Effects of Mung Bean (Phaseolus aureus L.) Supplementation on BUN and Hepatic Functional Enzyme Activities in Streptozotocin -induced Diabetic Rats (녹두(Phaseolus aureus L.) 급여가 당뇨성 흰쥐의 BUN 및 간 기능 효소 활성에 미치는 영향)

  • The purpose of this study was to investigate the improvement effect of mung bean (Phaseolus aureus L.) on the hepatic functional enzyme and catalase activity of streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley (SD) male rats were divided into four groups (n=6), and fed experimental diets containing mung bean meal [basal diet+5% mung bean (BM), basal diet+STZ+5% mung bean (SM)], and control (Basal Diet, BD), BS groups (basal diet+STZ). Serum concentrations of Blood Urea Nitrogen (BUN) and creatinine were significantly decreased (p<0.05) by 5% mung bean supplementation diet. The activities of aspartate transaminase (AST), alanine transaminase (ALT), akaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase and lipase were decreased in the BD, BM and SM group than BS group. The catalase (CAT) activity was significantly increased (p<0.05) in mung bean supplementation diet (BM, SM group) than diabetic group (BS). In vivo experiments with diabetic rats showed that ingestion of mung bean supplementation diet were effective in BUN concentration, and hepatic functional enzyme activities.

Metabolic Regulation of Homocysteine in Type 2 Diabetic Goto-Kakizaki Rats (당뇨병 Goto-Kakizaki 랫트에서 호모시스테인의 대사조절)

  • Elevation of homocysteine levels is a risk factor for cardiovascular diseases and liver diseases. It has been reported that both streptozotocin-induced type I diabetic rats and obese type II diabetic rats have plasma total homocysteine lower than each control rats. We determined the effects of lean type II diabetes on homocysteine levels using type 2 diabetic Goto-Kakizaki rats. The concentrations of serum glucose were increased to ${\sim}two-fold$ of control levels and the total cholesterol levels were also increased in GK rats. Hepatic aspartate, histidine, threonine, alanine and methionine levels were significantly increased in GK rats. Plasma aspartate and glutamate levels were elevated, but threonine and arginine levels were decreased in GK rats. Plasma total homocysteine levels were not changed in GK rats, but hepatic total homocysteine levels were increased to ${\sim}three-fold$ of control levels. These results suggest that hepatic metabolism of sulfur-amino acid may be altered in diabetic condition.

Effects of Dietary Folate Supplementation on the Homocystine Diet-Induced Hyperhomocysteinemia and Hepatic S-Adenosylmethionine Metabolism in Rats (엽산 보충이 호모시스틴 식이에 의해 유발된 고호모시스테인혈증과 간의 S-Adenosylmethionine 대사에 미치는 영향)

  • We investigated the effects of dietary folate supplementation on plasma homocysteine, vitamin B$_{12}$ and hepatic levels of S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH) in diet-induced hyperhomocysteinemic rats. All animals were fed 0.3% homocysteine diet for 2 weeks, then they were placed either on a 0.3% homocystine or no homocystine with or without 8 mg/kg folate diet for 8 weeks. Homocystine diet induced hyperhomocysteinemia up to 3.5-fold at 10 weeks (28.0 $\pm$ 4.8 $\mu$mol/l vs. 7.9 $\pm$ 0.3 $\mu$mol/l). Dietary folate supplementation caused a significant decrease in plasma homocysteine levels which had been increased by a homocystine-diet. Also, dietary folate supplementation made them return to control levels at 4 wk when the diet was free of homocystine. Plasma folate levels were markedly decreased with homocystine diet with no folate supplementation. Plasma vitamin B$_{12}$ did not differ between groups. Dietary homocystine increased hepatic levels of SAM in folate supplementation group at 10 weeks (p<0.05). Dietary folate supplementation increased hepatic levels of SAM/SAH ratios in homocystine group (p<0.05). In conclusion, dietary folate supplementation can effectively ameliorate the detrimental effects of hyperhomocysteinemia.mia.

