Anti-inflammatory Effect of Herbal Complex HP-04 on Degenerative Osteoarthritis (해동피를 포함한 생약복합제제 HP-04의 퇴행성 골관절염에 대한 항염효과)

  • To investigate the anti-inflammatory effect on Degenerative Osteoarthritis, Male sprague-Dawley rats were randomized and classified into three different concentration groups. We measured weekly weight change, dietary intake, drinking water intake, blood analysis like TNF-α (tumor necrosis factor-α), IL-6 (interleukin 6), TIMP-1 (tissue inhibitor of metalloprotease-1), MMP-2 (matrix metalloprotease-2), MMP-9 (matrix metalloprotease-9) and micro CT analysis. The results suggest that the treatment with herbal complex HP-04 improved the Monosodium iodoacetate (MIA) induced degenerative osteoarthritis and it could be applicable for the improvement of arthritic symptoms as a new therapeutic agent.

Effects of Herbal Complex on Atopic Dermatitis in BALB/c Mice (BALB/c 마우스에서 생약복합제의 아토피 치료 효능)

  • Atopic dermatitis (AD) is a common chronic inflammatory disease of preceding the development of allergic disorders. The incidence of AD is increasing and it causes problems with administrative costs. Therefore, no side effects, easyto- use development of AD treatment is needed. The aim of this study was to evaluate whether PBMCAT, a functional extract from herbal complex was effective to treat the AD mice. AD was induced by patching ovoalbumin on the backside in BALB/ c mouse and then mice were topically treated with PBMCAT. Elidel $cream^{(R)}$ (pimecrolimus, PL) was used as a control. Scratching counts (SC) and clinical skin severity (CSS) were measured, and total serum IgE level was also measured. After inducing AD, SC and CSS were increased. The total serum IgE level was also increased in AD-induced mice. Treatment with PBMCAT significantly decreased SC, CSS, and serum IgE concentration in mice. Especially, treatment of PBMCAT 0.1% in BALB/c mice more effected than PL. These results suggest that the ointment of PBMCAT may enhance the process of AD healing by influencing phase of allergic reacting.

How to establish the definition of Conventional medicine and Korean herbal medicine? (한약과 양약의 개념 설정 어떻게 할 것인가?)

  • Objectives : The purpose of this study is to find a reasonable solution to a current status of drug classification between the drugs of western conventional (allopathic) medicine and Korean medicine. A clear and distinct concept on the drugs of allopathic medicine and Korean medicine based on reasonable concepts and broad consensus is a pressing issue in Korea, and will facilitate the development of herbal medicinal products and pharmaceutical industry. Methods : Considering the issue of drug classification from domestic and international regulations, we reviewed the current Drug Law of Korea and China, Guidance for Industry, Botanical Drug Products of USA, Directive 2004/24/EC of the European parliament and of the council. Results : In Korea, the drug classification of allopathic medicine and Korean medicine is quite vague even though differential licensure system is enforced for the clinicians of allopathic medicine and Korean medicine field. According to the definition in the Drug Law, the scope of Korean medicine drug is so broad that even a drug made of single-compound material, as well as herbal extract of crude mixture, is regarded as a drug of Korean medicine, as long as the material may be separated from medicinal herbs, animal tissues, or mineral resources. Only new compound not found in natural resources are outside of the scope of Korean medicine drug. In USA and EU, medicinal products manufactured from herbs are approved by separate regulations for the herbs with special waivers. In China, the category of new medicine and the definition of allopathic medicine and traditional chinese medicine are clearly specified and classified. Conclusions : As medicines are validated therapeutic materials for efficacy and toxicity, we suggest that generally the concept of conventional medicines is based on a single compound that has been synthesized and individually validated and that of Korean medicines is based on a compound extracted from natural materials or a complex of compounds that has been validated as a whole in its totality.

A Study on the Distinction of Registration Regulations for Herbal Medicines (생약제제의 등록규정 차별화에 관한 연구)

  • Herbal medicines have been used since ancient times as medicines to treat and relieve the symptoms of many different human diseases. However, so far, relatively few herbal medicines have been evaluated scientifically to prove their safety, potential benefits and effectiveness. This study was conducted to provide the groundwork for improving the current registration regulations for herbal medicines in distinction from synthetic medicines. The study was performed based on the literature research and individual interviews with 5 experts who had extensive experience in registration of herbal medicines. When compared with synthetic drugs, herbal medicines exhibit some marked differences, namely the active principles are frequently unknown, standardization, stability and quality control are not easy, they are usually mixtures of complex compounds. Second, the current regulations for herbal medicines are reviewed by comparison of foreign regulation systems like the one in China. The regulation requirements of herbal medicine in China are in distinction from synthetic drugs. The authors conclude that new registration requirements for the herbal medicines should be changed as follows; the toxicity and efficacy data should be submitted as mixed herbal preparation and the documents and other research on the reproduction and generation toxicity need to be shown for the proof of reproduction and generation toxicity, if needed.

