철분의 과잉섭취시 셀레늄 섭취수준이 철분과잉 축적에 대한 예방효과에 관한 연구 (Preventive Effect of Selenium Supplementation on Iron Accumulation of Rats Fed Diets Containing High Levels of iron)

  • The purpose of this study was to investigate the effect of selenium supplementation of iron accumulation of rats fed diets containing high levels or iron. Sixty male Sprague-Dawley weaning rats were fed with diets containing various levels of iron(adequate : 35ppm, 2-fold : 70ppm, 4-fold : 140ppm) and selenium(adequat : 0.05ppm and high : 0.05ppm) for 12 weeks. Feed intakes of 2-fold and 4-fold iron groups were higher than that of adequate iron group. There was no difference body weight gain across iron and selenium containing diet groups. Hemoglobin level was increasd with iron increment and decreased with selenium supplementation. Iron contents in serum and tissues were increased as iron intake was increased. Liver iron content was decreased with selenium supplementation. Selenium content in liver was decreased with iron increment and increased with selenium supplementation. In the case of iron balance, iron excretion through urine and feces was significantly increased as iron intake was increased. However, apparent absorbability and retention rate of iron were not significantly affected by dietary iron or selenium.

고농도 Heme-iron의 생산을 위한 Hemoglobin의 가수분해 및 분리 조건 (Conditions of Hemoglobin Hydrolysis and Separation for the Production of Enriched Heme-iron)

  • Hemoglobin(Hgb)의 효소 가수분해액으로부터 고농도 heme-iron의 분리에 영향을 주는 기질의 농도, 가수분해도 (DH, degree of hydrolysis) 및 분리 pH의 효과 등을 조사하였으며, pilot scale에서 고농도 heme-iron을 생산하였다. 최적 반응조건(pH 10.0, $50^{\circ}C$, 5 hr)에서의 Esperase에 의한 Hgb의 DH는 $24{\sim}25%$였다. Hgb의 농도는 heme-iron의 분리에 영향을 미쳐서 Hgb의 농도가 2.5%, 5.0%, 10.0%로 증가할수록 heme-iron/peptide(heme-iron의 분리도)는 각각 47.4%, 35.6%, 21.8%로 감소하였으며, 이에 따라 농축율도 감소하였다. Hgb의 가수분해액으로부터 고농도 heme-iron의 분리에 효율적인 pH 영역은 DH에 따라 달라졌으며, DH가 6.3%의 경우는 pH $5.0{\sim}6$에서 heme-iron/peptide의 비가 8.92%, DH가 14.5%인 경우는 pH $4.0{\sim}6.0$에서 $12.1{\sim}19.5%$였으며, DH가 24.3%인 경우는 pH $3.0{\sim}5.0$에서 $34.7{\sim}36.4%$로 가수분해도가 높을수록 고농도 heme-iron의 분리시 최적 PH 범위가 산성 영역으로 이동되었으며, 이 영역의 pH에서 peptide의 용해도 증가로 분리도가 증가되었다. 가수분해액으로부터 분리된 heme-iron의 분자량은 약 1 kDa정도로 hematin과 유사한 분리패턴을 보였다. Pilot scale로 제조한 고농도 heme-iron 제품에서의 heme-iron 함량은 27.1%, heme-iron/peptide 비는 38.7%, 생산수율은 9.3%이었다.

목향과 향부자 추출물이 흰쥐의 만성 스트레스에 미치는 실험적 효과 (Experimental Effects of Aucklandiae Radix and Cyperi Rhizoma Extract on Chronic Stress in Rats)

  • This study aimed to investigate the anti-stress function of Aucklandiae Radix (AR) and Cyperi Rhizoma (CR). The essential oils used in the experiment were extracted from AR and CR using Steam Distillation Extraction and Super critical CO2 extraction. To observe the effects of sample administration, we measured feed intake, leukocytes, red blood cells, hemoglobin, platelets, serum serotonin content, immobility time, climbing time, and swimming time in mice subjected to chronic restraint stress as behavioral changes. The average body weight of all experimental groups increased than the average body weight of the control group. The immobility and climbing times of experimental groups A and B administered with supercritical extraction samples were shorter than those of the other experimental groups and the control group, and the swimming time was longer. The serotonin content in the blood of all experimental groups decreased compared to the normal group, and the serotonin content of the control group was increased. The authors suggest that Korean herbal medicines AR and CR may be utilized as anti-stress flavoring agents based on the above results.

