Bioavailability of Iron-fortified Whey Protein Concentrate in Iron-deficient Rats

  • An iron-fortified whey protein concentrate (Fe-WPC) was prepared by addition of ferric chloride to concentrated whey. A large part of the iron in the Fe-WPC existed as complexes with proteins such as ${\beta}$-lactoglobulin. The bioavailability of iron from Fe-WPC was evaluated using iron-deficient rats, in comparison with heme iron. Rats were separated into a control group and an iron-deficiency group. Rats in the control group were given the standard diet containing ferrous sulfate as the source of iron throughout the experimental feeding period. Rats in the iron-deficiency group were made anemic by feeding on an Fe-deficient diet without any added iron for 3 wk. After the iron-deficiency period, the iron-deficiency group was separated into an Fe-WPC group and a heme iron group fed Fe-WPC and hemin as the sole source of iron, respectively. The hemoglobin content, iron content in liver, hemoglobin regeneration efficiency (HRE) and apparent iron absorption rate were examined when iron-deficient rats were fed either Fe-WPC or hemin as the sole source of iron for 20 d. Hemoglobin content was significantly higher in the rats fed the Fe-WPC diet than in rats fed the hemin diet. HRE in rats fed the Fe-WPC diet was significantly higher than in rats fed the hemin diet. The apparent iron absorption rate in rats fed the Fe-WPC diet tended to be higher than in rats fed the hemin diet (p = 0.054). The solubility of iron in the small intestine of rats at 2.5 h after ingestion of the Fe-WPC diet was approximately twice that of rats fed the hemin diet. These results indicated that the iron bioavailability of Fe-WPC was higher than that of hemin, which seemed due, in part, to the different iron solubility in the intestine.

  • 성숙 합토글로빈(haptoglobin, Hp)은 혈장 당단백질로서 혈중의 유리 헤모글로빈(hemoglobin, Hb)을 제거하고 항산화 작용을 한다. 프로합토글로빈(prohaptoglobin, proHp)은 Hp 전구체이며 혈중에 낮은 농도로 존재하지만 그 생리적 기능은 아직 확실하지 않다. 본 연구에서는 proHp와 Hp의 구조적, 기능적 차이를 연구하기 위하여 재조합 proHp를 제조하여 시알산과 Hb-결합력을 조사하고 성숙 Hp와 비교하였다. 비환원 조건의 Western blot 분석에서 proHp1은 약 130 kD의 하나의 밴드를 보였고 proHp2는 200 kDa 이상의 다중 밴드를 보였는데 이것은 대응되는 성숙 Hp1-1과 Hp2-2와 각각 동일한 양상이었다. 하지만 비환원 및 비변성 조건 하에서 수행된 전기영동에서는 proHp가 Hp보다 더 느리게 이동하였다. 렉틴을 이용한 ELISA 분석에서 proHp는 Hp보다 산성 당인 시 알산 함량이 더 낮음을 알 수 있었다. 또한 proHp는 Hb과의 결합력도 더 낮았다. 이러한 결과들로부터 proHp는 Hp와 같은 양상으로 중합체를 형성하지만 시알산 함량과 Hb-결합력이 Hp와 다름을 알 수 있었고 혈중에서 전구체인 proHp는 Hp와 다른 기능을 수행할 것임을 시사한다.

Effects of Ammonia Concentration on Histological and Physiological Status in Black Seabream (Acanthopagrus schlegeli)

  • The histological changes of gill, liver, spleen and muscle, and respiration and blood variables and liver glycogen content were examined in black seabream, Acanthopagrus schlegeli. Fish were exposed to a high level of total ammonia nitrogen (10.4 mg/l) and recovered from exposure $(0.4{\pm}0.2mg/l)$ in a closed recirculating seawater system. In the process of exposure, mortality was $9\%$, and hyperplasia, necrosis or inflammation appeared in all tissues except for muscle. Oxygen consumption was decreased by $49\%$, and red blood cell (RBC) number, hematocrit and hemoglobin concentration were significantly decreased, while plasma glucose contents, activities of glutamate-oxaloacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT) increased. Liver glycogen content significantly increased from $6.6\%\;to\;10.4\%$. A large amount of hemosiderin was observed in the splenic tissue. During the recovery period, RBC number, hematocrit, hemoglobin concentration, GOT and GPT activities were returned to the normal status. Histological status of liver tissue was returned to the normal, but liver glycogen content was not recovered. During the recovery period, spleen melanin-macrophages temporarily increased, but subsequently decreased to the normal status.

