Time - and Concentration - Dependent Effects of Resveratrol on miR 15a and miR16-1 Expression and Apoptosis in the CCRF-CEM Acute Lymphoblastic Leukemia Cell Line

  • Background: Chemotherapy is one of the common approaches in treatment of cancers, especially leukemia. However, drug resistance phenomena reduce the likelihood of treatment success. Resveratrol is a herbal compound which through complicated processes makes some selected cells sensitive to treatment and induction of apoptosis. In the present study, the effects of resveratrol on the expression of miR 15a and miR16-1 and apoptosis in the CCRF-CEM cell line were investigated. Materials and Methods: The CCRF-CEM cell line was cultured under standard conditions and changes in miR 15a and miR 16-1 expression were analyzed by real time-PCR technique, with attention to reveratrol dose and time dependence. Also, apoptosis is evaluated by flow cytometry using annexin V and PI. Results: CCRF-CEM cells underwent dose-dependent apoptotic cell death in response to resveratrol. MiR 15a and miR 16-1 expression was up-regulated after 24 and 48 hours resveratrol treatment (p<0.05). Conclusions: The results of our study indicate that resveratrol induces apoptosis in a time and dose-dependent manner in CCRF-CEM cells. Also, increased expression level of miR 16-1 and miR 15a by means of resveratrol in CCRF-CEM cells might have a role in apoptosis induction and predisposition. According to our results resveratrol can be regarded as a dietary supplement to improve efficacy of anti-leukemia therapies.

Comparison of Packaging Methods to Prolong the Freshness and Quality of Korean Head Cabbage (Brassica rapa). (알배추 포장 방법에 따른 품질 및 선도 비교)

  • The effect of packaging and storage methods in enhancing the shelf life and improving the postharvest quality of the Korean head cabbage (Brassica rapa) used for wrapping vegetables was studied at 10℃. The wrapping cabbage was packed using four types of packaging and storage materials: (A) Perforated PP film; (B) Non-perforated PP film; (C) PVC film for wrapping; and (D) non-packaged as control. The quality parameters, such as fresh appearance, weight loss, hue angle, moisture content, hardness, and SSC of wrapping cabbage were investigated. The weight loss of wrapping cabbage showed a significant difference between the one packaged with film and the non-packaged as control. The general appearance of Korean cabbage stored at 10℃ was not significantly affected by the packaging treatments. However, Korean head cabbage packaged with perforated film tend to show a better external appearance compared with those exposed to the other packaging during three weeks of storage at 10℃. The inside appearance, hue angle, moisture content, hardness, and SSC, gradually decreased during the storage period. No remarkable change in the measured items were observed in Korean cabbage packaging methods. In this experiment, the Korean cabbage packaged inside a PP film with holes, and stored at 10℃ temperature had the most desirable outcome of extending the head cabbage's shelf life and appearance quality. Results suggest that perforated packaging treatment combined with low storage temperature could be an effective method in prolonging the shelf life of Korean cabbage for wrapping vegetable.

The Effects of Armeniacae Arnarum Semen and Platycodi Radix on IL-4, IL-5, IL-6 in Asthma Model (행인(杏仁)과 길경(桔梗)이 Asthma model 내(內)의 Cytokine IL-4, IL-5, IL-6에 미치는 영향(影響))

  • Background : Nowadays asthma is considered to be the inflammatory disease characterized by airway hyperresponsiveness and pulmonary eosinophilia, and mediated by Th lymphocytes expressing the Th2 cytokine pattern. In many recent studies, molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of asthma. Objective : We aimed to identify the effect of Armeniacae Arnarum Semen and Platycodi Radix on the transcriptional activities of cytokine IL-4, IL-5 and IL-6 involved in asthma model. Materials and Methods : RBL-2H3 cell lines were used. Cells were stimulated with calcium inophore for maximal gene expression. After 24 hours of Armeniacae Arnarum Semen and Platycodi Radix-treatment, total cellular RNAs were collected using Trizol solution method. Then transcriptional activities of IL-4, IL-S and IL-6 were measured by RT-PCR with electrophoresis. Results : In IL-4 study, Armeniacae Arnarum Semen treated group showed 48.4% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 45.4% of transcriptional activities compared to the control group. In IL-5 study, Armeniacae Arnarum Semen treated group showed 52.7%of transcriptional activities compared to the control group and Platycodi Radix treated group showed 60.2% of transcriptional activities compared to the control group. In IL-6 study, Armeniacae Amarum Semen treated group showed 42.3% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 69.1% of transcriptional activities compared to the control group. Conclusion : This study shows that Armeniacae Arnarum Semen and Platycodi Radix have the inhibitory effect on the transcription of IL-4, IL-5 and IL-6 gene expression in RBL-2H3 cell lines. Advanced studies are required to investigate the mechanisms of inhibition by herbal medicine in asthma model.