황칠(黃漆)이 만성 스트레스 유발 백서의 스트레스 및 수면 호르몬에 미치는 영향 (Effect of Resina Dendropanacis Morbiferus on Stress and Sleep Hormone in Chronic Mild Stress-Induced Rats)

  • Objectives: To evaluate effects of Resina Dendropanacis Morbiferus (RDM) on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats. Methods: Twenty-five healthy rats were randomly divided into 5 groups; Normal, CMS (Control), Resina Dendropanacis Mobiferus 50 mg/kg (RDM 50), 100 mg/kg (RDM 100), and 200 mg/kg (RDM 200). All rats except the normal group were exposed to unpredicted stress conditions such as water deprivation, empty bottles, forced thread mill, etc. according to timetable of CMS for 3 weeks. After a week starting experiments, rats in RDM 50, RDM 100, and RDM 200 groups were fed orally once a day for 2 weeks. Afterward, blood samples were taken from rats to analyze complete blood count, AST, ALT, and glucose. Noradrenalin, GABA and Melatonin were measured by ELISA kit. BDNF, CREB and TrkB were measured by RT-PCT. Results: 1. In Noradrenalin content, RDM 100 and RDM 200 groups revealed significant decrease compared to the control group. 2. In GABA content, RDM 50 and RDM 100 groups revealed significant decrease compared to the control group. 3. In Melatonin content, RDM 100 and 200 groups revealed significant decrease compared to the control group. 4. In Activity of BDNF, RDM 100 and 200 groups revealed significant increase compared to the control group. 5. In Activity of CREB and TrkB, RDM 100 group revealed significant increase compared to the control group. 6. In Erythrocyte and Thrombocyte changes, red blood cells and hematocrit significantly increased in RDM 50 and RDM 200 groups than the control group. Hemoglobin and platelet significantly increased in all experimental groups and the control group. 7. In Weight gain content, all RDM groups revealed insignificant increase compared to the control group. 8. In Glucose content, RDM 50, RDM 100, and RDM 200 groups revealed significant decrease compared to the control group. Conclusions: Results suggest that RDM have effects on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats.

종류가 다른 고 섬유질식이가 흰쥐의 납 흡수 및 체내대사에 미치는 영향 (Effect of High Dietary Fiber Diet on Lead Absorption and Metabolic Changes in Growing Rats)