산양의 실험적 납중독에 관한 임상병리학적 관찰 I. 임상학적 관찰(증상, 혈액, 뇨) (Clinico-pathological studies on the experimental lead poisoning in goats I. Clinical observations(Clinical, hematological and urinary findings))

  • Present experiments were undertaken in order to clarify the clinico-pathological characteristics of lead poisoning in goats. Twenty goats were divided into three experimental groups(A, B and C) and a control(D). The three experimental groups received diets contaminated artificially with 10(A group), 200(B group) and 1,000(C group) ${\mu}g/$ of lead, for 70 days respectively. The control group received normal diets. Blood samples were collected 1 or 2 weeks interval and were examined for anemia(erythrocyte counts, hemoglobin concentrations and hematocrit values) and lead contents of erythrocyte and serum. Urine samples collected similarly with blood were examined for delta-aminolevulinic acid and lead content. Collected samples were analyzed for lead content by atomic absorption spectrophotometry. From these experiments following results were obtained : In group B and C, marked decreases in body weight and feed intake, and diarrhea were observed from the $30^{th}$ day of experimental periods. The B and C groups showed pronounced anemia(decrease in erythrocyte count, hemoglobin concentration and hematocrit value) from the $21^{st}$ or $42^{nd}$ day. In group B and C, the lead content of erythrocytes was increased significantly from the $28^{th}$ or $14^{th}$ day. The lead content of serum was increased significantly from the $42^{nd}$ or $14^{th}$ day in B and C groups. The lead content of urine was increased significantly from the first day in both B and C groups. The urinary delta-aminolevulinic acid content was increased significantly from $14^{th}$ day in both B and C groups.

고본건양단(固本健陽丹)이 백서(白鼠)의 성(性)호르몬 및 조혈기능(造血機能)에 미치는 영향(影響) (The Effects of Gobongeonyangdan on the sex hormones and hematogenesis in rats)

  • It has been known that "Gobongeonyangdan" has a certain effects on male sterility in the field of oriental medicine. This thesis purposes to prove such effects of the medicine especially concerning sex hormones and hematogenesis. In the study were used three groups of rats: two of those were treated with Gobongeonyangdan with a variety of quantity and the other untreated. The experiment spans for 35 days: tests upon those subjects were done three times on the 7th, 21st, and 35th day checking the change of testosterone, estradiol, LH, FSH and iron in the serums, and RBC, hematocrit and hemoglobin levels. The results obtained from the study are as follows: 1. The content of testosterone in the serums marked a significant increase in both treated groups in comparison to the untreated group. 2. The content of estradiol in the serums showed a significant increase in the treated groups as to the untreated. 3. The content of LH in the serums increased significantly in the treated groups as to the untreated. 4. The content of FSH in the serums also increased remarkably in the two treated groups as to the untreated. 5. The content of iron in the serum was seen to be increased in the treated groups rather than the untreated group. 6. The RBC was observed to show a tendency toward a significant increase in both the treated groups in comparison with the untreated group. 7. The hemoglobin levels were also increased significantly in the treated groups as to the untreated. 8. The hematocrit levels were decreased significantly in one of the two treated group(sample A group) and there was no difference in the other group(sample B group) in comparison with the untreated group. Consequently, "Gobongeonyangdan" may well be a favorable medicine for patients two have sexual functional disorder and it is also effective on hematogenesis.

