Sixteen recommended soybean cultivars in Korea were tested for resistance to Heterodera glycines. The nubmer of females which developed on roots of cultivars were fewer on 'Hwanggeum kong' and greater on 'Geumgangdaelib'. But there was no resistant cultivar to Heterodera glycines. Four field populations of Heterodera glycines tested for ability to reproduce on five host differentials. Geographical differentiations of H. glycines were newly founded in Korea. These were identlified to race 1 at Hwasun, race 5 at Yangsan and Suweon and Unknown race(or race C) at Seonsan.
Influence of susceptible and resistant soybean varieties to the development of the soybean cyst nematode(Heterodera glycines) was examined. It does not seem likly that there are any differences between varieties in attracting the Second Stage larvae of the nematode. In susceptible varieties, there were large populations and three generations during a ten·week examination period, while the resistant varieties maintained very low populations and had only two generations.

A new soybean cyst nematode, H. sojae was reported from Korea in 2016. This species is closely related to H. glycines which has the same host plant. Morphological observations of the cyst, female, male and second-stage juveniles indicated that this species is differed from H. glycines. Especially, cysts of H. sojae appeared more round, shining and darker than that of H. glycines. The distribution of this new cyst nematode in soybean field in Korea is unknown so far. In 2016, 270 soil samples were collected from soybean fields and examined the existence of H. glycines and H. sojae. Total of 111 samples contained cysts (41.1%). Among them 77% were H. glycines and 23% were H. sojae. H. sojae is future threatening in soybean production area.
The soybean cyst nematode, Heterodera glycines, is a major plant-parasitic nematode that has caused important economic losses to Korea's soybean production. Four species of cyst nematodes, H. schachtii, H. glycines, H. trifolii, and H. sojae, all belong to schachtii group are coexist in field soil in Korea. The rapid identification of the nematode is crucial for preventing crop damage and in decision making for controlling this nematode. This study aimed to develop a species-specific primer set for quantitative PCR (qPCR) assay of H. glycines. The specific primer set (HGF1 and HGR1) for H. glycines was designed based on the cytochrome c oxidase subunit I (COI) sequence of mitochondrial DNA. After optimization, it is possible to identify the H. glycines using a qPCR assay with DNA extracted from a single cyst and single second-stage juvenile (J2). The specificity was confirmed by the absence of SYBR fluorescent signals of three other Heterodera species. A serial dilution of DNA extracted from a single cyst was obtained for the sensitivity test. The result showed that the standard curve of the test had a highly significant linearity between DNA concentration and Ct value (R2 = 0.996, slope = -3.49) and that the detection limit concentration of DNA of the primer set was 10 pg of DNA per reaction. Our findings suggested that H. glycines could be distinguished from H. sojae and other Heterodera species when a qPCR assay is used with a specific primer set.

