Utilization of Hypothalamic Hormones for Maturational Induction in Seawater Chum Salmon, Oncorhynchus keta (해면연어의 성숙을 유도하기 위한 시상하부호르몬의 이용)

  • Freshwater-adapted maturing chum salmon (Oncorhynchus keta) involuntarily captured by stationary nets in Yang-yang seashore areas were transferred to freshwater in an outdoor raceway tank at Yeongdong Inland Fisheries Research Institute, NFRDI, Yang-yang, Gangwon, Korea and kept over 1 day until the start of the experiments. The freshwater-adapted females were single-injected intraperitoneally with gonadotropin-releasing hormone analogue, (GnRH-a: $70\;{\mu}g/kg$ body weight, BW) alone or combined with a dopamine receptor antagonist, pimozide($700\;{\mu}g/kg$ BW). Although gonadosomatic indices [GSI, (gonad weight/BW)${\times}100$] did not show significant changes in both 2004 and 2005, GSI of GnRH-a-injected fish during the 2005 trial slightly increased on the 5th and 7th days post-injection compared to those of vehicle treated fish. Hepatosomatic indices [HSI, (liver weight/BW)${\times}100$] of fish injected with GnRH-a alone and combined with pimozide decreased significantly on the 7th day post-injection in 2004(P<0.05). In 2005 trials, HSI was significantly reduced in GnRH-a treated fish on the 7th day post-injection (P<0.05). Pimozide-injected fish showed a pattern with increase of GSI and decrease of HSI, without significant differences. Taken together, these results suggest that at least in part hypothalamic hormones and dopamine receptor antagonist may induce sexual maturation in freshwater-adapted maturing chum salmon. It remains to evaluate these preliminary results by further researches.

Reproduction of the Goby Fish Periophthalmus magnuspinnatus in Mud Flat of Suncheon Bay, Korea (한국 순천만 갯벌지역 큰볏말뚝망둥어(Periophthalmus magnuspinnatus)의 생식)

  • Reproduction of the goby fish Periophthalmus magnuspinnatus was examined using 298 specimens collected from April to October 2005 in a mud flat of Suncheon Bay, Korea. Specimens ranged in body length(BL) from 1.3 to 9.1 cm. The gonadosomatic index(GSI) of females was highest in June and decreased until August. The hepatosomatic index(HSI) and fatness index of females were high in April and then decreased to nadirs in July. Spawning season lated from April to August and the ratio of females to males did not significantly differ($x^2$-test, p>0.05). The first spawning length was 4.5 cm BL, and the size of 50% maturity was estimated at 4.98 cm BL. Fecundity(F) ranged from 1,316 to 4,768 eggs, and the relationship between F and BL was estimated as $F=0.1562BL^{1.4068}(R^2=0.59)$.

  • The maturation and spawning of Shuttles hoppfish Periophthalmus modestus were studied from April to October, 2005 in the mud flat of Suncheon Bay, Korea. The specimens ranged in body length (BL) from 1.5 to 8.4 cm. The gonadosomatic index (GSI) of females was highest in May and decreased until September. The hepatosomatic index (HSI) and fatness index of females were high in April and then decreased to a nadir in July. The data indicate that the spawning season was from April to September. There was no significant difference in the ratio of females to males $(X^2-test,\;p>0.01)$. Over 50% of the females over 5 cm in BL were sexually mature. Fecundity ranged from 1,664 to 13,428. The relationship between fecundity (F) and body length (BL) of the fish was expressed as $F=0.026BL^{2.8638}\;(R^2=0.79)$. The first spawning length was 4.5 cm BL.