Anti-oxidative and Anti-inflammatory Effect of 7 Herbal Extracts and Methods of Herbal Formula Compositioning (7종 한약재 추출물의 항산화 및 항염증 효과와 이를 이용한 방제 구성에 대한 고찰)

  • Objectives : Herbal formulas are complicated to analyze and difficult to compose because of its mixed, complex features. So we discussed about how to compose herbal formulas effective and efficient by analyzing various effectiveness of each herbal extracts. Methods : To evaluate the effectiveness of herbal formula, anti-oxidative and anti-inflammatory effectiveness were mathematically analyzed. DPPH, superoxide anions, Nitric Oxide scavenging activity was measured to evaluate the effectiveness of 7 herbal extracts. And next, cytotoxic activity of extracts on RAW 264.7 cells were measured using MTS assay. To asses anti-inflammatory effect, nitric oxide and $PGE_2$ production were measured. Based on these anti-oxidative and anti-inflammatory experiment result, mathematical analyzation were carried out with constOptm function, and determined efficacy-maximizing ratio. Results : This mathematical analysis based formula showed significantly outstanding effectiveness than other formulae. And estimated tendency of anti-inflammatory effectiveness was matched with real effectiveness. Conclusions : So, mathematical analysis can be available to evaluate and estimate the effectiveness of herbal formulae.

Optimum Concentration of the Cd(II)-Quercetin Complexation Reaction (Quercetin의 카드뮴 착물반응에 대한 최적농도)

  • The interaction of cadmium (II) ion with quercetin was investigated in aqueous solution at different pH. The quercetin/cadmium stochiometries for cadmium (II) binding have been determined by UV-vis spectrophotometric method. The complexation of Cd(II) ion with 54.72 ${\mu}M$ quercetin (A=1.00793) was formed in 0.2 M $NH_3-0.2$ M $NH_4Cl$ (pH 8.0) buffer solution. 1:1 Cd(II)-complex had a maximum absorbance and showed the bathochromic shift of the long-wavelength band of the UV-vis spectra in the alkaline pH when interacted with quercetin in buffer solution. These results suggest that Cd(II)-quercetin complex has the optimal condition of chelation in basic buffer solution.

A Case Report of Clinical Effects of Complex Korean Medicine Treatment in Patients with Scleroderma Caused by Juvenile Idiopathic Arthritis (소아 류마티스 관절염으로 인한 경피증에 대한 한방복합치료 증례보고 1례)

  • Objectives This study is to report a case on a patient with scleroderma caused by juvenile idiopathic arthritis. The patient's symptoms were improved by the complex Korean medicine treatment. Methods The patient was treated for 20 days with acupuncture, moxa, physical therapy, herbal medicine (Ganghwalbinsosan, 羌活檳蘇散), and herbal dressing therapy(Dohongsamultang, 桃紅四物湯). Results NRS and mRSS were used evaluate patient's symptoms daily to assess the changes. After 20 days of the treatments, the patient showed improvement of the symptoms including pain, pruritus and scleroderma of the left leg. Conclusions This case study showed that symptoms in the patient with scleroderma caused by juvenile idiopathic arthritis were dramatically improved by the Korean medicine treatment which includes acupuncture, moxa, herbal medicine, and herbal dressing therapy.

A Review of the Korean Experimental Studies on the Antidepressant Effect of Herbal Medicines (한약의 항우울 효과에 대한 국내 실험연구 고찰)

  • Objectives: The present study aims to review the antidepressant effect of herbal medicines reported in Korean local journals. Methods: We searched in electronic databases (Koreantk, KISS, OASIS, NDSL) for studies, published in Korean national journals, that assessed herbal medicine effect of depression model. The search term was 'depression' in the abstract or whole text. Depression model, herbal material, experimental results, mechanisms were extracted. Results: We included 43 articles in which 38 studies were in vitro experiments, and the rest 5 were in vivo experiments. The most common experiment subject model was a rat and the most widely used method to induce depression was Despair behavior test. 21 studies used simple herbal medicines, and 22 studies used complex herbal medication. Glycyrrhizae Radix was the most commonly used herbal material to improve depression model. The most common mechanisms of herbal medicine with antidepressant effect were inhibition of Monoamine activation mechanism and depression related neurohormone secretion. Conclusions: Herbal medicines may be a promising resource for treating depression.

Analysis of Research Articles on Oriental Herbal Medicines for Hypertension (고혈압의 한약 치료에 대한 연구동향 분석)

  • Objectives : This study aims to provide the fundamental information for future researches on combined medications of the oriental-western medicines or development of new Oriental medicine for hypertension. Methods : Every article related to hypertension was initially searched at journal web site, 'OASIS' and we finally analyzed 118 papers according to study types, kinds of herbal medicine, frequencies of herbal medicine, and results of researches. Results : Most studies were experiment researches which were composed of in vivo or in vitro. The formulas divided into 41 single and 99 complex prescriptions and total 218 medicinal materials were used in complex ones. The major effects of single prescriptions were decrease of the control of ACE activity and ANP concentration, while the effects of complex ones were various such as decrease of aldosteron concentration, decrease of epinephrine concentration, decrease of the heart rate, and decrease of dopamine concentration. Conclusions : More clinical studies based on scientific evidences should be done and the outcomes of this pilot study will contribute to development of Oriental medicines as substitute or combination for the western-hypertension remedies.