  • $CO_2$ enrichment systems have been recently used to shorten the growth period of plants and the improvement of harvest and its quality. To accomplish these goals, manifold should be designed to supply the same amount of $CO_2$. In this study, CFD approach has been used to understand the effects of geometric parameters, such as tube and hole diameters. An optimized geometry has been derived through pipe and tube part, respectively. As a result, the deviation of flow rate less than 0.1 g/s was expected at all holes of the $CO_2$ enrichment system.

Kimchi attenuates fatty streak formation in the aorta of low-density lipoprotein receptor knockout mice via inhibition of endoplasmic reticulum stress and apoptosis

  • BACKGROUND/OBJECTIVES: Endoplasmic reticulum (ER) stress is positively associated with atherosclerosis via elevating macrophage cell death and plaque formation, in which oxidative stress plays a pivotal role. Antioxidative, lipid-lowering, and anti-atherogenic effects of kimchi, a Korean fermented vegetable, have been established, wherein capsaicin, ascorbic acid, quercetin, 3-(4'-hydroxyl-3',5'-dimethoxyphenyl)propionic acid, and lactic acids were identified. In this study, mechanisms of action of kimchi methanol extracts (KME) on fatty streak formation via suppression of ER stress and apoptosis in aorta were examined in low-density lipoprotein receptor knockout mice. MATERIALS AND METHODS: Mice fed a high cholesterol diet with an oral administration of KME (KME group, $200 mg{\cdot}kg-bw^{-1}{\cdot}day^{-1}$) or distilled water (control group) for 8 weeks (n = 20 for group). Plasma lipid and oxidative stress levels were evaluated. Protein expression was measured by western blot assay. Fatty streak lesion size and the degree of apoptosis were examined in the aorta. RESULTS: Compared to the control group, in the KME group, plasma lipids levels were decreased and oxidative stress was alleviated (P < 0.05). Protein expression levels of nuclear factor (erythroid-derived 2)-like 2-mediated antioxidants in aorta were increased whereas those for ER stress markers, glucose regulated protein 78, phospho-protein kinase RNA-like ER kinase, phospho-eukaryotic initiation factor 2 subunit ${\alpha}$, X-box binding protein 1, and C/EBP homologous protein were decreased in the KME group (P < 0.05). Moreover, apoptosis was suppressed via downregulation of phospho-c-Jun N-terminal kinase, bcl-2-associated X protein, caspases-9, and -3 with a concomitant upregulation of anti-apoptotic protein, B-cell lymphoma 2 (P < 0.05). Fatty streak lesion size was reduced and the degree of apoptosis was less severe in the KME group (P < 0.05). CONCLUSIONS: In conclusion, antioxidant activity of KME might prevent fatty streak formation through, in part, inhibition of ER stress and apoptosis in aortic sinus where macrophages are harbored.

Phytobiotics and Organic Acids As Potential Alternatives to the Use of Antibiotics in Nursery Pig Diets