  • Reproductive biology of the slimy, Leiognathus nuchalis was investigated by means of histological methods. Sex ratio was 1.81 : 1 in female to male. Developmental pattern of oocytes was group-synchronous. Testicular structure was restricted spermatogonial testis-type of tubular testis. The size of first group maturity is 7.5 cm in total length. Gonadosomatic index(GSI) of female was the highest in July(12.83) and the lowest in September(1.08). GSI of male was the highest in June(19.0) and the lowest in October(0.24). Hepatosomatic index(HSI) of female showed to be positively correlated with GSI. Thoracic spot index(TSI) showed to be the minimum value from May to July when the maturation and ripe season of gonad. Reproductive cycle of female could be classified into the growing(March~May), maturation(May~June), ripe and spent(June~August), recovery(August~November), and resting stage(November~March). Reproductive cycle of male could be classified into the multiplicative and growing(January~April), maturation(April~May), ripe and spent(June~August), recovery(August~October), and resting stage(October~December).

Physiological and Histological Changes of Overfeeding-induced Obese Rainbow Trout Oncorhynchus mykiss (사료 과다공급으로 유도된 비만 무지개송어(Oncorhynchus mykiss)의 생리·조직학적 변화)

  • Obesity could cause immune-physiological disorders in fish. Yet, little is known about the impact of obesity on stress and histological responses. This study aimed to determine histological and physiological changes in and vulnerability of overfeeding-induced obese rainbow trout Oncorhynchus mykiss exposed to stress condition. Control, intermediate and overfed groups were fed at 1.5, 2.5 and 3.09% of their body weight per day, respectively, for eight weeks. Weight gain, body mass index, hepatosomatic index and serological parameters, and histology of liver were measured in five fish from each group at week 0, 2, 4, and 8. At week 8, 20 fish from each group were exposed to heat stress by increasing water temperature at a rate of 3℃ per day from 15 to 25℃ and maintaining the final temperature for 10 days. Overall, overfed fish showed significantly higher weight gain, body mass index, and serological parameters than those of fish in the other groups. Fish in the overfed and intermediate groups displayed multifocal infiltration of inflammatory cells in hepatic parenchyma. Mortality rate and serological parameters of fish in the overfed group exposed to heat stress were significantly higher than those of fish in the other groups, indicating increased vulnerability to environmental stress.

Effect of dietary glucose, dextrin and starch on growth and body composition of juvenile starry flounder Platichthys stellatus

  • A 10-week feeding trial was conducted to investigate the effects of dietary glucose, dextrin and starch on growth and body composition of juvenile starry flounder. Triplicate groups of fish (average weight, 9.7 g) were fed iso-nitrogenous (53% CP) and iso-caloric (3.8 kcal/g diet) diets containing 20% glucose, 20% dextrin and 5-25% alpha-potato starch with 5-14% lipid levels. Survival was not affected by dietary carbohydrate. Weight gain, feed efficiency and protein efficiency ratio of fish fed the diet containing 20% glucose were the lowest among all groups. The best weight gain was observed in fish fed the diets containing 20% dextrin. Growth and feed efficiency were not affected by dietary -potato starch level. Lipid contents of whole body and liver were not affected by dietary glucose, dextrin and starch at the same level. However, the lipid contents tended to decrease with increasing dietary starch level and those of fish fed the diets containing 5% alpha-potato starch were significantly higher than those receiving 10-25% alpha-potato starch. Liver glycogen content and hepatosomatic index tended to increase with increasing dietary starch level. These results indicate that juvenile starry flounder are able to efficiently utilize dextrin and -potato starch compare to glucose in diets and that alpha-potato starch could be incorporated up to 25% in the diet for optimum growth by juvenile starry flounder.

Effect of Fermented Food Garbage in Diet on Growth and Body Composition of Juvenile Flounder (Paralichthys olivaceus)

  • Food garbage fermented with microbial starter was formulated to diet for the growth of juvenile flounder (Paralichthys olivaceus). Two replicate groups of fish, an average weight of 4.0g, were fed the four isocaloric (19.5 MJ/kg diet) diets with different fermented food garbage levels $(0,\;5,\;10\;and\;15\%)$ for 45 days. Survival, feed efficiency, hepatosomatic index and protein efficiency ratio of fish were not affected by dietary fermented food garbage level (P>0.05). Weight gain of fish fed the diets with 5, 10 and $15\%$ fermented food garbage was significantly higher than that of fish fed the control diet (P<0.05). Condition factor of fish fed the diet with $10\%$ fermented food garbage was significantly higher than that of fish fed the control diet (P<0.05). Daily feed intake of fish fed the diets with 5 and $15\%$ fermented food garbage was significantly higher than the control diet (P<0.05). Proximate composition of whole body and plasma glucose concentration were not affected by dietary fermented food garbage level (P>0.05). These findings indicate that fermented food garbage could be utilized as a feed ingredient for juvenile flounder.