Analyses of intermediate products during degradation of pyrene in soil by hemoglobin-catalyzed reaction

  • Hemoglobin (Hb) is a member of heme-protein that can perform catalytic non-specific chain reaction in the presence of hydrogen peroxide ($H_2O_2$). Catalytic ability of Hb to degrade pyrene was demonstrated using soil contaminated with $^{14}C$ pyrene and 10 mg pyrene /kg soil. The composition of soil was similar to previously used soil except that it had lower organic carbon content. Bench scale laboratory tests were conducted in the presence of buffer only, $H_2O_2$ only, or Hb with $H_2O_2$ for 24 h. After 24 h reaction, 0.1 and 1.3% of $^{14}C$ pyrene in contaminated soil were mineralized with $H_2O_2$ only or Hb plus $H_2O_2$. No mineralization to $^{14}CO_2$ was detected with buffer only. Approximately 12.2% of pyrene was degraded in the presence of $H_2O_2$ only while 44.0% of pyrene was degraded in the presence of Hb plus $H_2O_2$ during 24 h of catalytic reaction. When degradation intermediate products were examined, two chemicals were observed in the presence of $H_2O_2$ only while 25 chemicals were found in the presence of Hb plus $H_2O_2$. While most degradation products were simple hydrocarbons, four of the 27 chemicals had aromatic rings. However, none of these four chemicals was structurally related to pyrene. These results suggest that Hb catalytic system could be used to treat pyrene-contaminated soil as an efficient and speedy remediation technology. In addition, intermediate products generated by this system are not greatly affected by composition change in soil organic matter content.

고식이섬유쌀의 급여가 KK 당뇨 마우스의 혈당에 미치는 영향 (Supplementary Effect of the High Dietary Fiber Rice on Blood Glucose in Diabetic KK Mice)

  • This study was carried out to investigate the supplementary effects of Suwon 464, which has over two times of dietary fiber content compared with normal rice (Ilpum), on blood glucose in diabetic mice. We supplied 5 kinds of experimental diets (corn starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight and contents of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, l.4, and 2.0% respectively. Body weight was lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentration of blood glucose in diabetic mice was lower in SB group than the other groups during the supplementary period of experimental diets. The hemoglobin Ale and serum insulin levels were lower in SP and SB groups. These results suggested that the brown rice of Suwon 464 with high dietary fiber can control diabetes in diabetic mice by reducing the blood glucose and hemoglobin Ale. HEMATOLOGICAL STUDIES ON CATTLE EXPOSED TO Fasciola gigantica INFESTATION

  • Hematological value were determined from 140 Fasciola gigantica infested and 138 non-infested apparently healthy (control) zebu cattle from eight different sites of Bangladesh. The F. gigantica infested cattle have experienced of normocytic normochromic anemia indicated by the significant (p<0.01) decline of total erythrocyte count, hemoglobin level, packed cell volume and mean corpuscular hemoglobin concentration and significant (p<0.01) increase of erythrocyte sedimentation rate, as compared with non-infested control animals. Values of mean corpuscular volume and mean corpuscular hemoglobin content were not differed significantly between the infested and control animals. There observed significantly (p<0.01) higher eosinophil and neutrophil counts and significantly (p<0.01) lower lymphocyte counts in fasciola infested cattle in comparison with the control animals.

Pb acetate의 13주 반복투여가 랫드의 조혈기능, 정소 및 신장에 미치는 영향 (Effects of Repeated Exposure to Pb Acetate on Hematopoietic Function, Testis and Kidney in Male Rats)

  • Male Sprague Dawley rats were exposed to 0, 0.04, 0.2, and 0.8% Pb acetate in drinking water for 13 weeks and fed a commercial diet. Dose-related adverse effects observed at the end of the Pb acetate exposure in the drinking water were as follows: decrease in body weight gain, decrease in hemoglobin, hematocrit(HCT), mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH), increase in serum glucose, decrease in serum testosterone, increase in lead accumulation and $\delta$-ALA release in urine, and decrease in $\delta$-ALAD activities DNA content and histopathlogy (intranuclear inclusion body in kidney proximal tubule cell). Taken together, repeated exposure of lead acetate induced toxicities in hematopoietic system, especially testis and kidney.

