Effects of the different hydrogen peroxide ($H_{2}O_{2}$) treatment level on physiological and biochemical responses of olive flounder (Paralichthys olivaceus) (넙치 (Paralichthys olivaceus)에서의 과산화수소;($H_{2}O_{2}$) 처리 농도가 생리.생화학적 반응에 미치는 영향)

  • This study was conducted to investigate the change of antioxidant enzyme activity (catalase and superoxide dismutase) and variation of blood physiology in olive flounder (Paralyticus olivaceus) by hydrogen peroxide (H2O2) treatment. Blood parameters were measured 1, 3 and 5 hours after H2O2 treatment with 0 (control), 100, 300 and 500 ppm for 1 hr. The value of hematocrit was decreased significantly dependently on treatment concentrate and elapsed time in the treatment of H2O2. Hemoglobin concentration in the test groups were lower than that of the control group. Red blood cell value in the test groups were significantly lower compared to that of the control group, but recovered to the level of the control group after 5 hr. Protein concentration was significantly lower compared to that of the control group at 0 and 1 hr, but recovered after 3 hr in 500 ppm treatment group. The superoxide dismutase (SOD) and catalase (CAT) enzyme activities were observed to be increased. Heat-shock protein 70 (HSP70) was significantly increased compared to that of control group in all of the test groups. HSP70 mRNA groups was highly expressed in 500 ppm treatment.

Experimental Studies on Extracorporeal Circulation by Sarns Heart-Lung Machine with Total Prime of Hartman's Solution (Sarns 심폐기의 혈희석 체외순환에 관한 실험적 연구)

  • Total body perfusion using Sarns Heart-Lung-Machine, five head pump motor system with Travenol disposable bubble oxygenator was attempted in the dogs by the hemodilution method with total prime of buffered Hartman`s solution under moderate hypothermia. The first of all, the functions of Sarns Heart-Lung-Machine and effects of the hemodilution perfusion by buffered Hartman`s solution was studied. At the same time the changes of pressure of artery and vein, gas contents of the blood, and influence on the blood pictures were observed before, during, and after perfusion in 1-2 days. Hemodilution rates were the ranges of 85.0ml/kg to 97.3ml/kg and perfusion flow rates were maintained with the average 80. 5ml/kg/min [the ranges of 73.3ml/kg/min to 92.8ml/kg/min]. Hypothermia was employed between $35^{\circ}C$ and $31^{\circ} of the esophageal temperature. The total body perfusion was continued for 50-60 minutes. In the total cardiopulmonary bypass, atriotomy, ventriculotomy, and atrioventriculotomy were performed respectively. Arterial pressure was ranged approximately between 50 mmHg and 140 mmHg, but generally, it was maintained over 75 mmHg. Venous pressure was measured between 3.8 cm$H_2O$ and 16.0 cm$H_2O$. Optimum oxygenation could be achieved when oxygen flow into the oxygenator was maintained approximately at 5. 5L/min. In this way, the $pO_2$, $pCO_2$, and oxygen saturation were measured before, during, and afterperfusion in 1-2 days. The $pCO_2$ ranged approximately between 26.0 mmHg and 38.5 mmHg, but generally, it was maintained in the average 30.9-32.5mmHg. The $pO_2$ was ranged between 73.0mmHg and 332.2 mmHg, but it was maintained in the average 103.0-219.0 mmHg. Oxygen saturation was measured over 95. 0% during and after extracorporeal circulation respectively. Erythrocyte count, hemoglobin, hematocrit, and leucocyte count were decreased to 49.2%, 49.0%, 49.4%, and 21. 1% of the preoperative value during extracorporeal circulation respectively and these reductions were not recovered until 1-2 days after perfusion. These. resulted from relatively high degree of hemodilution rate and operative bleeding during these experimental studies. The platelets count was also decreased about to 71% during perfusion, on the contrary, it was increased progressively after perfusion and in 1-21 days after perfusion, the value was returned to preoperative contro1 level. Three dogs were all recovered after extracorporeal circulation.