Protective effects of Korean red ginseng extract on cadmium-induced hepatic toxicity in rats

  • Korean red ginseng is known to regulate the immune system and help the body struggle infection and disease. Cadmium is widely distributed in the environment due to its use in industry. Exposure to cadmium is problematic causing organ dysfunction. This study was conducted to evaluate the protective effect of Korean red ginseng extract (RGE) against cadmium-induced hepatotoxicity in rats. In experiments, animals were orally administrated with RGE (25, 50 mg/kg) for 7 d and then intravenously injected with cadmium ($CdCl_2$, 4 mg/kg) to induce acute hepatotoxicity. Cadmium caused the elevated levels of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase in serum. In contrast, pretreatment with RGE significantly reduced those serum indexes related with liver damage. In histopathological analysis, RGE decreased the centrilobular necrosis around central veins and the peripheral hemorrhage around portal triads. Moreover, RGE restored the deficit in hepatic glutathione level resulting from cadmium treatment. RGE also inhibited the increase in the expression of Bad, a representative apoptosis marker protein, induced by cadmium treatment. Collectively, these results demonstrate that RGE can reduce the cadmium-induced hepatic toxicity, partly via anti-oxidative and anti-apoptotic process.

Diagnostic laparoscopy in a leopard cat (Prionailurus bengalensis) with intercostal abdominal hernia and hepatic lipidosis

  • Intercostal abdominal hernia in the 11th intercostal space was identified in a leopard cat. Although mild leukopenia was found in laboratory examinations, no remarkable abnormality was revealed in medical imaging. To investigate abdominal organs, diagnostic laparoscopy was performed after hernia repair. In laparoscopic view, closure of the herniation site and a lesion with whitish discoloration in the liver (left medial lobe) were observed. Subsequently, laparoscopic liver biopsy was performed against the affected hepatic tissue. Histologically, the sample was diagnosed as mild hepatic lipidosis. Laparoscopy is considered useful for abdominal visceral examination and liver biopsy in a leopard cat patient.

  • This study was undertaken to determine the effect of dietary fat to carbohydrate ratio on plasma glucose. free fatty acid level and hepatic glucokinase activity in normal or insulin treated diabetic rats. Sprague-Dawley male rats were fed with 3 different but isocaloric diets for 5 weeks. Diet 1 made to have low fat(4% corn oil and 65.8% corn starch wt/wt) : diet 2 medium fat (12% : 47.8%) : diet 3 high fat (20% : 29.8%) In the normal rats an apparent increase of GK activity was observed from the animal fed low fat diet when compared with other groups. GK activities were decreased in all the alloxan-diabetic rats than the normal rats. When insulin was injected the GK activities in all the livers of alloxan-diabetic rats restored to normal level and GK activity was highest in low fat group. In the entire group significant relationships were seen between the plasma glucose and GK activities(r=-0.6, p<0.001) FFA levels and GK activities(r=-0.63 p<0.001) Both in normal and insulin treated diabetic rats significantly depressed level of hepatic GK activity was observed in the livers of animals fed high fat diet for 5 weeks and depressed level of GK activity may be related to insulin resistance.

  • 8-Acyl-2-hydroxyphenzaine -5, 10-dioxides and 8-acyl-2-aminophenazine-5, 10-dioxides bearing n-butanoyl, n-hexanoyl and n-octanoyl groups were synthesized. It was attempted to observe the effect of phenazine dioxide derivatives on the hepatic xanthine oxidase activity in this study. As the activity of xanthine oxidase, the key enzyme in the generation of superoxide anion radical ($O_2$), was measured in the presence of phenazine dioxide derivatives, the action of 8-acyl-2-hydroxyphenazine-5,10-dioxide derivatives inhibited with increase of numbers of carbon atom bearing 8-acyl group. Moreover, when plotted on double reciprocal form, the Vmax value decrease as increase of numbers of carbon atom bearing acyl groups without affecting the Km value. However, the hepatic xanthine oxidase activity was not changed by 8-acyl-2-aminophenazine-5,10-dioxide derivatives.