  • This study was performed to investigate nutritional effect of various dietary fibers on lead absorption, and protein and lipid metabolisms in growing rats. Sixty male rats of Sprague-Dawley strain weighing 140$\pm$1.1g were blocked into 10 groups according to body weight and fed 10 kinds of diet different with fiber sources [non-fiber, cellulose, pectin, guar gum or carboxymethylcellulose(CMC)] and lead levels (0 or 1%) for 4 weeks. Results were summerized as follows : 1) Food intake, weight gain, FER and PER were remarkably decreased in lead(Pb)-added groups. Weight gain, FER and PER in Pb-added pectin group were significantly lower than those in Pb-added non-fiber group. 2) Liver and kidney weights, femur weight and length, hematocrit and hemoglobin content were decreased in Pb-added groups. Especially femur and liver weights in pectin groups were the lowest among groups. 3) Total protein content in serum was significantly decreased in Pb-added groups but was not different with dietary fiber sources. Total lipid content in serum was not different with dietary Pb levels and fiber sources, but cholesterol content in serum of guar gum group was significantly decreased by Pb addition. 4) Nitrogen, lipid and cholesteol contents in liver were significantly decreased in Pb-added groups, and lipid content in liver of pectin and CMC groups was lower than other groups. 5) Daily urinary and fecal excretions of nitrogen, kipid and cholesterol were decreased in Pb-added groups, and fecal nitrogen was significantly increased in Pb-added groups, and fecal nitrogen of cellulose and guar gum groups was significantly higher than other groups. Fecal excretions of lipid and cholesterol were increased by dietary fibers, and especially fecal lipid was remarkably increased in pectin and guar hum group. 6) Pb contents in liver and femur were decreased by dietary fibers. Especially Pb contents in liver, kidney and femur were significantly decreased in guar gum group. 7) Daily urinary and fecal excretions of Pb were significantly increased in cellulose and guar gum groups, and fecla excretion of Pb in guar gum group was twice of non-fiber group. Pb absorption ratio was significantly decreased in guar gum group. In conclusion, dietary fibers have effect on protein and lipid metabolisms, and decreased intestinal absorption of Pb by increasing fecal excretion. But the degree of effect was different with dietary fiber sources.

  • The present study was conducted in order to get the normal hematologic values of Korean Jindo dogs. Blood samples were taken from 328(♂ 92, ♂ 236) healthy Jindo dogs in Jindo area. The results obtained in this study were summarized as follows: 1. The mean values of red blood cell(RBC) count, hemoglobin(Hb) content and packed cell volume(PCV) in the age group of less than one year were 6.77(♂ 6.80, ♂ 6.76)${\times}10^6/{\mu}l$, 13.14 (♂ 13.53, ♂ 12.99)g/100ml and 43.28(♂ 44.47, ♂ 42.79)ml/100ml, respectively, whereas the RBC count, Hb content and PCV in the age group of one year and more were 7.42(♂ 7.44, ♂ 7.42)${\times}10^6/{\mu}l$, 14.98(♂ 15.56, ♂ 14.76)g/100ml and 47.18 (♂ 48.43, ♂ 46.71)ml/100ml, respectively. The values of RBC count, Hb content and PCV appeared to be higher in the male than in the female and it increased with aging in the age group of less than one year. 2. The mean values of mean corpuscular volume(MCV), mean corpuscular hemoglobin(MCH) and mean corpuscular hemoglobin content(MCHC) in the age group of less than one year were 63.93(♂ 65.40, ♂ 63.30)fl, 19.41(♂ 19.90, ♂ 19.22) pg and 30.36(♂ 30.43, ♂ 30.36)g/100ml respectively, whereas the MCV, MCH and MCHC in the age group of one year and more were 63.58(♂ 65.09, ♂ 62.95)fl, 20. 19(♂ 20.91, ♂ 19.89) pg and 31.75(♂ 32.13, ♂ 31.60)g/100ml respectively. The values of MCV, MCH and MCHC appeared to be higher in the male than in the female. No differences were found in the MCV and MCH between age groups. The MCHC appeared to be higher in older age group. 3. The mean values of white blood cell(WBC) count in the age group of less than one year were 14,356(♂ 13,878, ♂ 14,551)/${\mu}l$, whereas the values of WBC count in the age group of one year and more were 13,394(♂ 12,656, ♂ 13,672)/${\mu}l$. The WBC count appeared to be higher in the female than in the male. No differences were found between age groups in the WBC count. 4. In WBC differential count the mean percentage of lymphocyte, monocyte, neutrophil and eosinophil of the age group less than one year were 33.45(♂ 31. 64, ♂ 34.18), 2.77(♂ 3.00, ♂ 2.68), 57.40(♂ 58.18, ♂ 57.08) and 6.38(♂ 7.18, ♂ 6.06)% respectively, while those of the age group one year and more were 30. 22(♂ 28.38, ♂ 30.90), 3.03(♂ 3.23, ♂ 2.95), 60.93(♂ 63.34, ♂ 60.03) and 5.82(♂ 5.04, ♂ 6.11)% respectively. Lymphocyte count appeared to be higher in the female and in the age group below one year than in the male and in the age group of one year and more, whereas the reverse was the case with neutrophil and monocyte counts. 5. Mean platelet count in the age group of less than one year was 377,391(♂ 398,778, ♂ 368,721)/${\mu}l$, whereas mean platelet count in the age group of one year and more was 354,657 (♂ 373,660, ♂ 347,512)/${\mu}l$. The platelet count appeared to be higher in the male than in the female and especially lower in the age group of three years and more($305,513/{\mu}l$) than in the other age groups.