Nutrition Survey of Young Children of A Day Care Center in the Low Income Area of Seoul City (일부(一部) 도시저소득층(都市低所得層) 유아원원아(幼兒園園兒)의 영양실태조사(營養實態調査))

  • A survey of 113 children, aged 4 to 6 years, of the San Cheon a day care center, located in the upland, over populated, poor area of Seoul City, was conducted from July 28 to September 6, 1982, to investigate the dietary and nutritional status, and prevalence of parasitic inpection. House size was 10.1 Pyung; number of households per house, 3.1; number of family member per household, 4.8; and number of family members living together per room, 3.7; on the average. Only 49% of households possess own houses and the others live in the rented houses. About 40% of mothers were engaged income-producing activities within or outside of the home. Mean values of height, weight, arm circumference, girth of chest, and girth of head ranged from 98.4 to 102.2% of KIST mean. However, 13% of the subjects were assessed as undernutrition for 'weight for height'. Mean value of hemoglobin was $12.9{\pm}2.9\;g/dl$ and mean hematocrit value was $35.2{\pm}5.5%$; 17.9% of the subjects were proven to be anemic according to the criteria of the WHO. The positive prevalence of parasitic inpection was 3.3%, significantly lower than that observed in any other area, probably because of community parasitic control project. Intakes of energy and nutrients except thiamin and ascorbic acid were far below RDAs. Carbohydrates for energy provided 71. 2 to 73.8% of total energy intake; protein accounted 11.3 to 12.2%; fat provided 14.5 to 16.6%. Energy intake was divided among breakfast, lunch, evening meal and snack, on the average, in a percentage of 22.0; 23.4; 24.4; 30.2. There was significant correlation between pattern of food consumption and nutrient intakes. The nutrient intakes and quality of foods were highly correlated to the maternal factors, especially mother's educational level. The study suggested that development of nutrition guides for mother and children would be needed to improve nutritional status of those young children.

Relationship between Vitamin E and Polyunsaturated Fat - A comparative animal study emphasizing perilla seed oil as a fat constituent - (비타민 E와 불포화 지방과의 관계 - 들깨유(油)를 중심으로 한 동물의 비교 연구 -)

  • Perilla (frutescens) seed oil, which is widely used as a source of vegetable oil in Korea, contains a strikingly large amount (58.4% of total fatty acids) of polyunsaturated linolenic acid (18 : 3) which is one of the essential fatty acids. Our hypothesis was that vitamin E contained in this oil would not be enough to prevent peroxidation of this polyunsaturated oil. A comparative study was carried out using rats and chicks devided into seven groups with various diet combinations emphasizing fat sources for the period of four weeks. The level of fat in each diet was 15% and animals were fed ad libitum. Various diet combinations were as follows; perilla seed oil and sesame seed oil with and without vitamin E supplementation, tallow as a saturated fat source and perilla seed hull group (10% at the expense of carbohydrate). The fat constituents of control group were consisted of 50% vegetable oil and 50% animal fat. A few important findings are as follows: 1. Rats fed perilla seed oil lost their hair focally around the neck and suffered from a bad skin lesion at the same place. In chicks, yellow pigmentation both of feather and of skin was clearly observed only in groups fed perilla seed oil with or without vitamin E supplementation. The basis of biochemical mechanisms of this phenomena remains as an important research interest. 2. The mean value for hematocrit was significantly lower for the chicks fed perilla seed oil than for those fed control diet. This result seems to be attributable to the effect on the red cell membrane known as peroxidation-hemolysis of vitamin E deficiency. 3. The serum cholesterol level was higher for the rats fed perilla seed oil than for those fed control diet, whereas in chicks the group fed perilla seed oil showed lower value than the control group indicating that different animal species could vary in their responses to the same diet. 4. In pathological examinations, the sign of hepatic fibrosis was seen in the perilla seed hull group and it was noticeable that the level of hepatic RNA was significantly increased in the rat recovering from vitamin E deficiency. It is hoped that more detailed studies on perilla seed oil and hulls will soon be carried out in many aspects especially i) at various levels of fat in the diet, ii) in relation to dietary selenium level and iii) to find an optimum level of dietary essential fatty acids in terms of P/S ratio using various animal species. In the mean time, the public should be informed to preserve this particular oil with care to minimize fatty acid oxidation and should be discouraged from overconsuming this oil. Analysis of Bone Mineral Density according to Hemoglobin in University Students (혈색소 농도에 따른 대학생의 골밀도 분석)

  • This study was performed to evaluate the effect of hemoglobin (Hb) on bone mineral density (BMD) in university students by performing a quantitative analysis. The subjects included healthy university students aged 20 to 30 years. Although osteoporosis has traditionally been considered as a disease of aging women, it is becoming an increasingly concerning male health problem. Diagnosis of osteoporosis is calculated with a quantitative assessment of BMD. Laboratory blood and urine tests are mainly used with low BMD or fragility fractures to identify any possible causes of bone metabolism disorders. In this study, there was no difference in BMD according to gender. The average red blood cell (RBC), Hb, and Hematocrit (HCT) were significantly higher in males (p<0.01). The correlation between lumbar spine, skeletal muscle mass (SMM), and basal metabolic rate (BMR) was statistically significant (p<0.01). Hb showed a 51.7% statistical influence on BMD by multiple regression analysis. These findings are useful to understand the relationship between BMD and Hb; lower Hb level is associated with lower BMD. The Hb level was the strongest predictor of abnormal BMD. In conclusion, this study showed that a low Hb value was significantly correlated with low bone mass, suggesting that a low Hb value is a risk factor for changes in bone turnover that leads to a decrease bone density.