Effect of 2-Methylaminoethyl-4,4'-Dimethoxy-5, 6, 5' ,6'-Dimethyl­enedioxybiphenyl-2-Carboxylic Acid-2'-Carboxylate Monohydro­chloride (DDB-S) on Indocyanine Green (ICG) Clearance in Rats

  • The clearance of ICG, a known hepatic blood flow marker was investigated in rats in order to examine whether DDB-S influences hepatic blood flow. The effect of DDB-S on the protein binding and blood-to-plasma partition of ICG was measured. The steady-state plasma concentration of ICG was monitored before and after co-administration of various concentration of DDB-S, and ICG clearance was estimated from the steady-state concentration and the infusion rate of ICG. There was no significant difference in protein binding and blood-to-plasma partition of ICG with and without addition of DDB-S (10, 20, and 40 ${\mu}g/mL)$. When ICG was infused into DDB-S pretreated rats, the steady-state concentrations of ICG decreased and the calculated ICG clearance increased. However, no dose-dependency of ICG Css on DDB-S Css was observed. Since DDB-S did not affect the protein binding and blood-to-plasma partition of ICG, the increased clearance of ICG with co-administration of DDB-S seems to be due to the increased hepatic blood flow by DDB-S.

Hepatic Portojejunostomy for Biliary Atresia (담도폐쇄증에서 간문부-장 문합술의 성적)

  • The results of hepatic portojejunostomy in 34 patients with biliary atresia operated upon by one surgeon between May 1989 and December 1997 were analyzed. Eleven (32.3 %) patients were 60 days or younger, 14 patient (41.2 %) were between 60 and 90 days, and 9 (26.5 %) were over 90 days of age. Jaundice cleared in 20 cases (58.8 %). Three patients died of liver insufficiency, 2 were anicteric but died from esophageal variceal bleeding. Three patients died as a result of sepsis, heart failure and left kidney agenesis. Five patients were lost to follow-up. The five-year survival rate was 73.8 %. Two patients over 90 days of age, survived more than 5 years. Survival rates were not significantly related to the age at operation. We conclude that hepatic portojejunostomy should be considered as a primary surgical modality for biliary atresia, even at age 90 days or more. Early detection of esophageal varices and sclerotherapy may be necessary. Liver transplantation is necessary if hepatic failure develops.

Inhibitory Effect of Leek Greek Juice on $CCl_{4}$-induced Hepatotoxicity in Rats (부추녹즙이 사염화탄소 투여에 의한 흰쥐의 간손상에 미치는 영향)

  • To investigate the effects of leek green juice on the damaged liver of $CCl_{4}$-treated rats, Sprague-Dawley male rats weighing about 100g, were divided into 4 groups ; control group (CON), leek green juice-administered group (LGJ), $CCl_{4}$-treated group (CCL) and leek green juice and $CCl_{4}$-treated group (LCL). After 6 weeks, the activities of sGPT and sGOT, and content of hepatic TBA-reactants, elevated by $CCl_{4}$ treatment, were markedly decreased by administering leek green juice, compared to CCL. It was also observed that activities of hepatic SOD and GSH-Px were elevated by $CCl_{4}$-treatment as compared to CON, but concomitant treatment of leek green juice and $CCl_{4}$ decreased those levels adjacent to CON, whereas catalase activity did not show significant decreasing effects compared to CCL. The hepatic content of glutathione, decreased by $CCl_{4}$, was more abundantly increased by leek green juice administration than by CCL. These results suggest that leek green juice is believed to b a possible protective effect for the carbon tetrachloride-induced hepatotoxicity in rat liver.