한국 미취학 아동의 영양성 빈혈에 관한 연구 (A Study on Nutritional Anemia of Pre-school Children in Korea)

  • Nutritional anemia is an important nutritional problem affecting large population groups in most developing countries. Nutritional anemia is caused by the absence of any dietary essential involed in hemoglobin formation or by poor absorption of these dietary components. The most likely causes are lack of dietary iron, and folate, vitamin $B_{12}$ and high qualify protein. Anemia is considered to be a late mainfeastation of nutritional deficiencies, and even mild anemia is not the earilest sign of such a deficiency. Therefore, the object of therapy is to correct underlying deficiency rather than merely its manifestation. Iron deficiency anemia is generally much the most common form of anemia. And it is very prevalent particularly in pregnant women and young children, especially under five year of life. According to the rapid growth rate of infants, dietary iron should he provided for infants over three months of age in adequate amounts for the synthesis of hemoglobin required by the increasing blood volume and for the demands of newly formed cells. The principal causes of iron deficiency anemia are an inadequate dietary iron content, interference with absorption of iron from the intestine, excessive losses of iron from the body, disturbance of iron metabolism by infection, and social and cultural environments. The present study is planned to obtain informations concerning nutritional anemia through anthropometric and biochemical determinations for the assessment of nutriture in pre-school children. Determination was taken in 226 pre-school children in ruraI arae in 1968, 122 pre-school children in 1970, and 1526 hospitalized pre-school children in 1970. The results of this study are as follows; (1) According to Iowa Malnutrition Borderline (85 percentile) for weight, the proportions of underweighed pre-school boys and girls in rural area were 47.2% and 46.2% in1968, and were 36.1% and 51.8% in 1970. According to Iowa Malnutrition Borderline for height, the proportions of underheight boys and girls in rural area were 30.5% and 33.7%, and were 26.2% and 21.8% in 1970. Malnutrition scores of underweight for height values of boys and girls in rural area were 19.3 and 17.3 in 1968, and the scores of boys and girls were 15.6 and 15.5 in 1970. (2) The mean hemoglobin values of boys and girls in rural area were $11.2{\pm}1.8g/100ml\;and\;11.4{\pm}1.6g/100ml$ in 1968. In 1970, the mean values of boys and girls in rural area were $11.3{\pm}1.3g/100ml\;and\;11.7{\pm}2.4g/100ml$. The mean hemoglobin values of hospitalized boys and girls were $11.9{\pm}2.2g/100ml\;and\;11.7{\pm}2.4g/100ml$ in 1970. It is found that 92 of 215 children (42.7%) in rural area had concentrations of hemoglobin less than 11.0g/100ml in 1968. In 1970, 55 of 121 children (45.4%) in rural area and 559 of 1526 hospitalized children (36.6%) had concentrations of hemoglobin less than 11.0g/100ml. (3) The mean hematocrit levels of hospitalized boys and girls were $35{\pm}26.8%\;and\;35.4{\pm}6.4%$ in 1970. And 443 of 1334 hospitalized children (33.2%) had hematocrit values below 33%. (4) The average mean corpuscular hemoglobin concentration levels of hospitalized boys and girls were $32.4{\pm}2.2\;and\;32.3{\pm}2.2$ in 1970. And 1016 of 1352 hospitalized children (75.1%) had the mean corpuscular hemoglobin values below 34. (5) The mean iron values of young children in rural area and hospitalized children were $62.0{\pm}6.3{\mu}g/100ml\;and\;60.7{\pm}22.8{\mu}g/100ml$. The proportions of anemia cases below $50{\mu}g/100ml$ in rural area was 37.9%, and 34.3% in hospitalized children. (6) The mean total iron binding capacity of young children in rural area was $376{\pm}57.88{\mu}g/100ml,\;and\;342.2{\pm}6.15{\mu}g/100ml$ in hospitalized children. (7) The average transferrin saturation percentage of young children in rural area was $16.9{\pm}4.7%,\;and\;18.0{\pm}8.4%$ in hospitalized children. The proportions of anemia cases below 15% of young chi1dren in rural area and hospitalized children were 48.3% and 41.2%. Therefore, authors wish to recommend that the following further studies should be undertaken: (1) Standardization of simplied laboratory examination of nutritional anemia. (2) The prevalence of nutritional anemia and the requirements of iron, folate, and vitamin $B_{12}$ of pre-school children. (3) The content and absorption of iron in Korean food. (4) The pathogenesis of nutritional anemia and prevention of parasitic disease. (5) Maternal health and nutrition education.