A Survey of Nutrition and Parasitic Infection of Rural Young Children in the Family Health Project Area in Korea (농촌(農村) 가정보건사업지역(家庭保健事業地域)의 어린이 영양(營養) 및 기생충조사)

  • A survey of nutrition and incidence of parasites was conducted from January 23 to 31 and February 14 to 18, 1981, to determine the nutritional status of young children under the age of 6 years, in one of the rural family health project areas: Whaseoung-gun, Kyunggi-do, Korea. This study covered a total of 233 children. The results are summarised as follows: Approximately one-half of these families had an annual income between \1,000,000 and \2,500,000. Nearly 84% of the households were involved in farming. Energy intake of children met 70.6% of the requirement: carhohydrates provided 77%, protein provided 11%, and fat provided 12% of the total energy intake. Low intake of iron; calcium; and vitamins, excluding thiamin, was also found. Mean hemoglobin value was $10.6{\pm}1.0gm/100ml.;\;42%$ of subjects were categorized as low group, 22% were in the deficient group, and only 36% were at an acceptable level as indicated by O'Neal. Thus, 22% of the subjects were proven to be anemic by the hemoglobin criteria. Mean hematocrit value was $30.1{\pm}2.8%$, and 80 of 208 children, or 38.4% of subjects, were below 30% of the standard established by O'Neal. The positive prevalence of parasitic infection was 20% , significantly higher than that observed in the Yowido compound (5.5%) in 1980. Hematologic findings had no significant correlation to parasitic infection. The majority, 95.2% of the subjects, met or surpassed 90% of the KlST's standard of height:whtile 28. 6% of the subjects were below 90% of the KIST's standard of weight. The ratio of weight to height for boys 4 to 6 years old was 15.8 to 17.5, in contrast to 16.3 to 16.9 for boys of KHDI project areas, and also in contrast to 17.7 to 17.9 for boys of the same age group from the Yowido apartment compound in Seoul. According to the results of the assessments, the authors found a clear difference in growth values, hematologic findings and dietary patterns between rural areas and advantaged urban areas of Seoul. Our results suggest the need to develop the nutrition and parasite control training of village women for better nutritional care of their young children.

  • Blood test is a useful tool in establishing medical treatment for livestock. It provides information such as disease diagnosis, treatment effects, prognostic judgment, and health status. This study compared the value of erythrocytes and leukocytes among conventional, transgenic miniature, and transgenic conventional pigs aged six months to 24 months. Further, it analyzed the aspects of hematological value changes according to the pigs' ages. As a result, the number of red blood cells (RBC), which include hemoglobin, and hematocrit, and the number of white blood cells (WBC), which include neutrophils, and lymphocyte, were high among transgenic miniature pigs, compared with the conventional and transgenic conventional pigs. Conventional pigs showed similar values of RBC and WBC regardless of transgenesis. In comparing their age, the RBC decreased as the age increased compared with the pigs among all the three groups aged of 6~12 months. On the other hand, WBC and neutrophils showed no significant difference regardless of different ages among all the three groups. However, various counts in RBC and WBC were mostly found to be higher in each age in transgenic miniature pigs than in conventional and transgenic conventional pigs. The results of this study show that the values of RBC and WBC were generally higher in transgenic miniature pigs than in conventional and transgenic conventional pigs. Based on this research, hematological values can be widely used in diagnosing diseases or checking the health status of transgenic pigs that are used as disease models, organ transplant source and alike.

Relation between Various Body Fluid Volumes and Body Weight or Lean Body Mass in the Rats (흰쥐의 체액량과 체중 및 무지방 체중 사이의 관계)