  • Two experiments were conducted to determine the effect of phytobiotics and organic acids on growth performance of nursery pigs as an alternative to antibiotics. Phytobiotics refer bioactive compounds from plant materials including essential oils and herbal extracts. In Exp. 1,144 pigs, weaned at 23.4${\pm}$0.3 d age, were allotted to three dietary treatments. Treatment diets were: 1) NC (no antibiotics and no phytobiotics); 2) PC (NC+carbadox, 50 mg/kg); and 3) PB (NC+phytobiotics; 0.1% PEP1000-$1^{(R)}$. Each treatment had six replicates with eight pigs per pen. Pigs were fed the experimental diets for 5 wks in 3 phases (phase 1 for 2 wk; phase 2 for 2 wk; phase 3 for 1 wk). In Exp. 2, 192 pigs, weaned at 19.2${\pm}$0.3 d age, were allotted to three dietary treatments: 1) NC; 2) PC; and 3) PBO (NC+phytobiotics; 0.2% or 0.1% PEP1000-$1^{(R)}$ and organic acids; 0.4% or 0.2% $Biotronic^{(R)}$for the phase 1 and 2, respectively) with eight replicates per treatment and eight pigs per pen. Pigs were fed the assigned diets for 5 wks in 2 phases (phase 1 for 2 wk; phase 2 for 3 wk). Body weights were measured at the beginning of the experiment and at the end of each week in both Exp. 1 and 2. Feed intake was measured at the end of each week in both Exp. 1 and 2. Diarrhea score was measured daily during the entire period for Exp. 1 and during the phase 1 for Exp. 2. In Exp. 1, the PC had a higher (p<0.05) overall ADG than the NC, but the overall ADG of the PB did not differ (p>0.05) from the NC or the PC. In Exp. 2, the overall ADG did not differ (p>0.05) among all the treatments during the entire experimental period. The overall ADFI and the overall gain:feed ratio did not differ (p>0.05) among all the treatments during the entire experimental period in both Exp. 1 and 2. The PC had a higher (p<0.05) overall diarrhea score (harder stools) than the NC and the PB in Exp. 1, and a higher (p<0.05) overall diarrhea score than the NC in Exp. 2. The overall diarrhea score of the PB and the PBO did not differ (p>0.05) from the NC or the PC in Exp. 1 and 2. Results from this study show that the growth of pigs fed the diets with phytobiotics or the combination of phytobiotics and organic acids did not differ from those both with antibiotics and without antibiotics when tested in an environmentally controlled research facility. Further experiments are required to study the growth performance in disease challenged conditions.

The Steroid Sparing Effects of Cheongsangboha-tang in Asthmatic Patients (청상보하탕(淸上補下湯)의 기관지천식환자에 대한 스테로이드 절약효과)

  • Background : In recent years, the glucocorticoid hormone has become a fundamental medication for asthma. However, a long period of hormone administration will result. in general. side effects on many body parts as well as hormone dependence, which has become a serious problem for western physicians. Objectives : We aimed to identify the clinical effects of Cheongsanghoha-tang and the steroid sparing effects of Cheongsanghoha-tang on. Materials and Methods : A subject group consists of 36 asthmatics who had been treated with Cheongsanghoha-tang for four weeks. Cheongsanghoha-tang is a herbal decoction, which has been used of the traditional therapeutic agent of asthma. PFT, QLQAKA, blood eosinophil, serum IgE, Serum IL-4. IL-5, IFN-${\gamma}$ were checked before and 4weeks after the treatment. Results : The only FVC% in ICSG among asthmatic patients was increased significantly compared to NICSG. Treatment of Cheongsanghoha-tang for four weeks resulted in significant increase in QLQAKA. The NICSG treated with Cheongsangboha-tang for four weeks were no significant difference in the blood eosinophil, serum IgE, IL-4 and IL-5. The PFT and QLQAKA in NICSG were increased significantly after 4 weeks treatment. But the serum IFN-${\gamma}$ in NICSG was decreased significantly after 4 weeks treatment. Discontinuation of treatment with inhaled corticosteroid in ICSG resulted in insignificant changes in PFT, the blood eosinophil, the serum IgE, IL-4, IL-5 and IFN-${\gamma}$ and significant increase in QLQAKA. As a result. 8 of 13 cases were cured with hormones completely and the rest of ICSG reduced the dose of ICS. Conclusions : This study shows that Cheongsanghoha-tang has the effects on the improvement of pulmonary function and cures asthmatic patients. These findings demonstrate that Cheongsanghoha-tang has the steroid sparing effect. Some satisfactory therapeutic results have been obtained in treating hormone-dependent asthma by Cheongsanghoha-tang. However. the concept and mechanism of hormone-dependent asthma have not been fully defined yet, and the standard for judging therapeutic effects have not been established. Obviously further researches concerning all these are still necessary.