The purpose of this study was to develop rapid methods for distinguishing between Heterodera schachtii and H. glycines detected from chinese cabbage fields of highland in Gangwon, Korea. To do this, we performed PCR-RFLP and PCR with the primers set developed in this study for GC147, GC408 and PM001 population, H. schachtii, and YS224, DA142 and BC115 population, H. glycines. Eight restriction enzymes generated RFLP profiles of mtDNA COI region for populations of H. schachtii and H. glycines, repectively. As a result, treatment of two restriction enzymes, RsaI and HinfI, were allowed to distinguish H. schachtii from H. glycines based on the differences of DNA band patterns. The primer set, #JBS1, #JBG1 and #JB3R, amplified specific fragments with 277 and 339 bp of H. schachtii, 339 bp of H. glycines, respectively, while it did not amplify fragments from three root-knot nematodes and two root-lesion nematodes. Thus, the primer set developed in this study could be a good method, which is used to distinguish between H. schachtii and H. glycines.
This study was conducted to investigate the yield loss of soybean by initial density of Heterodera glycines in soil. A soybean cultivar named 'Cheongja5ho' was cultivated as test material in Dangjin city, Chungcheongnam-do, Korea. Each seed was planted with distance of 25 cm in 35 plots by initial density of egg of the cyst nematode on June 21st, and allowed to harvest on November 12st in 2019. As a results, the yield of soybean decreased sharply when as the initial density of eggs of cyst nematode increased. The initial density of eggs in soil affected largely on the decreased yields of the soybean by more than 20% in above 2,000 eggs, and 30% in above 6,000 eggs of the cyst. Beyond the 6,000 eggs, the yield loss was more slowly decreased compared with the above cases. Eggs of H. glycines are widely distributed in Korea, resulting in the serious soybean yield loss by H. glycines.
A survey on the geographical distribution of two soybean cyst nematodes, Heterodera glycines and Heterodera sojae, was conducted in soybean cultivation areas in the Gangwon-do and Chungcheongbuk-do provinces. In 67 fields of 10 counties or cities in Gangwon-do province, H. glycines was detected in 13 fields, H. sojae was detected in five fields, and both species were found in two fields. In 84 fields of nine counties or cities in Chungcheongbuk-do province, H. glycines was detected in 27 fields, H. sojae was detected in four fields, and both species were found in five fields. The diversity and dominance of the plant parasitic nematodes in the Gangwon and Chungbuk provinces were not significantly different among the soybean cultivars. The diversity and dominance index of the plant parasitic nematodes according to the presence or absence of soybean cyst nematodes did not show any significant difference between the soybean cultivation areas of both provinces. The relationship between the soybean cyst nematodes and soil chemical properties showed a low coefficient of determination, and the density of soybean cyst nematodes tended to increase as the pH and available phosphate content in soil samples increased. The soil texture of cyst nematode-detected fields was mostly sandy soils, which amounted to about 60% of the total. As the clay and silt content increased, the density of soybean cyst nematodes tended to decrease, and as the sand content increased, the density of cyst nematodes tended to increase.

Soybean cyst nematode (Heterodera glycines Ichinohe) is one of the serious soybean [Glycine max (L.) Merr.] pests in major soybean producing countries. The objective of this study was to investigate of Heterodera glycines type using the five SCN infested soybean field soils and was to evaluate resistance to the soybean cyst nematode HG 2.5.7 type on soybean varieties and germplasms. The five SCN contaminated soil samples were collected from the three provinces on November 2011 in Korea, and eggs were cultured on early spring season in 2012. For the second study, a total fifty nine soybean varieties and germplasms were tested by infestation of HG type 2.5.7 in the greenhouse. Soybean cyst nematode HG types were investigated from five locations, HG 2 (race 1) type at Donghae, HG 2.5 (race 1) type at Jeongseon and Hapcheon, HG type 2.5.7 (race 1 or 5) at Yeongwol, and HG 1.2.7 (race 5) type at Haenam locations in present study. No Korean soybean varieties and germplasms were observed with SCN resistant trait to the HG type 2.5.7. Average SCN female index were calculated with 82.7% in 59 plant materials. Our results could be provided useful information to develop a SCN resistant cultivar in Korea.

A total of 75 soybean cultivars developed in Korea was screened against soybean cyst nematode (SCN), Heterodera glycines HG type 2.5. Cysts developed on soybean cultivars ranged from 104 to 624 cysts per pot. There was no resistant cultivar but 'Jangyeopkong', 'Saealkong', 'Miryangkong', and 'Mansukong' were moderately resistant; 33 cultivars were moderately susceptible and the other cultivars were susceptible. 'Jangyeopkong', 'Saealkong', 'Miryangkong', and 'Mansukong' could be recommended for soybean fields infested with SCN until developing SCN resistant soybean.
Soybean has been cultivated in China for 5,000 years. The soybean cyst neamtode (SCN), Heterodra glycines, was recongnized in Northeastern China in 1899. Currently, it is known to occur in 12 provinces. The biology of SCN was investigated in several provinces. Six races of SCN were identified (race 1, 2, 3, 4, 5 and 7). About f10,000 soybean germplasm lines were evaluated for their resistance to race 1, 3, 4 and 5 of SCN. At least two black-seeded cultivars are resistant to all four races. Several tolerant soybean cultivars with yellow seed coat were released and are in production. Additional resistant cultivars are being developed. Nematicides were not applied in production Potential biocontrol agents and related aspects are being investigated.