  • Relationships between red ceil volume $(^{51}Cr-cell)$, total blood volume (red cell volume divided by hematocrit ratio), and extracellular fluid volume (SCN distribution space) and body weight (ranging between 73 and 384 grams) or lean body mass were studied in 59 nembutalized rats. Lean body mass was determined by means of underwater weighing method on rats clipped and eviscerated. There were positive correlations between body weight or lean body mass and the absolute values (in milliliters) of body fluid volumes. Body fluid volumes expressed on the body weight or lean body mass basis, however, showed negative correlations between body weight (grams) or lean body weight (grams) with one exception. Red cell volume expressed as % lean body mass showed a positive correlation with lean body mass. The other results are summarized as follows: 1. Body density of rats was 1.0561 $(range:\;1.0123{\sim}1.0781)$ and 19.8% body weight of total body fat was obtained. The mean value of lean body mass was 80.2% body weight 2. The correlation between body weight and lean body mass was high, namely, coefficient of correlation was r=.99. 3. The correlation between the absolute value of red cell volume (ml) and body weight showed a high correlation, namely, r= 92 and between the lean body mass coefficient of correlation was r=.93. On a weight basis, red cell volume was 2.67 ml/100 gm body weight or 3.48 ml/100 gm lean body mass. The coefficient of correlation between body weight (grams) and red cell volume (% body weight) was r=-. 30. The coefficient of correlation between lean body mass (grams) and red cell volume (% lean body mass) was r=. 50. Thus, the following regression equation was obtained. Red cell volume (% lean body mass)=. 00243 Lean body mass (gm)+3. 12. 4. Total blood volume was 6.06% body weight or 7.83% lean body mass. The correlation between these blood volume values and body weight or lean body mass were negative, namely, r= -.43 and r=-.42 respectively. 5. Extracellular volume (SCN space) was 30.0% body weight or 37.2% lean body mass. These percentage values showed negative correlations between body weight or lean body mass and coefficients of correlation were r=-.40 and r=-.54 respectively. 6. The rate of increase in body weight or lean body mass is accompanied by a smaller rate of increase in blood volume and extracellular fluid volume. The rate of increase in red ceil volume paralled that of lean body mass.

  • Parallel testing means ordering a number of tests at the same time so abnormalities in any of the tests can be found quickly and used in making the diagnosis. This is a good medical strategy to eliminate diseases and it is relatively inexpensive if all the tests are potential sources of information and performed on the same analyzer. In regression, the equation for the straight line is recast as y = bx + a. This change in terminology leads to confusion. Here a is the y-intercept or constant and b is the coefficient or slope of the line. A few more words of caution about regression - as in all of statistics there are certain assumptions: the x value is a true measure, both X and Y distributions are normal, and homoscedasticity, i.e., the variance of y is the same for each value of x. In this study the linearity classification made by different scientists were always in agreement. Typical examples of curves that were considered linear are presented in Fig. 1-5. Because these automated procedures values were usually within five percent of each other the curvature could be easily detected. The plot of the WBC, RBC, hemoglobin, hematocrit and platelet concentrations from approximately 74.4 to $0{\times}10^3/{\mu}L$ and $80.4-0{\times}10^3/{\mu}L$, $5.6-0{\times}10^6/{\mu}L$ and $6.1-0{\times}1106/{\mu}L$, 18.3-0 g/dL and 19.0-0 g/dL, 54.1-0% and 56.8-0% and 642.0 to $0.03{\times}10^3{\mu}L$ and $754.0-0{\times}10^3/{\mu}L$ on the ADVIA 2120 C Versus and A and B typical of an acceptable linear study as shown in Fig. 1-5. The grand mean of R2, intercept and slope is 0.99898, 0.99459 and 1.54626.

Statistical Analysis of Quality Control Data of Blood Components (혈액성분제제 품질관리 자료의 통계학적인 비교)

  • According to increase of domestic blood components use, the quality control of blood components is necessary to support good products. The purpose of this study is used to provide the producing index of the good product as compared with the accuracy and validity for the distribution of the quality control data. The value of mean, standard deviation, 95% confidence interval and degree of normal distribution of data were calculated by univariate procedure, the value of monthly mean of each blood centers per items were compared by Analysis of Variance(ANOVA) test for the degree of distribution. When there was difference among the mean values, the Duncan's multiple range test was done to confirm the difference. Finally, methods for accessing accuracy and validity of the quality data was done by the Contingency table test. The quality data of five blood centers was showed to the normal distribution and it was in a acceptable range. For each blood centers, the monthly means of Hematocrit(Hct), Platelet(PLT) and pH were not significantly different except Hct of C center, PLT of B, D center and pH of A center. The quality data per items was graded according to quality to six level. As a result of the comparative analysis, the monthly means of Hct of C and E center was significantly different higher than that of D, B and A center. The monthly means of PLT of A center and pH of C center was significantly different higher than that of the others. In the accuracy and validity of the quality control data, C center for Hct, A center for PLT and C center for pH were better than the other. The C blood center was most satisfiable and stable in the quality control for blood component. If the quality control method used in C blood center is adopted in other blood centers, the prepared level of the blood component of the center will be improved partly.