Comparison of Xenobiotic Metabolism and Antioxidative Status in Cultivated and Stocked Rockfish Sebastes schlegeli (양식산과 방류산 조피볼락(Sebastes schlegeli)의 간장 중 해독 및 항산화 효소계의 비교)

  • This study compared the composition and texture of muscle and levels of detoxification and antioxidant enzymes in the livers of stocked and cultivated rockfish Sebastes schlegeli released after the primary culture stage in Tongyoung, South Korea. The crude lipid content of muscle was significantly higher ($P$<0.05) in cultivated rockfish than stocked rockfish, while the texture did not differ significantly ($P$>0.05). The condition factor (CF) and hepatosomatic index (HSI) did not differ significantly and the growth of stocked and cultivated rockfish was similar. The levels of the detoxification enzymes cytochrome P450 (CYP) and Ethoxyresorufin-O-deethylase (EROD) were significantly lower in the livers of stocked rockfish, perhaps because of their reduced exposure to xenobiotic compounds. In addition, stocked rockfish had a significantly ($P$<0.05) lower CAT and higher GR than cultivated rockfish, but similar levels of tGPx, SOD, GSH, and GSSG. The present study shows that the growth rates of stocked and cultivated rockfish are similar and that stocked rockfish are exposed to fewer xenobiotic compounds and less oxidative stress.

Haematological Parameters Induced by Benzo(a)pyrene Exposure as a Toxicity Biomarker in the Fanned Red Sea Bream, Pagrus major

  • Farmed red sea breams, Pagrus major, were fed for 60 days with pellets containing different concentrations of benzo(a)pyrene (0, 0.2, 2, 20 mg/kg) to generate a biomarker of the chemical toxicity in the fish. The fish exposed to the chemical concentrations did not show any significant difference in the weight gain, conditioning, factor, and hepatosomatic index. However, some haematological parameters, such as glucose, calcium, magnesium, GOT (glutamic oxalate transaminase), and GPT (glutamic pyruvate transaminase) were influenced by the chemical exposure. Of them, two enzymes, GOT and GPT, increased significantly 60 days after the exposure in a way of concentration dependence (P<0.05). In the study of ecotoxicological biomarker, sensitivity to adverse environments is one of the key available factors. The fish changes in GOT and GPT were an earlier and reliable sign of the fish response against the chemical exposure, rendering the two enzymatic factors as a useful biomarker at least to benzo(a)pyrene exposure in the farming waters.

Effects of Dietary Protein Concentration on Growth and Feed Utilization of Juvenile Haddock (Melanogrammus aeglefinus) (사료내 단백질 수준이 해덕(Melanogrammus aeglefinus)의 성장과 사료이용효율에 미치는 영향)

  • The effects of dietary protein level on growth, feed utlization and liver size were studied with juvenile haddock (Melanogrammus aeglefinus) of 6.88$\pm$0.54 g (mean$\pm$SD). Five isoenergetic diets containing 44.4 to 64.7% protein were fed to triplicate groups of 50 fish in a flow-through system for 6 weeks. All fish showed a relative increase of more than 300% in final weight. There were, however, no significant differences (P>0.05) in weight gain (16.1 to 17.3 g) and specific growth rate (2.9 to 3.0) among fish groups. A higher protein diets resulted in a lower feed intake. Feed: gain ratio linearly improved (P<0.05) when protein level increased from 44.4 to 59.7%. The lowest protein efficiency ratio was observed in fish fed the highest protein diet. Hepatosomatic index of fish significantly decreased from 10.7 to 7.3% with an increase in protein level. Results from this study suggest that haddock could grow well even when fed the diet with protein lower than 44.4%.