Cadmium 투여 흰쥐에 있어서 Metallothionein 합성과 항산화적 해독기구에 미치는 식이 Selenium의 영향 (Effect of Dietary Selenium of Metallothionein Synthesis and Antioxidative Detoxificantion Mechanism in Cadmium Administered Rats)

  • In order to investigate the effect of selenium (Se) on the liver damage, metallothionein synthesis and hepatic antioxidative detoxification system in cadmium(Cd) administered rats. Sprague-Dawley male rats(60\\5g) were divided into two diet groups, depending on with (CdS groups) or without (Cd groups) 0.5ppm Se supplementation and fed experimental diets ad libidum for 4 weeks. And then each group was again subdivided into five groups, depending on injection number of Cd, i.e., 0, 1, 2, 3, and 4 times of 2.5mg Cd/kg of body wt once a day. Hemoglobin concentration, hematocrit values, superoxide dismutase, glutathione peroxidase and glutathione S-transferase activite were decreased progressively with increasing number of Cd injection, but increased by the supplementation of Se. The reduced form of glutathione (GSH) contents in blood and liver and vitamin E content were decreased and oxidized form (GSSG) increased in Cd groups, but these of Se supplemented groups were not very different from controls. Cd reduced liver vitamin E content which was not restored by Se supplementation. Liver lipid peroxide values were elevated with increasing doses of Cd, but Se supplementation reduced these elevated levels. Accumulation of metallothionein in liver and kidney was increased with increasing number of Cd injection, but Se did not affect on them. Histological examination revealed that lysosomes were significantly increased and mitochondria and Golgi apparatus were enlarged by Cd, however, these changes were reduced by Se. It was concluded that Se administration promoted antioxidative detoxification and alleviated peroxidative damage in rat liver by Cd.

Antioxidant and free radical scavenging activities of Cleome rutidosperma

  • The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.

식이내 Cadmium과 단백질 수준이 흰쥐의 체내 단백질 대사 및 Cadmium 중독에 미치는 영향 (Effects of dietary Cadmium and Protein Levels on the Body Protein Metabolism and Cadmium Toxicity in Growing Rats)

  • This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.

Is It Possible to Predict the Iron Status from an Infant's Diet History?

  • Purpose: Iron deficiency remains a very common nutritional problem despite the improvement in nutrition and increased understanding of methods for its prevention. Thus, we try to create a new method for screening iron nutrition through infant nutrition history. Methods: Among the children who visited Inha University Hospital from March 2006 to July 2012, 181 children with iron deficiency anemia (IDA) and 52 children without IDA ranging from 6 to 36 months of age were reviewed in this study. We used the age when they began to wean food, the type of sort weaning foods, the time required for successful weaning, iron content in weaning foods, and the duration of breastfeeding for scoring infant nutrition history based on a questionnaire. Results: The mean score of the IDA group was $7.8{\pm}2.6$ points, which was significantly higher than that of the control group ($5.6{\pm}2.1$) (p=0.000). If we set up the cutoff value at 6 points, this screening has 86.8% sensitivity and 36% specificity. In addition, as the IDA score increased, there was a falling trend of hemoglobin. Conclusion: The IDA score does not have high specificity or high sensitivity. However, this study conveys that those patients who record a high score have low hemoglobin. Therefore, we suggest this score system for screening more IDA patients via nonpainful techniques.