Antioxidant Effects of Gagam-jangwon-hwan(jiajianzhuangyuanwan) on Hydrogen Peroxide-Induced Oxidative Stress in PC 12 Cell Lines ($H_2O_2$로 유도된 산화적 스트레스에 대한 장원환가감방(壯元丸加減方)의 PC 12 cell 에서의 항산화 효과)

  • Objectives : Antioxidant effects of Gagam-jangwonhwan(LMK01 and 02) water extract against $H_2O_2$-induced oxidative damage and cell death were investigated in rat pheochromocytoma line PC 12. Methods : The cells were treated with LMK01 and 02 water extract and $H_2O_2$, oxidative damage-inducing materials for 24 h. The cellular viability was assessed by WST-1 assay, oxidative damages of the cells by 8-OHdG quantitation, apoptosis by Hoechst 33342 staining assay and activity of antioxidant enzymes by catalase and glutathione peroxidase assay. Results : 1. LMK01 and LMK02 water extracts improved significantly cell viability in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells 2, LMK02 suppressed significantly oxidative damage in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells but LMK01 didn't. Meanwhile, difference of oxidative damages in conditions treated with LMK01 or LMK02 was not significant, 3. The $H_2O_2$ induced-apoptosis in PC 12 cell lines was inhibited effectively by LMK01 and LMK02, and especially the features of apoptosis were obviously reduced in LMK02-treated cells. 4. LMK01 and LMK02 increased significantly activities of both catalase and glutathione peroxidase than those of $H_2O_2$-alone treated group and moreover, LMK02 showed significantly higher activities than those of LMK01. Conclusions : As shown, LMK01 and LMK02 suppressed $H_2O_2$-induced oxidative damage and cell death in PC 12 cell effectively. And they increased activity of major antioxidant enzymes in PC 12 cell line. Therefore, this study suggests the possibility of clinical usage over oxidative stress-induced neurodegenerative disease such as Alzheimer's disease.

Ginsenoside Re Inhibits Osteoclast Differentiation in Mouse Bone Marrow-Derived Macrophages and Zebrafish Scale Model

  • Ginsenosides, which are the active materials of ginseng, have biological functions that include anti-osteoporotic effects. Aqueous ginseng extract inhibits osteoclast differentiation induced by receptor activator of NF-${\kappa}B$ ligand (RANKL). Aqueous ginseng extract produces chromatography peaks characteristic of ginsenosides. Among these peaks, ginsenoside Re is a major component. However, the preventive effects of ginsenoside Re against osteoclast differentiation are not known. We studied the effect of ginsenoside Re on osteoclast differentiation, RANKL-induced tartrate-resistant acid phosphatase (TRAP) activity, and formation of multinucleated osteoclasts in vitro. Ginsenoside Re hampered osteoclast differentiation in a dose-dependent manner. In an in vivo zebrafish model, aqueous ginseng extract and ginsenoside Re had anti-osteoclastogenesis effects. These findings suggest that both aqueous ginseng extract and ginsenoside Re prevent bone resorption by inhibiting osteoclast differentiation. Ginsenoside Re could be important for promoting bone health.

The Influence of Materials for Surface Mulching on Soil Temperature and Vegetative Growth of Apple Nursery Trees (지표면 멀칭재료가 지온과 사과나무 묘목의 수체생육에 미치는 영향)

  • This study was conducted to investigate the influence of different polyethylene film (P.E. film) for mulching on the changes in soil temperature and the production of good feathered apple nursery trees. M.9 rootstocks with stem diameter of 9.1~11.0 mm were planted in plots covered with different P.E. film (i.e., transparent P.E. film, black P.E. film, and bare soil as control). Three weeks after planting, the rootstocks were veneer grafted with 'Sinano Sweet' apple cultivar. In the middle of June, BA was sprayed to nursery for inducing feathering during the growing season. The soil temperature of the control was higher than air temperature by about $0.7^{\circ}C$ from mid April to early October, and that of P.E. film mulching was about $1{\sim}5^{\circ}C$ higher than that of the control. The soil temperature under transparent P.E. film was about $2{\sim}3^{\circ}C$ higher than that under black P.E. film. The diurnal range of soil temperature under the black P. E. film was lowest among all treatments. The P.E. film mulching induced better tree growth and feathering than bare soil. Percentage of good feathering apple nursery of black P.E. film was highest among all treatments because the soil temperature unuder black P.E. film in the early growing season was higher than that of the control and the number of days when the maximum soil temperature was over $35^{\circ}C$ in the summer was lower than that under the transparent P.E. film.
