient. Results: The mean systolic blood pressure of participants was 133.11 mmHg, and HDL cholesterol was lower than the recommended level, although rest of physiologic risk factors were within normal. Of the participants, 82% did not do regular physical activity and 75% did not control fat intake persistently. Mean stroke risk points and knowledge scores were 4.48 and 29.15, respectively, which were significantly different between women with and without persistent controlling fat intake. Conclusion: Results indicate stroke prevention intervention for prehypertensive women should address comprehensively modifying lifestyle as well as physiologic factors, and information focusing on stroke risk factors and warning symptoms.

Preparation of Ceramic Paper Containing Zeolites and Its Characterization (제올라이트를 함유하는 세라믹종이의 제조 및 이의 특성평가)

  • The ceramic paper, which could adsorb VOC's effectively, was made by paper-making method with zeolite and ceramic fiber as main constituents. By this experiment, the optimal composition of the slurry for the ceramic paper formation was established. SEM observation showed that zeolite powder was uniformly dispersed and adhered to the paper by applying inorganic binder. The two type zeolites content in the ceramic paper was 26 wt$\%$ and its BET surface area was 131 $m^{2}$/g. The thickness and the basis weight of the ceramic paper were 0.2 mm and 130 g/$m^{2}$ respectively, and it had sufficient tensile characteristics to withstand tensile stress without tearing during corrugation. The total inorganic content of the paper was 78 wt$ \% $ and organic content was 22 wt$\%$. The equilibrium loading amount of toluene at the toluene at the toluene partial pressure of 0.2 mmHg was 3.2 wt$\%$.

Initial Alignment Algorithm for the SDINS Using an Attitude Determination GPS Receiver (자세 측정용 GPS 수신기를 이용한 SDINS의 초기정렬 알고리즘)

  • Since the stationary alignment process of the SDINS is not completely observable, some furls of the aided alignment have been applied. The purpose of this paper is to propose a new initial alignment algorithm, which utilizes the attitude output from the AGPS(Attitude Determination GPS) receiver and to demonstrate the feasibility of the proposed algorithm with several experimental results. A Kalman filter is designed for utilizing the attitude output as well as the zero velocity information. Also analyzed is the observability of the SDINS error model. To show the feasibility of the proposed scheme, we implement an alignment system where HG1700AE IMU (Inertial Measurement Unit) from Honeywell and an AGPS receiver designed at Chungnam National University are used. Test trials are done to evaluate the performance of the proposed alignment scheme. The proposed algorithm provides as good initial alignment performance as a high accurate navigation system, MAPS(Modular Azimuth Positioning System) INS.

Real-time Blood Pressure Monitoring in Porcine Tibial Artery Using LC Resonant Pressure Sensor (LC 공진형 압력 센서를 이용한 돼지 경골 동맥의 실시간 혈압 측정)

  • We have developed an implantable wireless sensor for real time pressure monitoring of blood circulation system. MEMS (micro-electro-mechanical system) technology was adopted as a sensor development method. The sensor is composed of photolithographically patterned inductors and a distributed capacitor in gap between the inductors. A resulting LC resonant system produces its resonant frequency in range of 269 to 284 MHz at 740 mmHg. To read the resonant frequency changed by blood pressure variation, we developed a custom readout system based on a network analyzer functionality. The bench-top testing of the pressure sensors showed good mechanical and electrical functionality. A sensor was implanted into tibial artery of farm pig, and interrogated wirelessly with accurate readings of blood pressure. After 45 days, the sensor's electrical response and histopathology were studied with good frequency reading and biocompatibility.

Partial Characterization of α-Galactosidic Activity from the Antarctic Bacterial Isolate, Paenibacillus sp. LX-20 as a Potential Feed Enzyme Source

  • An Antarctic bacterial isolate displaying extracellular ${\alpha}$-galactosidic activity was named Paenibacillus sp. LX-20 based on 16S rRNA gene sequence analysis. Optimal activity for the LX-20 ${\alpha}$-galactosidase occurred at pH 6.0-6.5 and $45^{\circ}C$. The enzyme immobilized on the smart polymer Eudragit L-100 retained 70% of its original activity after incubation for 30 min at $50^{\circ}C$, while the free enzyme retained 58% of activity. The enzyme had relatively high specificity for ${\alpha}$-D-galactosides such as p-nitrophenyl-${\alpha}$-galactopyranoside, melibiose, raffinose and stachyose, and was resistant to some proteases such as trypsin, pancreatin and pronase. Enzyme activity was almost completely inhibited by $Ag^+$, $Hg^{2+}$, $Cu^{2+}$, and sodium dodecyl sulfate, but activity was not affected by ${\beta}$-mercaptoethanol or EDTA. LX-20 ${\alpha}$-galactosidase may be potentially useful as an additive for soybean processing in the feed industry.

Purification and Characterization of Fibrinolytic Enzyme from Armillariella mellea (뽕나무버섯으로부터 Fibrinolytic enzyme의 정제 및 특성 연구)

  • A fibrinolytic enzyme has been isolated from the edible honey mushroom, Armillariella mellea and purified. The apparent molecular mass of purified enzyme was estimated to be 19800Da by SDS polyacryl amide gel electrophoresis and 19900Da by gel filtration, indicating that it was a monomer. The enzyme was optimal at pH 7, suggesting that the purified enzyme was a neutral proteinase. It shows the maximum fibrinolytic activity at $55^{\circ}C$, is completely inactivated above $65^{\circ}C$, and still indicates 40% of activity at $37^{\circ}C$. The fibrinolytic activity has been decreased by the addition of EDTA. Fifteen amino acid sequence was determined by protein sequencing techniques.
